Proceedings of International Symposium on East-West Medicine, Seoul. 212-230, 1999. -Hepatitis B is caused by hepadnavirus. Hepatitis B virus replicates through 3.5kb pregenomic RNA intermediate which is regulated by core promoter. Pathogenesis of hepatitis B virus has been bilieved the result of host immune response. But recently many studies have reported that high level of viral replication caused by mutation in core promoter might result in severs hepatitis. But these studies were performed in vitro, not in vivo. So there is yet debate about which factor, viral of host factor, is more important in pathogenesis of hepatitis B virus. So we measured real viral replication level in 204 chronic hepatitis B patients by quantifying HBV DNA from sera by our novel PCR-based more sensitive method, and compared these results with ALT level measured from same sera, which indicates liver cell damage. Surprisingly there are no significant correlation between HBV DNA quantity and ALT level. Then we cloned core promoter region. In SSCP, we found that many viral mutants coexist in one patient. Base on SSCP result, we chose main viral core promoter type in each patients, which is thought to determine overall viral replication level in this patient. Main type of core promoter region of each 41 patients were directly sequenced. And with these we measured promoter activity by luciferase assay system and compared promoter activity with on another. We found tha there were some differences in promoter activity according to core promoter sequences. And we constructed replication-competent viral constructs with core promoter from 41 patients and Transfected these into HepG2 cell and measured HBV DNA by southern blot. There were also differences in HBV DNA quantity according to core promoter sequences. On these all results we investigated correlation between the effect of HBV core promoter on viral replication in vitro and HBN DNA quantity, ALT level from sera of each patients. We found there is no significant correlation among them. As a result, we concluded that in determining severity chronic hepatitis B patients, host factors of each patient is more important rather than replicative activity of virus itself.

      • 실리카흄을 흔입한 고강도 콘크리트 개발

        콘크리트 구조물이 대형화됨에 따라 설계단면이 증대되어 상대적으로 많은 경제적 손실을 부담하게 된다. 따라서 단면을 감소시키면서도 소요의 하중에 안전한 구조물을 건설하기 위해서는 우선적으로 구조물 건설에 기초가 되는 고품질, 고내구성의 고강도 콘크리트 개발이 절실히 요구된다 본 연구에서는 일정 시멘트비 및 혼화재 비율하에서 소요의 워커빌리티가 확보되는 고강도 콘크리트를 시간과 온도의 변화에 따라 증기 양생하여, 압축, 인장, 휨강도 뿐만 아니라 파괴특성을 실험적으로 연구하였다. 시료 제작시 시멘트 입자 사이의 공극 및 불연속 영역을 충전하여 고밀도화하기 위한 콘크리트용 혼화재로 시멘트 비표면적이 상당히 작은 초미립 분말인 실리카흄을 이용하였다. 또한 AE감수제 및 고성능 유동화제를 사용하여 혼화재의 첨가에 의하여 발생될 수 있는 워커빌리티의 감소를 방지하였다. 실험결과에 의하면 일정 양생 지속 시간하에서 온도의 증가에 따라 콘크리트의 압축, 인장 및 휨강도가 전반적으로 증가하였다. 동일하게 일정 온도하에서 양생 지속시간이 커짐에 따라 강도들이 증가됨을 알 수 있었다. 따라서 상대적으로 높은 온도와 긴 지속 양생 조건하에서 파괴에 대한 저항력이 크게 나타났다. The compressible, tensible, and flexibleresistance of the high strength concrete is analyzed by the experimentation in the present study. For the test, we cure several samples with the silica fume as a mixture being become dense the spaces between the particle of cement under the variation of both the temperature and the curing-interval. Then, the superplasticizer and the ezcon are also used to satisfy the required workability for construction. The compressible, tensible, and flexible resistances to a stress are increased as increasing the temperature and the time interval for the curing. Therefore it is concluded that the overal fractural and mechanical properties is improved by mixing the silica fume into the cement.

        편측성 하악과두증식증에서 수술방법에 의한 술후 골격구조의 대칭성에 관한 비교연구

        In patients with unilateral condylar hyperplasia, increased length of the condylar neck causes deviation of the mandible to the opposite site, resulting in facial asymmetry, but the arch form and general contour of the displaced mandible is approximately symmetric. So, the unilateral condylectomy have been prefered surgical method among the surgeons. The purpose of this study is to analyze the postoperative results comparatively following two different surgical methods, condylectomy and ramus surgery, in unilateral condylar hyperplasia. Case of 6 patients with the unilateral condylar hyerplasia were analyzed in this study, 3 case of the condylectomy only, 3 cases of the ramus surgery, Clinical and radiographic evaluations were tried for comparison. In conclusion, comparative results between two groups did not show any significant difference,but we confirmed that unilateral condylectomy could be a excellent surgical methods in unilateral condylar hyperplasia thinking over the unique morphological characteristics of it.

