        알부민이 첨가된 시험관내 약역학 감염모델을 이용한 폐렴알균 치료에서 Ceftriaxone 일일 1회 요법

        배경 : Penicillin 내성 폐렴알균이 증가하는 지역에서 대안 중의 하나로 사용되는 ceftriaxone (CTR)의 적절한 용법, 용량을 파악하는 것이 중요하다. 본 연구는 임상에서 분리된 폐렴알균을 대상으로 시험관내 약역학 감염모델을 이용하여 알부민이 첨가된 경우와 첨가되지 않은 경우 CTR의 일일 1회 요법의 효과를 비교해 보고자 하였다. 재료 및 방법 : Penicillin 감수성(SM24), 중간내성(SM47), 내성(SM60)주를 대상으로 2-구획 시험관내 약역학 감염모델을 적용하였다. CTR 주입은 2 g씩 24시간마다로 사람의 약동학을 모의하였다. 알부민은 모델에서 4g/ dL가 유지되도록 하였다. 살균효과는 0, 2, 4, 6, 24, 30, 48시간째 집락수 변화로 측정하였다. 결과 : 모든 균주에서 알부민이 첨가되지 않은 경우 6시간 이내에 살균력이 관찰되었다. 최저억제농도 및 알부민 첨가유무에 관계없이 3주 모두 24시간째 살균력이 관찰되었고, 48시간째는 모두 측정한계 이하의 집락수로 감소하였다. 전 실험과정에서 CTR에 대한 내성주는 출현하지 않았다. 결론 : CTR의 일일 1회 요법은 알부민이 첨가되었을 때, 감수성, 중간내성, 내성주 모두에서 살균력 발현이 지연되었으나 최종효과는 알부민이 없을 때와 다르지 않았다. 앞으로 알부민 결합효과와 관련된 기초적인 추가 연구가 필요할 것으로 생각된다. Background : During the era of increasing penicillin resistant Streptococcus pneumoniae, it is important to have knowledge about adequate dosage and dosing interval of ceftriaxone (CTR). We examined efficacies of once-daily CTR and compared results in an in vitro pharmacodynamic infection model (IVPDIM) supplemented with albumin and those without albumin. Methods : Using three clinically isolated S. pneumoniae that were susceptible (SM24), intermediate (SM47) and resistant (SM60) against CTR, we utilized a two-compartment IVPDIM. CTR 2 g was administered intravenously every 24 h. Human albumin was added with concentration of 4 g/dL. Samples were removed at multiple time points over a 48-h period to determine the colony counts. Results : In SM24 and SM60, bactericidal effects were observed within 6 hours in groups without albumin. The number of colonies during 1st 6 hours were more decreased in albumin-free groups than in albumin-supplemented groups (P<0.05). In SM47, similar results were found during 1st 6 hours (P=0.03). But, regrowth was observed in albumin supplemented group at 30 h. Irrespective of results of minimal inhibitory concentrations and albumin supplementation, bactericidal effects were shown at 24 h in all 3 strains. All groups were decreased below the detection limit at 48 h. Development of resistance was not detected throughout the entire study period in either strain. Conclusions : Although extents of killing in albumin supplemented broth of once-daily CTR dosing were delayed in all 3 strains compared with those of albumin free broth, final efficacies were not different between the two groups.

