        제2형 당뇨병 환자에게 엽산과 아스코르브산 보충이 혈장 호모시스테인 농도와 산화 스트레스에 미치는 영향

        In patients with type 2 diabetes, oxidative stress could be increased by their metabolic changes. Elevated plasma homocysteine is considered as one of markers of enhanced oxidative stress. Due to oxidative stress, some complications like cardiovascular or renal diseases may develop in type 2 diabetes patients. Plasma homocysteine concentration may be increased if folate status were inadequate. Protective effects against oxidative stress may be diminished if the status of anti-oxidative nutrient as vitamin C was poor. It is, therefore, important to maintain adequate status of folate and vitamin C in type 2 diabetes patients. Thus, this study was performed to determine the effects of supplementation of folate and/or ascorbate on blood glycated hemoglobin (HbA1c) level, serum concentrations of homocysteine and cholesterol, plasma oxidized low density-lipoprotein (LDL), concentration and plasma glutathione peroxidase (GSH-Px) activity in the patients with type 2 diabetes. A total of 92 type 2 diabetes patients participated voluntarily with written consents. They were divided into one of the four experimental groups; Control (C), Folate-supplemented (F), Ascorbate-supplemented (A), and Folate plus ascorbate-supplemented (FA). The subjects in C were taken placebo, those in F were supplemented 1 mg of folate, those in A received 1,000 mg of ascorbate, and those in FA were given 1 mg of folate plus 1,000 mg of ascorbate daily for 4 weeks. Supplementation of folate or ascorbate resulted to increase serum folate level or plasma ascorbate concentration apparently, respectively. Folate supplementation not ascorbate seemed to decrease plasma concentrations of homocysteine and oxidized LDL and reduce plasma GSH-Px activity. There might not be synergic effect of the supplementation of folate plus ascorbate. The results indicate that oxidative stress in the patients with type 2 diabetes may lower mainly by folate supplementation.

        흰쥐 부분 근육 절제 모델에서 지방유래줄기세포의 치료 효과

        Adipose tissue derived stem cells(ASCs) are multipotent cells capable of forming bone, cartilage, and other connective tissues. In this study we evaluated the effect of stem cells derived from adipose tissue on the muscle regeneration in partial removed skeletal muscle in rats. Male sprague-dawley rats were randomly divided into three groups: control, PBS-injected and ASCs-injected group. Stem cells were isolated from rat abdominal adipose tissue by collagenase digestion and ASCs labeled with superparamagnetic iron oxide(Feridex) were used. Muscle injury was subjected to rat bypartial removal of quadriceps femoris muscle. The labeled cells were directly injected into the muscle injured area after muscle injury an hour. After 2 weeks, histological analysis was performed using H&E staining and prussian blue staining. The muscle regeneration markers were examined by immunohistochemistry and RT-PCR. ASCs labeled with Feridex were detected by prussian blue staining in injured muscle area. Microscopical muscle regeneration and neovascularization were more increased in ASCs-injected group compared to PBS-injected group. The expression level of Pax7, MyoD and myogenin were more significantly increased in ASCs-injected group compared with the PBS-injected group. This preliminary observation shows that ASCs may be directly involved in the regeneration process of injured skeletal muscle; therefore the possibility of using ASCs as a stem cell therapy for muscle regeneration need to be further investigated for the purpose of clinical trials.

        여드름 흉터의 한의학 치료에 대한 국내 임상 연구 동향

        Objectives : The purpose of this study is to analyze various Korean medical treatments on acne scar and to propose better methods on clinical studies and effective treatments. Methods : Key word 'Acne Scar', published year '2010-2018' were searched on three domestic database systems (RISS, KISS, NDSL). Results : Among 9 studies, 8 studies were case reports and 1 study was comparative study. The treatment used in 9 studies were acupuncture(Subcision, Microneedle Therapy System, laser), pharmacopuncture, external application, dermabrasion, high frequency therapy. 7 out of 9 studies used more than 2 different treatments. The most popular treatment method was 'subcision', followed by 'MTS'. The most popular methods of assessment was QGASC. All studies using assessment methods measuring the severity of acne scar reported improvement, though only 1 study was proved to be statistically different. 1 study reported the side effect, and other 4 studies reported flushing, itching, pain. Conclusions : This study shows the effectiveness of Korean medical treatment on acne scar. It will be expected to use this various treatments and study methods for treating acne scar with Korean medicine, and conducting further clinical studies with higher credibility and reproducibility.

