      • 항암제 처리한 백혈병 세포주에서의 Apoptosis 발현

        The bcl-2 proto-oncogene encodes a 26 kD protein that promotes cell survival by blocking apoptosis. The bax protein is a member of the bcl-2 familly, now known to form heterodimers with the bcl-2 protein. The ratio of bax to bcl-2 is be critical in determining the fate of the cell in response to stimuli that can induce apoptosis. Extract of Pulsatilla Koreana (SB-31) showed promising antitumor activity in vitro with Topo I inhibitory action. In the present study, the relationship between apoptosis and the apoptosis related proteins, bcl-2 and bax were investigated in human leukemic cell lines HL-60, U-937 and CEM-CM3. All anticancer drugs(adriamycin, etoposide, camptothecin, SB-31) induced extensive apoptosis in HL-60, U-937 cells and CEM-CM3 cells. The expression of bcl-2 and bax protein were determined in cell lines by western blotting before and after incubation with anticancer drugs at different time points. 1) In HL-60 or U-937 cell lines, down regulation of bcl-2 and up-regulation of bax were found after incubation with ADR, VP-16 or camptothecin. 2) In HL-60 or U-937 cell lines, no significant change in bcl-2 or bax protein expression resulted after incubation with SB-31. 3) In CEM-CM3 cells, virtually no change was noted in bcl-2/bax expression after incubation with ADR, VP-16, camptothecin or SB-31. It is suggested that different leukemic cell lines use different pathways of apoptosis activation and a given cell may utilize different pathways of apoptosis activation in response to different cytotoxic agents.

      • 냉동 제대혈 세포의 체외 증폭

        Background : Cord blood(CB), which has no HLA restriction, is an alternative to bone marrow for hematopoietic stem cell transplantation. The use of cord blood, however, is limited by the number of progenitor/stem cells necessary to reconstitute the older child or adult. Therefore, ex vivo expansion of CB could have tremendous impact on diverse clinical settings. We studied the ex vivo expansion of isolated population of CD34_(+) cells from cryopreserved CB cells. Methods : CD34 cells were isolated from cryopreserved CB mononuclear cells. Purified cells were cultured with various combinations of hematopoietic growth factors including erythropoietin(EPO), stem cell factor(SCF), granulocyte-colony-stimulating factor(G-CSF), gra-nulocyte, macrophage-colony-stimulating factor(GM-CSF), interleukin-1β(IL-1β), 1L-3, and IL-6. After 7, 10 or 14 days of culture, the fold increases of colony-forming unit- granu-locyte, macrophage(CFU-GM), burst-forming unit-erythroid(BFU-E), colony-forming unit-mix (CFU-Mix), and high proliferative potential colony-forming cell(HPP-CFC) were evaluated. Results : Ten-day culture with the combination of EPO, SCF, G-CSF, IL-1β, and IL-3 resulted in a median of 60-fold increase of CFU-GM, which was greater than those with the combinations of less than 5 growth factors. The addition of IL-6 or GM-CSF to this combination did not enhance CFU-GM expansion. Ten-day culture was significantly superior to 7-day culture for CFU-GM expansion. Prolongation of culture to 14 days, however, revealed decreased expansion of CFU-GM compared to 10 days. BFU-E and CFU-Mix were expanded to 2~5 folds in 7-day culture with the combination of EPO, SCF, and G-CSF. Further expansion was not achieved in 10-day culture and colonies disappeared in 14-day culture. HPP-CFC was expanded to a median of 7.5 folds in 7-day culture with the combination of EPO, SCF, G-CSF, IL-1β, IL-3, and IL-6. Neither 10-day or 14 day-culture enhanced expansion of HPP-CFU. Conclusion : Cryopreserved cord blood cells maintain ex vivo expansion potential. In our system, 10-day culture with the combination consisting of EPO, SCF, G-CSF, IL-1β, and IL-3 seems to be adequate for hematopoietic progenitor/stem cell expansion from cryopreserved cord blood cells.

