        Heme oxygenase-1 induced by desoxo-narchinol-A attenuated the severity of acute pancreatitis via blockade of neutrophil infiltration

        <P><B>Abstract</B></P> <P>Heme oxygenase-1 (HO-1) has an anti-inflammatory action in acute pancreatitis (AP). However, its mechanism of action and natural compounds/drugs to induce HO-1 in pancreas are not well understood. In this study, we investigated the regulatory mechanisms of HO-1 during AP using desoxo-narchinol-A (DN), the natural compound inducing HO-1 in the pancreas. Female C57/BL6 Mice were intraperitoneally injected with supramaximal concentrations of cerulein (50 μg/kg) hourly for 6 h to induce AP. DMSO or DN was administered intraperitoneally, then mice were sacrificed 6 h after the final cerulein injection. Administration of DN increased pancreatic HO-1 expression through activation of activating protein-1, mediated by mitogen-activated protein kinases. Furthermore, DN treatment reduced the pancreatic weight-to-body weight ratio as well as production of digestive enzymes and pro-inflammatory cytokines. Inhibition of HO-1 by tin protoporphyrin IX abolished the protective effects of DN on pancreatic damage. Additionally, DN treatment inhibited neutrophil infiltration into the pancreas via regulation of chemokine (C-X-C motif) ligand 2 (CXCL2) by HO-1.       • UV/Persulfate 산화 및 전기분해 방법을 이용한 TMAH 용액 및 부산물 처리 연구

        현재 TFT-LCD를 제조하는 과정에서 다양한 종류의 화학약품을 사용하며 이로 인해 다양한 종류의 폐수가 다량 발생되고 있다. 이 중 TMAH(Tetra methyl ammonium hydroxide)는 유입되는 질소의 주요 성분으로 TFT-LCD 제조 공정에 있어서 핵심 공정 중의 하나인 Photo 공정 중 포토 레지스트(photo resist)의 현상액으로 사용되는 강알칼리의 유기 아민계 용액으로, 무색, 투명의 강염기성 물질이고, 수용성, 가연성의 성질을 나타낸다. 또한 그 농도가 증가하면서 강한 독성을 배출하여 생물학적 분해가 어렵고, 분자 구조 속에 질소를 포함하고 있어, 분해 시 암모니아성 질소를 생성하는 특성을 가지고 있다. 이에 따라 자연 수중 생태계에 영향 등의 문제가 발생되기 때문에 TMAH 폐수의 효율적인 처리방법 도출이 필요한 사항이다. 현재 TMAH를 함유한 폐수는 호기성 생분해, 혐기성 생분해, 촉매 산화, 광촉매 분해(UV 조사) 또는 이온 교환 기술 등으로 처리되고 있습니다. 그 중 UV 조사는 과황산염 활성화를 위한 에너지의 한 형태로 적용되며 난분해성 분해에 많이 적용되고 있다. 과황산음이온(S<sub>2</sub>O<sub>8</sub> <sup>2-</sup>)은 산화환원전위가 2.01 V인 강한 산화제이며 토양과 지하수에 존재하는 유기오염물질을 분해하는데 효과적이라고 한다. 과황산염 반응 메커니즘은 다양한 과황산염 활성화를 통한 강한 황산염 라디칼(SO<sub>4</sub><sup>-·</sup>) 및 하이드록실 라디칼(HO<sup>·</sup>)의 생성에 기반을 두고 있으며 1 mole의 황산염은 자외선 조사를 통해 2 mole의 황산염 라디칼을 생성하도록 활성화 될 수 있다고 보고되어 다양한 난분해성 유기 오염 물질을 분해에 적용되고 있다. 전기화학적 산화 처리공정은 고분자량을 가지는 화합물을 파괴하며 난분해성 유기물질의 독성을 감소시킬 수 있어 효과적인 것으로 제시되고 있으며, 고도처리공정들 가운데 오염물질 생분해성과 관계없이 높은 제거율과 처리시간이 짧은 운전특성을 나타내고 있어 유기화합물 또는 질소화합물의 제거에 있어서 응용가능성이 높은 것으로 평가되고 있다. 따라서, 본 연구에서는 TMAH 모형폐수를 제조하여 먼저 과황산염(S<sub>2</sub>O<sub>8</sub> <sup>2-</sup>)을 이용한 UV 산화방법으로 TMAH를 분해하고 그에 따른 부산물을 전기분해 방법으로 처리하고자 하였다.

