        갈화 ( Puerariae flos ) 추출물이 Rat 혈중 Ethanol 농도에 미치는 영향

        Ethanol concentration in blood, brain and liver of rats was shown to be effectively lowered by arrowroot flower extract. The lowering effect for ethanol concentration in blood was maximum when measured after 1 hour from ethanol feeding. Hot water extract was more effective than 80% ethanol extract. The treatment of extract at 10 min. before ethanol feeding gave a better result than that at 10 min after or 1 hour before ethanol feeding. The ethanol concentration in brain and liver was lowered as found in the blood ethanol concentration. Acetaldehyde was not detected either in blood or the tissues. The optimal amount of the Puerariae flos was 55.7㎎/㎏·body weight. The newly developed analytical method using dichloromethane as extracting solvent was proven to be very effective in terms of speed and simplicity.

      • 都市主婦의 MAP와 %Fat 調査分析

        The purpose of this study is to analyze a maximal aerobic power(MAP) and % fat of the urban female. The subjects are 73 urban females who were devided into three groups(27 young femeles, 35 middle females, and 11 old age females) I measured Margaria bench stepping, VO_2 max calculated by Margaria's estimation expression, and % fat was calculated by using Nagamine's estimation expresstion. Therefore, The results of this study is to be compared to the cross sectional analysis among three groups. The results are as follows : 1. 「MAP」 Young female groups MAP was 41.4±13.99㎖/㎏·min, middle age females 39.2±13.95㎖/㎏·min, and old age females, 38.2±10.4㎖/㎏·min. This data showed that 3 groups were in fitness. 2. 「% Fat」 %fat in young female groups % fat was 31.4±5.46%, middle age females, 32.5±8.09%, and old age females, 31.3±6.92%. According to the results, 3 groups have no statistical significance(P>.05). These data showed that three groups were over weight(obesity).

      • Crosslinked anion exchange membranes with primary diamine-based crosslinkers for vanadium redox flow battery application

        <P><B>Abstract</B></P> <P>A series of polysulfone-based crosslinked anion exchange membranes (AEMs) with primary diamine-based crosslinkers has been prepared via simple a crosslinking process as low-cost and durable membranes for vanadium redox flow batteries (VRFBs). Chloromethylated polysulfone is used as a precursor polymer for crosslinked AEMs (CAPSU-x) with different degrees of crosslinking. Among the developed AEMs, CAPSU-2.5 shows outstanding dimensional stability and anion (Cl<SUP>−</SUP>, SO<SUB>4</SUB> <SUP>2−</SUP>, and OH<SUP>−</SUP>) conductivity. Moreover, CAPSU-2.5 exhibits much lower vanadium ion permeability (2.72 × 10<SUP>−8</SUP> cm<SUP>2</SUP> min<SUP>−1</SUP>) than Nafion 115 (2.88 × 10<SUP>−6</SUP> cm<SUP>2</SUP> min<SUP>−1</SUP>), which results in an excellent coulombic efficiency of 100%. The chemical and operational stabilities of the membranes have been investigated via ex situ soaking tests in 0.1 M VO<SUB>2</SUB> <SUP>+</SUP> solution and in situ operation tests for 100 cycles, respectively.       • Loop와 HPLC Purification 방법보다 더 높은 비방사능을 보여주는 카트리지 Methylation과 Purification을 이용한 손쉬운 [ <sup>11</sup>C]PIB 합성

