      • Effect of intense pulsed light on the deactivation of lipase: Enzyme-deactivation kinetics and tertiary structural changes by fragmentation

        <P><B>Abstract</B></P> <P>The effect of intense pulsed light (IPL) irradiation on <I>Chromobacterium viscosum</I> lipase was investigated with a primary focus on catalytic activity and molecular structure. During IPL irradiation, lipase activity decreased significantly with increasing pulse fluence (F<SUB>p</SUB>) and exposure time (t<SUB>e</SUB>). IPL-induced deactivation kinetics were further elucidated based on a two-step series-type deactivation model (constant deactivation rate <I>k</I> <SUB>1</SUB> ><I>k</I> <SUB>2</SUB>). F<SUB>p</SUB> was found to be the dominant variable affecting the degree of lipase deactivation, and residual activity was not associated with increasing t<SUB>e</SUB> below a certain F<SUB>p</SUB> energy density (2.66 mJ/cm<SUP>2</SUP>), implying a critical threshold for IPL-induced deactivation of lipase. From the results of fluorescence spectroscopy and sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), we determined that IPL-induced deactivation was caused by fragmentation, leading to lipase tertiary structural changes. Furthermore, the results of FindPept analysis and matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) indicated that the internal sensitive bonds of lipase were cleaved preferentially by IPL, such that IPL irradiation induced site-sensitive fragmentation and peptide bond cleavage.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Intense pulsed light (IPL) irradiation deactivated <I>Chromobacterium viscosum</I> lipase. </LI> <LI> Lipase deactivation was elucidated by the two-step series type deactivation kinetics. </LI> <LI> Pulse fluence (F<SUB>p</SUB>) was the dominant variable affecting the IPL-induced deactivation. </LI> <LI> The lipase deactivation was caused by fragmentation of protein structure. </LI> <LI> IPL irradiation induced site-sensitive fragmentation and peptide bond cleavage. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Background/Aims: Westernization in many Asian countries have changed lifestyles and diets, so once rare diseases have now become prevalent. The aim of this study is to investigate the changing pattern of digestive and liver disease in Korea, from 1990 to 2006. Methods: We extracted data specific gastrointestina (GI) disease based on the International Classification of Diseases code from the in-patients records at the Kangnam St. Mary`s Hospital in 1990, 1996 and 2006. This hospital is a tertiary-care hospital in Seoul, Korea, which has a capacity of 800 beds. Results: The admission rate for GI disease increased between 1990 and 2006. Overall in-patients were 1,623 persons in 1990, 2,368 persons in 1996 and 4,166 persons in 2006. The mean age of in-patients increased as time went by. A stomach cancer was the most common diagnosis during all periods, but its prevalence has decreased. Colon cancer ranked the 7th in 1990, but markedly increased and now ranks the second. The third was a hepatocellular carcinoma. The bile duct and gallbladder cancer, pancreas and esophageal cancer ranking followed with little interval change. In cases of cancer patients, a regular admission dramatically increased for chemotherapy. The number of patients admitted with pre-malignant neoplasm of stomach and colon increased remarkably with the development of endoscopy. The liver transplant, inflammatory bowel discase, and reflux esophagitis emerged form mid-1990s with greater frequencies, yet much below the levels found in the West. The admission rate for peptic ulcer, especially ulcer bleeding remained relatively stable, despite a decreased rate for ulcer perforation. Liver cirrhosis, hepatitis, cholelithiasis with cholecystitis-cholangitis, appendicitis, hemorrhoid and gastritis all decreased. Conclusions: The stomach cancer is the leading cause of admission, despite a recent decline. Colon cancer showed a marked rise. The liver transplant, inflammatory bowel disease and reflux esophagitis were new diagnosis with an increased tendency.

        We experienced a 25 year-old male patient with typhoid fever complicated with massive hemoptysis. Pulmonary complication in typhoid fever is very rare and to our knowledge, there has been no report of hemoptysis as a main cause of death with this disease. We herein report a rare case of typhoid fever.

