      • 효모운반체 내에 β-glucanase 유전자의 클론닝 및 Escherichia coli에서의 발현

        B. subtilis유래의 β-glucanase 유전자가 클로닝된 pBGA3 재조합체에 있는 효소유전자를 효모클로닝 운반체인 YEp36과 pKAC 2에 재클로닝하여 E.coli HB101에 형질전환 후에 발현 및 체외분비를 연구했다. YEp36의 SphI효소 절단 부위에 β-glucanase 유전자를 재클로닝을 하였을 때는 효모 유래 유전자인 LEU2, 2 플라스미드 DNA등이 삭제된 pBAG4와 LEU2, 2 플라스미드DNA와 구리 저항성 유전자(CUP1)가 모두 삭제된 재조합 플라스미드 pBAG5를 만들었으며, 두 클론은 유전자가 발현되었고, 체외로 분비되는 것을 관찰했다. 또한 CUP1유전자가 들어있는 pKAC2 운반체를 제조하여 BamHI부위에 β-glucanase 유전자를 재클로닝하여 pBAGC를 만들었다. 상기의 운반체에 클로닝된 β-glucanase 유전자를 대장균에서 형질전환시킨 결과는 대장균에서 형질발현이 되어 β-glucanase 활성을 나타냈고, 체외로 분비되는 것을 확인했다. The β-glucanase gene clone(pBAG3) derived from Bacillus subtilis was subcloned in YEp36 and pKAC 2 vectors, and then transformed and expressed in E.coli HB101 strain. The β-glucanase gene was recloned in the SPhI site of the YEp36, which resulted in pBAG4 and pBAG5 recombinant plasmids that expressed and extracellulary secreted the enzyme. Also the gene was recloned in the BamHI site of the pKAC2 which was newly constructed, and the new recombinant plasmid named pBAGC and then it was transformed in E.coli HB101, which expressed and extracellularly secreted the enzyme.

        Discordance in Spirometric Interpretations Based on Korean and Non-Korean Reference Equations

        Objectives: Korean regression models for spirometric reference values are different from those of other ethnic groups. The purpose of this study was to evaluate discordance in spirometric interpretations based on using Korean reference equations versus non-Korean reference equations. Methods: Spirometry was performed on 825 Korean male workers from April 2009 to November 2011. The spirometric patterns and disease severity were evaluated using two Korean equations (Choi’s and Lee’s) and three equations for Caucasians (NHANES III, Morris’s, and Knudson’s), and the results of Choi’s equation were compared with the non-Korean equations. The spirometric patterns were defined as normal, restrictive, and mild and moderate obstructive. Results: The mean differences in the FEV1% and FVC% between the two Korean equations were 2.0 ± 1.3% and 3.5 ± 2.2%, respectively. Morris’s equation had the greatest difference in the FEV1% from Choi’s equation: 32.9 ± 8.5%. Knudson’s equation had the greatest difference in the FVC% from Choi’s equation: 10.5 ± 6.5%. Conclusions: The two Korean equations shared similar characteristics of spirometric interpretation. The spirometric interpretations of Choi’s equation were significantly different from those of Morris’s equation and Knudson’s equation.

        림프종 환자에서의 갈륨, TI-201 그리고 Tc-99m MIBI 섭취의 비교

        Purpose; Ga-67 scintigraphy has been used for the evaluation of tumors, especially lymphoma. Recently, Tl-201 and Tc-99m MIBI were also used to tumor imaging. Tl-201 and Tc-99m MIBI had better physiologic characteristics than Ga-67, so we studied 32 biopsy proven lymphoma patients (male 24, female 8, mean age 46 years) with Ga-67, Tl-201 or Tc-99m MIBI and compared the scan findings. Materials and Methods: Twenty-three of 32 patients were injected 74-111 MBq (2-3 mCi) of Tl-201, before chemotherapy and imaged with dual-headed SPECT (Prism 2000, Picker, USA) at 30 minutes after injection. Delayed images were obtained after 3 hr in 8 patients. Twenty seven of 32 patients were injected 740 MBq (20 mCi) of Tc-99m MIBI and imaged at 30 minutes after injection. 111-185 MBq (3-5 mCi) of Ga-67 was injected in 12 patients and imaged at 48 and 72 hours after injection. Twenty eight patients were diagnosed as non-Hodgkin's lymphoma and others were Hodgkin's lymphoma. Results: Twenty patients were positive on Tl-201 scan and 3 patients showed negative findings. One of these 3 patients, Tc-99m MIBI and Ga-67 scan were positive. Twenty two patients were positive on Tc-99m MIBI scan and 5 patients showed negative findings. One of these 5 patients, Tl-201 was positive and 2 were positive on Ga-67 scan. Ten of 12 patients showed positive findings on Ga-67 scan. The sensitivity of these agents were 83.3%, 87.0% and 81.5% for Ga-67, Tl-201 and Tc-99m MIBI, respectively. The sensitivity was highest in Tl-201 scan, but there were no significant differences among three tests. In this study, there was no significant difference of uptake ratios between early and delayed images of Tl-201. Conclusion: Scintigraphy with Tl-201 and Tc-99m MIBI in lymphoma patients have similar sensitivity with Ga-67.

