        한국인 주요우울장애 환자에서 혈중 ACTH, Cortisol 농도와 해밀턴 우울 평가 척도의 신체증상 항목과의 상관관계

        Objectives:Hypothalamic-Pituitary-Adrenocortical(HPA) system dysregulation plays an important role in the pathophysiology of major depressive disorder. Medically unexplained somatic symptoms comprised the predominant complaints of korean patients with major depressive disorder. It might be related to dysregulation of HPA system and somatic symptoms. The aim of this study was to investigate the association between Hamilton Depression Rating Scale severity and the Plasma Adrenocorticotropic Hormone(ACTH) & Serum Cortisol level in Korean patients with major depressive disorder. Methods:Our study design was prospective. A total 111 hospitalized patients with major depressive disorder (DSM-IV) at Anam Hospital, Korea University Medical Center between Jan 2009 and May 2011 were selected. Hamilton Depression Rating Scale was measured by psychiatrist. Plasma ACTH, Serum Cortisol levels were analyzed at the time of admission. Results:There are significant correlations(p<0.05) between Hamilton Depression Rating Scale of somatic component(HAM-D 11-15 item) and Plasma ACTH and Serum Cortisol level. However, other HAM-D sub items were not significantly correlated with Plasma ACTH and Serum Cortisol level except somatic component. Conclusion:Our Study results suggest that hyperactivity of HPA axis might be correlation with somatic symptoms in korean patients with major depressive disorder.

