        광합성세균에 의한 미생물막의 형성

        홍색비유황 광합성세균인 Rhodopseudomonas capsulata를 선택하여 packed-bed reactor에서 미생물막을 형성할 때, porous ceramic bead가 다른 담체에 비해 우수하였고, 일정한 유입농도하에서 체류시간(hydraulic retention tiem, HRT)이 짧을수록 미생물막 형성이 양호하였으며, 그 때 반응기내의 세포농도는 11,400mg/l로 현탁처리시의 세포농도에 비하여 3~8배 증가하였다. PBR에서 미생물막의 형성은 cell attachment, microcolony formation, biofilm formation의 단계를 거쳐 형성되는 것으로 관찰되었으며, PBR이 FBR보다 안정적인 미생물 부착을 보였고 특히 PBR에서는 BOD용적 부하가 15gBOD/ℓ·day 이상이 되어도 미생물막의 부착비율은 90% 이상을 유지하였다. 전자현미경으로 담체의 표면 및 내부에 고정화된 광합성세균을 확인할 수 있었다. The formation of microbial films(biofilm) by a non-sulfur phototrophic bacteria, Rhodopseudomonas capsulata, on inorganic media was studied. Porous ceramic beads(PCB) were superior to other immobilizing media for the biofilm formation in a packed-bed reactor. It was found that the formation of microbial films favored a lower hydraulic retention time, showing a higher ratio of cells attatched to the media to those suspended in the solution. The cell concentration in the biofilm reactor was as high as 11,400 mg/ℓ, which is 8-folds of the cell concentration in a ordinary suspended treatment. It was observed that the formation of microbial film by R. capsulata followed a general serial process of cell attachment, microcolony formation, and biofilm formation. The microbial films thus formed was very stable even for an extremely high volumetric BOD loading rate of 15 gBOD/ℓ·day. The scanning electron micrographs of the microbial films showed that the cells were attached to both the surface and pores of the media.

      • D-Amino Acid Aminotransferase 활성보유 고온성미생물의 탐색 및 분류학적 특성 연구

        To acquire an industrially useful biocatalyst for the enzymatic synthesis and production of various D-amino acids, 1,300 thermophiles isolated from the soil of Korea were analyzed for D-amino acid aminotransferase (D-AAT) activity. The enzyme activity was found from 110 strains of isolated thermophiles revealing its wide occurrence in thermophiles. Enzyme activity and thermal stability of the D-AAT producers were compared. Finally we have selected four thermophiles as producers of potent biocatalysts for the D-amino acid production; two therrnophiles, Bacillus sp. LK-I and LK-2, having higher specific activity and two thermophiles, B. stearothermophilus KL-01 and Bacillus sp. KLS-0l, having higher thermal stability than the D-AAT producers. Taxonomic and physiological characteristics of the four isolated thermophiles were described herein.

        정신분열병 환자들의 自己灣入에 관한 예비 연구

        목 적 : 정 신분열병 환자의 자기만입 정도에 영향을 미치는 인구학적 변인들 및 질병연관 변인들을 규명해 보고자 하였다. 또한. 환자들의 자기만입과 병식 사이의 연관성. 자기만입과 자존심 사이의 연관성을 알아보고자 하였다. 방 법 : 정신과 의사가 병록지와 면담을 통하여 대상 정신분열병 환자들의 인구학적 변인들 및 질병연관 변인들에 관한 자료를 얻고 환자들의 병식 정도를 평가한 후, 환자들로 하여금 자기보고형의 자기만입 척도와 자존심 척도를 시행하게 하였다. 총 111명의 환자가 연구대상으로 선택되었다. 결과: . 1) 여자가 남자보다 유의하게 높은 자기만입 척도 점수를 나타내었다 2) 질병연관 변인들 중 하위집단 사이에 자기만입 척도 점수의 유의한 차이를 보이는 변은 없었다. 3) 자기만입 척도 점수와 병식 점수 사이에 유의한 상관관계가 없었다. 4) 자기만입 척도 점수와 자존심 척도 점수 사이에 유의한 역상관 관계를 보였다. 결 론 : 본 예비 연구의 결과로 보아 정신분열 병 환자의 병식이 부족한 상태에서도 자기만입이 진행될 수 있으며. 자기만입이 심할수록 환자의 자존심이 저하되는 것으로 생각된다. 또한 여자 환자들이 남자 환자들보다 자기만입에 더 취약한 것으로 생각된다. 따라서 정신분열 병 환자의 이해와 치료를 위하여 자기만입에 관한 지속적인 연구가 필요할 것으로 생각된다. objective : This study was conducted to identify demographic variables and illness related variables which may affect the severity of self-engulfment in the patients with schizophrenia. We also studied the relationship between self-engulfment and insight, and the relationship between self-engulfment and self-esteem. Methods : Data on demographic variables and illness related variables for the subjects were gathered from hospital records and clinical interviews by the psychiatrists. Degree of insight for the subjects was assessed through clinical interviews by the psychiatrists. Degree of self-engulfment and self-esteem for the subjects was assessed from the self-engulfment scale and the self-esteem scale respectively. A total of 111 patients with schizophrenia were selected for statistical analysis. Results 1) The females exhibited significantly higher scores than the males on the self-engulfment scale. 2) There was no illness related variable, which exhibited a significant difference among subgroups on the self-engulfment scale. 3) There was no significant correlation between the self-engulfment scores and the insight 4) There was a significantly inverse correlation between the self-engulfment scores and the self-esteem scores. Conclusion : The results of this preliminary study suggest that self-engulfment may develop despite lack of insight and that there was inverse relationship between self-engulfment and self-esteem in the patients with schizophrenia. It also suggests that females are more subject to self-engulfment than males. So, authors suggest that it is valuable to do further studies of self-engulfment in the patients with schizophrenia.

