      • Effect of ER signal peptides (KDEL) on expression and function of anti-virus monoclonal antibody in plant

        Plant genetic engineering has led to the production of plant-derived mAb (mAbP), which provides a safe and economically feasible alternative to the current antibody expression systems. In this study, the expression levels of mAbP SO57 with or without ER-signal peptides (Lys-Asp-Gly-Leu;KDEL) in transgenic tobacco plants were analysed in transgenic plant. PCR and Reverse Transcription-PCR analyses showed existence of heavy and light chain genes of mAb with or without KEDL and their transcription in plant, respectively. Western blot showed that the expression levels of mAbP SO57 with KDEL were significantly higher than that without KDEL. Flow cytometry analysis showed that the Fc domains of both purified mAbP and mammalian-derived mAb have similar binding activity to the FcγRI receptor. High-performance liquid chromatography analysis showed that the mAbP SO57 with KDEL had glycan profile with both oligomannose and golgi type, whereas the mAbP SO57 without KDEL had only golgi type glycans. Neutralizing analysis with rabies virus CVS-11 showed the similar neutralizing activity between mAbP SO57 with and without KDEL. These results suggest that the potential of mAbP SO57 for rabies immunotherapy is regardless of plant specific glycan structures.

        빅데이터의 형사법적 활용에 관한 연구

        범죄가 언제, 어디서 일어날지 미리 예측할 수 있다면 얼마나 좋겠는가? 힘들게 범인을 잡으러 다닐 필요 없이 범죄가 예측된 장소에서 기다리고 있다가 범인으로 지목된 사람을 잡아오기만 하면 될 것이다. 모든 범죄를 예측할 수 있다면 말 그대로 ‘범죄없는 사회’를 이룰 수도 있을 것이다. 미국의 대표적 과학소설(SF, Science Fiction)작가인 필립 K. 딕이 쓴 <마이너리티 리포트>라는 소설은 동명의 영화로도 잘 알려져 있는데, 2054년의 미래를 그린 이 이야기에는 범죄를 예측하는 세 명의 예언자가 등장한다. 범죄가 일어나기 전에 미리 범죄를 예측하는 최첨단의 치안시스템의 명칭은 말 그대로 ‘Precrime’ 이다. 즉 범죄가 일어나기 전에 대처한다는 것이다. 사회가 안정화되고 삶의 수준이 높아질수록 안전에 대한 갈구는 더 커지게 된다. 그런데 소설에서 또는 영화에서 공상과학적인 소재로 쓰이던 범죄의 예측이 현실에서도 가능하게 되었다면 어떨까? 미국 등에서 개발하여 사용하고 있는 범죄 예측 프로그램은 범죄를 예측하는 예언자 대신, ‘빅데이터’라고 불리는 거대한 규모의 데이터 분석을 통하여 범죄를 예측하고 있다. 빅데이터는 2012년 전 세계적으로 정보통신기술(IT, Information Technology) 분야에서 가장 주요한 화두이며, 기술의 진보를 통해 지금까지 불가능했던 일들을 가능하게 만들어 인간의 삶을 바꾸는 기회가 될 것으로 예상되고 있다. 우리나라에서도2012년 11월 ‘스마트국가 구현을 위한 빅데이터 마스터플랜’을 마련하고 데이터의 창조적 활용을 통해 스마트 강국을 실현할 것을 목표하고 있다. 본 논문은 이렇게 기업과 국가기관 등에서 빅데이터의 중요성을 강조하고 그 활용을 모색하고 있는 시점에서 형사법적으로 빅데이터를 활용하여 범죄로부터 국민을 보호하고, 재판단계에서 범죄자에게 적정한 형벌과 보호관찰 등을 부과하며, 범죄를 예방할 수 있는 방안을 연구해보고자 한다. 이를 위하여 먼저 빅데이터란 무엇이며, 어떤 방식으로 어떻게 활용할 수 있는 것인지를 살펴본다. 그리고 빅데이터를 법학 분야에서 이용한다면 지금까지와는 다른 여러 분석과 그에 따른 적용이 가능함을 생각해보고, 범죄예측단계와 재판에서의 양형단계 그리고 보호관찰과 수강명령·사회봉사명령 등 사회내 처우 단계에서 빅데이터가 활용될 수 있음을 차례로 알아보고자 한다. 그러나 빅데이터를 수집·분석·활용하고자 할 때 간과해서는 안 될 문제가 있다. 흔히 ‘프라이버시’라고 일컬어지는 개인의 사생활 보호와 관련한 문제가 그것이다. 빅데이터로 인한 사생활의 침해 우려가 ’빅브라더’의 형태로 실현되지 않도록 그에 대한예방책도 함께 강구해야 할 것이다. What if we could predict the right time and the right place in which a crime would occur? If that would be a reality, there would be no needs to chasing suspect hardly. All the police would have to do is waiting the suspect at the predicting time and the predicting place. When the potential criminal would show his appearance, then the police could arrest the potential criminal and the crime would never occur actually. Philip K. Dick, the famous science fiction novelist in U.S.A, wrote <Minority Report> and it was made into the movie of same title by Steven Spielberg. The movie showed the world where "Precrime", a specialized police department, arrest criminals based on predictions provided by three psychics at future of 2054. So the public order system of the movie is so-called pre-crime, because according to the "Precrime" the police could apprehend the criminals before the actual crime. In the recent years, the development of Information Technology actualized the imagination of the movie in real world. And the technology is called the "Bigdata" technology. It is a analysis of enormous, various and fast-created data. Until now very huge amount of datas have been thrown out, so we could not use that data. But the advances of technology made many wonderful things possible, for example "PredPol", calculates its forecasts based on times and locations of previous crimes, combined with sociological information about criminal behavior and patterns in crime prediction, sentencing guideline , probation, attendance center order , community service order the city of Santa Cruz of U.S.A. So we can apply the ``Bigbata`` technology and analysis into the crime prediction, sentencing guideline, probation, attendance center order and community service order. Although ``bigdata`` has the concerns about invasion of privacy, if we use it with care it could bring us to the insight for the better future.