        Uncoupling Protein 3의 골격근 세포내 과발현이 OLETF 백서 및 배양된 골격근 세포에서 포도당대사에 미치는 영향

        연구배경:Uncoupling protein(UCP)는 미토콘드리아의 내막에 위치하는 단백질로 세포내의 과다한 에너지를 열로 발산시키는 기능을 가진다. 최근 동물의 갈색지방조직에만 존재하는 UCP와 유사성을 가진 아형들(UCP2,3)이 사람에게도 존재함이 알려져 큰 관심을 끌도 있는데 이중 UCP3는 그 발현이 골격근세포와 갈색지방조직에만 국한된다. 본 연구에서는 UCP3가 체내 인슐린 감수성을 결정하는데 가장 중요한 조직인 골격근에 국한되어 발현되는 점에 착안하여 UCP3를 골격근세포에 과발현시켰을 때 포도당 대사에 어떠한 영향이 나타나는 지를 조사하였다. 방법:25주령의 8마리의 OLETF 백서를 대상으로 하여 4마리는 골격근에 adenovirus 2mL(1×10¹²pfu/mL)를 주사하여 대조군으로 하였고 4마리는 골격근에 재조합법으로 제작된 adenovirus­UCP3 2mL(1×10¹²pfu/mL)를 주사하였다(UCP3 과발현군). UCP3를 투여한 백서에서 먹이섭취가 증가하는 경향이 있어 그 전날 대조군이 먹은 야의 먹이만큼 투여하였다. 골격근에 adenovirus를 주사한 10일 후에 euglycemic hyperinsulinemic clamp를 시행하였다. Adenovirus­UCP를 C2C12 골격근 세포에 transfection시켜 UCP3를 C2C12 골격근 세포에 transfection시켜 UPS3­C2C12를 만들고 C2C12 골격근 세포와 UPS3­C2C12 골격근 세포에서 포도당 수송 및 당원합성을 측정하였다. 결과:UCP3 과발현 OLETF에서 체중이 감소하는 경향을 보였고 인슐린 감수성이 증가하였다. C2C12세포에서 기저상태 포도당 수송은 1.28±0.17μmol/L/min였고 100nM 인슐린으로 2시간 처리한 후 2.67±0.20 μmol/L/min로 증가하였다. UCP3­C2C12 세포에서는 기저상태 포도당 수송이 3.98±0.13μmol/L/min로 증가되었고 인슐린 처리 후 5.74±0.44μmol/L/min로 증가하였다. 인슐린을 처리한 UCP3­C2C12 세포에 P13K 억제제인 wortmannin을 첨가하였을 때 포도당 수송활성이 3.81±0.20μmol/L/min로 감소하였다. 기저상태 당원합성은 C2C12 세포에서 0.25±0.01μmol/L/min였고 인슐린 처리 후 0.45±0.01μmol/L/min로 증가하였다. UCP3­C2C12 세포에서는 기저상태 당원합성이 0.62±0.01μmol/L/min였고 인슐린 처리 후 1.26±454μmol/L/min로 증가하였다. UCP3­C2C12세포에 wortmannin을 첨가하였을 때 당원합성율이 0.80±0.04μmol/L/min로 감소하였다. 결론:UCP3 과발현이 OLETF 백서에서 인슐린 감수성을 증가시켰고 골격근세포에서 포도당 수송 및 당원합성을 증가시켰다. wortmannin을 첨가하였을 때 포도당 수송 및 당원합성이 감소함으로 보아 이 과정이 인슐린 신호전달체계인 P13K에 일부 의존함을 알 수 있었다. Background : UC P3 is a mitochondrial membrane protein expressed selectively in the skeletal muscle and brown adipose tissue. Since the skeletal muscle is the main organ determining insulin sensitivity in the body, it was hypothesized that UCP3 overexpression in skeletal muscle cells would improve glucose metabolism. Methods : An adenovirus-UCP3 was produced by a recombinant DNA method. OLETF rats were divided into 2 groups. Four rats were injected with the adenovirus-UCP3 (UCP3 group) and others were injected with the adenovirus(control group) in the skeletal muscle. The UCP3 group was provided with the same quantity of food as that consumed by the control group on the previous day. Insulin sensitivity was evaluated by the euglycemic hyperinsulinemic clamp method. In a separate experiment, glucose transport and glycogen synthesis we evaluated in C2C212 cells transfected with ether an adenovirus or the adenovirus-UCP3. Results : The insulin sensitivity improved significantly and the body weight decreased in the UCP3 group. The glucose transport and glycogen synthesis were higher in the UCP3-C2C12 skeletal muscle cells at the basal state. After insulin treatment, glucose transport and glycogen synthesis were also higher in the UCP3-C2C12 cells but the increments were reduced after treatment with wortmannin, a PI3K inhibitor. Conclusion : Insulin sensitivity was higher in the UCP3-overexpressed OLETF rats in the in vivo study. UCP3 transfection also increased glucose transport and glycogen synthesis in the cultured skeletal muscle cells by a PI3K dependent mechanism(J Kor Diabetes Asso 25 :460~468, 2001).

      • 달걀버섯(Amanita hemibapha subsp. hemibapha DGUM 28001)의 균사체 생육을 위한 배양조건의 최적화

        In many recipes, Amanita hemibapha subsp, hemibapha has been regarded and used as an excellent edible mushroom. However, so far, no artificial production of this mushroom has been reported. It has usually found in the mountain or the plateau from summer through autumn. The culture condition and media composition for the enhanced mycelial growth of A hemibapha subsp. hemibapha DGUM 28001 were investigated. Optimal temperature and pH for the mycelial growth were 30℃ and pH 5.0, respectively. Among the complex media tested, SYSP(starch yeast extract soytone pyridoxine) was the most favorable complex medium for enhancing the mycelial growth. When Czapek-Dox medium was used as a minimal medium, starch of polysaccharide was very excellent as a carbon and energy source. Sucrose and xylitol were very good carbon source for disaccharide and monosaccharose, respectively. In general, an organic complex nitrogen source was better than an inorganic one. Among the organic complex nitrogen sources tested, yeast extract was the best as a source of nitrogen. As a phosphorus source for the mycelial growth, the most favorable one was NH_4H_2PO_4. When folic acid, vitamin, was supplemented to the minimal medium, mycelial growth was enhanced. These results could be used to produce biologically-active substances from the mycelial culture and for the artificial production of A. hemibapha subsp. hemibapha.

