        Free Radical Scavenging Activity of β-chitooligosaccharides

        The free radical scavenging activity of β-chitooligosaccharides (β-COSs) was evaluated by employing an electron spin resonance (ESR) spectrometer on 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl and alkyl radical. We prepared three kinds of β-COSs with different molecular weights: higher molecular weights (HMWβ-COSs: 50-10 kDa), medium molecular weights (MMWβ-COSs: 10-5 kDa), and lower molecular weight (LMWβ-COSs: below 5 kDa). The DPPH radical scavenging activity of HMWβ-COSs was the highest among the three kinds of β-COSs, and the IC50 value was 0.73 mg/mL. In addition, all β-COSs scavenged hydroxyl radical, and the IC50 values of LMWβ-COSs, MMWβ-COSs and HMWβ-COSs were 0.49, 0.62 and 0.47 mg/mL, respectively. Also, all β-COSs showed the alkyl radical scavenging activity, and the IC50 values of LMWβ-COSs, MMWβ-COSs and HMWβ-COSs were 0.35, 0.13 and 0.17 mg/mL, respectively. These results indicate that β-COSs possess antioxidative activity.       • 민들레 물추출물의 항산화 및 자유라디칼 소거활성

        민들레 잎과 뿌리로부터 물추출물을 제조하여 항산화 활성 및 라디칼 소거활성을 분석하기 위하여 linoleic acid에 대한 과산화물 생성저해율, DPPH radical에 대한 소거활성, chemiluminescence를 이용한 hydroxyl radical에 대한 소거활성, EPR을 이용한 superoxide anion radical에 대한 소거활성 및 hydrogen peroxide에 대한 소거활성을 측정하였다. 그 결과, 민들레 잎의 물추출물의 뿌리의 물추출물 보다 지방산에 대한 과산화물 생성 저해율이 높았고, DPPH radical, hydroxyl radical, superoxide anion radical 및 hydrogen peroxide에 대한 소거활성 역시 매우 높은 것으로 나타났다. The antioxidative and free radical scavenging activity of water extracts of dandelion were investigated. Antioxidative and radical scavenging activity were assessed by means of different tests; inhibition of peroxidation on linoleic acid model system, scavenging DPPH radical, scavenging of hydroxyl radical by chemiluminescence assay, scavenging of superoxide anion radical by EPR spectroscopy and scavenging of hydrogen peroxide. The leaf extract showed strong antioxidant activity in linoleic acid system. The antioxidant activity of water extracts of dandelion increased with increasing concentrations of extracts. The scavenging activity of the dandelion extracts, on inhibition of the DPPH radical, was related to the reaction time. Hydroxyl radical were generated by fenton reaction and dandelion extract was found to scavenge OH in a concentration-dependent manner. The water extract of leaf had effective scavenging activities on hydrogen peroxide and superoxide anion radical. From the teses data, it is evident that water extract of dandelion leaf is an effective scavenger for OH, O_2^--, DPPH^-, hydrogen peroxide. And, the antioxidative effect observed is believed to be partly due to this radical scavenger activity.