      • 위성자료를 이용한 한반도 주변해역의 기후적 특성 연구

        Satellite data, with sea surface temperature(SST) by NOAA and sea level(SL)by Topex/poseidon, are used to estimate characteristics on the variations and correlations of SST and SL in the East Asian Seas from January 1993 through May 1998. We found that there are two climatic characteristics in the East Asian Seas: the oceanic climate, the eastern sea of Japan, and the continental climate, the eastern sea of China, respectively. In the oceanic climate, the variations of SL shown the high values in the main current of Kuroshio and the variations of SST shown not the remarkable seasonal variations because of the continuous compensation of warm current by Kuroshio. In the continental climate, SL shown high variations in the estuaries(the Yellow River, the Yangtz River) with the mixing the fresh water in the mouth of estuaries of the saline water in the coasts of continent and SST shown highly the seasonal variations due to the climatic effect of continents. In the steric variations in summer, the eastern sea o f Japan, the East China Sea and the western sea of Korea shown the increment of sea level with 10~20cm. But the Bohai bay in China shown the relatively the high values of 20 ~30 cm due to the continental climate. Generally the trends of SST and SL increased during all periods. That is say, the slopes of SST and SL presented 0.29℃/year and 0.84cm/year, respectively. The annual and semi-annual amplitudes shown a remarkable variations in the western sea of Korea and the eastern sea of Japan, In the case of the annual peaks, there appeared mainly in the western sea of Korea and the eastern sea of Japan because of the remarkable variations of SL associated with Kuroshio. But in the case of the semi-annual peaks, there appeared in the eastern sea of Japan by the influence of current, and in the western sea of Korea by the influence of seasonal temperature, respectively. From our results, it should be believed that SST and SL increase gradually in the East Asian Seas concerning to the global warming. So that, It should be requested to the international co-operation against to the change of the abnormal climate.

      • 아가리쿠스버섯에서 분리한 조단백다당류의 막분리 및 분무건조 특성

        η_(evap)분자량에 따라 다양한 생리활성을 나타내는 아가리쿠스버섯에서 분리한 조단백다당류를 고부가가치 기능성 소재로 개발하기 위하여 막분리 및 분무건조 공정에 따른 특성을 조사하였다. 아가리쿠스버섯 조단백다당류를 분자량 크기에 따라 여과하는 동안 공정온도에 따른 투과플럭스의 변화는 10 kDa와 150 kDa 모두에서 유사하였는데, 막분리 온도 및 압력이 증가함에 따라 비례적으로 증가하였으며,40℃에서의 투과플럭스가 가장 높았다. 시간에 따른 투과플럭스의 변화는 막분리 30분 경과시 급속히 감소하였으며, pore size가 10 kDadhk 150 kDa일 때 공정압력이 각각 1.2 kg_(f)/㎠ 와 2 kg_(f)/㎠에서 가장 높았다. 한외여과 후 아가리쿠스버섯 조단백다당류의 점도는 여과 전 13.9cP에서 여과후 10 kDa이하와 10∼150 kDa구간은 각각 10.8, 11.9cP로 점도가 저하되었으나, 150 kDa이상은 20.1cP로 점도가 증가되었다. 분자량 크기가 다른 3가지 분획물에 대한 분무건조 공정에서의 열효율성은 가열공기온도, 시료공급속도 및 농도가 증가할수록 열효율성이 증가함을 알 수 있었으며, 시료공급속도가 가장 중요한 영향인자임을 확인하였다. This study was conducted to investigate the characteristics of ultrafiltration and spray drying process for crude protein bound polysaccharide(CPBP) isolated from Agaricus blasei Murill. In ultrafiltraion process, the permeate flux increased with the increase of operating pressure and temperature. The permeate flux declined continuously while the fouling materials were accumulated on the membrane as the operation time increased. In comparing of raw CPBP and filtered CPBP, the viscosity of CPBP treated UF was decreased and AE value of treated samples was increased. Thermal efficiencies of spray drying process were increased by increasing inlet temperature, feed rate and feed concentration.

        원자로압력용기장 용접열영향부의 미세조직 변화가 Magnetic Barkhausen Noise 거동에 미치는 영향

        최근 자기적 특성의 변화가 재료의 미세조직적 상태에 민감하다는 보고가 있다. 재료의 미세조직은 상(phase), 결정립의 크기, 석출물 등의 다양한 인자로 구성되며, 본 연구에서는 이러한 인자들을 이용하여 재료의 특성과 Barkhausen noise(BN)의 관계를 연관시키기 위한 시도를 하였다. ASTM A 508 Gr.3 원자로압력용기강을 대상으로 용접열영향부의 미세조직적인 변화를 일곱가지 대표적인 영역으로 구분하였고, 각 영역에서의 미세조직과 기계적특성의 변화를 파악한 후, 각 조건별로 BN을 측정하였다. 통상의 보자력이나 잔류자화 같은 자기적 성질은 큰 변화가 없었지만, BN은 미세조직의 상태에 따라 현격한 변화를 보였다. 결정립과 석출물의 크기가 증가함에 따라 BN은 증가하였고, tempered martensite 보다는 tempered bainite 조직에서 높은 BN을 나타냈다. Recent study has demonstrated that some magnetic properties are sensitive to the microstructural state of material. The ASTM A 508 Gr. 3 reactor pressure vessel steel has various microstructural changes including martensitic and bainitic phases, and various sizes of grain and precipitates in the weld heat-affected zone (HAZ). To correlate the microstructural state with Barkhausen noise (BN), specimens were prepared through simulating various weld thermal cycles using a thermal simulator. The conventional magnetic properties, i.e. coercive force, remanence and maximum induction, did not change significantly. whereas the BN amplitude and energy during a magnetization cycle changed markedly with microstructural state. The BN increased with increasing grain and carbide sizes, and the tempered bainite structure showed higher BN parameter than tempered martensite.

      • 유압관로내의 동특성을 이용한 비정상 유량계측에 관한 연구

        This paper describes an approach for estimating the unsteady flowrate through a hydraulic pipe lines and a components in real time. Concerning the dynamic characteristics of a hydraulic pipe lines, experimental research from the view point of focusing on the unsteady flowrate through a pipe is insufficient because of the lack of adequate flowmeters. In this experiment, a new instantaneous flowrate measurement system making use of a sharp-edged cylindrical choke and LDV has been proposed and developed by the authors. The unsteady flowrate is obtained from the measurements center-line velocity of a circular pipe using LDV and the pressure difference using a sharp-edged cylindrical choke, and experiments on the frequency characteristics are carried out. The results show a good agreement between the estimated and directly measured flowrate waveforms illustrates the validity of the method proposed here.