        Adrenomedullin Deficiency Increases the Susceptibility of Liver Fibrosis Induced by CCl<SUB>4</SUB>

        아드레노메둘린은 모든 조직에서 발현되는 펩타이드로 이 펩타이드 수용체가 간경화 유발 시 증가된다. 이번 연구에서는 사염화탄소(CCl4) 투여로 유도되는 간경화에서 아드레노메둘린의 효과를 아드레노메둘린 유전자 한쪽이 결핍된 생쥐에서 관찰하였다. 사염화탄소를 일회투여 받은 급성 군과 6주 동안 지속적으로 투여 받은 만성군으로 나누어 효과를 관찰하였다. 간경화 정도는 병리조직적 검사와 웨스턴 블롯 방법을 사용하여 측정하였다. 급성에서는 결핍된 생쥐와 야생형 생쥐에서 별다른 차이점이 없었다. 만성의 경우 아드레노메둘린이 결핍된 생쥐에서 심각한 간경화가 관찰되었다. 아스파르테이트 아미노전이요소(AST)와 알라닌 아미노전이요소(ALT) 수치가 아드레노메둘린이 결핍된 생쥐 군에서 높게 관찰되었다. 간경화 마커 단백질인 콜라젠, 알파-SMA과 TGF-beta가 아드레노메둘린이 결핍된 생쥐 군에서 높게 관찰되었다. 또한 아드레노메둘린 결핍 생쥐 군은 심각한 지질 과산화 반응이 확인되었고 이는 글루타시오 펄옥시다아제(GSH)의 감소가 원인이었다. 따라서 이 연구는 아드레노메둘린의 결핍은 사염화탄소로 유도된 간경화 감수성을 증가시키는 것으로 결론할 수 있다. 이로써 아드레노메둘린은 간경화 새로운 치료 타겟으로 사용될 수 있음을 나타낸다. Adrenomedullin (AM) is a peptide expressed in all body tissues, and its related receptors are increased in liver fibrosis. In this study, we evaluated the effect of AM deficiency on liver fibrogenesis induced by CCl4 using AM heterozygous (HT) mice. The animals received a single injection of CCl4 or olive oil for the acute experiment, and received CCl4 or olive oil three times a week for 6 weeks for the chronic experiment. Fibrosis was accessed using histopathological analysis and the western blot. The AM HT mice showed mild pericentrilobular degeneration when compared to the AM wild type (WT) mice. In the acute experiment, there was no significant difference between the AM WT and AM HT mice. However, in the chronic experiment, the CCl4-treated AM HT mice showed more severe liver fibrosis than that of the CCl4-treated AM WT mice. The AST and ALT levels of the AM HT CCl4 group were higher than those of the AM WT CCl4 group. Additionally, the collagen deposition, α-SMA protein and TGF-β protein were increased in the AM HT CCl4 group when compared to the AM WT CCl4 group. The AM HT mice also exhibited severe lipid peroxidation through the GSH decrement. Taken together, our data suggest that AM deficiency increases the susceptibility to liver fibrosis induced by CCl4, indicating a novel therapeutic target for patients with liver fibrosis.

        There has been and will be continuous increasing in the use and demand of toxic gases in accordance with the rapid development of domestic semiconductors and related high-tech industries. With the number of facilities handling high-risk toxic gases continuing to skyrocket, the need for a toxic gas safety management agency has emerged for the safe management and use of toxic gases. As part of the safety management policy, Industrial Gas Safety Technology Support Center, a world’s first toxic gas comprehensive safety management center, was founded. The task of Industrial Gas Safety Technology Support Center comes to 3 major businesses : toxic gas disposal business that safely removes toxic gas residue to prevent potential hazards from industrial sites, performance evaluation of toxic gas safety devices to improve the performance and reliability, and education specialized in toxic gas to train toxic gas safety management professionals and to secure technology. In addition, according to the 2nd Basic Plan for Gas Safety Management, the Center performs tasks such as developing toxic gas certified reference materials, establishing a comprehensive education infrastructure, developing certification standards and expanding certification obligations, establishing as toxic gas container disposal system and distributing guidelines. As a toxic gas safety management control tower, the Center is to lead the toxic gas safety management technology, contribute to prevention of toxic gas accident and enhance safety management capabilities through continuous expansion of business areas such as the establishment of an Virtual Reality based educational system and training ground for toxic gas and the construction of and electromagnetic wavetest infrastructure for safety equipment.