        Since most program execution time is spent in the loop structure, the researches of transforming sequential programs into parallel programs have been focused on the method of a loop structure transformation. But, most programs have implicit parallelism of interprocedure. Most parallelization of loop with procedure calls just focus on the uniform code. This paper suggestions a way of procedure analysis (an interprocedural analysis and an intraprocedural analysis). We extend the previous loop transformation algorithms for procedure transformation analysis. By using the suggested transformation analysis, we show the examples of parallelization using uniform code, nonuniform code, and complex code, and then evaluate the performance of a sequential and a parallel code using CRAY-T3E.

        The reactions of nitric oxide with hydrogen, carbon monoxide and hydrogen-carbon monoxide mixture have been investigated over the catalysts supported on alumina. In NO-H_2 System the relative activity of catalysts was decreased in the order as Pd>Ru>Pt>Os, while it was Ru>Pd<Pt<Os in NO-CO system. The relative activity sequence of catalysts in NO-H_2-CO system was similiar to that in the mixture of nitric oxide and carbonmonoxide. The inverstigation on the effectiveness of air-fuel ratio for the purification of exhaust gases has also been carried out over alumina supported ruthenium and palladium. Of exhaust gases the nitric oxide conversions were decreased slowly with increasing the air-fuel ratio, while the carbon monoxide conversions were increased slowly under the same condition. In this experiment the best value of air-fuel ratio was 14.5. In the nitric oxide conversions, the ruthenium and palldium of catalysts used were not so much inhibited by carbon monoxide.

        원격 체육수업 콘텐츠 개발 방향 연구

        The purpose of this study is to provide a direction for developing content to be utilized in online physical education( PE) class situations. Four faculty members and four school sports content production experts produced examples of content development after identifying the forms and difficulties of PE curriculum analysis, online PE class content analysis, and online PE classes. The principles and methods for restructuring the areas of PE curriculum and achievement criteria were adjusted from five to four, and achievement criteria were restructured in the categories of ‘maintenance’, ‘integration’, ‘modification’ and ‘movement’. Examples of physical activities were selected to develop content for online PE classes, and details of content development examples were defined, including topics for each unit, major learning activities, teaching and learning strategies, and evaluation methods. The direction of future development of contents for online PE classes is as follows. First, the PE curriculum should be restructured following the proportion of online PE classes. Second, while utilizing ‘modified physical activity’ to be carried out in online PE classes, it should be aligned to existing physical activities. Third, it should be developed as ‘real-time interactive’ content rather than one-way video content. It is required to establish an online evaluation system in the future and adopt diversified ways to reflect performance evaluations. 본 연구의 목적은 원격 체육수업 상황에서 활용할 콘텐츠 개발 방향을 제시하는 데 있다. 4명의 교수진과 4명의 학교체육 콘텐츠 제작 전문가가 체육과 교육과정 분석, 원격 체육수업 콘텐츠 분석, 원격 체육수업의 형태 및 어려움 등을 파악하고 콘텐츠 개발 예시를 제작하였다. 체육과 교육과정의 영역과 성취기준의 재구성 원칙과 방법을 구안하여, 영역은 5개에서 4개로 조정하고, 성취기준은 ‘유지’, ‘통합’, ‘수정’, ‘이동’ 의 방식으로 재구성하였다. 원격 체육수업 콘텐츠 개발을 위해 신체활동 예시를 선정하고, 단원별 주제, 주요 학습 활동, 교수·학습 전략, 평가 방법이 포함된 콘텐츠 개발 예시를 상세화하였다. 향후 원격 체육수업 콘텐츠 개발 방향은 첫째, 체육과 교육과정은 원격 체육수업 비중에 따라 재구성해야 한다. 둘째, 원격 체육수업에서 수행할 ‘변형 신체활동’을 활용하되, 기존 신체활동과 연계성이 있어야 한다. 셋째, 일방향의 동영상 콘텐츠 활용형보다는 ‘실시간 쌍방향형’ 콘텐츠로 개발해야 한다. 향후 온라인 평가 시스템을 구축하고, 수행평가를 반영할 수 있는 방법을 현재보다 다양화해야 한다.