      • Lipopolysaccharide, Dexamethasone, 및 N-Nitro-L-Arginine Methyl Ester가 흰쥐 간 조직의 프리라디칼 발생과 제거에 미치는 영향

        Lipopolysaccharide (LPS) stimulates various immune and inflammatory reactions to induces the generation of reactive oxygen intermediates (ROI) and reactive nitrogen intermediates (RNI). ROI and RNI are free radicals with very high reactivity, oxidize cellular constituents including proteins and lipid membranes, and thus play important roles in the pathogenesis of tissue damages in LPS-induced septic shock. The present study examined effects of dexamethasone (DexM), and N-nitro-L-arginine methyl ester (L-NAME) on the generation of ROI and RNI, and on the activities of enzymes removing ROI in the liver of rat model of septic shock. DexM has widely been used as an immunosuppressant. L-NAME has known to block synthesis of nitric oxide (NO), the main RNI, by inhibiting NO synthase (NOS). LPS (10 mg/kg body weight) markedly enhanced NO generation in serum and liver. The increased NO generation was completely blocked by pretreatment with DexM (10 mg/kg) and effectively inhibited by L-NAME (10 mg/kg). Induction of NOS protein expression by LPS was demonstrated by a Western analysis. The NOS expression was completely blocked by DexM pre-treatment, and markedly inhibited by L-NAME. However, the generation of superoxide radical, an important ROI, was not enhance in rat liver by LPS, rather it was decreased a little. Superoxide radical production was increased by L-NAME treatment or combined treatment of DexM and LPS. The level of lipid peroxidation, an index for free radical-induced cell damage, was increased by either LPS or DexM. The enhancement of lipid peroxidation by DexM was abolished by adding LPS. DexM markedly enhanced the activities of ROI removing enzymes, superoxide dismutase, glutathione peroxidase and catalase. However, rats were treated with LPS in addition to DexM, the elevated activities of superoxide dismutase, glutathione peroxidase and catalase were abolished. These results provide information on the generation and removal of ROI and RNI by LPS, DexM and L-NAME in rat liver, and on development of drugs to inhibit free radical-induced liver damage in sepsis.

      • 방사선에 의한 폴리아닐린/은 복합제의 제조 및 전기전도 특성

        The conducting Ag/PANI composites were prepared by ^(60)C_(o) γ-irradiation using aniline monomer, hydrochloric acid and sulfuric acid as dopant, and AgCl₂ for nano-sized Ag particle. And the effect of experimental conditions such as species and concentrations of dopants, and Ag/PANI mole ratio on the electrical conductivity was studied. Also, we investigated the electrical conductivity according to the absorbed dose so Ag/PANI compoistes. It showed that electrical conductivity of Ag/PANI composite was increased with the increase of the Ag/PANI mole ratid and concentration of popant in case that H₂SO₄ was used as dopant. On the other hand, electrical conductivity showed maximum value and decreased with the increase of the Ag/PANI mole ratio and concentration of dopant in case of HCl dopant. The electrical conductivity of γ-ray irradiated Ag/PANI was increased up to 30kGy irradiation and decreased above 30kGy.

        들깨잎의 품종에 따른 성분분석 및 생리활성물질 탐색

        The biochemical components of Namcheondlggae, Miryangdlkkae 25, Boradlggae and Ipdlkkae 1 were measured. The samples were extracted with hot water, 60% acetone or 80% ethanol for screening physiological activity. The crude protein content (4.36%) was found in the Miryangdlkkae 25 and calcium content (497.5 mg%) was found in the Namcheondlggae among the tested 4 perilla leaves. Fructose was 30.86 mg% in the Namcheondlggae and free amino acids at all perilla leaves was detected seventeen. In Boradlggae, glutamic acid and alanin were 25.37 and 11.91 mg%. Totally nine non-volatile organic acids were also detected and the contents of malic acid and glutaric acid were 28.34 and 14.57 mg% in Boradlggae. The Miryangdlkkae 25 had the highest vitamin C amount which was 113.24 mg%. Angiotensin converting enzyme (ACE) inhibition activity of 80% ethanol extract of Boradlggae was 46.71%. Electron donating activity of 60% acetone extract from Namcheondlggae was the strongest inhibition activity as 98.19% when 200ppm level of the sample extracts were added.
