        Antioxidant and hepatoprotective effects of Korean ginseng extract GS-KG9 in a D-galactosamine-induced liver damage animal model

        BACKGROUND/OBJECTIVES: This study was designed to investigate the improvement effect of white ginseng extract (GS-KG9) on D-galactosamine (Ga1N)-induced oxidative stress and liver injury. SUBJECTS/METHODS: Sixty Sprague-Dawley rats were divided into 6 groups. Rats were orally administrated with GS-KG9 (300, 500, or 700 mg/kg) or silymarin (25 mg/kg) for 2 weeks. The rats of the GS-KG9- and silymarin-treated groups and a control group were then intraperitoneally injected Ga1N at a concentration of 650 mg/kg for 4 days. To investigate the protective effect of GS-KG9 against GalN-induced liver injury, blood liver function indicators, anti-oxidative stress indicators, and histopathological features were analyzed. RESULTS: Serum biochemical analysis indicated that GS-KG9 ameliorated the elevation of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), and lactate dehydrogenase (LDH) in GalN-treated rats. The hepatoprotective effects of GS-KG9 involved enhancing components of the hepatic antioxidant defense system, including glutathione, glutathione peroxidase (GPx), superoxide dismutase (SOD), and catalase (CAT). In addition, GS-KG9 treatment inhibited reactive oxygen species (ROS) production induced by GalN treatment in hepatocytes and significantly increased the expression levels of nuclear factor erythroid-2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) proteins, which are antioxidant proteins. In particular, by histological analyses bases on hematoxylin and eosin, Masson"s trichrome, α-smooth muscle actin, and transforming growth factor-β1 staining, we determined that the administration of 500 mg/kg GS-KG9 inhibited hepatic inflammation and fibrosis due to the excessive accumulation of collagen. CONCLUSIONS: These findings demonstrate that GS-KG9 improves GalN-induced liver inflammation, necrosis, and fibrosis by attenuating oxidative stress. Therefore, GS-KG9 may be considered a useful candidate in the development of a natural preventive agent against liver injury.

        Phosphodiesterase inhibitor improves renal tubulointerstitial Hypoxia of the Diabetic rat Kidney

        Background/Aims: Renal hypoxia is involved in the pathogenesis of diabetic nephropathy. Pentoxifyllin (PTX), a nonselective phosphodiesterase inhibitor, is used to attenuate peripheral vascular diseases. To determine whether PTX can improve renal hypoxia, we investigated its effect in the streptozocin (STZ)-induced diabetic kidney. Methods: PTX (40 mg/kg, PO) was administered to STZ-induced diabetic rats for 8 weeks. To determine tissue hypoxia, we examined hypoxic inducible factor-1α (HIF-1α), heme oxygenase-1 (HO-1), vascular endothelial growth factor (VEGF), and glucose transporter-1 (GLUT-1) levels. We also tested the effect of PTX on HIF-1α in renal tubule cells. Results: PTX reduced the increased protein creatinine ratio in diabetic rats at 8 weeks. HIF-1α, VEGF, and GLUT-1 mRNA expression increased significantly, and the expression of HO-1 also tended to increase in diabetic rats. PTX significantly decreased mRNA expression of HIF-1α and VEGF at 4 and 8 weeks, and decreased HO-1 and GLUT-1 at 4 weeks. The expression of HIF-1α protein was significantly increased at 4 and 8 weeks in tubules in the diabetic rat kidney. PTX tended to decrease HIF-1α protein expression at 8 weeks. To examine whether PTX had a direct effect on renal tubules, normal rat kidney cells were stimulated with CoCl2 (100 μM), which enhanced HIF-1α mRNA and protein levels under low glucose conditions (5.5 mM). Their expressions were similar even after high glucose (30 mM) treatment. PTX had no effect on HIF-1α expression. Conclusions: PTX attenuates tubular hypoxia in the diabetic kidney.

        Objective : The purpose of this study was to find the relationship between the follow-up management status for medical problem to be treated and various characteristics of some workers at transportation business. Methods : A questionnaire survey was made toward 183 workers with medical problem(C and D2) among 945 examine in 1997. Results: Sixty four point five percent of surveyee received the examination result report and 40.5% were explained about the result 80.3% of surveyee answered that they knew the result of health examination. Most workers felt the necessity of the health examination, but 54.1% were dissatisfied with present health examination system and 66.7% pointed it out as nominal The rate of follow-up management for the medical problem was 62.8% By the multiple logistic regression the follow-up management rate for those who knew the health examination result were 2.98 times higher than those who didn't know the result (OR=2.98, 95% CI 1.41~6.29) The rate for those who perceived the medical problem already were 2.64 times higher than those who perceived the problem for the first time (OR=2.64, 95% CI 1.13~6.15). The rate for those who had subjective symptoms were 2.83 times higher than those who didn't have subjective symptoms. Conclusion : sixty two point eight percent of workers with medical problem performed the follow-up management and perception of health examination result, perception of the problem in the past and experience of subjective symptoms were related with the follow-up management.