      • 林地利用에 따른 溪流水의 水質變化

        Table 2. Evaluation of stream water quality in three stands by Drinking water quality standard <원문참조> 임지이용에 따른 수질변화 및 오염정도를 구명하여 향후 비점오염원 관리를 위한 기초자료를 제공하고자 경남 진주시 정평리에 위치한 3개 임분(소나무, 밤나무Ⅰ, 반나무Ⅱ임분)에서 2000년 7월부터 9월까지 계류수의 수질변화를 분석하여 얻은 결과는 다음과 같다. 1. 세 임분의 계류수 pH는 소나무 임분(pH6.59)>밤나무 Ⅱ임분(ph 6.53)>밤나무 Ⅰ임분(pH 6.47)계류수 순으로 나타났다. 2. 전기전도도는 밤나무 Ⅰ임분>소나무 임분>밤나무 Ⅱ임분 계류수의 순으로 높게 나타났다. 3. 양이온은 세 임분의 계류수 모두 Ca^2+, Na^+, Mg^2+, K^+, NH_^+의 순으로 높게 나타났다. 4. 음이온은 소나무 임분과 밤나무 Ⅱ임분 계류수에서는 SO_4^2, Cl, NO_3의 순으로 높게 나타났고, 밤나무 Ⅰ임분 계류수에서는 NO_3, SO_4^2, Cl 의 순으로 높게 나타나 차이를 보였다. 5. 소나무, 밤나무 Ⅱ임분 계류수는 pH, NH_4+, NO_3, Cl, SO_4^2이 먹는 물 수질기준 범위내에 있었다. 그러나 밤나무 Ⅰ임분 계류수에서 5항목 중 NO_3는 먹는 물 수질기준을 넘어서는 범위에 있어 임지이용에 따른 하류수질의 오염이 우려되었다. 따라서 이러한 상류지역에서 오염원 차단을 위한 수질정화시설 등의 대책이 마련되어야 할 것으로 사료된다. This study was carried out to clarify the change characteristics of stream water quality by land-use of forest from July to September, 2000 in three stands(Pinus densiflora, Castanea crenata(Ⅰ), Castanea crenata(Ⅱ) stand) of Jeongpyeong-ri, Jinju-si, Gyeongnam. The mean pH of rainfall results in acid rain of 5.3. The pH of stream water in three stands was hugh in order of Pinus densiflora (pH 6.59), Castanea crenata(Ⅱ)(pH 6.53) and Castanea crenata(Ⅰ) stand(pH 6.47). The electrical conductivity of stream water was high in order of Castanea crenata(Ⅰ), Pinus densiflora and Castanea crenata(Ⅱ) stand. Cations of three stand in stream water high in order of Ca^2+, Na^+, Mg^2+, K^+ and NH_4^+. But anions of stream water in Pinus densiflora stand and Castanea crenata(Ⅱ) stand were high in order of SO_4^2, Cl and NO_3. In Castanea crenata(Ⅰ) stand cations of stream water were high in order of NO_3, SO_^2 and Cl. The level of pH, NH_4^+, NO_3, Cl and SO_4^2 of stream water in Pinus densiflora stand and Castanea crenata(Ⅱ) stand reached within the level of domestic use standard for drinking water But the level of NO_3 of stream water in Castanea crenata(Ⅰ) stand was higher than that of domestic use standard. Therefore, non-point sources like forest watersheds which are fertilizer application lands should be taken to the appropriate mitigation measures.