      • The Antioxidant Effect of the Rhaponticum uniflorum Extracts on the Oxidative Stress in H₂O₂ induced Human Keratinocytes

        In this study, To investigate the antioxidant activities of Rhaponticum uniflorum extracts (RUE), we assessed scavenging activities on DPPH, ABTS radicals, and reducing power. The DPPH and ABTS radical scavenging activities (SC50) of RUE were 96.97±1.19 μg/mL and 31.26±0.13 μ g/mL, respectively, and the reducing power significantly increased dose-dependent manner. To study the antioxidant effecters of RUE in H₂O₂ induced human keratinocytes, expression of Nuclear factor erythroid-2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) were determined by Western blot and qRT-PCR. RUE induced the expression of HO-1, a critical target enzyme of Nrf2, in a concentration-dependent manner. The expression of HO- 1 mRNA was also induced in H₂O₂-treated HaCaT cells but no significant difference in the level of HO-1 mRNA was observed. Taken together, these results suggest that RUE may be useful as a potential ingredient for anti-aging products.

        8년간(1992~1999) 산업안전보건연구원에 의뢰된 직업병 심의 사례 분석

        Objectives : Pneumoconisis and noise-induced hearing loss (NIHL) have been reported as main occupational diseases by the Special Health Examination. The Industrial Accident Compensation Insurance has reported various work-related diseases, however, these two diseases occupied almost a half of compensated cases. Therefore, it was not well known about the status of occupational diseases other than pneumoconiosis, NIHL, and cardio-cerebrovascular accident (CVA). This study was conducted to analyze claimed cases as an occupational disease, that was requested to the Korea Industrial Safety and Health Agency (KOSHA). Methods : The local office of the Korea Labor Welfare Corporation (KLWC) has asked the KOSHA for confirmation of claimed cases as an occupational disease. We analyzed 379 cases requested from KLWC, the Ministry of Labor, employers, unions and occupational health agencies from 1992 to 1999. Results : Male was 80.7 % of the requested cases. Their mean age was 42 years old and 75.5 % of them were more than 35 years old. The requested cases were increased rapidly from 25 cases in 1992 to 108 cases in 1999 and the accept rate was 50.7 %. The majority of requested cases were respiratory diseases(22.4%), cancers(18.5%), Neuropsychiatric problems(14.5%), and musculoskeletal problems(13.5%). The accept rate was high in reproductive, respiratory, musculoskeletal and digestive disorders and low in neuropsychiatric, renal and otologic problems and occupational cancers. 73.6% of them were caused by chemical agents, especially 28.5 % were by organic solvents. 67% of them were clinically confirmed at university hospitals. A half of the cases were from KyongIn area, even the request came from the whole country. Conclusions : A claim was common in workers whose age was over 35 years old and exposure history was over 10 years. The respiratory diseases and neuropsychiatric disorders were still main problems in occupational health and occupational cancers was increasing even though its accept rate was not high yet.

      • 흰쥐 십이지장 점막의 Polyamine 대사, [^(14)C]Spermine 단백 결합능 및 [³H]Thymidine DNA 섭취에 대한 Calcitriol과 Prednisolone의 상호작용

