Clq-coated ELISA법을 이용한 정맥용 면역글로불린제제의 항보체성 측정

The quality of an intravenous immunoglobulin preparation (IVIG) is reflected by the degree of nonspecific activation of complements, the so-called anticomplementary activity (ACA). ACA of aggregates in IVIG was investigated using method by the European Pharmacopoeia and Clq-coated microtiter enzyme-linked immunosorbent assay (ELISA). Both the EP method and the ELISA method showed a dose response curve with the amount of complements bound increasing with the percentage content of aggiegates in immunoglobulin standard. The correlation between the two tests was good (r=0.96, r=0.99). However the correlation was not found when the ACA (EP method) of IVIG product was compared with its aggregate percentage. These results emphasize that the method of aggregate formation affects ACA and that estimation of the percentage distribution of aggregates by HPLC may not reflect ACA. In analysing IVIG product for Clq binding activity test with the ELISA, the result by using Protein A-HRP correlated with aggregate percentage (r=0.84). But the correlation decreased (r=0.48) when the result used Protein A-AP(having poorer sensitivity than HRP) was compared with aggregate percentage. As a result, some variation between the two methods, due to differences in assay principles, is to be expected. However ELISA technique has the advantage in that it is easier to perform, more precise and less subject to reagent variability and is the more suitable screening method than HPLC analysis.

The clotting assay was replaced by the chromogenic substrate assay which is recommended by the European Pharmacopoeia (EP) and the Scientific and Standardization Committee of the International Society on Thrombosis and Haemostasis based on the reliability convenience and simplicity of the chromogenic assay: A correlation study was carried out with a one-stage factor VIII : C clotting assay and the perfonnance of the chromogenic assay was evaluated using two test kits that fulfilled the requirements of EP for factor VIII concentrates test. Although chromogenic assay has partly differences in measurement principle and standardization, this assay has a high correlation with clotting assay in various types of factor VIII concentrates and factor Vlll standard. We conclude that the chromogenic assay for factor VIII : C con- centrates correlates well with the clotting assay and shows good analytical performance.

AICD(후천성면역결핍증:acquired immunodefiency syndrome)는 사람면역부전바이러스(HIV)의 만성지속성감염에 의하여 일어나는 면역결핍상태를 중심으로 전신에 퍼지는 기회감염증, 악성종양, 정신신경계장해를 수반하며, 일단 감염이 된 후에는 효과적인 치료제도 거의 없을 뿐 아니라 결국에는 죽음에 이르게 하는 질환으로 그의 발증 예방, 치료의 어려움과 높은 발증률, 사망률등으로 세계 공통의 문제로서 지대한 관심사가 되고 있다. 따라서 HIV에 감염되지 않도록 예방을 최우선적으로 해야 하며 또한 일단 감염된 사람으로부터 더 이상의 전파를 방지하기 위해서는 HIV의 감염여부의 조기진단이 필수적이라고 하겠다. 현재 HIV에 감염된 사람을 검사하는 일상진단법으로 viral epitope에 대한 항체검사방법이 있으나 1) HIV에 감염되어 있으나 그 혈액 내에 항체가 형성되지 않는 window period가 있는 점. 2) 태반을 경유한 모체로부터 이행한 IgG항체가 생후 15개월까지 소아의 혈중에 머무르고 있으므로 HIV항체를, 감염의 유무를 나타내는 신뢰성 있는 지표로서 사용할 수 없는 점. 3) 신생아, 유아로부터 채취할 수 있는 혈액량은 대부분의 경우 극히 제한되어 있다는 점. 등으로 완전한 검사방법이라고는 말할 수 없다.
국내 유통 소금 중의 포타슘시아니이드와 페로시아나이드염의 분석에 관한 연구

