대복, Gomphina veneriformis 아가미의 조직학적 변화와 산소소비율에 미치는 TBTCl의 독성

This study was conducted to find out biological response of bivalves exposed to tributyltin chloride (TBTCl). The results of the study confirmed that TBTCl induce the reduction of oxygen consumption rate and histopathological feature in the gill structure of equilateral venus, Gomphina veneriformis. The experimental groups consisted of a control and 3 TBTCl exposure groups (0.4 ㎛ TBTCl L-¹, 0.6 ㎛ TBTCl L-¹, 0.8 ㎛ TBTCl L-¹) and the experimental period was 36 weeks. For histological analysis, gill tissues were fixed in Bouin's fluid and then stained H-E stain, AB-PAS (pH 2.5) reaction and Masson's trichrome stain after having serially sectioned the tissue by paraffin method at thickness of 4~6 ㎛. The oxygen consumption rate was not significantly different between the control and exposure groups at 4 weeks, but in all exposure groups at 28 weeks, it was significantly different to the control. Gill of G. veneriformis had demibranch that attached two sheets of lamellae and a lamella was composed of numerous filaments, numbering 25 on average. The frontal filament zone had three types of cilia; frontal, latero-frontal and lateral depending on locations while the lateral cilia were the longest and largest in number. The mucous cells observed in filaments were more abundant in the frontal filament zone than abfrontal filament zone and mucous substances were showed blue color (542c) in AB-PAS (pH 2.5) reaction. Gill exposed to TBTCl was extended hemolymph sinus and increased hemocytes at 4 weeks, and then it showed increases of mucous cells and partially disappearance of frontal cilia. In the group of 0.8 ㎍ TBTCl L-¹ at 12 weeks, hypertrophy of frontal and latero-frontal epithelia was observed. Also it observed m decrease of mucous cell containing weekly acid mucosubstance and appeared partially destruction muscle fiber bundle. In the groups of 0.4 and 0.6 ㎍ TBTCl L-¹ at 36 weeks, it appeared partially modification of epithelia and in 0.8 ㎍ TBTCl L-¹ group, observed filaments that come out chitinous rod from disappearance of frontal and latero-frontal epithelia.

This study was conducted to find out biological responses of bivalves exposed to organotin compound. The results of the study confirmed that tribultyltin chloride (TBTCl) induce reduction of survival rate and burrowing activity, and histopathological feature in the foot structure of the equilateral venus, Gomphina veneriformis. The experimental period was 36 weeks. The experimental groups consisted of a control and 3 TBTCl exposure groups (0.4 ㎛ TBTCl L-¹, 0.6 ㎛ TBTCl L-¹, 0.8 ㎛ TBTCl L-¹). The survival rate and activity were recorded daily. For histological analysis, foot tissues were fixed in Bouin’s fluid and then stained H-E stain and AB-PAS (pH 2.5) reaction after having serially sectioned the tissue by paraffin method at thickness of 4~6 ㎛. The survival rate was not significantly different among the control and exposure groups for 20 weeks, but in 0.8 ㎛ TBTCl L-¹, it was on the decreased ever since the exposure. The burrowing activity was not significantly different in the exposure group compared to the control up to 12 weeks, but in 0.6, 0.8 ㎛ TBTCl L-¹, it measured the lowest level after 20 weeks. The foot is composed of the epidermal layer, connective tissue, and muscular layer. The epidermal layer is composed of simple columnar, cuboidal epithelium and mucous cells. The cilia were well developed on the apical surface of epithelium. Circular, longitudinal and transverse muscle bundle were well developed in the muscular layer. The majority mucous cells showed blue color (542c) when it subjected to AB-PAS (pH 2.5) reaction. No histopathological alterations in the foot were observed before 12 weeks of exposure. After 20 weeks of exposure to 0.8 ㎛ TBTCl L-¹, the foot samples of exposed G. veneriformis showed disappearance of cilia and striated border partially and extension of hemolymph sinus. The mucous cell increased in the marginal of foot. At 28-week of exposure to 0.4 ㎛ TBTCl L-1, it observed weekly acid (564c), neutral (264c) and mixed mucous cell. At 36-week of exposure to 0.6 ㎛ TBTCl L-1, it showed fragmentation of the muscle and collagen fiber bundle, and also diappearance of cilia on epithelia and edema of epithelium in 0.8 ㎛ TBTCl L-¹.