        $[^{11}C]PIB$는 베타아밀로이드($A{\beta}\;plague$)라는 변성 단백질에 결합하여 뇌의 기능과 기억력을 서서히 감퇴시키는 비가역적인 질환인 치매를 조기에 감별할 수 있는 대표적인 방사성의약품이다. 지금까지 많은 실험실에서 $[^{11}C]PIB$는 자동화합성장치에서 $[^{11}C]methyl\;iodide$나 $[^{11}C]methyl\;triflate$를 만든 다음 loop나 vial 방법을 사용하여 methylation을 한 다음 HPLC로 정제를 하는 것이다. 하지만 기존의 보고된 방법은 시간이 오래 걸리며, HPLC와 같은 복잡한 시스템을 필요로 하여 소규모 실험실에서 합성하기에 적합하지 않으며, 최종 product에서 에탄올 함량이 높다는 단점이 있었다. 이러한 단점을 보완하기 위하여 카트리지만을 사용하여 카트리지에서 methylation과 purification을 동시에 실시함으로써 합성 시간을 단축하고, 비방사능이 높고, 낮은 에탄올 함량을 가진 $[^{11}C]PIB$를 합성 가능한지 확인하고자 하였다. 가장 널리 사용하는 카트리지 6종(CM, HLB, Alumina, C18, tC18, tC18 environmental을 선택하여 screening test를 실시하였다. 6-OH-BTA-0 1 mg을 c-HXO에 녹인 다음 6개의 카트리지에 loading를 한 다음 0.5 M MSP(pH 5.1) 20 mL로 정제를 한 다음 최종 fraction을 받아서 analytical HPLC로 전구체 잔류량을 측정한 결과 hydrophobicity가 낮은 계열(CM, HLB, Alumina)의 카트리지에서는 완충액으로 정제를 하였을 때 잔류전구체의 양이 많았으나, 탄소함량이 많은 계열의 카트리지(C18, tC18, tC18 environmental)에서는 잔류전구체의 양이 CM, HLB, Alumina 카트리지에 비하여 상대적으로 적었다. 완충액의 정제 농도와 부피를 최적화 하기 위하여 screening test에서 가장 좋은 결과를 나타낸 C18 series cartridge를 가지고 추가 실험을 진행하였다. 인산완충액 농도를 10 mM, 20 mM, 30 mM, 40 mM, 50 mM, 250 mM, 500 mM로 변화시켰으며, 에탄올 함량은 20%와 30%로 하여 용출액을 분석하여서, $[^{11}C]PIB$를 카트리지로 합성하기 위한 최적의 조합은 tC18 environmental cartridge와 0.5 M MSP 20 mL인 것을 알 수 있었다. 기존에 보고된 방법과 cartridge를 비교한 결과, 합성시간에서는 각각 15 ~ 18min, 8 ~ 9 min이 소요되었으며, product activity는 각각 $4.1{\pm}1.4\;GBq$ (n=41), $3.8{\pm}0.9\;GBq$ (n=3), 방사화학적 수율(based on HPLC analysis of the crude product)에서는 $13.9{\pm}4.4%$ (n=41), $12.3{\pm}2.2%$ (n=3)로 별다른 차이가 없었으며, 비방사능에 있어서는 HPLC purification method가 $78.7{\pm}39.7\;GBq/{\mu}mol$ (n=41), cartridge method가 $420.6{\pm}20.4\;GBq/{\mu}mol$ (n=3)로 카트리지 방법이 기존 방법보다 더 좋은 결과를 나타내었다. 또한, 잔류 용매(c-HXO)도 vial or loop method와 별다른 차이가 없었으며, 에탄올 함량에 있어서는 70%(기존 방법)에서 30%(카트리지 방법)로 두 배 이상 함량이 적다는 사실을 알 수 있었다. 지금까지 알아본바와 같이 cartridge method는 reported method(HPLC purification)에 비하여 더 향상된 결과를 보여준다는 사실을 확인하였다. $[^{11}C]PIB$ synthesis has been performed by a loop-methylation and HPLC purification in our lab. However, this method is time-consuming and requires complicated systems. Thus, we developed an on-cartridge method which simplified the synthetic procedure and reduced time greatly by removing HPLC purification step. We compared 6 different cartridges and evaluated the $[^{11}C]PIB$ production yields and specific activities. $[^{11}C]MeOTf$ was synthesized by using TRACERlab FXC Pro and was transferred into the cartridge by blowing with helium gas for 3 min. To remove byproducts and impurities, cartridges were washed out by 20 mL of 30% EtOH in 0.5 M $NaH_2PO_4$ solution (pH 5.1) and 10 mL of distilled water. And then, $[^{11}C]PIB$ was eluted by 5 mL of 30% EtOH in 0.5 M $NaH_2PO_4$ into the collecting vial containing 10 mL saline. Among the 6 cartridges, only tC18 environmental cartridge could remove impurities and byproducts from $[^{11}C]PIB$ completely and showed higher specific activity than traditional HPLC purification method. This method took only 8 ~ 9 min from methylation to formulation. For the tC18 environmental cartridge and conventional HPLC loop methods, the radiochemical yields were $12.3{\pm}2.2%$ and $13.9{\pm}4.4%$, respectively, and the molar activities were $420.6{\pm}20.4GBq/{\mu}mol$ (n=3) and $78.7{\pm}39.7GBq/{\mu}mol$ (n=41), respectively. We successfully developed a facile on-cartridge methylation method for $[^{11}C]PIB$ synthesis which enabled the procedure more simple and rapid, and showed higher molar radio-activity than HPLC purification method.

      • Pseudomonas sp. BCNU 171균에서의 유기용매 shock반응과 heat shock반응의 연관성

        The response of Pseudomonas sp. BCNU 171 to various forms of stress was examined by using O'Farrell two-dimensional gel electrophoresis. The toluene resistance of mid-growth cells of Pseudomonas sp. BCNU 171 grown at 30℃ was enhanced by exposing the cells to a prior toluene shock in the presence of 0.05% toluene for 30 min. The heat tolerance of the cell was also induced by the same condition. Individual protein expression was examined at 30 min and 60 min by silver staining after the addition of 0.05%(v/v) toluene to mid-growing cultures of the strain BCNU 171. The toluene stress response increased the expression of eighteen proteins between 30 and 60 min toluene exposure. At 60 min after toluene addition, a toluene specific protein was detected. A few proteins induced by toluene and heat were overlapped on 2D-PAGE.

