Purpose: This study evaluated the effect and safety of photodynamic therapy (PDT) with verteporfin retreatment on patients with corneal neovascularization. Methods: Corneal neovascularization was induced with a silk suture of the corneal stroma in 24 white rabbits (48 eyes). Four rabbits were examined histologically before performing PDT. Ten rabbits were chosen randomly, one eye from each rabbit was treated with PDT at 50 J/cm2, and the other eye was used as a control. Both eyes of the remaining 10 rabbits were treated with PDT at 150 J/cm2. One week later, one eye was further retreated with PDT at the same intensity. The area of corneal neovascularization was measured and evaluated histologically using light and electron microscopies. Results: The least neovascularized area was observed in the retreated group (P=0.04). The histologic examination revealed fewer new corneal vessels in the retreated group, but the corneal epithelium, stroma, and endothelium showed a normal appearance. Results from electron microscopic examination demonstrated thrombi as well as destruction and nuclear fragmentation of the vascular endothelial cells. However, there were no other changes in the cornea except for vascular endothelial cells, even in the retreated group. Conclusions: In conclusion, these results suggest that PDT with verteporfin retreatment appears to be safe and significantly more effective in patients with regression of corneal neovascularization. Growth of fish is controlled by various environmental factors, including water temperature (WT). WT is also a major factor that affects the eating behavior of fish. In this study, we studied the relationship between WT and expression profiles of factors involved in appetite regulation and digestion in red spotted grouper. Fish were divided into three groups with 30 fish per group and reared for 2 weeks in a recirculating system at three different WT (natural WT, 20 °C, and 25 °C). After 2 weeks, they were sacrificed immediately after food intake (+ 0 h), 3 h after (+ 3 h), 6 h after (+ 6 h), and 3 h before being fed the next day (− 3 h). Fish samples were obtained for analysis of mRNA expression levels of cholecystokinin (CCK), leptin A (LepA), leptin B (LepB), and neuropeptide Y (NPY) in the brain, liver, and intestine. Expression of CCK and NPY mRNA was significantly higher at 25 °C compared to that at natural WT and 20 °C, but no difference was observed for leptin. This suggests that the red spotted grouper shows characteristics typical of subtropical fish and has higher levels of feeding and digestion when reared at 25 °C water temperature condition.
Production of Tagatose by Whole-cell Bioconversion from Fructose Using Corynebacterium glutamicum

Tagatose is a rarely occurring sugar found in food and sweet fruits. It is a potential sweetener with low calories but a sweetness similar to that of sucrose. In this study, we developed a whole-cell biocatalyst using Corynebacterium glutamicum for direct bioconversion of fructose to tagatose. First, we constructed a biological conversion pathway for the conversion of fructose to tagatose by expressing tagatose 4-epimerase (TN) from Thermotoga neapolitana in C. glutamicum. Next, to increase the expression level of the enzyme, we engineered copy number of plasmid. Plasmid library was constructed by randomizing the cgrI antisense RNA region in the plasmid, and the plasmid with high-copy number was isolated using fluorescence-activated single cell sorting (FACS)-based high-throughput screening. The isolated plasmid, pHCP7, had 2-fold higher copy numbers than the original plasmid. A higher expression level and conversion yield could be achieved using pHCP7. Finally, we examined a novel tagatose 4-epimerase TN(KNF4E) isolated from Kosmotoga olearia. The gene expression level was further increased (33.9% of total fraction) through codon optimization and expression in pHCP7, and a conversion yield as high as 21.7% was achieved.
메콩강 수력발전댐 개발에 대한 하류국가 캄보디아 정부의 태도 변화: 그 배경과 영향 요인

전 세계적인 물 부족 현상이 심화됨에 따라 국제 하천을 둘러싼 국가 간의 갈등이 심화되고 있다. 메콩강은 6개 국가가 공유하는 동남아시아의 대표적인 국제하천으로, 경제가 성장함에 따라 수 력발전은 메콩 유역국들의 증가하는 전력 수요를 충족시키기 위 한 주요한 전력 공급방식이 되었다. 그러나 강 상류 지역의 수력발 전은 강 생태계 전체에 부정적인 영향을 미치고 특히 하류지역민 들의 삶에 상당한 영향을 미치기 때문에 메콩강의 댐 건설은 지속 적인 논란의 대상이 되고 있다. 국제하천에서 발생하는 논란을 이 해하기 위해서는 이를 공유하는 여러 국가의 각각 다른 입장을 모두 고려해야 한다. 그러나 지금까지 메콩 수력발전에 대한 특정 국가의 입장이나 태도를 분석한 연구는 중국과 라오스를 중심으 로이루어져왔다. 이에 이 연구에서는다른유역국들 중 메콩강 하류에 위치하여 수력발전으로 인한 이익을 얻기보다는 비용을 부담하게 될 가능성이 높은 캄보디아 정부의 입장을 연구하였다. 캄보디아 정부는 메콩 상류의 수력발전에 대해 어떠한 태도를 보 이고 있으며 그러한 태도는 어디에서 기인하는가? 연구를 통해 메 콩강에 건설되는 댐에 대한 캄보디아 정부의 지지 입장을 확인할 수 있었다. 또한 이러한 입장의 형성에는 1) 중국과의 경제적 비대 칭성, 2) 라오스에 대한 전력 의존도와 지리적 비대칭성, 3) 캄보디 아 국내의 전력 계획 상 수력발전의 중요성이 크게 영향을 미쳤을 것으로 파악되었다.