아치형재배 시설장미에서 점박이응애(응애아강: 잎응애과)의 밀도추정 및 방제의사결정을 위한 표본추출법 개발

아치형재배 시설장미(Rosa sp., "Rote")에서 점박이응애(Tetranychus urticae koch)의 정확한 표본추출법 개발과 평가를 위한 실험이 1997년 7월부터 1998년 1월까지 이루어졌다. 5개엽으로 이루어진 복엽을 장미주 상, 중, 하단에서 채취하였으며, 각 복엽은 relative net precision값을 비교하기위해 3개의 서로다른 단위(1엽, 3엽, 5엽 단위)로 나뉘어졌다. 그 결과 1엽 단위가 다른 단위들 보다 정확도와 효율적인 면에서 우수했다. 대부분의 점박이응애의 밀도는 장미의 상단과 하단에서 얻어졌다. Taylor의 power law가 Iwao의 patchiness regression보다 자료를 잘 설명해주었으며, 점박이응애 개체군의 장미주내의 Taylor의 Power law계수인 기울기와 절편은 상단과 하단이 차이를 보이지 않았다. 따라서 표본정시선을 구하기 위한 Taylor의 power law 계수는 장미주 상단과 하단의 장미 1엽 단위에서 점박이응애 밀도 자료를 합친 뒤 재계산하여 사용하였다. 유리온실 장미의 점박이응애 발생과 관련된 방제의사결정은 장미주 상단과 하단에서 피해를 주는 점박이응애의 밀도를 각각 임의로 정하여 구하였다. 정하여 구하였다. Greenhouse rose (Rosa sp., cultivar "Rote" ) grown by arching method was sampled from July, 1997 thorough January 1998 to develop and evaluate the two spotted spider mite (TSSM), Tetranychus urticae Koch, sampling programs. A compound leaf consisted of five leaflets was collected from the top, middle and bottom plant canopies, and evaluated 3 different sampling units (1-leaflet, 3-leaflet and 5-leaflet) for their relative net precision. Relative net precision values indicated a 1-leaflet unit was more precise and cost-efficient than either the 3-leaflet unit or 5-leaflet unit. Taylor's power law provided a better fit to the data than Iwao's patchiness regression. Slopes and intercepts of Taylor's power law did not differ between TSSM populations from the top and bottom canopies. A fixed- precision- level sequential sampling plan was developed using the parameters of Taylor's power law generated from the combined data of the top and bottom canopies in a 1-leaflet sampling unit. A sequential classification sampling plan was also developed using the action threshold of 0.5 and 10 mites for the top and bottom canopies, respectively.pectively.

The objective of this study was to evaluate several types of uterine bacteria in Hanwoo. uterine bacteria from randomly selected 5 uterus was collected by flushing methods into a sterilized 1.5 ml centrifuge tube and was inoculated onto MacConkey agar and blood agar, respectively. After being incubated for 5% CO2, aerobic or anaerobic condition at 37℃ during 48h, bacterial colonies were selected and re-inoculated onto blood agar plates. Re-cultured colonies were identified by Gram staining and finally identified using Vitek system. The identified bacteria were Staphylococcus lentus, Staphylococcus sciuri, Staphylococcus vitulinus, Staphylococcus warneri of Gram (+) and Rhizobium radiobacter, Sphingomonas paucimobilis of Gram () bacteria. Although, pathogenicity of identified bacteria was unclear, the bacteria can have an effect on the uterine microenvironment. Therefore, repetitive research will be required to determine the effects of bacteria in cattle exposed to a various environment.
한우에 Ov-synch 처리시 배란시기와 수태율 구명에 관한 연구

