      • Characterization of a Constitutive β-Lactamase from a Pseudomonas aeruginosa

        Objective : We analyzed some physicochermical chracteristics of a constitutive β-lactamase from a clinical isolate of a Pseudomonas aeruginosa which resistant to various β-lactams except imipenem and susceptible to aminoglycosides the proportion of its contribution to such resistance profile. Method : 1. Antimicrobial susceptibility test was done by the broth dilution method. 2. In hibition profile was done using EDTA, HgCl₂ and p-chloromercuribenzoate (pCMB). We also measured 50% inhibitory concentration(IC??) using β-lactamase inhibitors, such as clavulanic acid, sulbactam. 3. Kinetics study for substrate profile was performed by spectrophotometric assay using crude extracts and various antimicrobial agents. Relative hydropysis rates were determined. 4. The isoelectric point(pI) was determined by isoelectric focusing. The molecular weight was estimated by SDS-polycrylamide gel electrophoresis. Results : 1. The strain was resistant to ceftriaxone, sulperazon, piperacillin, ceftazidime, and aztreonam, while it was susceptible to gontamicin, tobramycin, amikacin and imipenem. 2. The β-lactamase was not inhibited by EDTA, and pCMB, whereas it was inhibited by HgCI₂. The IC?? of clavulanic acid was over 100μM and that of sulbactam was 22μM. 3. The relative maximal hydrolysis rate was calculated by setting V_(max) for cephaloridine at 100. The enzyme hydrolyzed cephalosporins more rapidly than penicillins. 4. The pI was 9.3 by isoelectric focusing. The molecular weight was estimated to be 40kDa. Conclusion : According to its substrate profile, pI and molecular weight, the β-lactamase from a clinical isolate of P. aeruginosa was a cephalosporinase belonging to group 1 in the Bush-Jacoby-Medeiros classification scheme.

        한국여성노인들의 노년기 적응과 노년기 삶의 모습에 대한 질적 연구

        The purpose of this study is to explore what makes the elderly women resilient in old age and how they reconstruct their later lives. This study used life histories extracted from tape-recorded interviews of 13 elderly women and analyzed key issues focused on how to adapt in old age. Main results are following: First, emerging themes to maintain life satisfaction in old age are related to both informants' inner and outer life worlds; subjective reinterpretation of life, feeling rich, family, social activities, overcoming social stigma. With supporting and supplementing one another, these five dimensions are important contributors to better later life. Second, where my informants search for meaning of life in old age is the critical clue of understanding the later life styles. Some of them are still searching meaning of life in roles as a 'mother', while others are finding their aense of worth outsild the family or in themselves. These results show that the elderly are interacting with themselves and their surroundings in adapting to old age and different life styles according to different sources of life satisfaction manifest the changes among the eldery and Korean society.

      • 석이로부터 분리한 페놀성 화합물의 phospholipase A_2 저해활성

        호알카리성 진균 Cephalosporium sp. RYM-202가 생산하는 alkaline xylanase (CX-III)의 작용에 의해 xylan 기질로부터 생성되는 주요 가수분해 산물은 xylobiose와 중합도가 4 이상인 xylooligosaccharides이었다. 이 효소는 xylobiose에 대한 분해능을 가지고 있지 않지만 xylotriose로부터는 xylobiose를, xylotetraose로부터는 xylobiose와 xylotriose를 주산물로 형성하였다. 이러한 결과들은 CX-III가 transglycosidase 활성을 소유하는 전형적인 endo-type xylanase임을 보여준다. N-bromosuccinimide에 의한 CX-III의 화학적 변화 실험 결과 효소 1분자 당 2개의 tryptophan 잔기가 활성에 관여하는 것으로 나타났다. 그러나 iodoacetamide 및 diethylpyrocarbonate에 의한 효소활성의 저해효과는 나타나지 않음으로써 이 효소의 활성부위에 cysteine과 histidine 잔기가 필수적이지 않음이 확인되었다. The hydrolysis products formed from birchwood xylan by the action of an alkaline xylanase (CX-III) from alkalophilic Cephalosporium sp. RYM-202 were xylobiose and xylooligosaccharides polymerized with more than 4 sugar molecules. This enzyme was not active on xylobiose but readily attacked xylotriose accumulating xylobiose as a major product. The predominant end-products from xylotetraose by CX-III were xylobiose and xylotriose. These results indicate that the enzyme is typically endo-type xylanase possessing transglycosidase activity. Chemaical modification of CX-III with N-bromosuccinimide revealed that two tryptophan residues per molecule of CX-III were essential for its catalytic activity on xylan. On the other hand, iodoacetamide and diethylpyrocarbonate did not influence the activity of the enzyme, suggesting that cysteine and histidine residues are not involved in the active site of this alkaline xylanase.