본 연구는 국내 유통 소금중의 안전성을 규명하기 위하여 천일염 54건, 재제염 보건, 가공염 33건 등 총 143건에 대하여 숲행하였다. HPLC, ICP, Mercury Analyzer 등을 이용하여 KCN, 페로시아나이드 염, 납 등 6개 성분의 항량을 측정하였으며 그 결과는 다음과 같다. [rdn-max(mean) ; m곱3 ; f(CN : ND-ND(ND), 페로시아나이드염 : ND-ND(ND), Pb . ND-1.30 (0.331), Cd :ND-0.23(0.070), Hg ND-0.35(0,035), As :ND-1.52fo.062). 또한, 신속하고도 효율적이 면서 기존의 환경부 시안분석법의 단점을 보완할 수 있는 KCN과 페로시아나이드염의 분석 조건을 계시하였으며, 천일염, 재제염, 가공염 뿐만아니라 소금이 첨가된 식품까지도 적용하기 위한 기초 자료로 활용이 가능할 것으로 사료된다. This study was conducted to confirm the safety in a total of 143 samples of salts. The contents of potassium cyanide(KCN), potassium ferrocyanide(K_(4)Fe(CN)_(6)), lead(Pb), cadmium(Cd), mercury(Hg), and arsenic(As) were determined using a high performance liquid chromatograph(HPLC), inductively coupled plasma spectrometer(ICP), and mercury analyzer. We have measured CD4+T lymposite , β²-MG) and p24 antigen level in 188 HIV patients Estimated duration presumptive HIV inpection last tested CD4+T lymphocyte counting was 55.2±23 months in group with Zidovudine (Acidothymidine, AZT) treatment(Tx) and 38.4±20 months in group without AZT. There was no CD4+T lymphocyte of less than 400/mm³in 56 normal persons but 46% (86) were CD4+T lymphocyte of less than 400/mm³in 188 HIV patients. Annual decrase of CD4+T lymphocyte counts was 70/mm³in group without AZT tx and 38/mm³in group with AZT tx months after tx start. In the distribution of serum β₂-MG level in 182 HIV patients and 48 normal control, 90% (43) of control were below 2.0 mg/L and 76%(139) of HIV patients were over 2.0mg/L (p=0). β₂-MG level was negatively correlated with CD4+T lymphocyte (r=-0.394,<0.001). HIV p24 core antigen was detected in 26 (14.9%) out of 174 HIV/AIDS patients. Of the 26 patients, 21(81%) were CD4+T lymphocyte of less than 400/mm³and 46% (12) of less than 200/mm³. In was confirmed that 3 tests mentioned above are useful markers for prognosis of HIV infected persons.

본 연구는 패류중 중금속함량 측정을 위한 분석효율이 높고 간편한 분해법을 찾아내어 그 방법에 의끓 유종중인 패류중 중금속함량을 파악하고자 수행하였다. 시료를 분해시키는 Microwave와 습식, 건식회법을 비교한 결과 변형습식회화르가 가장 적합하였다. 변형습식회차딘는 려린0,3+C104+H2SOf의 혼찰산을 쪼 · j 1 (v/v)의 비율로 혼합하여 사용하였 을 때 회수을 Pb린틴. Cd 97.8, Cu 94.2, Zn 92.8%를 나타내었근 분해시간은 2~2.5시간 소요되었다. 유통중인 패류중 중금속함량 」최소~최대(평균), mgAgl 은 Pb 0.028~o.528(0.153), Cd 0.OO7~1,832(0.587), Cu 0.360~37.375(4.6691, Zn 6.317~233.2iO(38.881), Hg 0.001 ~O.151(0.012)mg/ltg으로 나타났다. 또한 유통중인 5종의 패류중 중금속평균항량이 생산지 하재지역에서 구입한 패류에 비해 모든 중금속언써 약간 높은 함량을 보였으나 오염된 수준이 아닌 자연함유량수준이었다. This study was conducted to improve the digestion method for the determination of heavy metals in shellfishes and to estimate the contents of heavy metal in commercial shellfishes by their digestion method. This digestion method was compared microwave with wet and dry ash digestion for shellfish samples, obtained results showed that the modified wet ash Ⅱ digestion was convenient and easy to use. For the modified wet ash Ⅱ digestion, The mixed acid of HNO₃+HClO₄+H₂SO₄(25:5:1, v/v) was used. Under this condition, Recoveries were Pb 94.3, Cd 97.8, Cu 94.2 and Zn 92.8% and digestion time was ∼2.5 hours. The contents of heavy metal [minimum∼maximum (mean), mg/kg in commercial shellfishes were as follows ; Pb:0.028∼0.528(0.153), Cd:0.007∼1.832(0.587), Cu:0.360∼37.375(4.669), Zn:6.317∼233.250(38.881), Hg:0.001∼0.151(0.013). The contents of heavy metal in commercial shellfishes of five species were slightly greater than those of shellfishes purchased at Hajae. There was no great difference between the contents of heavy metal of Hajae and those of shellfishes purchased at Whole market.