본 연구는 한우에 GnRH + PGF+GnRH (Ov-synch)를 처리하여 배란동기화를 시켰으며, 2차 GnRH 투여후 배란시간, 수태율, 계절별 수태율, 산차별 수태율을 조사하였으며, 시험축은 2산 이상의 개체를 무작위로 선발하여 실험에 공시하였으며, 배란동기화 처리후 1회 인공수정을 실시하고 수태율을 환산하였다. 호르몬 처리방법으로는 GnRH + PGF + Gn-RH(Ov-synch)법을 이용하였으며, 배란시간의 조사는 초음파를 이용하여 2차 Gn Recently a protocol was developed that precisely synchronizes the time of ovulation in Hanwoo. Cows were treated with GnRH on Day 0, PGF 7 d later, GnRH 2 d later, and then time-inseminated approximately 24 h after this second treatment with GnRH. Ovarian morphology was monitored cows by trans-rectal ultrasonography 6.5MHz linear transrectal probe(Sonovet - 600., Medison co. Korea) from 24 hr to 31 hrs after second GnRH injection. The result obtained summarized as follows - 1. Induced ovulation were 24 to 31hr after the second GnRH injection, but high induced ovulation was 28hr. 2. Conception rate with HML(High meat lin) and HIL(High milk lin) treatment were 48.1%(38/79) and 43.9%(40/91), respectively. 3. Conception rate of 1∼2 parity and 3∼4 parity was 44.3% and 55%, respectively. 4. Conception rate of spring, autumn was more increased, 47.3% than summer.

본 연구는 한우에 PGF₂α와 GnRH+PGF₂α+GnRH(Ov-synch)를 처리하여 발정 및 배란을 동기화 시켰으며, 2차 GnRH 투여후 배란시간, 2차 GnRH 투여후 시간 경과에 따른 수태율과 발정ㆍ배란 동기화법에 의한 수태율을 조사하고자 실시하였다. 시험축은 총 4개 농가에서 경산우 150두를 무작위로 선발하여 시험에 공시하였으며, 발정ㆍ배란동기화 방법에 따라 발정을 유기한 후 1회 인공수정을 실시하고 수태율에 미치는 영향을 조사하였다. 호르몬처리 방법으로는 GnRH+PCF₂α+GnRH(Ov-synch)와 PGF₂α를 이용한 발정동기화 방법을 사용하였다. 2차 GnRH 투여 후 배란시간을 알아보기 GnRH 투여 후 24시간 후부터 32시간까지 2시간 간격으로 초음파 Sonovet-600(Medison. Korea)를 이용하여 난소를 촬영하였다. 1. 호르몬 투여후 발정동기 화율은 PGF₂α 투여구에서 40.0%와 GnRH+PGF₂α+GnRH(Ov-synch)처리구에서 91.3%로 나타났다. 2. 2차 GnRH 주사후 24시간에 배란이 시작되어 32시간에 배란이 종료되었으며, 배란율은 28시간째에 46.6%가 배란되어 가장 높게 나타났다. 3. 2차 GnRH주사후 6∼24시간에 수정한 군이 6시간 이전과 30시간 이후에 수정시킨 군에 비해 높은 수태율을 나타냈다. 4. 호르몬 처리별 수태율은 PGF₂α, CIDR 및 GnRH+PGF₂α+GnRH(Ov-synch)에서 각각 50.0, 36.0와 76.9%로 GnRH+PGF₂α+CnRH(Ov-synch)군에서 가장 높은 수태율을 나타냈다. This objective of this experiment were to evaluate the effect of various estrus synchronization programs on estrus detection rate and pregnancy rate in Hanwoo. After Postpartum 60 Days, a total of 150 cows divided into 2 groups. Cows Group 1 were treated with one luteolytic dosage of PGF₂α(25 mg, im; lutalyse. USA) on Day 0, and with a second dosage 14 d later; cows in Group 2 were treated with GnRH(l00 ㎍, im; Conceral. Korea) on Day 0, PGF₂α 7 d later, GnRH 2 d later, and then time-inseminated approximately 16 h after this second treatment with GnRH. Ovarian morphology was monitored cows by trans-rectal ultrasonography from 24 hr to 32 hrs after second GnRH injection. The result obtained summarized as follows: 1. Cows synchronization of estrus with GnRH+PGF₂α+GnRH(Ov-synch) and PGF₂α were 91.3 and 40.0%, respectively. 2. Induced ovulation were 24 to 32hr after the second GnRH injection, but high induced ovulation was 28hr. 3. High conception rate were 24hr insemination after the second GnRH injection. 4. Conception rate with PGF₂α, CIDR and GnRH treatment were 50.0, 36.0 and 76.9%, respectively.
일본재첩, Corbicula japonica 아가미의 정상구조와 고염분 급성노출에 의한 조직병리학적 변화