      • 수출용 돼지고기 부산물의 냉장저장 중 미생물, pH, VBN 변화 및 유통기한 설정

        본 연구는 저장기간동안 수출용 돼지 부산물(간, 신장)의 미생물, pH 그리고 VBN을 측정하고 이를 토대로 유통가능 기간을 예측함으로서 수출 부산물에 품질과 취급에 대한 기초자료를 얻고자 실시하였다. 총균수는 간과 신장 모두 저장 4일까지 3 logCFU/㎠ 수준을 나타내었으며, 저장6일까지는 6 logCFU/㎠을 유지하였다. 내냉성 균수도 총균수와 비슷한 양상을 보여 주었고, 신장은 간보다 초기오염도가 높고 미생물 증식도 빨라 저장기간이 상대적으로 짧을것으로 사료된다. 저장 1일차의 pH는 간에서 6.31이었고, 신장은 6.78이었다. 저장기간 중 pH의 변화는 간이 6.31∼6.37, 신장은 6.70∼6.78의 범위로 신장이 간보다 pH가 다소 높게 나타나 미생물과 pH와의 관계에서 볼 때 신장의 저장성이 상대적으로 낮을 것으로 사료된다. 저장기간 중 VBN의 변화를 보면 신장은 초기 17.93 mg%에서 8일차에 이르러 40.12 mg%를 나타내었지만, 간은 초기 30.26 mg%에서 저장 6일차에 50.82 mg%에 도달하였다. 유통기한 설정에 있어서는 5.8일∼6.9일까지의 결과가 나왔으나 부패 판단시점은 모두 7일 이상으로 나타났다. 그러므로 초기 오염이 일반적인 식육의 초기 미생물 수준일 경우 일주일 정도가 돼지의 간, 신장에 있어서의 적절한 유통기한이라고 사료된다. This study was conducted to determine the changes in microbe, pH, Volatile Basic Nitrogen (VBN) of exportation by-products of pork and establish their shelf-life during storage at 4℃. Exportation pork by-products of pork were obtained from commercial slaughter house. The number of total plate count (TPC) and psychrotrobes increased (P<0.05) with storage periods. Both TPC and psychrotrobes were significantly (P<0.05) higher in kidney, pH of kidney was singnificantly (P<0.05) higher than liver, whereas VBN of liver was significantly (P<0.05) higher in all storage periods. The shelf-life in both kidney and liver were 7 days at 4℃.

      • 산삼 배양액 급여가 돈육의 이화학적 특성에 미치는 영향

        Berkshire를 이용하여 70일간 산삼 배양액 급여 시 육의 이화학적 특성에 미치는 영향은 다음과 같다. 산삼 배양액의 급여에 의해 육의 수분, 조단백, 조지방, 회분, pH, 가열감량, 콜레스테롤 함량, 육색, 조직감의 응집성과 검성, 포화지방산 및 불포화지방산의 비율은 차이를 보이지 않았다. 보수력, 지방색 L^(*)값, 지방산 중 linoleic acid(18:2), arachidonic acid(20:4), 필수지방산의 함량, 아미노산 중 threonine, glutamic acid, valine, isoleucine, leucine, histidine, lysine 및 총아미노산 함량은 산삼 배양액의 급여에 의해 감소하였다. 전단가, 조직감의 표면경도, 경도, 씹힘성, 지방산 중 stearic acid(18:0), oleic acid(18:1) 함량, 아미노산 중 serine, glycine, cystine, methionine, tyrosine 및 phenylalanine의 함량은 산삼 배양액의 급여에 의해 증가하였다. 본 연구결과를 종합하면 산삼 배양액의 급여는 돈육의 경도를 증가시키는 효과 외에 뚜렷한 이화학적인 차이를 나타내지 않았다. 이러한 이유는 급여된 산삼 배양액 내에 함유된 생리활성 물질의 함량이 낮고, 급여수준이 낮은 결과로 사료되어진다. A total of 60 pigs (Berkshire) were used to investigate the effect of dietary artificial culture medium of wild ginsengs (CMG) on physico-chemical characteristics of pork. About 60 kg pigs were randomly alloted into one of two experimental diet groups (C:commercial diet feed; T:commercial diet + 1 L CMG per day for 70days). Pigs were slaughtered at approximately 110 kg live weight, and chemical composition and physico-chemical characteristics were measured in pork loin. Moisture, crude protein, crude fat and ash were not significantly different by dietary artificial culture medium of wild ginsengs. Water-holding capacity was decreased by dietary artificial culture medium of wild ginsengs, while shear force was increased. However, pH, cooking loss and cholesterol contents were not significantly different between two different treatments. In color, L^(*) of fat was decreased by dietary artificial culture medium of wild ginsengs. Brittleness, hardness and chewiness were increased in dietary artificial culture medium of wild ginsengs group. In fatty acid, stearic acid (18:0) and oleic acid (18:1) were higher in dietary artificial culture medium of wild ginsengs group, whereas linoleic acid (18:2) and arachidonic acid (20:4) were higher in control group. In amino acid, serine, glycine, cystine, methionine, tyrosine and phenylalanine were higher in dietary artificial culture medium of wild ginsengs group, whereas threonine, glutamic acid, valine, isoleucine, leucine, histidine and lysine were higher in control group.