        Calcitriol has been shown to induce the duodenal synthesis of the spermine-binding protein. and hydrocortisone was reported to increase the intestinal polyamine contents. This study examined whether prednisolone might enhance or reduce the calcitriol effects on the polyamine contents, [^(14)C]spermine-protein binding activity, and [^(3)H] thymidine DNA synthesis of the rat duodenum. The mucosal contents of putrescine. spermidine. and spermine were increased by 198.8%, 84.0%, and 78.3%, respectively, within 6 hours after calcitriol. 5 ㎍/kg (CT) and fell down to the control values at 12 hours. And prednisolone. 10 mg/kg (PD) also increased the polyamine contents in similar patterns. They did not counteract each other in the polyamine metabolism. CT increased the [^(14)C]spermine-binding activity of the cytosol protein fraction from the duodenal mucosa by 71.1%. But PD increased the binding activity by 28.0% and did not affect the CT-induced increase. CT increased the [^(3)H]thymidine DNA synthesis of the duodenal mucosa by 42.3%. but PD reduced it slightly. The CT-dependent increase of the synthesis was significantly inhibited by PD. These results suggested that the inhibitory effect of glucocorticoids on the vitamin D dependent increases of the duodenal calcium absorption and mucosal proliferation seems not to be associated with the metabolic changes of polyamines in the duodenal mucosa.

      • Drosophila melanogaster에 미치는 Kojic acid의 독성에 관한 연구

        수종의 Aspergillus 및 Pencillum 속 균에 의해 생산되는 진균애사 산물인 kojic acid (5-hydroxy-2-hydroxymethyl-γ-pyrone) 가 Drosophila melanogast에 미치는 독성 효과에 대한 기초 조사를 실시하였다. Kojic acid의 처리 농도가 증가할수록 발생단계상의 발육속도의 지연과 성체에 대한 높은 치사율의 증로 강한 독성을 나타내었다. 성체에서의 LC_50 값(the concentration at which 50% of treated filles died within 72hr)은 약 4.8mg/ml로 추정되었으며, 발생단계상의 처리로 인한 우화 후 성비에 대한 유의적인 영향은 주지 않았다. 그리고 본 실험에서 사용한 attached-X법에 의한 반성 치사 돌연변이 유발에서도 유의적인 돌연변이 유발 효과가 없는 것으로 나타났다. The present experiment was carried out to detect the basic toxic effect of kojic acid (5-hydroxy-2-hydroxymethyl-γ-pyrone), a fungal metabolite produced by some species of Aspergillus and Penicillium, in Drosophila melanogaster, Kojic acid was highly toxic on the developmental and adult stages, resulting in prolongation of the developmental time and lowering of the viability from larvae to adult, and high mortality of adults as dose 'increased, The LC_50 value at 72 hr i.e., the concentration at which 50% of treated flies died within 72hr, was about 4.8mg/ml. As to the sex ratio of flies fed kojic acid during the development, there was no differences between the control and the treated groups, And, kojic acid gave no a significant mutagenic effect on the inducbon of the sex-@ed lethal mutagenesis by the attached-X method used in this study.

        In Korea, fermented fish has been playing an important role as a preserved and flavor rich food. It is said that the digestion of fish protein is due to both action of intrinsic(autolytic enzymes) and bacterial enzymes in fish. The mass production of fermented fish has been impeded since traditional method of fermentation requires a long duration for a complete digestion. A high concentration of salt and unsanitary condition are also considered disadvantages of the old method. To improve the quality of the product and to develop mechanized process of fermentation, fermentors which have such control device as temperature, pH and agitation control system have been urgently needed, In this study, a model design of a fermentor is studied. The calculation was based on the optimum conditions for enzymatic hydrolysis of fish protein which involve temperature, pH, viscosity and other factors.

      • 테니스 포핸드 스트로크 시 왼팔의 이용 유무가 스트로크의 정확성에 미치는 영향

        The purpose of this study is to analyze and compare accuracy and kinematic characteristics of forehand strokes upon using the left-hand and non the using left hand in tennis. Six unskilled collegiate players who complied primary tennis course participated in this study. Subjects were requested to hit 10 balls with 4 types each: first, it was two directions(down the line, cross), second, it was two velocities(slow ball, fast ball) and then it was type of using left hand. Variables such as accuracy, time, distance, angle and velocity variables were measured by USPTA method and video cameras. The results of this study are as follows: 1) accuracy of hitting balls was not different according to the type of using left hand. While there was an interaction between ball velocity and hitting direction. 2) impact positions of hitting ball were not different according to the type of using left hand. While impact positions were done near by target direction in cross direction then down the line direction. 3) shoulder rotation angle was not different according to the type of using left hand. While shoulder rotation angle was grater in down the line direction than cross direction. 4) the maximum velocity of racket head was faster in using left hand than non using left hand.