        지역4-H조직의 내적 적합성에 관한 문화기술적 연구

        이 연구의 목적은 지역4-H조직의 내적 적합성을 탐색하고 이를 기술하는데 있었다. 2006년 8월부터 10월까지 약 3개월 동안 지역4-H후원회 회장 및 임원진 3명, 지역4-H연맹 회장 1명, 학교4-H 지도교사 4명, 지역의 영농4-H회장 1명, 시·군 농업기술센터 소장 및 4-H담당자 9명을 주요 연구 참가자로 하여 심층면담을 통해 자료를 수집하였다. 수집한 자료를 분석한 결과는 다음과 같았다. 첫째, 지역4-H조직 관련자들이 인식하는 지역4-H조직의 주요성과 격차로, ① 민간4-H조직을 중심으로 한 자율적이고 독립적인 활동 부재, ② 지역4-H조직 간 다양한 종류의 협력 부재, ③ 지역사회에서 4-H활동에 대한 가치 인식 부족이 나타났다. 둘째, 지역4-H조직의 핵심 과업으로 ① 4-H의 가치 및 이념에 부합하고, 지역의 실정에 맞는 프로그램 확보, ② 지역4-H 조직 간 협력적 네트워크 확립, ③ 4-H에 대한 지역사회의 관심 및 지지 확보가 나타났다. 셋째, 지역4-H조직의 내적 부적합성으로는 ① 인원 확보의 어려움과 핵심과업에 대한 전문성 결여, ② 관료화되고 경직된 낡은 문화, ③ 조직적 자원의 축소 및 조직 간 낮은 네트워크의 수준이 도출되었다. The purpose of this study was to describe and understand internal fit in Local 4-H Organization. The main participants of this study were one leader and two members of Local 4-H Society for the support, one leader of Local 4-H Association, four School 4-H teacher, one leader of Local farm 4-H, 9 local agricultural extension workers. The data were collected through in-depth interview with them from August, 2006 to October, 2006. The Results from this study were as follow: 1. Key performance gaps of local 4-H organizations were identified: ① lack of local 4-H based activities, ② lack of various types of partnership between local 4-H organizations, ③ lack of understanding value of 4-H activities in the communities. 2. Key tasks of local 4-H organizations were identified: ① developing local 4-H programs based on 4-H's values and ideas, ② establishing the cooperative networks among local 4-H organizations, ③ ensuring the interest and support about 4-H in the communities. 3. Internal misalignments of local 4-H organizations were identified: ① difference of recruitment and lack of expertise, ② bureaucratic and rigid cultures, ③ decrease in organizational support and low level of network among organizations.

      • n-CdS_(0.46)Se_(0.54)/p-Cu_92-x)S_(0.46)Se_(0.54) 이종접합 태양전지의 제작과 그 특성에 관한 연구

        승화방법에 의해 CdS_0.46Se_0.54 단결정을 성장하여 결정구조를 조사하고, Van der Pauw 방법으로 Hall effect를 측정하여 carrier density의 온도 의존성과 mobility의 온도 의존성을 조사하였다. 성장된 CdS_0.46Se_0.54 단결정을 치환반응하여 n-CdS_0.46Se_0.54/p-Cu_2-xS_0.46Se_0.54 이종접합 태양전지를 제작하였다. Spectral response, 전류-전압특성 및 전력변환 효율을 조사하여 그 결과로부터 개방전압은 0.48V, 단락 전류 밀도는 21mA/㎠, fill factor와 전력변환효율은 각각 0.75와 9.5%를 얻었다. CdS_0.46Se_0.54 single crystal was grown by a sublimation method. The crystal structure and the temperature dependence of carrier density and mobility of CdS_0.46Se_0.54 single crystal were studied. Heterojunction solar cells on n-CdS_0.46Se_0.54/p-Cu_2-xS_0.46Se_0.54 were fabricated by the substitution reaction. The spectral response, the J-U characteristics and the conversion efficiency of the n-CdS_0.46Se_0.54/p-Cu_2-xS_0.46Se_0.54 heterojunction solar cells were studied. The open-cricuit voltage, short-circuit density, fill factor and conversion efficiency of n-CdS_0.46Se_0.54/p-Cu_2-xS_0.46Se_0.54 heterojunction solar cells under 80mW/㎠ illumination were found to be 0.48V, 21mA/㎠, 0.75 and 9.5%, respectively.