        인터넷 쇼핑몰에서 서비스 회복 공정성이 고객의 만족과 재구매 의도에 미치는 영향: 오픈마켓과 종합 인터넷쇼핑몰을 중심으로

        The purpose of this study is to classify the service recovery fairness of internet fashion shopping mall depending on the type of shopping mall into 3 levels, and analyze their influence on customers` satisfaction and intention to repurchase. Data were collected from 271 customers who had made a complaint or raised a question about service among the customers, and was analyzed through AMOS 7.0. For hypothesis testing, a research model was set up in the form of structure equation model, and its suitability was investigated. The results of this research can be summarized as follows: First, in relation to the service recovery fairness at internet shopping mall, only the consequential fairness has a meaningful relationship with customer satisfaction. Second, it has been shown that the higher the customers` satisfaction with internet shopping mall, the more positive influence it has on intention to repurchase. Third, it has been shown that the influence of service recovery fairness on customers` satisfaction and intention to repurchase has a meaningful difference depending on the type of internet shopping mall. In other words, it has been shown that customer satisfaction is higher at open market than general internet shopping mall. Inaddition, it has been shown that in open market, only the consequential fairness has a meaningful influence, therefore it will be more effective to strengthen a strategy focused on economical rewards than other fields.

        상, 하악골 유래 조골세포의 골형성 능: 일차 연구

        Purpose: Maxilla and mandible have different patterns of cortical and trabecular bone and different bone mineral densities, even though both are components of the jaw bone. However, cellular differences between maxilla- and mandible derived osteoblasts (OBs) have rarely been studied. We hypothesize that maxilla- and mandible-derived OBs show different responses to $17{\beta}$-estradiol (E2), which is one of the critical factors for bone formation. This study compares skeletal site-specific cell responses between maxilla- and mandible-derived human OBs to E2. Methods: Maxilla- and mandible-derived OBs derived from an identical donor were separately isolated from a total of five normal healthy subjects aged 18~44 years old, cultured with a treatment of 100 nM estrogen. The responses between maxilla- and mandible-derived OBs to E2 were evaluated and compared using cell proliferation, alkaline phosphatase (ALP) activity and gene expression of osteoprotegerin (OPG), ALP, insulin-like growth factor-1 (IGF-1), and estrogen receptor ${\alpha}$ ($ER{\alpha}$). Results: E2 did not have any distinct effects on the proliferation of both types of OBs. Mandible-derived OBs exhibited higher ALP activity than maxilla-derived OBs in the non-treated condition, which was common in all tested individuals. ALP activities of both types of OBs showed a minor increasing tendency with the treatment of E2, even though there was no statistical significance in some specimens. The gene expression of OB by E2 was diverse, depending on the individuals. There was increased expression of OPG, IGF-1, or $ER{\alpha}$ gene in the part of subjects, which was more repeated in maxilla-derived OBs. In particular, OPG or ALP induction by E appeared less frequently in mandible-derived OBs. Conclusion: Current results revealed that E2 affects maxilla- and mandible-derived OBs into facilitating the osteogenic process despite individual differences. Mandible-derived OBs are less sensitive to bone-forming gene expression by E2.

        2023 파프리카 종자의 Fusarium oxysporum 오염에 의한 철원지역 파프리카 모잘록병 대발생

        In 2023, symptoms like damping-off disease were observed in 74 paprika growing in greenhouses in Cheorwon-gun, Gangwon-do, Korea. In this study, we tried to find the cause of the damping-off disease outbreak. We collected symptomatic seedlings and observed the typical crescent-shaped conidia of Fusarium oxysporum by microscope. To confirm the presence of F. oxysporum in the samples, polymerase chain reaction was performed using primers specific for F. oxysporum; the resulting sequence showed 99.11% identity with F. oxysporum. To confirm the pathogenicity of the F. oxysporum (CW) isolated from the samples, healthy paprika plants were inoculated with F. oxysporum CW and damping-off symptoms were observed 2 weeks later. To investigate whether the damping-off disease outbreak in Cheorwon-gun was caused by F. oxysporum-contaminated seeds, 100 paprika seeds were disinfected and placed in Murashige and Skoog medium. Typical pink F. oxysporum hyphae were found only in control non-disinfected seeds. An 18S rRNA-based and a TEF genebased phylogenetic analysis showed that the F. oxysporum CW isolate was not grouped with a F. oxysporum isolate reported from Cheorwon-gun in 2019. This study is the first report that an outbreak of damping-off disease in paprika in Cheorwon-gun, Gangwon-do, Korea, was caused by contamination of F. oxysporum seeds.