      • 논 비점오염 저감을 위한 최적관리기법(BMP) 보급 효과 분석

        본 연구에서는 최적관리기법(BMP)을 실제 영농지역에 보급하여, 광역단위 논에서 발생하는 비점오염물질의 저감효과를 분석하고자 하였다. 현재까지 선행 연구들을 통해 다양한 BMP 효과 검증이 이루어졌으나, 실제 영농지역 농민들에게 BMP를 보급하여 그 효과를 측정한 연구는 부족한 실정이다. 이에 본 연구에서는 광역단위 논 지역을 대상으로 물관리기술(물꼬)과 시비관리기술(완효성 비료)을 보급하고 강우시와 비강우 모니터링을 통해 BMP 효과를 측정하였다. 연구 대상지구는 전라북도 부안군 백산면 용계리 일대로 새만금 지역에 위치하여 있다. BMP 보급 효과를 분석하기 위하여 먼저 참여농민과 농지를 선정하였으며, 관개지역과 배수면적을 구분하여 모니터링 조사 지점을 선정하였다. 연구대상지구는 SO#1(완효성 비료 95%, 비참여 농가 5%), SO#2(완효성 비료2%, 물꼬 8%, 완효성비료 + 물꼬 40%), SO#M(완효성 비료 96%, 비 참여 농가 4%) 총 3개의 배출구로 구분하였다. 각 지구별 오염부하량을 측정한 결과, 대조구 대비 SO#1에서는 수질항목별로 10.6 ∼ 85.5%, SO#2는 8.1 ∼ 45.9%, SO#M은 10.7 ∼ 86.2%의 범위로 저감된 것으로 분석되었다. 특히, SS의 단위면적당 오염부하 저감율이 가장 큰 것으로 나타났으며, TOC의 단위 면적당 오염부하 저감율은 가장 작은 것으로 나타났다. 하지만 본 연구 결과는 7월부터 10월까지 측정한 데이터를 활용하여 산정한 결과로 써레질, 이앙기간에 발생한 단위면적당 오염부하량은 포함되지 않은 결과이다, 따라서 추가적인 연구를 통해 효과를 검증할 필요가 있을 것으로 판단된다.

        일 대학병원에 입원한 노년기 양극성 장애 환자의 인구학적 및 임상적 특징

        Objectives : As the elderly population is continuing to increase, psychiatric diseases of the elderly are becoming an important social issue. This study looks into the demographical and clinical features of the elderly patients diagnosed with bipolar disorder in a University hospital. Methods : The study subjects include patients admissioned in the closed wards of Soonchunhyang Seoul hospital and Cheonan hospital from March, 2000 to February, 2005 who met the DSM-IV-TR criteria for bipolar disorder at the time of discharge. A total of 146 patients (76 men, 70 women) medical records were studied retrospectively. Subjects older than the age of 50 at time of admission were grouped as the old age group, whereas those who were younger than 50 as the young age group. Also among the old age group, those who had the first onset of episode under the age of 50 were grouped as the early onset group, whereas those who had the first onset after the age 50 were grouped as the late onset group. Results : The number of bipolar disorder patients in the young age group and old age group were 73 and 73 respectively. The number of early onset group and late onset group were 46 and 23 respectively. The old age group had a relatively higher incidence of bipolar II disorder than the young age group. Also in the old age group there was a higher incidence of hypomanic or depressive episode rather than manic episode compared to the young age group. And in the old age group psychotic symptoms were less common and the mean admission length shorter than the young age group. Additionally in the old age group treatment with Lithium monotherapy or with no mood stabilizers at all were relatively common. Among the old age group, late onset group had a higher rate of bipolar II disorder than the early onset group. Conclusion : This study shows differences in the demographical and clinical features among different age groups. But additional research would be required to determine whether the bipolar disorders in the elderly or late onset group are actually a different type of disorder from those of the young.