        L-라이신 발효에 있어서 당밀전처리의 영향

        Can molasses, the most widely used carbon source for the industrial fermentation of L-lysine, usually contains a high concentration of calcium ions which tend to cause scaling problem in the recovery process. To remove the calcium ions, cane molasses was pretreated with sulfuric acid by adjusting the pH to 2.5~3.5. When the pretreated solution was directly heat-sterilized and used in the fermentation, a significant reduction in L-lysine production was observed. In this paper, we proved that sucrose is a superior substrate for L-lysine fermentation of that of glucose or fructose and that the above-mentioned decrease of L-lysine production was caused by the hydrolysis of sucrose in the molasses when the molasses was heat-sterilized at a low pH. The problem was overcome by adjusting the pH of molasses to neutral before sterilization.

        Temporal expression of AMP-activated protein kinase activation during the kainic acid-induced hippocampal cell death.

        <P>Kainic acid (KA)-induced seizure induces the hippocampal cell death. There are reports that AMP-activated protein kinase (AMPK), which is an important regulator of energy homeostasis of cells, has been proposed as apoptotic molecule. In this study, we investigated the altered expression of AMPK cascade in the hippocampus of mice during KA-induced hippocampal cell death. Mice were killed at 2, 6, 24 or 48 h after KA (30 mg/kg) injection. Histological evaluation of KA-treated hippocampus revealed hippocampal cell death first at 6 h and appearing prominently by 48 h after KA injection. Immunoreactivity of Ca(2+)/calmodulin-dependent protein kinase kinasebeta (CaMKKbeta) was increased after KA treatment. In Western blot analysis, AMPK activation was increased 2 h after KA treatment. The proteins of downstream AMPK, including those of glucose transporter1 (GLUT1) and phosphorylation of Acetyl CoA Carboxylase (ACC) were increased in the hippocampus after KA treatment. These results indicate that sustained AMPK activation might be a mechanism by which KA-induced seizure causes hippocampal cell death of mice.</P>

      • Silicone implants capable of the local, controlled delivery of triamcinolone for the prevention of fibrosis with minimized drug side effects

        <P><B>Abstract</B></P> <P>We propose silicone implants capable of the local, controlled release of a glucocorticoid drug, triamcinolone acetonide (TA), for the prevention of fibrosis. The shells of these silicone implants were coated with two different loading amounts of TA, which could release the drug in a sustained manner for 12 weeks. The drug-loaded implants were inserted into the subcutaneous space in living rats, and the tissues were biopsied at scheduled times during 12 weeks. For the drug-coated implants, the capsule thickness and collagen density decreased compared with those of the non-coated implant. Because of the effect of TA, inflammation and the expression of pro-inflammatory cytokines were downregulated, thereby decreasing the number of monocytes during acute inflammation. This effect in turn decreased the number of macrophages at the later stage of inflammation, leading to the expression of less TGF-β and consequently fewer fibroblasts and myofibroblasts. Our findings also revealed that with an appropriate dose control, skin and muscle atrophy, major side effects of TA, could be avoided while still effectively reducing fibrosis. Therefore, we conclude that the local, sustained release of an appropriate dose of a glucocorticoid drug can be a promising strategy for safely preventing fibrosis around silicone implants.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Intracellular transformation rates of fatty acids are influenced by expression of the fatty acid transporter FadL in <i>Escherichia coli</i> cell membrane

        <P>Fatty acids have a low permeability through the cell membrane. Therefore, the intracellular biotransformation of fatty acids can be slow due to supply limitations. The effects of expression level of the fatty acid transporter FadL in Escherichia coli on the biotransformations were investigated. The enhanced expression of FadL led to 5.5-fold increase of the maximum reaction rate V-max (i.e., 200 mu mol/min per g dry cells (200 U/g dry cells)) of the recombinant E. coli expressing a hydratase of Stenotrophomonas maltophilia in the periplasm with respect to hydration of oleic acid. The FadL expression level was also critical for oxidation of 12- and 10-hydroxyoctadecanoic acid by the recombinant E. coli expressing an alcohol dehydrogenase (ADH) of Micrococcus luteus. In addition, the multistep biotransformation of ricinoleic acid into the ester (i.e., (Z)-11-(heptanoyloxy) undec-9-enoic acid) by the recombinant E. coli expressing the ADH of M. luteus and a Baeyer-Villiger monooxygenase of Pseudomonas putida KT2440 was 2-fold increased to 40 U/g dry cells with expression of FadL to an appropriate level. The FadL expression level is one of the critical factors to determine whole-cell biotransformation rates of not only long chain fatty acids but also hydroxy fatty acids. This study may contribute to whole-cell biocatalyst engineering for biotransformation of hydrophobic substances.</P>