        Factors Influencing the Acrylamide Content of Fried Potato Products

        BACKGROUND: Acrylamide (CAS No. 79-06-1) is known to be a carcinogenic compound, and is classified as a Group 2A compound by the International Agency for Research on Cancer (IARC, 1994). Acrylamide can be generated during the browning process via the non-enzymatic Maillard reaction of carbohydrates such as reducing sugars and of amino acids such as asparagine, both of which occur at a temperature above $120^{\circ}C$. Potato tubers contain reducing sugars, and thus, this will affect the safety of processed potato products such as potato chips and French fries. In order to reduce the level of acrylamide in potato processed products, it is therefore necessary to understand factors that affect the reducing sugar content of potatoes, such as environmental factors and potato storage conditions, as well as understanding factors affecting acrylamide formation during potato processing itself. METHODS AND RESULTS: Potatoes were cultivated in eight regions of Korea; For each of these different environments, soil physico-chemical characteristics such as pH, electrical conductivity, total nitrogen, available phosphate, and exchangeable cation content were measured and correlations with potato reducing sugar content and potato chip acrylamide levels were examined. The reducing sugar content in potato during storage for three months was determined and acrylamide level in potato chip was analyzed after processing. The storage temperature levels were $4^{\circ}C$, $8^{\circ}C$, or $10^{\circ}C$, respectively. The acrylamide content of chips prepared from potatoes stored at $10^{\circ}C$ or $20^{\circ}C$ for one month was analyzed and the different frying times were 2, 3, 5, and 7 min. CONCLUSION: This study showed that monitoring and controlling the phosphate content within a potato field should be sufficient to avoid producing brown or black potato chips. For potatoes stored at low temperatures, a reconditioning period ($20^{\circ}C$ for 20 days) is required in order to reduce the levels of reducing sugars in the potato and subsequently reduce the acrylamide and improve chip coloration and appearance.

        벤조디아제핀 수용체 영상용 양전자 방출 핵종 표지 플루마제닐 유도체 [F-18](3-(2-Fluoro)flumazenil의 합성과 생체 내 분포

        Purpose: Radiotracers that bind to the central benzodiazepine receptor are useful for the investigation of various neurological and psychiatric diseases. [C-11]Flumazenil, a benzodiazepine antagonist, is the most widely used radioligand for central benzodiazepine receptor imaging by PET. We synthesized 3-(2-[F- 18]fluoro)flumazenil, a new fluorine-18 (t1/2=110 min) labeled analogue of benzodiazepine receptor imaging agent, and evaluated in vivo for biodistribution in mice. Materials and Methods: Flumazenil (Ro 15-1788) was synthesized by a modification of the reported method. Precursor of 3-(2-[F-18]fluoro)flumazenil, the tosylated flumazenil derivative was prepared by the tosylation of the ethyl ester by ditosylethane. [F-18] labeling of tosyl substitued flumazenil precursor was performed by adding F-18 ion at 85℃ in the hot cell for 20 min. The reaction mixture was trapped by C18 cartridge, washed with 10% ethanol, and eluted by 40% ethanol. Bidistribution in mice was determined after intravenous injection Results: The total chemical yield of tosylated flumazenil derivative was ∼40%. The efficiency of labeling 3-(2-[F-18]fluoro)flumazenil was 66% with a total synthesis time of 50 min. Brain uptakes of 3-(2-[F-18]fluoro)flumazenil at 10, 30, 60 min after injection, were 2.5±0.37, 2.2±0.26, 2.1±0.11 and blood activities were 3.7±0.43, 3.3±0.07, 3.3±0.09%ID/g, respectively. Conclusion: We synthesized a tosylated flumazenil derivative which was successfully labeled with no-carrier-added F-18 by nucleophilic substitution.

      • RF 마그네트론 스퍼터링 방법에 의한 (Ba, Sr)TiO₃박막의 특성

        A sputtering target(BaxSr1-xTiO3) is obtained by changing the mol% of Ba and Sr. Pt is evaporated on the SiO2/Si substrate as a bottom electrode, and subsequently the BaxSr1-xTiO3 thin film is sputtered. Or, for obtaining the MIS structure Pt is not used, and directly thin film is sputtered on the Si substrate. Then, the capacitor structures(MIM, MIS) have been obtained by evaporating Al as a top electrode after post-annealing. Our results show that produced thin firm is found to have Perovskite structure according to the XRD measurement and shows better crystal growing phenomena on SiO2/Si substrate than those on Si substrate. And, the existence of polarization is recognized by the hysteresis loop. The capacitor(MIM) structure of BaxSr1-xTiO3 thin film shows that the dielectric constant is increased as the mol% of Ba and the post-annealing temperature increase. MIS structure shows a increase of capacitance with increasing the mol% of Ba.

