      • 암세포주에 대한 우황 50% MeOH 엑스와 함암제의 병용효과

        The studies were conducted to investigate the combined effects of 50% MeOH extract of Be-zoar Bovis(MEB) and several anti-tumor drugs. The effects of MEB and several anti-tumor druge on the proliferation of HaLa, Hep G2, SNU-1, MOLT-4 and S 180 cells estimated by MTT colorimetric assays. MEB inhibited the proliferation of HeLa, Hep G2, SNU-1, MOLT-4 and S 180 cell. The inhibitory action of mitomycin C(MMC), cisplatin(CPT) or 5-fluorouracil(5-FU) on the proliferation of HeLa cell was increased by the combined of MEB and their drugs, and the inhibitory action of MMC or CPT on the proliferation of Hep G2 and S 180 cell was increased by the treatment of MEB and their drugs. MEB inhibited the proliferation of Balb/c 3T3 and Tlymphocyte at 10^-4g/ml. The 50% inhibitory concentration of MMC, CPT and 5-FU on T lymphocyte were 0.75, 0.57 and 28.35㎍/ml repectively. The inhibitory activity of MMC, CPT or 5-FU treated group, respectively, on the proliferation of T lymphocyte was increased by the combined treatment MEB and their drugs. MEB increased the number of leukocyte at 5th day. In the groups of the combined treatment of MMC and MEB, the number of leukocytes and the weight of thymus were increased significantly than MMC treated group. MEB increased the number of total peritoneal cells and inbibited the exudation of evans blue into peritoneum. In conclusion, this paper suggest that MEB inhibits the proliferation of HeLa, Hep G2, SNU-1, MOLT-4 and S 180 cells, and the combined treatment of anti-tumor drugs and MEB increase the inhibitory activity of their drugs on several tumor cells. MEB has the possibility of decreasing the side effect of anti-tumor drugs such as leukopenia and immunsuppression.

        수종보익제가 면역세포의 조절 및 Apoptosis에 미치는 영향

        The effects of several tonifying prescriptions on the apoptosis of transplanted L1210 cells and the regulation of immune cells in L1210-transplanted mice were studied. Sa-Mul-Tang(SMT), Sa-Kunja-Tang(SKT), Sipjeon-Daebo-Tang(SDT) accelerated the apoptosis of transplanted L1210 cells, and SMT and SKT reduced the mitochondrial transmembrane potential of transplanted L1210 cells. SMT promoted the proliferation of thymocytes and splenocytes in L1210-transplanted mice. Whereas SMT increased CD8+ T_c/T_S cell, Bojung-Ikki-Tang(BIT) decreased CD4+ T_H cell population in the thymocytes of L1210-transplanted mice. SMT and SKT increased the lucigenin chemiluminescence from peritoneal macrophage of L1210-transplanted mice. These results indicate that SMT and SKT have an inductive action of apoptosis of transplanted L1210 cells and peritoneal macrophage activation.

      • 複合生藥製劑의 止血作用및 摘出 子宮筋에 미치는 影響(第3報) : 加減當歸補血湯

        These experiments were performed to investigate the effects of the KAGAM-DANGKWI-BOHYUL TANG(KDBT) extract, clinically used in gynecology, on the hemostatis and on the contractile force of the isolated uterine muscle. The bleeding time and the plasma prothrombin time test were employed as the means to estimate the hemostatic effect of KDBT extract, and the contractile effect of the extract was measured. The experimental results were summarized as followings; 1. In mice treated with KDBT extract the plasm prothrombin time and the bleeding time were remarkably shortened compared with the control group. 2. The contractile effect of KDBT extract on the isolated uterine muscle was not blocked by atropine and cyproheptadine. 3. The contractile effect of KDBT extract on the isolated uterine muscle was inhibited by the pretreatment with verapamil. From these experimental results, it is concluded that the hemostatic effect of KDBT extract might be attributed to the stimulation of prothrombin formation and the direct contraction of the uterine muscle through the increasing uptake of calcium ions into the muscle.

      • ROSAE MULTIFLORAE RADIX의 藥理作用에 關한 硏究

        In an attempt to investigate the effect of ROSAE MULTIFLORAE RADIX(RME) on the diabetes in rats induced by alloxan(80 ㎎/㎏) or streptozotocin(40 ㎎/㎏) and the contractile force of isolated rabbit ileum, RME was administered per oral daily and the level of blood glucose, serum triglyceride and serum total cholesterol were measured by spectrophotometer and the contractile force of intestine was measured by physiograph. The following resuits were obtained: 1) RME did not change the level of blood glucose and serum total cholesterol, but decrease the serum triglyceirde on the 5th day in normal rats. 2) The level of blood glucose in diabetic rats induced by the alloxan was decreased on the 5th and 7th day of experiment (RME 100㎎/㎏), induced by streptozotocin was decreased on the 7th day of experiment (RME 10 ㎎/㎏) and 5th and 7th day of experiment (RME 100 ㎎/㎏). 3) The level of serum triglyceride in dibetic rats induced by the alloxan was decreased 7th day (RME 100 ㎎/㎏) and induced by streptozotocin was decreased 7th day (RME 10 ㎎/㎏) and 3th, 5th and 7th day (RME 100 ㎎/㎏) of experiment. 4) The level of serum total cholesterol in diabetic rats induced by the alloxan and streptozotocin was not changed by RME. 5) The contractile response of isolated rabbit ileum by RME was not affected by cyproheptadine, but inhibited by atropine and verapamil.