      • 벼(Oryza sativa L.) 統一型品種의 溫度에 따른 營養 生長期 主稈 出葉速度와 栽培時期 移動에 의한 主稈總葉數의 變化

        The response of leaf developing of rice plant to temperature and total number of leaves on mother stem under field condition are important to understand the characteristics of growth duration among the rice varieties. To investigate number of days required to develop a leaf at vegetative stage, 6.1-leaf age, nine indica/japonica Korean rices (Oryza saitva L.) were subjected to four levels of air temperature, at 13, 18, 23, and 28℃ of glass rooms in a phytotron for 168 hours. Also to examine variations of lifelong leaf number on the mother stem of the rices, a field experiment was conducted with transplanted five times from 11 May to 10 July at the 15-day intervals at Suweon, Korea in 1977. Significant varietal differences were observed in number of days required to develop a leaf. The differences were increased by decreased the temperature levels. The number of days required to develop a leaf, overall varietal means at 13, 18, 23, and 28℃ were 15.9, 6.7, 4.5, and 4.2 days respectively. A low temperature, at 13℃, an early maturing rice, Milyang 21 was required 6.8 more days than that in a late maturing rice, Geumgangbyeo. A relationship between daily mean air temperatures (X) and the number of days required to develop a unit leaf (Y) could be fitted significantly as a hyperholic equation: Y=X/aX-b. A base temperature, the threshold temperature for stoppage of leaf developing was estimated 12℃ by the equation. A minimum degree-days required to develop a leaf was 104℃ at 23℃ of mean air temperature in the rices. Total number of leaves on mother stem of overall rices and the transplanting dates in the field condition were ranged 13∼20 leaves of which the late maturing rices had more the loaves than those in early maturing rices, and in the same variety, early season transplanted rices also had more leaves than those in the late season transplanted rices. A negative correlation showed between the total number of loaves on mother stem of overall rices at transplanted in 26 May in the field and number of days required to develop a leaf at 13℃ only under the phytotron condition.

        This research presents a designing method of information systems for a company, constructing industrial plants, to introduce responsibility based accounting system. When the informations are designed, responsibility-focused points, suitable to a company, should be matched with implementing environment. When the information systems are implemented, it should be developed under consideration of developing procedures including design, analysis, verification, and constraints of the system, and connection to other information system. Two recognizing methods for profits are presented and compared to evaluate results each profit center under responsibility based accounting system as a study for a case of H company. This research can be utilized to other companies constructing industrial plants, when information systems for establishing responsibility based accounting system are designed.

        제주도 지하수 질산염 농도의 시·공간적 변화 특성: 장기(1993-2015) 모니터링 자료의 평가

        1993년부터 2015년까지 관측된 제주도 지하수 장기모니터링 관측정(N = 4,835)에서 수집된 지하수 수질자료(N = 21,568)를 기반으로 질산성질소의 시공간적 변동 특성을 평가하였다. 제주도 지하수의 질산성질소 농도의 중앙값은 2.5 mg/L로서 다른 국가나 대륙의 조사 결과에 비해 다소 높거나 유사한 것으로 나타났다. 또한 지하수 용도, 행정구역 및 고도 별로 유의한 차이를 보였다. 특히, 산간 지역에 비해 저지대 해안가에 위치한 농업 및 주거지역에서 농도가 높음을 확인하였다. Mann-Kendall 및 Sen’s slope 분석을 활용한 질산성질소 농도의 추세 분석 결과, 하류 저지 대에 비해 중산간지역에서의 질산성질소 농도 증가 경향이 뚜렷하였다. 제주도 내 토지 피복의 시계열 변화 특성과 결부 지어 보면, 중산간지역의 오염 증가 추세는 농업지역의 확장 등 인위적 활동 증가에 기인한 결과로 판단된다. 반면,기지정된 지하수자원특별관리구역에서는 전반적으로 질산성질소 농도의 감소 경향이 나타났는데, 이는 지하수 관리 측면에서 수질관리를 위한 적극적인 정책이 유효함을 시사한다. 본 연구에서는 제주도 지하수의 질산성질소 오염관리를 위한 적정 방안을 제안한다. The spatio-temporal variations of nitrate concentrations in groundwater of Jeju Island were evaluated by an analysis of time series groundwater quality data (N = 21,568) that were collected from regional groundwater monitoring (number of wells = 4,835) for up to 20 years between 1993 and 2015. The median concentration of NO 3 -N is 2.5 mg/L, which is slightly higher than those reported from regional surveys in other countries. Nitrate concentrations of groundwater in wells tend to significantly vary according to different water usage (of the well), administrative districts, and topographic elevations: nitrate level is higher in low-lying agricultural and residential areas than those in high mountainous areas. The Mann-Kendall trend test and Sen’s slope analysis show that nitrate concentration in mid-mountainous areas tends to increase, possibly due to the expansion of agricultural areas toward highland. On the other hand, nitrate concentrations in the Specially Designated Groundwater Quality Protection Zones show the temporally decreasing trend, which implies the efficiency of groundwater management actions in Jeju. Proper measures for sustainable groundwater quality management are suggested in this study.

      • 굵은 골재 종류에 따른 포장 콘크리트의 피로특성

        It is the purpose of this study to identify comprehensive fatigue characteristics of pavement concretes considering coarse aggregate types. The types of coarse aggregates considered are limestone, sandstone and granite, which are the most representative stones in Korea. The static strength tests were carried out to check the compressive strength, flexural strength at 7 and 28 days, and split tension strength at 56 day in order to minimize the various effect of strength during the test. The results are follows. The comprehensive fatigue models of pavement concretes were developed considering coarse aggregate types. The developed model showed a good relationship between aggregate types. The fatigue models with coarse aggregate types were shown, respectively. The combined fatigue models was also shown.