      • 기계적 합금화에 의한 Al-Fe 및 Al-Fe-Ce 합금의 고온 변형거동

        The deformation behavior of a mechanical alloyed Al-Fe and Al-Fe-Ce alloy was studied in the temperature range of 300℃ to 500℃. The steady state for Al-8wt.%Fe powder was obtained after 1000 minutes of mechanical alloying at the process condition of 2wt.% stearic acid, 46:1 of grinding media/powder weight ratio, and 300 rpm of impeller revolving velocity. The relative densities of Al-8wt.%Fe alloy powders reached almost 100% after 1 hour sintering at 470℃ under vacuum state. With increasing the amount of PCA, hardness of the sintered alloys increased but the relative densities were not affected. It was also found that decrease in hardness and tensile strength after high temperature aging treatment was reduced with increasing the amount of Ce addition.

      • C6 glia 세포에서 유도성 Nitric Oxide Synthase 유전자 발현조절에 관한 연구

        To investigate transcriptional regulation of iNOS gene by LPS and cytokines, the production of NO, expression of iNOS mRNA and protein, binding activity of nuclear factor-kappa B(NF-kB), and promoter activity of iNOS gene were examined in rat C6 glial cells. LPS, interferon-gamma(IFN-γ), and tumor necrosis factor-alpha (TNF-α) stimulated the production of NO, which was increased synergistically by co-treatment. By the treatment of LPS, iNOS mRNA expression was initiated at 1 h, markedly increased by 3 h, and decreased gradually afterward. iNOS mRNA expression was markedly enhanced by mixture of LPS, IFN-γ and TNF-α. iNOS protein synthesis was increased by the treatment of mixture LPS and cytokine mixture. Treatment of LPS stimulated NF-kB activation, and the activation reached to the maximum level at 30 min, and the treatment of mixture of LPS and cytokines increased the activation. To determine the effect of NF-kB binding activity on iNOS promoter activation, CAT assay was performed. iNOS promoter activity was increased by the treatment with LPS for 5.5 h, and further increased by the combined treatment with LPS and cytokines. These results suggest that NF-kB activation by LPS and cytokines may play a significant role in the induction of the iNOS gene.

      • LCA 기법을 적용할 제품의 우선순위 선정 방법론

        본 연구의 목적은 일정한 논리적 절차를 거쳐서 객관적인 전과정평가의 우선순위를 선정하기 위한 일반적인 방법론의 개발이다. 우선 전과정평가 대상이 되는 제품(산업군)의 우선순위를 평가하기 위해서 제품의 환경성과 관련된 환경규제, 국내외 전과정평가 수행사례, 유해물질 등을 평가항목으로 설정하였다. 설정된 평가항목들에 대한 자료조사 및 분석을 통하여 세분화한 데이터베이스를 생성하였으며, 또한 학계 및 산업계의 의견을 반영하기 위해 전과정평가 전문가 및 산업계 관계자들을 대상으로 설문조사를 실시하였다. 설문조사결과를 바탕으로 각각의 평가항목 및 세부평가항목의 가중치를 부여하였고, 각 평가항목별로 점수를 설정하였다. 본 연구에서 개발한 방법론은 우선순위를 비교해야하는 제품(산업군)들이 각각의 평가항목에서 설정해놓은 해당 점수를 부여받게 된다. 부여받은 각 평가항목의 점수들을 모두 합산하면 평가대상이 되는 제품(산업군)에 대해 하나의 우선순위 평가 점수를 갖게 된다. 이와 같은 과정으로 방법론이 구축되었으며, 사례연구를 통하여 방법론의 타당성을 검증하였다. 이렇게 산출된 우선순위 평가점수를 평가하고자하는 제품(산업군)들과 비교하여 높은 점수를 갖는 제품(산업군)이 전과정평가를 먼저 실시하는 것이 바람직하다는 결론을 얻을 수 있다. The purpose of this study is to develop a general method to decide the priority of LCA application through a logical procedure. First of all, evaluation categories were developed to decide priority of subject products(industrial groups). The evaluation categories consist of environment regulations related to subject product, LCA case studies, hazardous substances, etc. The subdivided database was constructed through researching and analyzing information about the selected evaluation categories. In addition, a questionnaire was conducted to reflect the opinions from academic and industrial circles. Based on the result of the questionnaire, each evaluation category and sub-evaluation category were weighed differently, and certain points were assigned to them. According to the method developed in this study, comparing industrial groups or products for prioriting will have its total score that is a sum of points assigned to each evaluation category. This results that an industrial group or product having higher score than the others becomes the subject to apply LCA.