        항암화학요법 관련 오심과 구토 완화를 위한 비약물요법에 관한 연구동향

        Purpose: Purposes of this study were to understand the current trends on complementary therapy in relieving chemotherapy-induced nausea and vomiting and to suggest the future research direction. Method: Subjects were selected on CINAHL, MEDLINE, Korean Academy Data Base from 1980 to 2001 which used nausea, vomiting, chemotherapy and complementary therapy as key words in experimental studies. Eight korean articles and twenty-one international articles were analyzed in terms of general characteristics, research methods, and types of complementary therapy. Data were analyzed by using descriptive statistical methods. Result: Since 2000, researchers have more actively used complementary therapy. In subject characteristics, mean age was 35.5 years old, 45% of the researchers were performed with high level of incidence of chemotherapy induced vomiting, 14% of them set limit of consecutive cycle during research, and 65% of them did not comment the selecting criteria of sample. About 60% of them were designed post-test only control group: 35% used INV by Rhodes, 31% used Likert scale, and 24% used VAS for dependent variable. Muscle relaxation therapy was mostly applied for relief of nausea and vomiting. Conclusion: Further studies will be needed to control extrinsic variables affecting nausea and vomiting in research design and to accumulate evidence with studies applying various complementary therapies.

        한국 기혼여성의 일상적 삶의 질 향상에 관한 연구 (3) : 한국 기혼여성의 삶의 질 향상을 위한 프로그램 개발 연구 Development of A Program for the Enhancement of Korean Married Women's Quality of Life

        The purpose of this study was to develop a program for the enhancement of Korean married women's quality of life(LQ) based on the results of the first year study(1996) and the second year research(1998). In order to construct a program, a psycological type test was conducted to 505 women and four preliminary programs were carried out focusing on self-esteem, understanding of one's own personality dynamics, efficient communication, positive emotion, social belongingness and expressions of feelings of unfinished business. For the final program, Jung's theory of psychological type(MBTI), Satir's family therapy approach and psychodrama were applied. An experimental and a control group were designed consisting 16 women for each group. The program was carried out for 6 weeks for the experimental group. The result of treatment were as follows: l)There was a difference statistically in LQ related to family harmony, self-achievement and improvement of relationship with in-laws. 2)There was a statistical change in psychological factors of expression of feelings, self-esttem, communication, and positive emotions. 3) The program participants self reports indicate that the program was effective for the reconstruction of family relationship, self-understanding, group coherence, and instillation of hope. The reports also shows that the program was effective for the perceptual changes in parent-child, wife-husband and siblings' relationships. In conclusion, the program developed for the enhancement of Korean married women's life quality(LQ) based on the Jung's psychological type theory, Satir's family therapy approach and psychodrama was effective for the improvement of LQ.

      • Interacelluar reprogramming of expression, glycosylation, and function of plant-derived anti-virus therapeutic monoclonal antibody

        Plant genetic engineering has led to production of plant-derived mAb (mAbP), which can provides a safe and economically effective alternative to the conventional antibody expression methods. In this study, the expression levels of mAbP SO57 with or without ER-retention peptide extensions signal (Lys-Asp-Glu-Leu; KDEL) in transgenic tobacco plants were analysed in transgenic plant. The expression levels of mAbP SO57 with KDEL were significantly higher than that without KDEL regardless of transcription level. mAbP SO57 with KDEL localized surround to the nucleus suggesting that the mAbP with KDEL is localized in ER. The mAbP without KDEL and mAbH had mainly Golgi type glycans, whereas the ER-localized mAbP with KDEL had glycan profile with both oligomannose type (47.6%) and Golgi type (52.4%). The Fc domains of both purified mAbP (with and without KDEL) and human-derived mAb (mAbH) had similar bingding activity to the Fcγ RI receptor (CD64). Both mAbP (with and without KDEL) had a shorter half-life than mAbH. However, both mAbP with and without KDEL was as effective at neutralizing activity of the rabies virus CVS-11 as the mAbH. These results suggest that ER localization of recombinant mAbP by KDEL reprograms glycosylation and enhances production of the functional antivirus therapeutic antibody in the plant.

      • 돼지고기육종의 방사선 유래 휘발성 물질 분석

        Bacillus cereus KCTC 3674 produced at least two kinds of extracellular proteases. Two major bands of the protease activity with molecular weights of approximately 100 and 38 kDa were obtained after gelatin-SDS-PAGE. The production of extracellular proteases occurred during the logarithmic growth phase and was greatest when cultures reached the stationary growth phase. On the other hand, the growth of B. cereus KCTC 3674 and the production of its extracellular proteases were severely inhibited by the addition of protonophore carbonylcyanide m-chlorophenylhydrazone (CCCP). Thus, it was concluded that the growth of B. cereus KCTC 3674 and the production of its extracellular pretenses were strongly affected by a H+ electrochemical potential.