      • 다운증후군유아의 운동발달과 트레드밀트레이닝 및 걸음 보조기의 중재효

        Down Syndrome is a representative hereditary disability which accompanies mental retardation, communication difficulties and motor development delay. In particular, the motor development delay that begins in infancy brings forth negative effects in various developmental domains such as emotional, social and cognitive areas as well as interfering with various aspects in everyday life. Therefore many researchers have tried to minimize such development delay in Down Syndrome by developing various early intervention programs and shown continuous attempts to prove its positive effects. The treadmill or foot orthoses intervention methods introduced in this study are being applied in America or Europe to infants with Down Syndrome in order to minimize their motor development delay. It is reported that treadmill training helps infants with Down Syndrome in movements related to balance such as standing or walking, and foot orthoses improves balance in ankles and motor skills when walking, as well as increase steady posture. For future studies, there is a need for researchers to discover the cause of motor development delay and abnormal development patterns in infants with Down Syndrome and to study the effects of such motor development delay on other developmental areas. Also, currently in Korea there is relatively insufficient development and supply of early intervention programs for infants with disabilities as well as studies regarding its effects. Therefore we need to develop early intervention programs according to the characteristics of each disability and evaluate the effects through experience. For this, we need to make the parents of infants with disabilities aware of the need for early intervention programs related to physical activity and its effects. 다운증후군은 정신지체와 의사소통의 곤란, 그리고 운동발달 지연이 동반되는 대표적인 유전적 장애영역 중 하나이다. 특히, 영유아기에서 시작되는 운동발달 지연은 이후의 다양한 발달영역 즉, 심동적, 사회적, 인지적 영역에 부정적인 영향을 미쳐 일상생활을 비롯한 다양한 영역에서의 수행에 곤란을 초래하게 된다. 따라서 많은 연구자들은 이러한 다운증후군의 발달 지체를 최소화하기 위해 다양한 조기중재 프로그램을 개발하고 그 효과를 증명하는 시도를 진행해 왔다. 본 연구에서 소개하고 있는 트레드밀이나 걸음보조기 중재방법은 미국이나 유럽에서 다운증후군유아의 운동발달지체를 최소화하기 위해 유아기부터 적용되는 방법이다. 트레드밀 트레이닝은 다운증후군유아의 서기와 걷기 등 주로 균형 및 보행에 도움을 주는 것으로 나타났으며, 걸음보조기는 보행 시 발목의 균형과 운동 기능을 향상시키고, 자세의 안정성을 높이는 것으로 보고되고 있다. 향후 연구자들에게는 다운증후군유아의 운동발달 지연과 비정상적인 발달 패턴을 유발하는 요인이나 그러한 운동발달 지연이 다른 발달 영역에 미치는 영향에 대해 알아보는 좀 더 포괄적인 연구가 요구된다. 또한 국내에서는 현재 장애유아를 위한 조기 중재프로그램의 개발과 보급, 그리고 효과 규명에 상대적으로 소홀한 실정이다. 따라서 장애유형별 특징을 고려한 조기 중재프로그램을 개발하고, 효과를 경험적으로 검토해 보는 시도가 필요하다. 이를 위해, 장애유아의 부모들에게 신체활동 관련 조기 중재프로그램의 필요성과 효과에 대한 올바른 인식을 전파하는 작업이 요구된다고 하겠다.

      • 농흉과 종격동염이 합병된 viridans streptococcus에 의한 인후농양 1예

        최근 저자들은 viridans streptococcus에 의한 인후농약으로 인하여 발생한 종격동염 및 농흉을 보인 47세 여자 환자를 적절한 배농과 항생제로 치료한 국내 최초의 1예를 치험하였기에 문헌 고찰과 함께 보고하는 바이다. Since the antibiotic era, retropharyngeal abscess became a rare disease entity nowadays. But it may often progress to lethal complications involving vital structure if diagnosis or treatment is delayed. We experienced a case of empyema and mediastinitis complicating retropharyngeal abscess caused by viridans streptococcus in a 47-year-old female patient with diabetes mellitus. She was successfully treated with appropriate antibiotic therapy combined with percutaneous abscess drainage. To our knowledge, such a complication of retropharyngeal abscess has not been reported in Korea yet and we present this unusual case herein followed by a review of the related literature.