      • 신경교세포 및 RAW 264.7 세포에서 Protein kinase의 활성에 의한 유도성 Nitric oxide synthase의 발현

        NO(nitric oxide) plays an important role as neurotransmitter or cytokine, and pathologic factor for some diseases by the large amount production with iNOS(inducible NO synthase) expression in macrophages or glial cells. The expression of iNOS is regulated by various cytokines, protein kinases and transcription factors. In this experiment, to investigate the roles of progein kinase and NF-kB for iNOS expression, the effects of PMA(phorbol 12-myristate 13-acetate), cAMP, and various protein kinase inhibitors on LPS(lipopolysaccharide)-induced iNOS mRNAN expression and nuclear NF-kB binding complex were examined in C6 glial cells and RAW 264.7 cells. In C6 glial cells, iNOS mRNA expression by LPS was induced from 1 hour and peak at 3 hour after treatment. In RAW 264.7 cells, the mRNA was observed from 3 hour and peak at 6 hour. PMA enhanced markedly LPS-induced iNOS mRNA expression and NF-kB binding complex in C6 glial cells, but did not much influence on LPS-induced iNOS mRNA expression in RAW 264.7 cells, in spite of increased LPS-induced NF-kB binding complex at 30 min. cAMP(dibutyryl cAMP) did not much influence on LPS-induced iNOS mRNA expression, by increased LPS-induced NF-kB binding complex in C6 glial cells. However, in RAW 264.7 cells, cAMP increased slightly LPS-induced iNOS mRNA expression without change of NF-kB binding complex. Staurosporine did not influence on LPS-induced iNOS mRNA expression and NF-kB binding complex in C6 glial cells, but in RAW 264.7 cells, decreased LPS-induced iNOS mRNA expression and NF-kB binding complex. Ro-31-8220 did not much influence on LPS-induced iNOS mRNA expression and NF-kB binding complex in C6 glial cells, but in RAW 264.7 cells, decreased significantly LPS-induced iNOS mRNA expression in spite of increased LPS-induced NF-kB binding complex for 3hours. G 6976 did not much influence on LPS-induced iNOS mRNA expression with decreased NF-kB binding complex in C6 glial cells, but in RAW 264.7 cells, decreased iNOS mRNA expression without influence on LPS-induced NF-kB binding complex. Genistein did not influence on LPS-induced iNOS mRNA expression and NF-kB binding complex in C6 glial cells, but in RAW 264.7 cells, decreased LPS-induced iNOS mRNA expression inspite of increased NF-kB binding complex. These results suggest that LPS-induced regulation of iNOS expression or NF-kB activity in C6 glial cells, might be different from RAW 264.7 cells through various protein kinases or other factors.

      • Current strategies for the control of COVID-19 in South Korea

        This study sheds light on the current strategies for control of coronavirus-19 (COVID-19) in South Korea in terms of the current status of the 2019 novel coronavirus infection, role of the government, infrastructures preparation, people's responses, etc. The South Korean government organized a response team in the early stages of the COVID-19 infection to quickly disclose information related to the infection and information regarding those in close contact with infected persons and to carry out a wide range of COVID-19 tests. The South Korean government established and operated a system for sending disaster text messages since 2003. If a virus infection occurs, this system can find all information of confirmed cases such as the infected area, age, gender, close contacts, and route of transmission through GPS tracking of cell phones, CCTV tracking, credit card usage details, and interviews to make it transparent to the people throughout the whole country. South Korea introduced the world's first “Drive Thru” and “Walking Thru” system to carry out rapid virus testing and hospitalize confirmed cases of COVID-19 at national designated hospitals where the negative pressure system is deployed. Nevertheless, South Korea did not implement a full-scale denied entry for all countries around the world and tried to block the transmission of virus by testing all suspected cases and people who were in close contact with them. In addition, the government managed the supply and sale of masks to allow the entire nation to purchase two masks every week, and most Koreans wore masks without panic and participated in the social distancing campaign.