      • 명태의 행동제어용 심전도 도출에 관한 연구

        In the walleye pollock. Theragra chalcogramma. which has a heart with a strong electromotive force, the ECGs were found to be recordable by electrodes placed in the water from fish's body. The day on the experiment, fish was operated on in order to install a leading-out bipolar electrode. This electrode was used for recording of the cardiosignal of the fish on a digital storage oscilloscope. The electrode was made from a fish book which was re-shaped according to the shaped of fish heart and body surface. This hook was then connected to the lead-code insulated with paint, and the connecting part was covered with paint. The pointed end was stripped off slightly for conduction of electricity and the another pointed edge was used for hooking the electrode into the chest cavity. QRS complex of the ECG was marked. The amplitude of QRS complex was about 0.1-lmV. This method enabled us to observe the heart beat rate of the walleye pollock under least disturbed state. Ordinary heart beat rate was not quite regular. Very slight stimulation was enough to bring about cardiac inhibition. which was also accompanied by a considerable reduction in the amplitude of the QRS. After rough treatment. such as sound stimulus or electric shock. the heart beat rate showed remarkable decrease. reaching a value a few times lower than the normal level.

        정신분열병 가계에서 6p24-22 염색체에 대한 연관 분석

        Objectives : Chromosome 6p24-22 has been identified as a disease locus with a high probability for schizophrenia based on several genomewide linkage scans with Caucasian families. The recent association studies suggest that the dysbindin gene located at chromosome 6p22.3 may be a candidate gene of schizophrenia. The purpose of this study was to investigate the linkage of chromosome 6p24.3-22.3 locus to schizophrenia in Korean families. Methods : We recruited one hundred fifty-seven family members from forty-six multiplex schizophrenia families. One hundred three of them were affected individuals. four microsatellite markers with 4.8 cM intervals on 6p24.3-22.3 were genotyped. Non-parametric linkage analysis was performed by evaluating the levels of allele sharing between the affected relative pairs. Results : In the single point analysis, no markers on chromosome 6p24.3-22.3 locus showed statistical evidence for linkage. Significant evidence for linkage was not found in the multi-point analysis. Conclusion : These results do not support the previous evidence from Caucasian families for a locus predisposing to Schizophrenia at 6p24.3-22.3, the locus of dysbindin gene. We conclude that if there is a susceptibility locus for schizophrenia in this region then its effect size is so small as to render our study insufficiently powerful to detect it and schizophrenia susceptibility loci in Korean families likey have different ethnicity-specific effects from Caucasian families.

      • 교내 수도꼭지형 냉온수기의 생수 배출 부위와 생수에 존재하는 미생물의 분포

        The distributed species and number of viable microorganisms in drinking water were monitored according to water dispenser maintenance. our purpose was to investigate the distribution of bacteria in drinking water dispenser. To analyze possible dispenser contaminations sampling were performed from 14 locations in Namseoul University for 3 months from March to May, 2010. The isolated bacteria were identified by Gram stain and biochemical test using VITEKⅡ systems. The microorganism concentrations around the water discharge areas were measured and the average of total microorganisms was determined. The number of bacteria were measured by McFarland turbidity. The water and water discharge areas were separated and identified. The isolation rates of Gram positive cocci and Gram negative bacilli were 56.5%, 17.4% respectively. The frequently isolated bacteria were Staphylococcus epidermidis, Enterobacter aerogenes, Enterbacter sakazakii, Alcaligenes faecalis, Acinetobacter baumannii, Staphylococcus hominis, respectively. Further systematic studies are necessary with an emphasis on species identification. Key Words : Microorganisms, Water Discharge Areas, Drinking Water, McFarland