      • Application of Artificial Insemination Technology for Dairy Breeding in Mongolia

        This study was focused on improvement of milk production in Mongolia dairy industry by artificial insemination (AI) technology, of which was supported from ODA project of KOICA in Republic of Korea. The study was started from January 2009 to present and 3rd years in this year. So, all data, especially synchronization and pregnancy of dairy cows (Holstein) will be summarized in final result in this year. For synchronization, total 81 dairy cows selected from 4 private farms that were 38, 30, 8 and 5 in Undarmal milk, Onjin (Enkhbayer), Jargalant, and BRM School, respectively. All the dairy cows were injected intramuscular (IM) of 5 ml PGF2α in the vulva and detected estrus 2 to 3 days after PGF2α injection. Total 78 out of 81 dairy cows (96.3%) were detected estrus by only 1 time injection of PGF2α. The dairy cows that were induced estrus, inseminated with 0.5 ml dairy frozen semen by conventional AI techniques. The pregnancy diagnosis of the AI dairy cows was detected by uterus palpation after 60 days of insemination. Total 75 from 78 inseminated dairy cows (90.1%) were diagnosis pregnant. The estrus induction and pregnant rate were very effective using PGF2α injection and conventional AI techniques in Mongolia dairy cow. The results indicated that AI after estrus induction in Mongolia dairy cows could be applied to dairy breeding technology to improve the breeding efficiency and milk production.

      • 민속놀이의 활성화 방안 연구

        As an effort to revive our folk tradition, I am suggesting that our folk plays, especially those with a competitive nature, be turned into a sporting event which youths can enjoy, and which can be taught in every school. This will enable our students to gain a better understanding of our traditions and come to value and love them. Above all, an organization should be created that will play a central role in implementing this agenda. Second, this organization should lead in turning various folk plays into sporting events. Third, those events should be exciting competitions that will gain peoples attention. Fourth, to promote the spread of folk plays, they should be included in school curricula and trained physical educators should help spread these competitions in the schools. In addition, these events should be actively publicized through the press and other mass media. Lastly, this organization ought to take the lead in seeking assistance from the Office of Education to hold a variety of these sporting events. If these projects are carried out successfully, we will have a future in which folk plays are always with us in everyday life.

      • 코크스 오븐 가스에 의한 중독사 1례

        코크스 오븐 가스(coke oven gas, COG)는 제철공장에서 흔히 취급하는 물질로 200여 성분을 함유하고 있다. 그 중 인체에 심각한 급성 중독을 초래할 수 있는 물질로는 일산화탄소, 청산 및 황화수소이다. 본 증례는 코크스 오븐 가스를 탈류 세정하는 작업장에서 보호구를 착용하지 않은 채 탈류 세정탑의 찌꺼기를 제거하던 중 이에 잔존하는 유해성 가스에 중독 되어 사망한 경우로, 황화수소가 가장 유력한 원인 성분으로 간주되었다. 본 증례의 경우로 보아 첫째, 코크스 오븐 가스를 처리하는 곳에서 황화수소 등의 각종 유해 물질의 폭로에 대하여 충분한 대책이 이루어져야 하고, 둘째, 부검시 혈액은 항응고제가 투여된 용기에 채취되어야 하며 냉장 보관되는 상태로 운송되어 가급적 빠른 시간 내에 검사되어야 한다고 생각한다. Coke oven gas is a main gas managed in steel plants, which contains 200 composites. Carbon monoxide, hydrocyanide, and hydrogen sulfide of them are serious gas incurring acute intoxication in human. Steel plants have considered of adverse effects of carbon monoxide and hydrocyanide, but because occupational exposure of hydrogen sulfide is uncommon, the measure against hydrogen sulfide is not enough. This case is that a male worker, during removing sludge packed in a tower purifying coke oven gas without a protective mask, expired from acute intoxication of remained toxication of remained toxic gas and that hydrogen sulfide intoxication is probably the cause of death. According to this case, it is thought that first, a complete measure for exposure of hydrogen sulfide is necessary in the plants managing coke oven gas, and second, the blood should be sampled with an anti-coagulinated container, delivered in the cool condition, and analyzed immediately.

