        Paricalcitol attenuates lipopolysaccharide-induced inflammation and apoptosis in proximal tubular cells through the prostaglandin E<sub>2</sub> receptor EP4

        Background: Vitamin D is considered to exert a protective effect on various renal diseases but its underlying molecular mechanism remains poorly understood. This study aimed to determine whether paricalcitol attenuates inflammation and apoptosis during lipopolysaccharide (LPS)-induced renal proximal tubular cell injury through the prostaglandin E<sub>2</sub> (PGE<sub>2</sub>) receptor EP4. Methods: Human renal tubular epithelial (HK-2) cells were pretreated with paricalcitol (2 ng/mL) for 1 hour and exposed to LPS (1 μg/mL). The effects of paricalcitol pretreatment in relation to an EP4 blockade using AH-23848 or EP4 small interfering RNA (siRNA) were investigated. Results: The expression of cyclooxygenase-2, PGE<sub>2</sub>, and EP4 were significantly increased in LPS-exposed HK-2 cells treated with paricalcitol compared with cells exposed to LPS only. Paricalcitol prevented cell death induced by LPS exposure, and the cotreatment of AH-23848 or EP4 siRNA offset these cell-protective effects. The phosphorylation and nuclear translocation of p65 nuclear factor-kappaB (NF-κB) were decreased and the phosphorylation of Akt was increased in LPS-exposed cells with paricalcitol treatment. AH-23848 or EP4 siRNA inhibited the suppressive effects of paricalcitol on p65 NF-κB nuclear translocation and the activation of Akt. The production of proinflammatory cytokines and the number of terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling-positive cells were attenuated by paricalcitol in LPS exposed HK-2 cells. The cotreatment with an EP4 antagonist abolished these anti-inflammatory and antiapoptotic effects. Conclusion: EP4 plays a pivotal role in anti-inflammatory and antiapoptotic effects through Akt and NF-κB signaling after paricalcitol pretreatment in LPS-induced renal proximal tubule cell injury.

      • 진해만 연안 퇴적물의 자성과 중금속에 대한 연구

        진해만 두 정점의 자기특성과 대자율의 비교를 바탕으로 하여 중금속 농도 변화가 자화강도 변화에 미치는 영향을 있는지를 살펴보았다 이를 위해 깊이에 따른 중금속 농도 변화 (산화환원 환경) 그리고 유기물' 의 함량을 조사하였다 본 연구에서는 5가지의 중금속(Al, Fe, Cr, Ni, Mn)의 농도가 측정되었으며, 자화강도와 중금속의 농도 변화는 대체적으로 유사한 경향을 나타내었고, 산화환원환경에서 각각 중금속과 자화강도의 상관성이 다르게 나타났다. 산화환경에서는 반대되는 경향을 보이고, 환원환경에서는 비슷한 상관성을 나타낸다. 총유기물(TOM)과 대자율을 비교해 보면 두 정점의 유기물 농도가 다르게 나타나며, 산화환경에서 반대되는 경향성을 나타내고, 환원환경에서 비슷한 경향을 보인다. 이것은 산화환원 환경에 따라 유기물 함량과 중금속 의 존재형이 변화하므로 서로 연관성이 있다고 생각된다. The impact of heavy metal concentrations on magnetization compared to magnetic susceptibility is investigated by the measurements of heavy metals (redox change) and organic matter contents in the sediment core samples at two stations of the Jinhai Bay. In the present study, five heavy metals (Al, Fe, Cr, Ni, Mn) concentrations were determined, and indicated generally similar trends with the corresponding magnetization. The relationship between heavy metal concentration and magnetization depend on redox-environmental conditions, showing negative correlation under the oxidation environment and positive correlation under reduction environment, respectively. Comparing total organic matter content with magnetic susceptibility, organic matter contents at both stations are different, and showed a opposite trend against magnetic susceptibility in oxidation parts, and a similar pattern in reduction parts. These results demonstrate that both organic matter contents and heavy metal speciation are influenced by redox-condition.

        This study provides marketing framework about how to operate strategic e-business successfully under rapidly changing environment. While considering the ultimate goal of e-business as profit securing through producing customer value, this study suggests future e-marketing strategy focusing in understanding and utilizing opportunities arising under rapidly changing environment. First, e-business should be customer-focused. One should escape from product-focused mear-sightedness to thinking about customer's desire and dream. Second, e-business should be future inclined. One should escape from the management that focuses too much on the existing market or present to the direction where one can utilize the future opportunity. Third, e-business should manage online and offline comprehensively so that they can produce synergy effect. Successful operation of e-business requires comprehesive and efficient utilization of value chain that online and offline are connected. Moreover, e-marketing process closely interacts with each other. It is difficult to establish effective e-business mission or model without ideas in opportunity analysis or STP strategy. Therefore, effectiveness of e-marketing process requires close interation between the processes.