        Starter 및 멸치액젓 첨가가 김치양념 및 겉절이 김치의 품질에 미치는 영향

        양념에 Leu. mesenteroides, Lac plantarum의 젖산균과 멸치액젓 첨가가 김치 양념의 대장균군 제어, 속성촉진여부 및 관능적 특성에 미치는 영향을 조사하였다. Leu. mesenteroides과 젓갈을 첨가한 경우 초기 숙성이 촉진되나 후기 산패는 완만하였으며 Lac. plantarum의 첨가시에는 초기 숙성에는 큰 차이가 없고 후기 산패가 현저하게 진행되었다. 대장균군은 산도 0.80% 이상에서 사멸되었는데 leu. mesenteroides의 경우가 숙성이 촉진되어 보다 신속히 사멸시켰다. 총균수와 젖산균수는 초기에 starter 첨가구는 10^6CFU/mL 정도인데 반해서 starter를 첨가하지 않은 않은 처리구에서 총균수는 10^6CFU/mL , 젖산균수가 10^4CFU/mL 이었고 숙성 후에는 첨가균이 높은 수준을 유지하였다. 하지만 대체적으로 스타터의 첨가가 숙성 및 대장균에 크게 영향을 주지 못하는 경향으로 동일하게 나왔다. 관능적 특성에서는 Leu. mesenteroides 첨가구가 Lac. plantarum의 경우보다 우수하였다. 한편, 시판 겉절이 김치는 산도가 0.09~0.18%, ph 5.46~5.89, 총균수 2.2×10^6~3.1×10^7CFU/mL, 젖산균수는 2.1×10^4~6.6×10^5CFU/mL, 대장균수는 1.8×10^4~7.1×10^5CFU/mL, 염분 1.89%~2.51%으로 스타터 첨가 겉절이에 비해 ph와 염도를 제외한 품질 특성에서 전반적으로 낮은 값을 나타냈다. This study was performed in order to investigate the effect of Leuconostoc mesenteroides, Lactobacillus plantarum and salt fermented anchovy extracts on Kimchi sauce. The sauce was fermented more rapidly by addition of Leu. mesenteroids or salt fermented anchovy extracts on the early fermentation stage than the control, but it was fermented slowly on the late fermentation stage. When L. plantarum was added to the Kimchi on the early fermentation stage, the acidity of Kimchi didn't show a significant difference from the control, but acidity was remarkably increased on the late stage. Coliform group was disappeared when acidity of sauce was higher than 0.8% during fermentation. It was controlled by Leu. mesenteroides but not by L. plantarum. Total count and lactic acid bacterial count of the sauces with starter were 6.30×10^6~1.0×10^7 CFU/mL and 1.04~2.04×10^6 CFU/mL, respectively, but those of the control sauce were 10^6 CFU/mL and 10^4 CFU/mL, respectively. Those count of the sauce with starter were higher than those of the control sauce on the later stage of fermentation. Organoleptic quality of the sauce with Leu. mesenteroides was superior to that with L. plantarum.

        구미산업단지 대기중 휘발성유기화합물(VOCs)의 농도특성

        This study focuses on the measurement of airborne Volatile Organic Compounds (VOCs) in the Kumi electronic industrial complex during the time periods of August and September, 2002 and January and February, 2003. This study was based on the US-EPA method TO-14 while the VOCs were analyzed with GC/MSD. The toluene level revealed high concentration at all measurement sites. The areal rank of average concentrations of VOCs is as follows : industry l>industry2>urban>middle>residential. Concentrations of VOCs in Kumi electronic industrial ones were generally higher than at Yeochon and Ulsan industrial complexes. Dichloromethane and trichloroethylene, which are used as a cleaner in the process of electronic industries, were observed 4 to 8 times higher than those of other areas. Among the aromatic compounds, toluene showed the highest level, while the concentrations of dichloromethane and trichloroethylene were higher than those of other halogen compounds. In Kumi, toluene, trichloroethylene, and dichloromethane were confirmed as the major compounds of VOCs by this research.
