      • cDNA Cloning and Genomic Structure of Glyceraldehyde-3-Phosphate Dehydrogenase from the Entomopathogenic fungus, Paecilomyces tenuipes-Jocheon

        Fungi belonging to the Paecilomyces spp. have recently been used as food and herbal medicines in Korea and are greatly popular as commercially available powdered supplement or dried fruiting body. Despite this acceptance and its use, little is known of the genes related to its reactive agents. Presently, We have constructed an olig-d(T) primed directional cDNA library from the silkworm Dongchunghacho, an entomopathogenic fungus, of which species is belonging to Paecilomyces spp. based on the previous identification of ITS1 and ITS2 at the molecular level and collected from Jocheon Miryang, Korea. To isolate and screen genes in the fungus, 626 expressed sequence tags(ESTs) were generated by a partial sequencing from the cDNA library. cDNA encoding the glyceraldehyde-3-phosphate dehydrogenase(Pt-GAPDH) of Paecilomyces tenuipes- Jocheon was cloned from the above cDNA library. The complete cDNA sequence of Pt-GAPDH is comprised of 1,014bp encoding 338 amino acid residues. The deduced protein sequence of Pt-GAPDH showed higher homology with Beauberia bassiana-GAPDH(93% amino acid identity). Hydropathy analysis revealed that Pt-GAPDH protein is hydrophilic. The major three amino acids in its composition of amino acid residues were alanine(11.54%), valine(9.47%) and glycine(8.88%). The Pt-GAPDH gene of Paecilomyces tenuipes entomopathogenic fungus consisted of three exons and two introns coding for 338 amino acid residues, and the genomic DNA length of the gene spans 1302bp. The accession number of the gene in GenBank are GU997099 for Pt-GAPDH cDNA and GU997102 for Pt-GAPDH genomic DNA. More investigation works including gene expression, immunological analysis etc. will be carried continuously without hesitation after this presentation.

The cDNA encoding Pt-GAPDH was expressed as a 37 kDa polypeptide in baculovirus-infected insect Sf9 cells. The Pt-GAPDH gene of Paecilomyces tenuipes entomopathogenic fungus consisted of three exons and two introns coding for 338 amino acid residues, and the genomic DNA length of the gene spans 1302bp. The accession number of the gene in GenBank are GU997099 for Pt-GAPDH cDNA and GU997102 for Pt-GAPDH genomic DNA.

      • Kalman-Szego-보조정리의 응용에 의한 제한된 제어입력을 위한 보상기 설계

        In this paper a method for design in order to use control input due to the saturation is proposed. This is done by designing a linear controller with the good property for reference input and control output without considering saturation or partially considering the saturation and is done by setting up a compensation loop which compensates for the lost portion of control input due to saturation. The Lyapunov function constructed by using the Kalman-Szego-Lemma was guaranteed for the stability problem arising due to the nonlinerarity of a compensation loop when a compensator is designed. Also, it is shown that this Lyapunov function was satisfied for Riccati equation.

      • HEC-RAS모형을 이용한 유량측정 검증에 관한 연구

        The basis of human life and being among the resources all of the earth is water and air. The water of modern city life is used to agricultural water, industrial water, production of energy and recreation. Also water was taken up unmoving position as natural resources. And it has been subject of boom just like a prevention of flood damage. Therefore Acquisition of discharge-data with confidence for effective management and division and rainfall-runoff relation of water resources as foundation work are very important. Discharge measurement of irrigation type is essential to pan of Dam, the improvement of river, construction plan of flooding-structure at river, establishment plan of water. And Flood control type is using to building of a bank, a flood system for prevention and warning. Both of them is important data. But Declination of discharge measurement data is very hard according to condition of weather, a river physical situation, a using situation of river water an upper stream and down stream. So it needs to verification. This study was selected a place of Naju at YoungSan river Andthen using HEC-RAS model. The result of correlation analysis about water-level of actual measurement and water-level ; the correlation coefficient is analyzed 0.998. In accordance with the result that confidence is recognized.