        This study aimed at determining if the types of cultural marketing affected corporate image, social responsibilities, and business ethics and which element they were more closely related to among such types of cultural marketing as cultural sales promotion, cultural support, cultural direction, and cultural businesses. First, Companies that participate more actively in cultural marketing have positive effects on their image, which shows that cultural marketing is an effective means of improving corporate image perceived by customers. Second, Consumers recognize that companies participating more actively in cultural marketing will fulfill their social responsibilities and conform to business ethics, which shows that cultural marketing can have effects not only on corporate image but also on their social responsibilitiesandbusinessethics. From the results mentioned above, cultural direction and cultural business have stronger correlation with each other than cultural sales promotion and cultural support among the types of cultural marketing; therefore, a company should participate in cultural marketing not for the corporate benefits or as visible support but for good corporate image perceived by consumersasaculturalbusinessthrough culture as living. Therefore, It is necessary to provide an effective method to improve corporate image and set a strategic plan by examining each property in the types of cultural marketing.

      • 12-O-Tetradecanoylphorbol-13-Acetate에 의한 HL-60 세포 분화유도중 Vimentin 유전자 전사조절에 대한 AP-1의 역할

        Purpose: To gain insight on the role of AP-1 in transcriptional regulation of vimentin gene during differentiation of HL-60 cells by 12-O-tetradecanoylphorbol-13-acetate (TPA), the levels of vimentin mRNA and AP-1 have been investigated with Northern blot hybridization and DNA mobility shift assay. Materials and Methods: HL-60 cells were grown in RPMI 1640 medium supplemented with 10% heat-inactivated fetal calf serum and antibiotics in a humidified 5% CO_2 at 37℃. Total RNA was prepared by a modification of the method of Karlinsey et al. Northern blot hybridization was performed by the method of Virca et al. EcoRI fragment of pVIM-GEM was used as probe for vimentin mRNA. DNA mobility shift assay was performed by the method of Lim et al. End labeled DNA probe(Upper strand, 5'-CGCTTGATGAGTCAGCCG- 3') for AP-1 binding activity was mixed with nuclear extracts in a 20 μl reaction volume containing 300 mM KCI, 60 mM HEPES, pH 7.9, 25 mM MgCI_2, 1 mM EDTA, 1 mM DTT, 60% glycerol, and 2 ㎍ of poly[dI-dC]. Results: TPA increased vimentin mRNA levels, with maximal stimulation reached at 24 hr. The level of vimentin mRNA was induced in proportion to the concentration of TPA. TPA-induced vimentin mRNA was almost reduced by actinomycin-D pretreatment. TPA-induced stimulation of vimentin gene was completely reduced by staurosporin pretreatment. In DNA mobility shift assay, AP-1 newly appeared at 24 hr during TPA-induced differentiation and was almost not detected after the pretreatment of staurosporin. Conclusions: These results suggest that the induction of vimentin mRNA during TPA-dependent differentiation in HL-60 cells may be mediated by protein kinases C signal transduction and AP-1 is important to transcriptional regulation.

        Spiritual Care Guide in HospiceㆍPalliative Care

        The Spiritual Care Guide in HospiceㆍPalliative Care is evidence-based and focuses on the universal and integral aspects of human spirituality—such as meaning and purpose, interconnectedness, and transcendence—which go beyond any specific religion. This guide was crafted to improve the spiritual well-being of adult patients aged 19 and older, as well as their families, who are receiving end-of-life care. The provision of spiritual care in hospice and palliative settings aims to assist patients and their families in finding life’s meaning and purpose, restoring love and relationships, and helping them come to terms with death while maintaining hope. It is recommended that spiritual needs and the interventions provided are periodically reassessed and evaluated, with the findings recorded. Additionally, hospice and palliative care teams are encouraged to pursue ongoing education and training in spiritual care. Although challenges exist in universally applying this guide across all hospice and palliative care organizations in Korea—due to varying resources and the specific environments of medical institutions—it is significant that the Korean Society for Hospice and Palliative Care has introduced a spiritual care guide poised to enhance the spiritual well-being and quality of care for hospice and palliative care patients.