      • Bacillus subtilis β-glucanase유전자의 효모세포에서의 발현

        The clone pABC1 inserted the β-glucanase gene of Bacillus subtilis was transformed and expressed in a polyploid yeast cell, S. uvarum OE2. The yeast/pABC1 produced the enzyme intracellularly, however not extracellularly. When the S. uvarum OE2 was disrupted, the cell extract showed β-glucanase activity which meant the expression of β-glucanase gene in S. uvarum OE2. To make a β-glucanase extracellular secretion in S. uvarum OE2, the promoter of β-glucanase region derived from B. subtilis in the pABC1 was substituted with the promoter of the alcohol dehydrogenase 1 gene in the pAMC1 vector originated from S. cerevisiae and named pABC1. The S. uvarum OE2 transformed with pABC1 extracellularly secreted the β-glucanase.

      • 흰쥐 내측 전정신경핵 뉴론의 자발적 활동성의 발생에 관여하는 이온전류에 관한 연구

        Medial vestibular nucleus (MVN) neurons are found to have spontaneous electrical activity in the absence of any detectable synaptic input. To investigate the contributions of intrinsic mechanisms to the spontaneous activity of MVN neurons, we examined the effects of various channel blockers on spontaneous firing by means of patch clamp recordings. Coronal slice (400 ㎛) of the vestibular nucleus region was sequentially treated with pronase 0.2 ㎎/㎖ and thermolysin 0.2 ㎎/㎖, then single neurons were mechanically dissociated. Almost all the MVN neurons recorded in neonatal rat were shown to have spontaneous resting activity. In 300 nM TTX, spontaneous firing was blocked in all cells tested. In 8 of 11 cells, underlying fluctuation or oscillations in membrane potential was not remained, and hyperpolarization did not produce rebound low-threshold calcium spikes. Although MVN neurons possessed hyperpolarization activated cation current (Ih), cesium had no effect on firing rates. The spike afterhyperpolarization (AHP) is calcium dependent. When Ca^2+ influx was blocked in external Ca^2+ free solution, repetitive firing was abolished and the cell rested at depolarized membrane potentials. Application of apamin (300 nM) caused a profound reduction in the amplitude of the AHP and produced rhythmic burst firing. These findings suggest that the spontaneous activities of MVN neurons is regulated by interactions between the membrane depolarization mainly due to a persistent sodium conductances and hyperpolarization due to the calcium-activated potassium conductances.

        STS 304 강의 저주기 및 고주기 피로에 있어 초기 마르텐사이트의 영향

        Zero to tension fatigue tests and strain controlled fatigue tests were carried out to find how initial strain induced martensite, α' affects low and high cycle fatigue behavior and fatigue crack growth mechanisms. Microscopic study and phase analysis were carried out with TEM, SEM, EDAX, Optical Microscope, Ferriscope, and X-ray diffractometry. The amount of Initial α' was controlled from 0% to 33% by controlling the temperatures for cold working and heat treatment. Lower contents of initial α' showed higher fatigue resistance in low cycle fatigue but lower fatigue resistance in high cycle fatigue because it is ascribed to the more transformation of α' martensite during low cycle fatigue and higher ductility. In high cycle fatigue, fatigue life is attributed to the strength and phase transformation of austenite into α' during fatigue was negligible. γ boundary, γ/twin boundary, and γ/α' boundary were found to be the preferred site of fatigue crack initiation.

      • Polymer Light Emitting Diode의 발광특성 연구 : A study on electoluminescence properties of PLED with poly [2-(2'-ethylhexyloxy)-5-methoxy-1,4-phenylenevinylene]

        In order to know how the [2-(2'-ethylhexyloxy)-5-methoxy-1,4-phenylenevinylene] (MEH-PPV) layer works in organic light emitting diode, two different types of devices were fabricated, which are MEH-PPV single layer devices and MEH-PPV/tris (8-hydroxy-quinoline)aluminium(Alq3) bilayer devices. On the indium-tin-oxide(ITO) glass anode MEH-PPV layer was first spin-coated, and the Alq3 layer and the aluminium(Al) anode were vapor-deposited sequentially. The devices shows a trend that has lower current, luminous efficiency in case the thickness of the device is larger. Some bilayer devices, however, show more improved performance than other devices of smaller total thickness. This results were discussed on the basis of improved exciton formation resulting from balanced carrier transport.
