네트워크 약리학을 활용한 메니에르병에 대한 二陳湯의 활성 성분과 치료 기전 연구(Ⅰ)

Objectives : This study used a network pharmacology approach to explore the active compounds and therapeutic mechanisms of Yijin-tang on Meniere's disease. Methods : The active compounds of Yijin-tang were screened via the TCMSP database and their target proteins were screened via the STITCH database. The GeneCard was used to establish the Meniere's disease-related genes. The intersection targets were obtained through Venny 2.1.0. The related protein interaction network was constructed with the STRING database, and topology analysis was performed through CytoNCA. GO biological function analysis and KEGG enrichment analysis for core targets were performed through the ClueGO. Results : Network analysis identified 126 compounds in five herbal medicines of Yijin-tang. Among them, 15 compounds(naringenin, beta-sitosterol, stigmasterol, baicalein, baicalin, calycosin, dihydrocapsaicin, formononetin, glabridin, isorhamnetin, kaempferol, mairin, quercetin, sitosterol, nobiletin) were the key chemicals. The target proteins were 119, and 7 proteins(TNF, CASP9, PARP1, CCL2, CFTR, NOS2, NOS1) were linked to Meniere's disease-related genes. Core genes in this network were TNF, CASP9, and NOS2. GO/KEGG pathway analysis results indicate that these targets are primarily involved in regulating biological processes, such as excitotoxicity, oxidative stress, and apoptosis. Conclusion : Pharmacological network analysis can help to explain the applicability of Yijin-tang on Meniere's disease.

Objectives : This study used a network pharmacology approach to analyze the treatment mechanisms of Yijin-tang on Meniere's disease, and comparative analysis the treatment mechanisms of drugs recommended in the Meniere's disease treatment guidelines. Methods : We collected information on the recommended drugs from the Meniere's disease treatment guidelines and their target proteins were screened via the STITCH database. The intersection targets were obtained through Venny 2.1.0. Gene Ontology(GO) analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway analysis were performed using ClueGO. Results : The 7 proteins(TNF, CASP9, PARP1, CCL2, CFTR, NOS2, NOS1) were associated with both Yijin-tang and Meniere's disease related genes. The 10 proteins(AQP2, KCNE1, AQP1, AVP, ACE, HRH1, HRH3, NOS1, CA1, CFTR) were associated with both the recommended drugs in the guidelines and Meniere's disease related genes. The 2 proteins(CFTR, NOS1) were common across all three groups. Further, GO/KEGG pathway analysis of the collected proteins revealed that the common mechanisms of action between Yijin-tang and the recommended drugs in the guidelines were related to pathways involving immune dysfunction and disturbances in lymphatic fluid homeostasis. In addition, the recommended drugs in the guidelines appeared to act through mechanisms that improve blood flow through vasodilation. Conclusions : Pharmacological network analysis can help to explain the treatment mechanisms of Yijin-tang on Meniere's disease. .
폴리비닐알코올 하이드로겔 캅셀을 삽입한 중공좌제로부터 염산프로프라놀롤의 조절 방출

Hollow type suppositories inserted polyvinyl alcohol (PVA) hydrogel capsule containing propranolol. HCl (PPH) were prepared using different bases, polyethylene glycol (PEG), Witepsol H-15(WH-15) and Witepsol W-35 (WW-35) to improve the controlled release of PPH. The release of PPH from the hollow type suppository inserted PVA hydrogel capsule was retarded than that from PEG, WH-15, or WW-35 hollow type suppositories in rat rectal cavity. When the suppositories were administered to rats, the controlled release of PPH was proved by the plasma concentration-time profiles of PPH. No significant difference (p<0.05) among the three different hollw type suppositories was observed in terms of AUC and MRT of PPH. WH-15 hollow type suppository inserted 12% of PVA hydrogel capsule caused irritation to rat rectal mucosa. However, the WH-15 hollow type suppository inserted PVA hydrogel capsule caused no severe irritation on rectal mucosa. The application of the hollow type suppositories using PVA in sustained rectal delivery of drugs might be feasible.
뇌척수액에서 실시간-이중-역전사중합효소연쇄반응을 이용한 장바이러스의 검출

배경 : 장바이러스(enterovirus)는 무균수막염, 호흡기질환, 피부병변, 심근염, 뇌염 등의 다양한 증상을 유발할 수 있다. 감염의진단은 항체검사와 숙주세포를 이용한 배양법 및 중합효소연쇄반응에 의존해 왔으나 낮은 민감도가 문제가 되어 왔다. 이러한 단점을 보완하기 위해 저자들은 실시간-중합효소연쇄반응의 장점을이용한 장바이러스 검출법의 유용성을 평가하였다.방법 : 무균수막염이 의심되어 뇌척수액 검체를 채취한 50명을대상으로 세 종류의 각기 다른 표적을 대상으로 시행한 역전사중합효소연쇄반응과 실시간-이중-역전사중합효소연쇄반응을 시행하였다. 이 중 4명의 환자에 대해 바이러스 배양검사를 시행하였다.결과 : 세 종류의 각기 다른 표적을 대상으로 시행한 중합효소연쇄반응의 결과 각 26.0%, 40.0%, 46.0%의 장바이러스 검출률을 보였으며 바이러스 배양법의 양성률은 6.8%였다. 실시간-중합효소연쇄반응의 장바이러스 검출률은 86.0%였다.

