        대왕범바리(Epinephelus fuscoguttatus ♀ × E. lanceolatus ♂) 수정란의장거리 수송을 위한 염분, 수온 및 발생단계에 따른 생존율 및 부화율

        We examined the effect of salinity and water temperature on hatching and survival rates of fertilized eggs of hybrid grouper (Epinephelus fuscoguttatus ♀ × E. lanceolatus ♂) at different developmental stages, determining optimal conditions for their long-distance transportation. Deformities and hatching rates of fertilized grouper eggs were observed at salinities of 24, 27, 30, 33, 36, and 39 psu. The optimal salinity was determined to be 36 psu, with a survival rate of 70.0 ± 2.0%. Transportation experiments at 36 psu were conducted at water temperatures of 21, 24, 27, and 30oC, different developmental stages such as morula, 5-myomere, and tail beating for hatching and survival rates. The optimal water temperature and developmental stage for transporatation were 30oC and tail beating stage and those hatching rates were 50.6 ± 1.9% and 86.3 ± 1.3%, respectively. At 21oC, the survival rate by lowest. Therefore, the hybrid grouper fertilized eggs (E. fuscoguttatus ♀ × E. lanceolatus ♂) can be most efficiently produced under long-distance transportation conditions during the tail beating stage and at a water temperature of 30oC.

        보문 : 섬강의 어류군집 및 멸종위기종 꾸구리 (Gobiobotia macrocephala)와 돌상어(G. brevibarba)의 서식현황

        The fish community structure and inhabiting status of the endangered species Gobiobotia macrocephala and G. brevibarba were investigated at 10 stations in the Seom River, Korea from spring to autumn in 2010. During the survey, 37 species belonging to 10 families were found as inhabitants. A dominant species was Zacco platypus (36.2%), subdominant species were G. macrocephala (13.2%) and G. brevibarba (12.2%), and predominant species were Coreoleuciscus splendidus (8.1%) and Rhinogobius brunneus (4.9%). In addition, 18 species were endemic species to Korea; G. brevibarba and G. macrocephala were endangered species. The similarity index based on species composition and numbers clearly separated fish communities in the Seom River according to main sections, i.e., upper section (St. 1~3), middle section (St. 4~6) and lower section (St. 7~10). Dominance index gradually decreased toward the lower section, while diversity and species richness index gradually increased toward the downstream section. G. macrocephala resided in a fast-running downstream riffle with pebble and cobble substratum, while G. brevibarba inhabited the entire riffle with cobble substratum.

        멸종위기어류 다묵장어 Lethenteron reissneri (Petromyzontiformes: Petromyzontidae)의 분포 및 서식지 특성

        우리나라 멸종위기어류 다묵장어의 분포 및 서식지 특징을 밝히기 위해 2011년부터 2012년까지 분포조사를 실시하였다. 조사기간 동안 모두 755개 지점을 조사한 결과 한강권역에서 8개 지점, 금강권역에서 4개 지점, 섬진강 및 영산강권역에서 32개 지점, 낙동강권역에서 16개 지점 등 모두 60개 지점에서 유생 208개체, 성어 435개체를 채집하였다. 과거의 출현지역과 비교하면 한강 및 금강, 동해안 독립하천, 삽교천 등에서 급격히 감소한 것으로 추정된 반면, 섬진강은 과거보다 많은 출현지점과 개체수가 확인되었다. 출현지역의 서식지 특징은 물이 맑고 소와 여울이 잘 발달한중상류 지역으로 수생식물이 많이 서식하는 곳이었다. 다묵장어의 분포양상을 세계자연보전연맹(IUCN) 적색목록 기준지침서에 따라 평가한 결과 일부 지역에서 서식지역이 급격히 감소하고 있고 출현 지점수와 채집된 성어 수 등을 고려할 때 취약(VU) B2ab(ii,iii,iv,v);D1으로 평가되었으며,그 밖에 다묵장어의 보전 방안에 대해 논의하였다. The distribution status and habitat characteristics of the endangered species, Lethen-teron reissneri were investigated in 2011~2012 in Korea. During the study period, L. reissneri were collected 60 stations among 755 sampling stations investigated. The total numbers of ammocoetes and adults were 208 and 435, respectively from eight stations of the Han River drainage system, four stations of Geum River drainage system, 32 stations of Seomjin and Yeongsan River drainage systems, and 16 stations of Nakdong River drainage system. The Han and Geum river drainage systems, the Sapkyo Stream and independent streams running toward the East Sea were estimated to have suffered sudden reductions of population density, whereas the Seomjin River drainage system show-ed increased population density compared with historic occurrence records. L. reissneri inhabited the middle-upper stream of clean water with well-developed rapids and lots of aquatic organisms. Accord-ing to IUCN Red List categories and criteria L. reissneri was assessed to be vulnerable (VU) [B2ab (ii,iii,iv,v);D1] because of the rapid reduction of the occupying areas and the small number of mature individuals. Besides the distribution status and habitat characteristics we discussed the conservation strategies of L. reissneri.

