        Differential Manifestation of Ocular Phenotypes in TALEN-mediated p19arf Knockout FVB/N and C57BL/6J mouse lines

        Background p19arf, primarily known as a tumor suppressor, has also been reported to play an essential role in normal development of mouse eyes. Consistently, lack of p19arf has been associated with ocular defects, but the mixed background of the knockout (KO) mouse strain used raised a concern on the accuracy of the phenotypes observed in association with the targeted gene due to genetic heterogeneity. Object We carried out a study to investigate into the efect of genetic background on the manifestation of p19arf KO associated phenotypes. Methods We characterized the phenotypes of novel p19arf KO mouse lines generated in FVB/N and C57BL/6J using a transcription activator-like efector nuclease (TALEN) system in comparison to the reported phenotypes of three other p19arf-defcient mouse lines generated using homologous recombination. Results Ninety-fve percent of FVB/N-p19arf KO mice showed ocular opacity from week 4 after birth which worsened rapidly until week 6, while such abnormality was absent in C57BL/6J-p19arf KO mice up to the age of 26 weeks. Histopathological analysis revealed retrolental masses and dysplasia in the retinal layer in FVB/N-p19arf KO mice from week 4. Besides these, both strains developed normally from birth to week 26 without increased tumorigenesis except for a subcutaneous tumor found in a C57BL/6J-p19arf KO mouse. Conclusion Our fndings demonstrated surprisingly variable manifestation of p19arf-linked phenotypes between FVB/N and C57BL/6J mice, and furthermore between our mouse lines and the established lines, indicating a critical impact of genetic background on functional study of genes using gene targeting strategies in mice.

        일본자료를 통해 본 ㆍ의 변천과정

        본고에서는 한국어를 일본문자인 가나로 기록한 18-19세기초(?)의 일본자료《전일도인》《물명》《교린수지》를 분석하여 ㆍ의 음성적인 면 및 그 변천과정을 고찰하였다. 이들 자료에 나타나는 어두의 ㆍ의 변천과정은 다음과 같다.《전일도인》에 나타나는 어두의 ㆍ는 선행자음에 따라 a단과 o단 가나로 전사되어 있으나,《물명》과《교린수지》의 음주에서는 모두 a단으로 기록되어 있어《물명》의 시기 이후에는 어두의 ㆍ>ㅏ의 변화가 완성되었음을 보여주고 있다. 그러나 세 자료에서 비어두의 ㆍ는 대부분 o단으로 기록되어 있어 u단으로 기록되어 있는 ㅡ와는 구별되어 있다. 만약 이 시기에 비어두의 ㆍ가 ㅡ와 합류된 상태라면 비어두의 ㆍ는 u단으로 기록되어 있었을 것이다. 이러한 가나전사는 어두에서 ㆍ>ㅏ의 변화가 일어난 18세기 이후의 시기에도 비어두의 ㆍ는 일정기간 음가를 유지하였을 가능성을 보여준다. The aim of this study is to clarify the process of the change of Korean Vowel ㆍ[Λ] in the analysis of the transcriptions of Korean source-materials (i. e. Korean textbooks for Japanese written in the 18-19th centuries, Jen'ichidojin(全一道人), Butsumyo(物名), and Korinsuchi(交隣須知)) which are written with the Japanese writing system. The results are as follows. 1) In Jen'ichidojin, the vowel ㆍ[Λ] in the first syllable of the lexical morphemes is transcribed with Japanese a or o according to preceding consonants. 2) In Butsumyo and Korinsuchi, the vowel ㆍ[Λ] in the first syllable of the lexical morphemes is transcribed with Japanese a, which means that the merger of the vowel ㆍ[Λ] and ㅏ[a] is completed. 3) The vowel ㆍ[Λ] which is not in the first syllable of the lexical morphemes is usually transcribed with Japanese o in the three textbooks. Also the vowel ㆍ[Λ] in the above condition is not merged with the vowel -[Ш] previous to the 18th century.