Gastritis is the most common disease among Korean. The demand for the development of gastritis drugs has been increasing. Currently, however, there is no guideline available for the clinical evaluation of gastritis drugs worldwide. As a consequence, domestic and international pharmaceutical companies make errors in the drug development processes, and it becomes difficult for them to establish the scientific validity and objectivity of newly developed drugs. The objective of this study was to develop the Guideline for Clinical Trials Evaluation of Gastritis which can be used in improving the quality and consistency of clinical trials. First, we collected and reviewed the clinical trials on gastritis drugs that were available from Japan Pharmaceuticals and Medical Devices Agency and Korea Food and Drug Administration (KFDA), and investigated the recent research trends on clinical trials of gastritis drugs. Reviewers from KFDA and National Institute of Food and Drug Safety Evaluation and scientific experts from the pharmaceutical industries developed the guidelines through regularly scheduled meetings. Opinions and consultation from academic fields and industry experts were also obtained. This project will provide the clinical trial practitioners, investigator and reviewers the scientific and rational guidelines for performance and evaluation of clinical trials for gastritis drugs. Furthermore, we hope this guideline contributes to establishing the national competitiveness, improving the quality of clinical trial, and encouraging researches on drug development for gastritis.

A new double cropping potato variety, ‘Gangseon’, was bred by the Highland Agriculture Research Institute, National Instituteof Crop Science (NICS), RDA, Korea, in 2016. ‘Gangseon’ was obtained from the cross between ‘Daegwan1-83’ and ‘Chubaek’ in 2006. Seedling and line selection was performed from 2008 to 2009, and preliminary and advanced yield trials were carried out with ‘Gangseon’from 2010 to 2013 in Gangneung city. The regional yield trials were conducted in Gangneung, Muan, and Jeju from 2014 to 2016. ‘Gangseon’has light purple flowers, green narrow leaflets and petioles with medium anthocyanin coloration, round tubers with yellow russet skin, andyellow flesh. The dormancy of ‘Gangseon’ stored in 20 ℃ was 60-70 days after harvesting, which is longer than those of ‘Dejima’ and‘Chubaek’, but shorter than that of ‘Superior’. The average yields of tubers in the spring and fall seasons were 36.6 and 21.2 ton⋅ha-1,respectively, in the regional yield trials. It is susceptible to common scab and potato virus Y, but has high resistance to potato leaf rollvirus, potato virus X, early blight, and late blight. The dry matter contents were 22.1% in spring and 19.8% in autumn. The adaptation areaof ‘Gangseon’ is the southern coastal area, including Jeju Island, Korea. The plant variety protection right of ‘Gangseon’ was registered inApril 2020, and its grant number is 8081.

We all human beings, should be reached the terminal of life in the world. There is the only difference between that comes suddenly or slowly. Persons who should be come the terminal stage suddenly due to disease, especially, malignancy, are Hospice patients. Hospice work is the work of all of us because anyone, anywhere, whenever can be suffered in terminal stage. The characters of Hospice-care are total care of wholistic human beings, comprehensive total assessment of the life and the team work composed of diverse team-members, for example, doctors, nurses, social workers, physical therapists, psychologists, ministers & volunteers. The care manager of the total care(the coordinator of Hospice care), should be worked systemically and, rationally. The comprehensive assessment concept should be entered to the infra-consultant of terminal care-program. The care manager should be have the ability of comprehensive assessment for terminal patients. It will also help standardization of Hospice, and application of medical insurance and social security.

This study has been conducted to investigate the amount of residual sulfur dioxide for herbal medicines of 30 spe-cies which are purchased in 13 different regions (Bonghwa, Busan, Chunju, Jecheun, Kwangju, Keumsan, Seoul, Taeku, Che-ungdu, Xian, Beijing, Tokyo and Osaka). The sulfur dioxide residues were determined in the collected 386 samples by themodified Monier-Williams method. The residues of sulfur dioxide in 386 samples ranged from ND (under detection limit) toArgyi Herba, Scolopendrae Corpus, Schizandrae Fructus and Cyperi Rhizoma were detected under the detection limit regardlessof the collected regions. The sulfur dioxide residues in herbal medicines collected in domestic regions were relatively lower thanthose in foreign regions. There are no standards for sulfur dioxide residual limits of herbal medicines in Korea. This results willprovide the scientific basis for the standardization of sulfur dioxide residues in Korea Pharmacopoeia. M.W. method