      • Sol-Gel 법으로 제작한 (Pb_xBa_₁-_x)TiO₃박막의 특성에 관한 연구

        We tried to find optimal condition of (PbxBa1-x)TiO3(x=0.0, 0.2, 0.5, 0.8, 1.0) thin films produced by sol-gel spin coating. Barium acetate, Lead acetate trihydrate and Ti-isopropoxide are used as the coating sol for producing the thin film, and acetic acid, methanol, and distilled water are used as the solvent. It is found that the manufactured sol was at the most stable condition when the component content satisfied H2O/Ba=65, after observing the state transition for a specified period. We coated sol for coating on the various kinds of substrate and pre-annealed on 68℃ and 460℃. Then we annealed them on 700℃ for 30 minutes and found that they showed a good growth of crystal by the XRD measurement. We selected Pt as the bottom electrode, using the substrate Pt/SiO2/Si in order, we raised the different conditional thin films and obtained the capacitor by evaporating Al as the top electrode. We measured the electric capacitance and dielectric loss by changing the frequency for three step as 1, 10, 100kHz for each and calculated the dielectric constant. The dielectric constant of the thin film (PbxBa1-x)TiO3 was decreased below x=0.8 and increased upto x=1.0 as x increases.

        한국판 아동용 Leyton 강박증 척도의 개발

        요 약 소아의 강박증 평가 도구인 아동용 레이톤 강박증 척도를 한국판 집단용 지필 검사로 개발하기 위하여 본 연구를 시행하였다. 국민학교 4학년부터 고등하교 3학년까지 127 명을 대상으로 한국어로 번역한 지필 검사 양식의 아동용 레이톤 강박증 척도를 실시하였고, 검사- 재검사 신뢰도 검증을 위하여 3주뒤에 82명에게 재검사를 시행하였다. 또한 전 대상 군에 신뢰도와 타당도가 이미 입증된 간이진단검사(SCL90-R)를 함께 실시하여 공존타당도가 있는 지를 알아 보았 고, 강박환아군과 정신과 환아대조군 그리고 정상대조군을 9명씩 비교하여 진단 변별력이 있는 지를 보 았다. 검사-재검사 신뢰도, 내적 일관성 등은 상당히 만족스러운 수준이었고, 간이진단검사와의 공존 타당도도 비교적 높게 나왔다. 강박환아군이 정신과 환아대조군이나 정상 대조군보다 유의하게 높은 점수가 나와서 변별력 또한 높은 것으로 나타났다. 요인분석상 문항들을 5개의 요인 으로 묶을 수 있었다. 한편 증상과 저항척도에서 여아가 남아보다 유의하게 높은 점수를 보고하였 으며, 특히 국민학교 여아에서 높은 점수를 보여 주었다. 결론적으로 한국판 아동용 레이톤 강박척도는 실시하기 간편하고, 신뢰도와 타당도가 좋은 척 도임 이 입증되 었다. 향후 전국 규모의 규준을 작성하고, 미흡한 부분을 보완한 후속 연구가 필요하다. ABSTRACT Objects : This study was carried out to develop the Korean form of Leyton Obsessional Inventory-Child Version(LOI-CV) designed to assess the obsessive compulsive symptoms in children and adolescents. The LOI-CV was modified into a paper-and-pencil form in order to administer it to group. Methods : We applied the Korean form LOI-CV to 127 elementary, middle and high school children, and retest it to 82 children with 3 weeks interval. Togather with LOI-CV, Korean form of SCL-90-R which had been proved its reliability and validity, were administered to the all subjects for examininig of concurrent validity And then we examined whether this scale discriminates between obsessive patients, psychiatric controls and normal controls in the level of obsessive symptoms. Results : Test-retest reliability, internal consistency were very satisfactory. Concurrent validity with SCL-90-R was moderate level. And obsessive patients was scored significantly higher than psychiatric and normal controls in the level of obsessive symptoms. In factor analysis, items are clustered to 5 factors. In the global obsessive symptom, obsessive scores of female elimentary school students were higher than that of male students. Conclusions : These results indicate that the Korean form of LOI-CV is a reliable and valid self-rating scale to assess the obsessive-compulsive symptoms in Korean children and adolesce- nts. Author suggests that future research is needed for Korean version standardization study in normal and clinic-refered population.