        정신분열병과 N-Methyl-D-Aspartate 수용체 유전자와 연관된 VNTR 표지자인 D9S158 다형성의 연합

        정신분열병과 NMDA 수용체와 연관된 D9S158 유전적 표지자와의 연관성을 알아보기 위해 75명의 정신분열병 환자군과 87명의 정상대조군을 대상으로 중합효소연쇄반응을 이용하여 연합연구를 시행하여 다음과 같은 결론을 얻었다. D9S158 유전좌위에는 모두 8개의 대립유전자가 있었고, 각 대립유전자는 cytosine-adenine염기가 반복되는 217염기쌍에서 233염기쌍의 형태였다. 먼저 정신분열병 환자와 정상대조군의 대립유전자의 빈도를 비교해 본 결과 통계적으로 유의한 차이를 보이지 않았다(p〉.05). 환자군을 좀 더 동질적으로 만들기 위해 정신분열병의 가족력 유무, 음성 및 양성증상, 연성신경학적 징후, DSM-IV 임상아형에 따라 환자군을 나누어 대립유전자의 빈도를 비교해 본 결과, 특정 임상양상과 관련이 있는 대립유전자를 발견하지 못하였다. 결론적으로 본 연구에서는 D9S158 유전좌위와 정신분열병의 연관성을 입증하지 못하였으나 NMDA 수용체 유전자주위의 다른 표지자나 좀 더 유전적으로 동질한 환자군을 대상으로 한 추후연구가 필요할 것으로 생각된다. An association study with Korean schizophrenic patients(N=75) and normal controls(N=87) was performed to find the relationship between D9S158 polymorphism and schizophrenia using polymerase chain reaction. Eight different alleles of a dinucleotide polymorphism on D9S158 locus were observed in both group. When we compared the frequencies of alleles between schizophrenics and normal controls, there was no significant difference between two groups. To increase homogeneity of schizophrenic group, we divided schizophrenic group by clinical phenotypes such as family history, negative and positive symptoms(PANSS), soft neurologic signs(NES-K) and DSM-IV diagnostic subtypes. Then we compared the frequencies of alleles among subgroups of clinical phenotypes, and there were no significant differences between subgroups(p〉.05). Although our findings fail to provide an evidence of association between schizophrenia and D9S158 locus, further investigation of other loci that are linked to NMDA receptor gene may be needed in genetically homogeneous subgroups of schizophrenia.

      • 황금(Scutellaria baicalensis)의 열수 추출물이 병원성 세균에 미치는 항균활성

        This study was investigated antibacterial activity of aqueous fractions from ScuteItaria baicalensis against various pathogenic bacteria. Extracts were obtained from the roots of 5. baicatensis which has shown selective antibactehal effects on various strains of bacteria. The aqueous extract of S. baicatensis was tested on 19 species of human pathogenic bacteria. The bacteria were cultured in nutrient broth, and YM broth then incubated aerobically at 37℃ for 18-24 hours. Antimicrobial activities tested by paper disc and radial diffusion assay methods. The aqueous extracts of 5. baicatensis inhibited the growth of most human pathogenic bacterial and showed 62.5㎎/㎖ of MIC values against StaphyIococcus aureus SG511 and 125㎎ /㎖ of MIC values against Staphytococcus aureus 285 and Staphylococcus aureus 503. These results suggest that S. baicaIensis extract can be used as an effective natural antibiotics.

        최근 개발된 cone beam computed tomography의 흡수선량 및 유효선량 평가

        Purpose : Cone beam computed tomography (CBCT) provides a lower dose and cost alternative to conventional CT, promising to revolutionize the practice of oral and maxillofacial radiology. The purpose of this study was to evaluate the absorbed and effective doses of Implagraphy and VCT (Vatech Co., Hwasung, Korea) and compare them with those of panoramic radiography. Materials and Methods : Thermoluminescent dosimeter (TLD) chips were placed at 27 sites throughout the layers of Female ART Head and Neck Phantom for dosimetry. Implagraphy, VCT units, and Planmeca Proline XC panoramic unit were used for radiation exposures. Radiation weighted doses and effective doses were measured and calculated using 1990 and 2005 ICRP tissue weighting factors. Results : Effective doses in Sv (ICRP 2005, ICRP 1990) were 90.19, 61.62 for Implagraphy at maxillay molar area, 123.20, 90.02 for Implagraphy at mandibular molar area, 183.55, 139.26 for VCT and 40.92, 27.16 for panoramic radiography. Conclusion : Effective doses for VCT and Implagraphy were only about 2.2 to 4.5 times greater than those for panoramic radiography. VCT and Implagraphy, CBCT machines recently developed in Korea, showed moderately low effective doses.