This study was performed to observe ultrastructure of the gill and to ascertain the effect of salinity on histopathological and ultrastructural changes in the gill of the Japanese clam, Corbicula japonica. Experimental period was 7 days. Experimental groups consisted of control, 5, 10, 20 psu. LC_(50) (96 h.) by the probit was 19.55 psu. Mortality was significantly different from the control (p < 0.05). Inner demibranch of the gill of C. japonica was wider 1.37 times than outer demibranch (p < 0.001). The filament zone on the plica can be distinguished by the six epithelial celll cell; frontal ciliated epithelium (7 μm), latero-frontal ciliated epithelium (5 μm), postlatero-frontal epithelim (3 x 8 μm), and lateral ciliated epithelium (5 μm) in the frontal zone, endothelial cellin the intermediate zone, and abfrontal cell in the abfrontal zone. It had one type of secretory cell that was filled with fibrous substances of low electron density. The gill of C. japonica exposed to 5 psu for 7 days was observed partially disappearance of the cilia, and glycogen granule in the filament. In the 10 psu, gill appeared partially modification of epithelial cell and destruction of the glycocalyx. Gill exposed to 20 psu was extended nuclus of the ciliated epithelial cell, destruction of the organelles, and observed glycogen granules infiltration and numerous vacuoles. Moreover, more than 50% filaments were observed that come out chitinous rod from disappearance of epithelial cell in the filament. Therefore, the destruction of the cilia and epithelial cell induce physiological activity and it may be leading directly to death. 96시간 동안 고염분에 노출시킨 일본재첩, Corbicula japonica의 LC_(50)은 19.550 psu였다. 0, 5, 10, 20 psu에 7일 동안 노출시킨 실험개체들은 실험종료시기에 각각 95%,80%, 35%, 10%의 개체들이 생존하였다. 일반적인 일본재첩의 아가미는 좌․우 한 쌍으로서 내부판의 면적은 외부판보다1.37배 넓었다 (p < 0.001). 아가미의 새엽에는 그 위치에 따라 상부에 정단섬모상피세포 (7 μm), 정단측면섬모상피세포(5 μm), 후정단측면섬모상피세포 (3 × 8 μm), 측면섬모상피세포 (5 μm) 가 존재하고, 새엽의 중간부분에는 혈림프동을 둘러싸고 있는 혈관상피세포가 존재하며, 하부에는 새엽하부상피세포가 존재하고 있었다. 새엽의 하부에 주로 존재하는분비세포들은 전자밀도가 낮은 섬유성의 분비과립을 가지고있었다. 5 psu에 7일 동안 노출된 일본재첩의 아가미는 부분적인 섬모의 탈락과 glycogen 과립이 다수 관찰되었다. 10psu에 노출된 개체들은 일부 새엽의 상피세포가 파괴되었으며, 미토콘드리아를 포함한 세포소기관 또한 파괴되었다. 섬모들은 원형질막이 팽창되었고 미세융모를 연결시키는 당질층의파괴도 관찰되었다. 20 psu에 노출된 일본재첩의 아가미는 새엽섬모상피세포 핵비대, 세포소기관의 파괴, 세포질내glycogen 과립의 침적과 공포형성이 관찰되었고, 50% 이상의새엽은 새엽상피층의 탈락으로 인하여 키틴질 기둥이 모두 노출되었다. 따라서 이러한 섬모와 상피세포의 파괴는 생리활동의 장애를 유발시키고, 개체 사망의 직접적인 원인으로 작용할것이다.