        미성숙 목화다래 메탄올 추출물의 항산화 효능 평가

        본 연구에서는 미성숙 목화다래 추출물의 항산화능을 알아보기 위해 다래를 크기별로 구분하여 총 폴리페놀 및 플라보노이드 함량과 DPPH radical의 소거능, 아질산염 소거능, ABTS radical 소거능 및 SOD 유사활성을 측정하였다. 추출물의 페놀성 화합물과 플라보노이드 함량은 목화다래의 크기가 작을수록 높게 나타났으며, DPPH radical 소거능 및 ABTS radical의 소거능 또한 작은 크기에서 상대적으로 높은 활성을 보였다. 이는 다른 선행연구들에서와 같이 페놀함량이 높은 실험군에서 DPPH radical 소거능과ABTS radical 소거능이 높게 나타나 유의성을 확인하였다. 아질산염 소거능은 인체 위 내부환경과 같은 pH 1.2에서는높은 활성을 보이다가 pH 4.2에서는 점차로 활성이 떨어지는 결과를 보였으며, 중성에 가까운 pH 6.0에서는 아질산염소거에 영향을 미치지 못하는 것으로 확인되었다. 또한 SOD유사활성은 대조군인 ascorbic acid에 비하면 낮은 활성이었지만, 기 보고된 다른 천연물들의 효소활성과 비교하였을 때 더 높거나 비슷한 결과를 나타냈다. 이와 같이 본 실험을 통해 미성숙 목화다래의 뛰어난 항산화 활성을 확인할수 있었으며, 이는 향후 화장품이나 기능성 식품 등에 있어서 새로운 소재개발의 가능성을 시사해주는 것으로서, 그동안 주로 천연 솜의 생산을 위해 재배되어왔던 목화의 제한적 활용성을 더욱 다양하게 해줄 것으로 기대된다. The results of the content of total polyphenol and flavonoid, DPPH (1-1-diphenyl-2-picryl-hydrazyl) radicalscavenging activity, ABTS [2,2’-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)] radical scavenging activity, nitritescavenging activity and SOD-like activity of methanol extracts from immature cotton boll are follows. The contents of totalpolyphenol and flavonoid compound were higher in small size cotton boll, and DPPH and ABTS radical scavenging activityalso showed a relatively high activity in the small size. These results indicate that there is a correlation between phenolcontent and DPPH radical scavenging, ABTS radical scavenging. The test concentrations of immature cotton boll extract formeasuring DPPH and ABTS radical scavenging activities were set as 1.25, 2.5, 5, 10 and 20 mg/ml. Immature cotton bollhas high radical scavenging activity at the concentration of 1.25~20 mg/ml and the result showed a tendency to increase ina concentration-dependent. The nitrite scavenging activity showed high activity in the pH 1.2, and the result in the pH 4.2showed progressively less active, and in the pH 6.0 near neutral was confirmed that does not affect the nitrite scavenging. Inaddition, SOD-like activity showed somewhat lower activity compared with ascorbic acid, but tended to be higher whencompared with the results of the other natural substances. Through this experiment, we confirmed that immature cotton bollwas excellent antioxidant activity. Therefore, it is demonstrated that the cotton suggest the possibility of development ofnew material for cosmetic product or functional food in the future, and is expected to make a greater usability.

        Effects of boron/nitrogen/phosphorus doping on the scavenging action of armchair single‑walled carbon nanotubes (armchair‑SWCNT) for OH radicals: a DFT study

        Hydroxyl radical (OH radical) is the most harmful free radical amongst the Reactive Oxygen Species (ROS) responsible for numerous diseases of DNA damage like mutagenesis, carcinogenesis and ageing. Therefore, it is important to find a suitable scavenger for OH radical. In the present contribution, we aim to investigate the ability of pristine armchair-SWCNT and B/N/P-doped armchair-SWCNT to scavenge OH radicals using DFT calculations. The calculations reveal that the B/Pdoped armchair-SWCNTs can act as a better scavenger for OH radical compared to pristine armchair-SWCNT but N-doped armchair-SWCNT does not act as a better scavenger for OH radical compared to pristine armchair-SWCNT. Furthermore, the developed scavenger is examined in terms of large-scale availability, biocompatibility, conductivity, stability and reactivity. For both in vivo and in vitro studies, the work is found to useful for enhancing SWCNT as a free radical scavenger.

        Antioxidant Effects of Salicornia bigelovii Seed Extracts

        The objective of this study was to compare antioxidant effects of different extracts of Salicornia bigelovii seed. DPPH radical scavenging activity, chelating activity of ferrous, superoxide anion radical-scavenging activity, hydroxyl radical scavenging activity, and contents of total polyphenol and flavonoid were measured for different extracts. Ethanol extract (10 mg/mL) was found to have the maximum DPPH radical scavenging and superoxide anion radical scavenging activities (65.26 and 62.36%, respectively). However, hot water extract (10 mg/mL) showed the highest hydroxyl radical scavenging and chelating activities (51.50 and 53.24%, respectively). Total phenolics and flavonoid contents in the ethanol extract were 522.8 and 480.35 μg /mg, respectively. They were 401.51 and 352.60 μg /mg, respectively, in the hot water extract. These results indicate that ethanol and hot water extracts of S. bigelovii seed have good potential to be used as a new source of antioxidants for functional foods, cosmetics, and pharmaceuticals.