      • 음향 스펙트럼을 이용한 기계식 판막의 혈전 현상 검출에 관한 연구 -In-Virto 및 In-Vivo 실험

        A Mechanical valve is one of the most widely used implantable artificial organs and its reliability is so important that is failure means the death of the patient. Thrombosis si the most common cause of mechanical valve failure, therefore, early noninvasive detection of mechanical valve thrombosis is essential. The objective of this research was to detect thrombosis formation by using spectiral analysis an a neural network. Using a microphone and an amplifier, we measured the soud form a mechanial valve which was attached to a pneumatic ventricular assist device. The should was sampled by an A/D converter(DaqBook 100), and the periodogram was the amin algorithm for obtaining the spectrum. We made the thrombosis models by uisng pellethance and silicon on thte valvular disk around the sewing ring and and on fibrous tissue growth across the orifice of the valve. The performance of the measurement system was tested by sung 1-㎾ sinusoidal wave. The measurement system properly detected 1-㎑ spectrum, as expected. The spectra of a nomal valve and 5 kinds of thrombotic valves were obtained, and primary and secondary peaks appeared in each spectral waveform. We found tat the secondary peak changed according to the thrombosis model. Thus, to distinguish the secondary peak of normal valve and form that of thrombotic valve quantatively, we employed a 3-layers, and 1 output. This trained neural network could distinguish betwen a normal and a thrombotic valve with more than 90% probability. In conclusion, noninvasive monitoring of an implanted mechanical valve is possible by analyzing the acoustical spectrum through the use a neural network algorithm. This method will be applied to the performance evaluation of other implantalbe artificial organs.

        의원성 사고로 발생한 타액선염 : 증례보고

        A case of sialadenitis is presented in a patient with painful swelling of the right mouth floor. The condition was caused by trauma on the right mouth floor during dental treatment, which had happened 15 days before admission. On aspiration, mucous secretion was found and ultrasonography showed obstruction of duct. Histopathological studies and surgical investigation established a definite diagnosis of obstructive sialadenitis caused by ductal laceration. As surgical treatment sialodochoplasty was selected. The case and relevant considerations are discussed.

        방사선사진에서의 골질과 임상적으로 평가한 골질 분류의 상관관계

        Purpose : To investigate the validity of digital image processing on panoramic radiographs in estimating bone quality before endosseous dental implant installation by correlating bone quality in radiographic images with clinical bone quality classification. Materials and Methods : An experienced surgeon assessed and classified bone quality for implant sites with tactile sensation at the time of implant placement. Including fractal dimension eighteen morphologic features of trabecular pattern were examined in each anatomical sites on panoramic radiographs. Finally bone quality of 67 implant sites were evaluated in 42 patients. Results : Pearson correlation analysis showed that three morphologic parameters had weak linear negative correlation with clinical bone quality classification showing correlation coefficients of -0.276, -0.280, and -0.289, respectively (p<0.05). And other three morphologic parameters had obvious linear negative correlation with clinical bone quality classification showing correlation coefficients of -0.346, -0.488, and -0.343 respectively (p<0.05). Fractal dimension also had a linear correlation with clinical bone quality classification with correlation coefficients -0.506 significantly (p<0.05). Conclusion : This study suggests that fractal and morphometric analysis using digital panoramic radiographs can be used to evaluate bone quality for implant recipient sites.       • 일부 자동차 공장 근로자들의 職業的 特性과 社會 心理的 Stress의 相關性 硏究