      • 처리방법에 따른 사과 중 잔류농약 제거효과

        농약은 농작물이 잡초나 해충, 세균으로부터 받는 피해를 예방하고 농작물의 생산성 증진 등으로 농업에서 중요한 역할을 하는 농자재이다. 친환경 인증 농산물 외 일반 농산물은 허용기준 이내라도 농약이 잔류할 가능성이 있어 소비자들은 농산물 섭취 시 잔류농약 노출에 불안감을 갖고 있다. 이러한 불안감을 줄이기 위하여 과일에 잔류하는 농약을 효과적으로 제거하는 방법을 규명하기 위한 연구를 진행하였다. 본 실험은 친환경 사과를 처리구당 3개씩 3반복으로 나뉘어 살균제(Carbendazim, Trifloxystrobin), 살충제(Acetamiprid, Diflubenzuron)를 살포한 후, 7일 후에 무처리, 껍질 제거(제거), 물 세척(세척), 물 침지 후 세척(침지), 식초물 침지 후 세척(식초), 소다물 침지 후 세척(소다)의 방법으로 처리한 후 잔류농약을 분석하였다. 침지시간은 5분, 세척시간은 20초로 하였다. 잔류농약 분석을 위한 전처리는 균질화 된 시료 10 g을 50 mL 원심분리관에 칭량하여 Acetonitrile을 10 mL 주가한 후 1분간 진탕하고, 4 g MgSO<sub>4</sub>, 1g NaCl, 1g NaCitrate, 0.5 g Disodium citrate sesquihydrate를 첨가한 후 다시 1분간 균질기로 진탕한 다음 3,000 rpm/min으로 5분간 원심분리를 진행하였다. 상등액 1ml를 Dispersive SPE(25mg PSA, 150 mg MgSO4) tube에 넣고 1분간 진탕하고, 10,000 rpm에서 1분간 원심분리하여 상등액을 0.2 μm 시린지 필터로 여과하여 LC/MS/MS, GC/MS/MS 분석시액으로 사용하였다. 분석방법은 「농산물 잔류농약 다성분 분석방법」에 따라 4가지 농약성분을 분석하였다. 분석 결과 Carbendazim(살균제)은 무처리 대비 제거 88%>소다 72%>침지 64%>식초 61%>세척 55% 순으로 제거되는 결과를 나타냈다. Trifloxystrobin(살균제)은 제거 87%>소다 79%>식초 75%>세척 63%>침지 60% 순으로 제거되었다. Acetamiprid(살충제)는 무처리 대비 제거 91%>식초 63%>소다 59%>침지 50%>세척 47% 순으로 제거되는 결과를 나타냈다. Diflubenzuron(살충제)은 소다 87% >제거 80%>침지 78%>세척 76%>식초 67% 순으로 제거되었다. 4가지 성분 모두 껍질을 제거한 방법에서 잔류농약이 효과적으로 제거되었다. 세척에 따른 결과는 소다침지 후 세척이 Acetamiprid 외 3가지 농약에서 제거효과가 가장 좋았다. 농약성분 별로 처리방법에 따른 제거효과가 다른 결과에 대하여서는 추가적인 연구가 필요할 것으로 사료되었다.

        세라밴드를 이용한 PNF 기법 적용이 65세 이상 여성노인의 균형과 보행에 미치는 영향

        Purpose: Study on the effect of the use of Proprioceptive Neuromuscular Facilitation(PNF) method by use of the direction and charge regulation which is the advantage of the Thera-band therapy on the walks and balances in old people and comparison with the result after applying the general PNF technique. Method: The study has been performed on 30 females over 65 years old. The study has been done by dividing the object group in 3 patterns, which are number 1, the comparison group of 10, two ones that are applied the PNF technique using Thera-band and third, the ones that are applied only the PNF technique. For the PNF and the Thera-band using PNF, we have divided the group into Combination of Isotonic technique and the Rhythmical stabilization technique according to the patient's acquaintance pattern and applied them to the patient's body. Evaluation was the balancing ability which was calculated by using the BIODEX Balance system / FRT and for the evaluation of walking ability, we have used the speed of walking for 10M / TUG. Result: In the comparison group of 10, the balancing ability and the walking ability did not change much before and after the experiment, which made it possible to compare the group with the other two easily(p>0.05). For the other two groups, we have recognized the enhancement both in the balancing ability and the walking ability, but they did not know much difference between themselves(p<0.05). Conclusion: Though there were not a big difference in the sense of improvement between the Thera-band using PNF and the PNF technique only, we could infer that these two therapy has enhanced much in the walking and balancing ability for people over 65 and through these result we can foresee that not only using the method shown in this study but also by using many advantages of Thera-band, we could diminish the tiredness of healer, enhance the efficiency of exercise in them and also by forming self training program for older people we could help them build the prevention program from falls.