        Antioxidative Activities of the Ethyl Acetate Fraction from Heated Onion (Allium cepa)

        Heated onion juice was partitioned using the solvents hexane, chloroform, ethyl acetate, and butanol. The ethyl acetate fraction showed the strongest scavenging effect on the ABTS radical. The antioxidant activities of the ethyl acetate fraction from raw and heated onion (120, 130, and 140℃) were evaluated using radical scavenging assays. Radical and nitrite scavenging activities were higher in heated onion than raw onion, and the higher the temperature of heat treatment, the greater the radical and nitrite scavenging activities. Heated onion (140℃, 2 hr) was more effective than raw onion, having higher DPPH radical scavenging (5.7-fold), hydroxyl radical scavenging (6.4-fold), superoxide radical scavenging (2.3-fold), hydrogen peroxide scavenging (11.8-fold), and nitrite scavenging (4.3-fold) activities. Onion increased its physiologically active materials after heating, and in this regard, heated onion can be used as biological material for the manufacture of health foods and supplements.

        In vitro Antioxidant Activity of Crude Extract from Rubus coreanus MIQ.

        Objectives: This research aimed to evaluate the effects of Rubus coreanus MIQ. Methods: These extracts from fresh R. coreanus MIQ. were investigated with radical scavenging activity, the content of total phenolics, ferric thiocyanate method and flavonoid contents Results: These results revealed that the ethyl acetate extract from fresh R. coreanus MIQ. possesses remarkable radical scavenging activity. The content of total phenolics of the ethanol extract from dried R. coreanus MIQ. (IC50=155.87±8.8μg/mL) is the highest among all of the samples. The stronger radical-scavenging activity (IC50=77.72±2.1μg/mL) of ethyl acetate extract from dried R. coreanus MIQ. should be related to its phenolic content as measured by gallic acid test.         Comparison of Hydroxyl Radical, Peroxyl Radical, and Peroxynitrite Scavenging Capacity of Extracts and Active Components from Selected Medicinal Plants

        The ability of 80% ethanol extracts from five medicinal plants, Aralia continentalis, Paeonia suffruticosa, Magnolia denudata, Anemarrhena asphodeloides, and Schizonepeta tenuifolia, to neutralize hydroxyl radical, peroxyl radical and peroxynitrite was examined using the total oxyradical scavenging capacity (TOSC) assay. Peroxyl radical was generated from thermal homolysis of 2,2'-azobis(2-methylpropionamidine) dihydrochloride (ABAP); hydroxyl radical by an iron-ascorbate Fenton reaction; peroxynitrite by spontaneous decomposition of 3-morpholinosydnonimine N-ethylcarbamide (SIN-1). The oxidants generated react with α-keto-γ-methiolbutyric acid (KMBA) to yield ethylene, and the TOSC of the substances tested is quantified from their ability to inhibit ethylene formation. Extracts from P. suffruticosa, M. denudata, and S. tenuifolia were determined to be potent peroxyl radical scavenging agents with a specific TOSC (sTOSC) being at least six-fold greater than that of glutathione (GSH). These three plants also showed sTOSCs toward peroxynitrite markedly greater than sTOSC of GSH, however, only P. suffruticosa revealed a significant hydroxyl radical scavenging capacity. Seven major active constituents isolated from P. suffruticosa, quercetin, (+)-catechin, methyl gallate, gallic acid, benzoic acid, benzoyl paeoniflorin and paeoniflorin, were determined for their antioxidant potential toward peroxynitrite, peroxyl and hydroxyl radicals. Quercetin, (+)-catechin, methyl gallate, and gallic acid exhibited sTOSCs 40~85 times greater than sTOSC of GSH. These four components also showed a peroxynitrite scavenging capacity higher than at least 10-fold of GSH. For antioxidant activity against hydroxyl radical, methyl gallate was greatest followed by gallic acid and quercetin. Further studies need to be conducted to substantiate the significance of scavenging a specific oxidant in the prevention of cellular injury and disease states caused by the reactive free radical species.