        주물사업장에서 발생하는 호흡성분진의 농도 및 석영함유량에 관한 연구

        This study was performed to measure and evaluate the concentration of respirable dust and quartz content at molding and shakeout process in the foundry. The 129 samples in 30 foundries were collected using 10mm Dorr-Oliver nylon cyclone and analysed for quartz concent by Fourier Transform Infrared Spectroscopy. The average quartz concentrations in the respirable dust was 0.0273mg/㎡ and ranged from 0.0007 to 0.3757mg/㎥. The average quartz concentrations of shakeout process wsa significantly higher than that of molding in the foundry. The average of respirable dust in foundry 102129mg/㎡ and ranged from 0.1524 to 7.0428mg/㎥. But there is no significant difference between molding and shakeout processes. The average contents of quartz for OEL-MOL was 2025% and ranged from 0.05 to 32.79% The non-compliance rates for quartz were 22.96% in Korea-OEL and 34.01% in ACGIH-TLV and NIOSH-REL, and 35.03% in OSHA-PEL. The airbome dust in the foundry was effectively controlled when local exhaust ventilation and/or wetting system were operated. The result of this study therefore showed that the engineering controls and periodic environmental suveillance in foundry will be very important for preventing the health effects of workers exposed to quartz.

      • 수질보전을 위한 영산강 유역의 TMDL 결정에 관한 연구

        In the present study, the Total Maximum Daily Load (TMDL) was introduced to the Young-San river basin for the multipurpose expansion of the aquatic-ecosystem recovery and the water resource utilization of the Young-San Lake. For the methodology, the Young-San river basin was divided into 12 sub-basins in which point sources and non-point sources were investigated. The representative stations from the Young-San river were selected to estimate the hydraulic coefficients (a, b, alpha, beta) of the respective stations using HEC-RAS Model with the profile data from 『Ministry of Construction and Transportation, 1998』 Water quality prediction was performed by the QUAL2E Model with the input data from the water-stage network data of the Young-San River Flood Control Office and the water quality network data of the Young-San River Basin Environment Office. Based on the results from the water quality prediction, TMDLs for the respective sub-basins were proposed so as to fit the target water quality which was estimated in the river channel by the reduction of load.

      • 충주 지역에서 10년간 급성심근경색의 임상관찰에 대한 연구

        Background: The current treatments of acute myocardial infarct(AMI) Include noninvasive method using thrombolytics and invasive methods such as primary percutaneous transluminal coronary angioplasty(PTCA) and coronary artery bypass graft(CABG)surgery. Although there are many studies comparing the effectiveness of noninvasive and invasive methods, when large clinical centers, 20% of totar hospitals, were excluded, most middle-sized hospitals lacked personnels and facilities for the invasive methods. Thus they opted for the noninvasive methods. Therefore, in this study, the clinical characteristics of AMI patients and the results of thrombolytic & conservative treatment were observed. Method: 137 patients with acute myocardial infarct, who visited KonKuk University Medical Center, ChoongJu Hospital during Sept. 1990 to Sept. 1999 were analysed retrospectively using medical records. Result: The sexual compositions of patients were 63.5% male, and 36.5% female. The mean age for male patients were 56±12.9, and for female patients 68.7±9.6 (p<0.05). The mean age for female patients was significantly older than male patients. The patients in the thrombolytic treatment group had mortality rate of 6.25% which is lower than that of the conservative treatment group at 12% (p>0.05). Conclusion: Although in our study, small number of acute myocardial infarct patients caused the statistical insignificance, rapid revascularization by thrombolytic therapy within 6 hours of onset of symptoms improved mortality rate and early ambulation. And we had good results on AMI with thrombolytic agent in ChungJu including near rural area as primary emergency hospital. Therefore a prospective research with greater number of subjects is needed to develop better treatments for AMI.

        PECVD법으로 증착된 전자소자용 thiophene 박막의 전기화학적 신뢰성에 관한 연구

        The corrosion failure of electronic devices has been a major reliability concern lately. This failure is an ongoing concern because of miniaturization of integrated circuits(IC) and the increased use of polymers in electronic packaging. In this paper plasma-polymerized thiophene films were considered as a possible candidate for an interlayer dielectric for multilever metallization of ultra large scale integrated (ULSI) semiconductor devices. The protective ability of above films as a function RF power in an 3.5 wt.% NaCl solution was examined by electrochemical methods and contact angle measurement. The protective efficiency of the film increased with increasing RF power, which induced the higher degree of cross-linking and hydrophobicity of the films.