        제2형 당뇨병 남성 환자에서 유리 테스토스테론과 성호르몬 결합 글로블린 농도

        연구배경:인슐린저항성은 제2형 당뇨병과 심혈관질환의 주요한 위험 인자로 성호르몬과 상호 관계가 있다고 알려져 있다. 그러나 여성과는 달리 남성에서는 연령과 인슐린저항성에 따른 유리 테스토스테론과 성호르몬 결합 글로블린 농도의 변화에 대한 연구가 부족한 실정이다. 그러므로, 본 저자등은 제2형 당뇨병 남성 환자에서 정산인과 비교해서 유리 테스토스테론과 성호르몬 결합 글로블린 농도를 측정하고 연령에 따른 변화 정도를 알아보고자 하였다. 방법:대상 환자 모두에서 연령과 체질량지수, 총 콜레스테롤, 중성지방, 공복혈당과 인슐린 농도를 측정하였다. 혈중 유리 테스토스테론 농도는 방사면역측정법(radioimmunoassay)을 이용해서 측정하였고, 혈중 성호르몬 결합글로불린은 면역방사계측측정법(immunoradiometric assay)을 이용해서 측정하였다(Diagnostic System Laoratories, Wbster, TX, USA). 결과:1)제2형 당뇨병 남성 환자에서 정상 대조군 남성에 비해 성호르몬 결합글로불린은 104.1±35.0 vs 25.7±3.5 mole×10??로 의미 있게 높았으나(p<0.001), 유리 테스토스테론은 13.7±9.5 vs 13.6±6.5 ng/dL로 차이가 없었다. 2)연령과 성호르몬 결합 글로블린 사이의 상관 계수는 제2형 당뇨병 남성 환자에서 0.40로 중등고의 양의 상관 관계를 보였고(p<0.001), 정상 대조군 남성에서 0.11로 유의한 상관 관계를 보이지 않았다. 연령과 유리 테스토스테론 사이의 상관 계수는 제2형 당뇨병 남성 환자에서 0.08, 정상 대조군 남성에서-0.17로 모두에서 유의한 상관 관계를 보이지 않았다. 3)연령과 체질량지수를 보정한 후에 제2형 당뇨병 남성 환자와 정상 대조군 남성에서 혈중 인슐린 농도, 유리 테스토스테론과 성호르몬 결합 글로블린 사이에는 상관 관계가 없었다. 결론:제2형 당뇨병 남성 환자에서 정상 남성에 비해 성호르몬 결합 글로블린 농도가 증가되어 있었으며, 유리 테스토스테론은 차이가 없었다. 연령이 증가함에 따라 성호르몬 결합 글로블린이 제2형 당뇨병 남성 환자에서 정상 남성에 비해 증가 폭이 의미있게 컸으며, 유리 테스토스테론은 변화가 없었다. Background: Insulin resistance is a risk factor for cardiovascular disease and type 2 diabetes mellitus. There are many previous studies indicating that insulin lowers serum sex hormone-binding globulin levels, and there is inverse correlation between insulin resistance and serum sex hormone-binding globulin levels in women. However, in men, a limited number of studies are available to explain the effect of sex hormone on age and insulin. Therefore, the present study was undertaken to investigate the relationship among free testosterone, hormone-binding globulin and age in type 2 diabetic men and control subjects. Method: Age, body mass index, total cholesterol, triglyceride, fasting blood sugar, and insulin concentrations were examined on 89 type 2 diabetic men and 47 control subjects. The free testosterone level was measured by commercially available double-antibody system (Radioimmunoassay). The sex hormone-binding globulin level was also measured by commercially available double-antibody system(Immunoradiometric assay). Results: 1) Sex hormone-binding globulin level was significantly increased in patients with type 2 diabetes. However, there was no significantly difference in free testosterone level between the two groups. 2) Sex hormone-binding globulin was positively correlated with age (r=0.4, p <0.001) in patients with type 2 diabetes. Sex hormone-binding globulin and free testosterone were not correlated with age in control sujects. 3) Free testosterone and sex hormone-binding globulin concentrations were not significantly related to serum insulin concentration after adjusting for age and body mass index. Conclusions: We observed increased sex hormone-binding globulin concentration in diabetes man, and was a positively related to age. Further studies are needed to understand the relationships between age, insulin resistance, testosterone, and sex hormone-binding globulin concentrations(J Kor Diabetes Asso 24:699~707,2000).

        With the recent change of paradigm and the influence of organic and digital architecture, a lot of studies and experiments on nonlinear shape are performed. Development of computer technology expedited these kinds of experiment. Though using techniques such as 3D modeling and VRML and so forth, expressing shapes like these has difficulties. The advanced solution for those difficulties is the application of RP technology using for designing machinery and metallic pattern, which is used by architects such as Frank Gehry, Coop Himmelblau, Greg Lynn. Making physical model is important in the point of the fact that it can embody model to a real building, not a media on monitor. In architectural design, using RP equipment is very important and a lot of studies for utilization are necessary.

        This study was carried out to analyze physical and chemical properties of soil and leaf analysis to estimating amount of tree growth in Chamaecyparis pisifera, Jinhae city, Gyeongnam province. The results were summarize as follows; As the results from analysis of relation of physical-chemical properties of soil and leaf component, the relationship p component inside soil and n component inside leaf showed positive correlation with determination coefficients(R^2=0.9986), and also relation of nitrogen component inside soil and component of Na_2O inside leaf showed positive correlation with determination coefficients(R^2=0.9987), but the relation of component Ca^2+ inside soil and component of Na_2O inside leaf showed negative correlation with determination coefficients(R^2=-0.9984), and also relation of p component inside soil and Fe component inside leaf showed negative correlation with determination coefficients(R^2=-0.7215). The results from analysis of relation of leaf component and tree growth, showed correlation of determination coefficient(R^2=0.4256) in height growth, determination coefficient (R^2=0.6248) in DBH growth, determination coefficient(R^2=0.6248,) in volume growth, respectively. In each tree growth of recent five years, N and P components showed positive correlation of determination coefficient(R^2=0.6170, 0.5780, respectively) in height growth, and also N and Na^+ components showed positive correlation of determination coefficient(R^2=0.6016, R^2=0.8727, respectively) in DBH growth, but K= component showed negative correlation in DBH growth As the results from analysis of stepwise regression to estimating amount of DBH growth, sequence of inputted variables entered in order of N, Na+, Mg++, ,and estimated equation was Y=44.791N-56.640Mg+0.6867Na-26.47, determination coefficients of 69%.