        〈Objective〉 This study were carried out to study to assess the relationship between job characteristics, general characteristics, health behaviors, and psychosocial distress and suggested the preventive oriental medicine approaches in occupational health area. 〈Methods〉 We measured job characteristics(job demand, job control, jop insecurity) by The Karasek's Job Content Questionnaire, health behaviors(smoking, drinking, exercise, coffee), general characteristics at work by a self-administered questionnaire. Psycho social distress was measured by PWI(Psychosocial Well-being Index), a 18-item self- administered instrument. 370 data for research were collected through the process of oriental health examination with questionnaire in workplace. Statistical analysis was done by t test, oneway ANOVA, and hierarchical multiple regression analysis with SPSSWIN (version 10.0) 〈Results〉 Among the 152 respondents, the prevalence of high job strain was 27%(41 persons). The Odds ratio of high job strain was 3.11(95 2.12~4.11), and those of passive group and active group were 2.01(95 0.83~2.76) and 1.80(95 0.83~2.76), respectively. Our results supported the association between psychosocial distress and job strain. 〈Conclusion〉 The odds ratio of high strain isolated group(high strain group+low social support) was 3.59(95 2.05 5.13), but the odds ratio of high strain collective group was 1.32(95 0.23 2.41). Social support at work was modifing factor of the association between psycho social distress and job strain. Therefore we reviewed the evidence that economic depression was associated with psycho social distress in this study, and that the strength of the social support was important in maintain health.

        상부위장관 출혈 원인의 변화 양상

        Upper gastrointestinal(UGI) bleeding was critical disease that was commonly found in emergency department and needed more early diagnosis and rapid treatment for decreasing mortality and morbidity. It's causes and frequencies were affected by many factors, but the majorities were caused by peptic ulcers and esophageal-gastric varices. With development of economy and medical services, new drugs and endoscopic interventions, the treatment of patients with UGI bleeding was more improved. So I would like to evaluate the change of causes and frequencies of UGI bleeding and the associated clinical findings. A total of 1,546 patients presented with UGI bleeding in emergency department of Yeungnam university hospital during the five years from Jan. 1991 to Dec. 1995 were clinically reviewed by chart and compared with previous seven years report from Jan. 1984 to Dec. 1990 in the same hospital. The ratio of male to female was 6.1:1 and slightly increased in comparison with previous seven years. The incidences in 6th and 5th decades were highest(54.4%)as like as previous seven years. Age distribution of duodenal ulcer bleeding, Mallory-Weiss tear and acute gastric mucosal lesion were developed in lower decades than the others. The causes of UGI bleeding were caused by esophageal varix(45.4%), peptic ulcer(38.8%), Mallory-Weiss tear(4.3%), gastric cancer(3.8%), other(3.1%), acute gastric mucosal lesion(2.7%), undetermined causes(1.8%) in descending order of frequencies. The proportion of esophageal varices was increased from 36.6% to 45.4%, but that of peptic ulcer was relatively decreased from 47.6% to 38.8%. And the annual distribution of varices was increased, however the annual distribution of peptic ulcers was increased. Although the ranking of annual distribution of gastric ulcers and duodenal ulcers were not changed compared to previous study and the frequency of gastric ulcers was increased since 1992, but additional follow-up was needed. Seasonal variations were found that upper gastrointestinal bleeding was increased in fall and peptic ulcer bleeding was increased significantly in summer and fall, however, varix bleeding was less frequent in summer time as like as previous seven years. Emergency endoscopic examination was performed within 6 hours for 54.5%, 24 hours for 90.8% and this results were to be like previous results, 57.5% and 90.7 respectively. Findings of emergency endoscopic examination were active bleeding focus(21.2%), blood clot(7.9%), exposed vessels(13.3%), flat blood spot(6.4%) and lesion without evidence of bleeding(51.3%). Endoscopic findings of active bleeding were easily seen when examination was perfomed within 24 hours. Blood transfusion was performed in 67.3% with average 2.1 units. When the causes of bleeding were due to varix and marginal ulcer, more amounts of blood were transfused than the others. In previous seven years, blood transfusions were performed in 71.2% with average 5 units and more amounts of blood were transfused in gastric cancer and varix bleeding.