        우리나라 미꾸리속 (genus Misgurnus) 알비노 개체의 미토콘드리아 및 핵 유전자 염기서열 분석에 의한 유전적 동정

        최근 우리나라에서 자연발생적인 미꾸리속 알비노 개체들이 낮은 빈도로 출현하고 있으나, 색소 결핍으로 인해 형태적 종 동정이 어렵다. 따라서 본 연구에서는 핵 유전자인 recombination activating gene 1 (rag1) 영역 및 미토콘드리아 유전자 cytochrome b (cytb) 영역을 이용한 분자계통학적 분석을 이용해 미꾸리속 알비노 개체의 분자동정을 수행하였다. 그 결과 rag1과 cytb의 분자계통도에서 미꾸라지, 미꾸리, 그리고 M. mohoity로 3개의 clade가 확인되었다. 확보된 M. mohoity의 염기서열을 유전자은행의 BLAST를 이용해 유사성을 검색한 결과 M. mohoity와 가장 유사하였다. 분자계통도를 기준으로 25마리의 알비노 미꾸리속 개체의 종 동정을 수행한 결과 빨간 눈 타입은 미꾸리 16마리, 미꾸라지 1마리로 판별되었고, 나머지 3개체는 미꾸라지♀×미꾸리♂ 잡종 1마리와 M. mohoity♀×미꾸리♂ 잡종이 2마리 판별되었다. 또한 검은 눈 타입 5마리는 미꾸리 1마리와 미꾸라지 3마리 및 M. mohoity 1마리로 판별되었다. 따라서 본 연구에 이용한 분자마커를 활용함으로써 미꾸리속 어류의 정확한 종 또는 잡종을 동정하기 위한 유용한 방법으로 이용될 수 있을 것이다. The spontaneous color mutant, albino individuals of genus Misgurnus, are rarely discovered in Korea and there are difficult to identify morphological species due to lack melanin pigmentation. In this study, we developed a genetic identification method for the species of albino Misgurnus individuals based on phylogenetic analysis by using recombination activating gene 1 (rag1) and cytochrome b (cytb) region of mitochondrial DNA. As a result of molecular phylogenetic analysis, three clades were identified as Misgurnus mizolepis, M. anguillicaudatus and M. mohoity. The homology of the cytb sequences of M. mohoity was best match to that of M. mohoity sequences in GenBank database. As a result of species identification of 25 albino Misgurnus individuals based on the phylogenetic tree, the red-eye type was identified as 16 M. anguillicaudatus and one M. mizolepis. The remaining three individuals were identified as one M. mizolepis ♀×M. anguillicaudatus ♂, and two M. mohoity ♀×M. anguillicaudatus ♂, respectively. In addition, the five black-eye type individuals were identified as one M. anguillicaudatus, three M. mizolepis and one M. mohoity. Therefore, this genetic identification method will be an useful techniques for species or hybrid identification in genus Misgurnus.

        금강 수계 지천의 어류군집 구조 및 멸종위기종 미호종개 Cobitis choii와 흰수마자 Gobiobotia naktongensis의 서식현황

        Community structure of fish and inhabiting status of endangered species, Cobitis choii and Gobiobotia naktongensis were investigated in the Ji Stream, a tributary of the Geum River Drainage System of Korea from May to October 2011. Ten to 23 fish species inhabited upper stream (St. 1~~St. 4) with a Aa-Bb river type composed of mostly pebble and cobble bottoms, 16~~28 species did middle stream (St. 5~~St. 7) with a Bb type composed of mostly cobble and boulder bottoms, and 20-29 species did lower stream (St. 8~~St. 10) with a Bb-Bc type composed of mostly sand bottoms. A total of 44 species belonging to nine families were found in the stream during the survay. The dominant species were in the order of Zacco platypus (37.2%), Pungtungia herzi (5.8%) and Pseudogobio esocinus (5.5%). Other abundant species included Acheilognathus lanceolatus (5.3%), Zacco koreanus (5.2%), Hemibarbus longirostris (4.9%) and Squalidus gracilis majimae (3.5%). Among residing species, 16 species were endemic to Korea, two (Cobits choii and Gobiobotia naktongensis) were endangered, and one (Micropterus salmoides) was non-indigenous. The similarity index based on species composition and abundance clearly delineated the fish community of the Ji Stream according to the three major sections, which were defined at the above. Dominance index gradually decreased toward downstream, while diversity, evenness and species richness indexes gradually increased toward downstream. The two endangered species, C. choii and G. naktongensis co-occurred at the lower stream due to the prevalence of a sandy substratum.