        Feline Diffuse Iris Melanoma in a Cat

        A 4-year-old spayed female British shorthair cat was referred for abnormal pigmentation on the right iris. The pigmentation was mainly located in the medial portion of the iris. No abnormalities except iris were detectedin a full ophthalmic examination. There was no evidence of metastasis after thoracic radiography and abdominalsonography. Enucleation was performed on the right eye and it was sent for a histopathological evaluation. It wasconfirmed as early stage of feline diffuse iris melanoma (FDIM) with involvement of iris stroma.

        Endovascular Treatment for Common Iliac Artery Injury Complicating Lumbar Disc Surgery : Limited Usefulness of Temporary Balloon Occlusion

        Vascular injury during lumbar disc surgery is a rare but potentially life-threatening complication. It has been managed by open vascular surgical repair. With recent technologic advance, endovascular treatment became one of effective treatment modalities. We present a case of a 32-year-old woman who suffered with common iliac artery injury during lumbar disc surgery that was treated successfully by endovascular repair with temporary balloon occlusion and subsequent insertion of a covered stent. Temporary balloon occlusion for 1.5 hours could stop bleeding, but growing pseudoaneurysm was identified at the injury site during the following 13 days. It seems that the temporary balloon occlusion can stall bleeding from arterial injury for considerable time duration, but cannot be a single treatment modality and requires subsequent insertion of a covered stent.

        Clostridium difficile Toxin A Inhibits Erythropoietin Receptor-Mediated Colonocyte Focal Adhesion Through Inactivation of Janus Kinase-2

        Previously, we demonstrated that the erythropoietin receptor (EpoR) is present on fibroblasts, where it regulates focal contact. Here, we assessed whether this action of EpoR is involved in the reduced cell adhesion observed in colonocytes exposed to Clostridium difficile toxin A. EpoR was present and functionally active in cells of the human colonic epithelial cell line HT29 and epithelial cells of human colon tissues. Toxin A significantly decreased activating phosphorylations of EpoR and its downstream signaling molecules JAK-2 (Janus kinase 2) and STAT5 (signal transducer and activator of transcription 5). In vitro kinase assays confirmed that toxin A inhibited JAK 2 kinase activity. Pharmacological inhibition of JAK2 (with AG490) abrogated activating phosphorylations of EpoR and also decreased focal contacts in association with inactivation of paxillin, an essential focal adhesion molecule. In addition, AG490 treatment significantly decreased expression of occludin (a tight junction molecule) and tight junction levels. Taken together, these data suggest that inhibition of JAK2 by toxin A in colonocytes causes inactivation of EpoR, thereby enhancing the inhibition of focal contact formation and loss of tight junctions known to be associated with the enzymatic activity of toxin A.

        Effect of the COVID-19 Pandemic on Headache in Pediatric Migraine Patients at a Single Tertiary Center

        Purpose: The aim of this study was to evaluate the impact of the coronavirus disease 2019 (COVID-19) pandemic on children and adolescents with migraine. Methods: This longitudinal cohort study enrolled children and adolescents with migraine from the Department of Pediatric Neurology at our hospital from January 2017 to June 2021. Self-reported data from individual headache diaries were used. The patients were questioned about their headache frequency and intensity, stress, physical activity, changes in mood and sleep, and their school and home lives during the COVID-19 pandemic. The Pediatric Migraine Disability Assessment (Ped-MIDAS) scoring system was applied to assess headache-related disability. Results: In total, 325 pediatric migraine patients (mean age 12.8±5.6 years, 62.5% female) were included in this study. The average monthly frequency of migraine headaches was 2.17±1.32 and 4.62±3.29 before and during the COVID-19 pandemic (P<0.001), respectively. The Ped-MIDAS score was obtained for 207 patients both before and during the pandemic, and the total score slightly increased from 13.8 to 14.7 points (P=0.295). Sixty patients (18.5%) showed significantly worsening migraine headaches. Younger age (P=0.017), mood deterioration (P<0.001), sleep problems (P<0.001), increased acute medication use (P=0.010), and larger changes in the Ped-MIDAS score (P=0.002) were significantly associated with worsening headache in the logistic regression analysis. Conclusion: Headache attacks in children and adolescents with migraine were more frequent during the COVID-19 pandemic than before it. Worsening headaches could be independently attributed to younger age, mood deterioration, and poor sleep during the COVID-19 pandemic.