        고성 배봉천의 어류군집과 멸종위기어종의 서식현황

        Fish community structure and inhabit status of endangered fish species were investigated during April to November 2011 in Baebong Stream, Gangwon do, Korea. The number of fish species inhabiting the upper section (Stations 1~3, 5) was 14 and the stream bottoms were mostly composed of cobbles and boulders (Aa-type). Meanwhile, the number of fish species inhabiting the lower section (Stations 4, 6, 7) was 26 and the stream bottoms were mostly composed of pebbles, cobles and sand (Bb-type). A total of 26 species belonging to ten families were found in the stream during the survey period. The dominant and subdominant species were Rhynchocypris steindachneri (31.1%) and Squalidus multimaculatus (13.3%), respectively, and others such as Pungitius kaibarae (11.0%), Zacco koreanus (7.8%), Misgurnus anguillicaudatus (7.0%), Iksookimia pacifica (5.5%) and Z. platypus (5.3%) were followed in the order. Four species of Squalidus multimaculatus, Z. koreanus, I. pacifica and Liobagrus andersoni were endemic to Korea. And, Lethenteron japonica, L. reissneri, Pungitius sinensis and Cottus hangiongensis were endangered fish species in this stream. Oncorhynchus keta, L. japonica and Tribolodon hakonensis were anadromous species, and Plecoglossus altivelis was an amphidromous species. The similarity index based on species composition and abundance clearly divided the fish community structure of the Baebong Stream into two sections fragmented by a weir installed into the stream. It is necessary to install a fish ways on the weirs for a fish migration and movements.

        점몰개 Squalidus multimaculatus (Gobioninae)의 난 발생 및 자치어 형태 발달

        한국고유어종 Squalidus multimaculatus의 난 발생 과정 및 초기 생활사를 연구하였다. 성숙된 암컷 친어에 HCG (human chorionic gonadotropin)를 10 IU/g의 농도로 주사하여 성숙란을 얻었으며, 습식법으로 인공수정하였다. 수정란의 직경은 0.8~0.9mm로 원형으로 투명한 황색을 띈 침성 점착란이었으며, 유구는 없었다. 부화는 수온 평균 24±1°C에서 수정 후 65 시간을 전후하여 시작하였다. 부화자어는 전장이 2.5~3.1m로 입과 항문은 열려있지 않았다. 부화 후 5일째 자어는 전장이 4.0~4.2 mm로 난황이 완전히 흡수되며 입과 항문이 완전히 열리면서 후기자어로 이행하였다. 부화 후 30일째 개체의 전장은 11.2~15.7 mm로 모든 지느러미의 기조수가 정수에 달하며 몸의 무늬가 어미와 유사한 치어 단계로 이행하였다. 부화 45일 후에는 전장이 18.8~22.5mm로 자라 외형이 성어와 유사하였고, 80일 후에는 25.7~35.9 mm로 자라 외형과 반문이 성어와 동일하였다. The egg development and early life history of Korean endemic fish, Squalidus multimaculatus (Gobioninae), were investigated. The eggs from the females were obtained by injecting 10 IU/g of human chorionic gonadotropin and inseminated by wet method in the laboratory. The fertilized eggs were 0.8~0.9 mm in diameter and had no oil globules. The embryo began to hatch about 65 hrs after fertilization under water temperature of 24±1°C. The newly-hatched larvae were 2.5~3.1 mm in total length, and their mouth and anus were not opened. Four days after hatching, the postlarva were 4.0~4.2 mm in total length, and their york sacs were completely absorbed. They entered the juvenile stage when all fin-rays were formed at 30 days after hatching, and their total length were 11.2~15.7 mm. At 45 days after hatching, the external from of juveniles were similar to those of adults (total length were 18.8~22.5 mm), and 80 days after hatching, the external characteristics from of juveniles were same to adults (total length were 25.7~35.9 mm).
