Differential Manifestation of Ocular Phenotypes in TALEN-mediated p19arf Knockout FVB/N and C57BL/6J mouse lines

Background p19arf, primarily known as a tumor suppressor, has also been reported to play an essential role in normal development of mouse eyes. Consistently, lack of p19arf has been associated with ocular defects, but the mixed background of the knockout (KO) mouse strain used raised a concern on the accuracy of the phenotypes observed in association with the targeted gene due to genetic heterogeneity. Object We carried out a study to investigate into the efect of genetic background on the manifestation of p19arf KO associated phenotypes. Methods We characterized the phenotypes of novel p19arf KO mouse lines generated in FVB/N and C57BL/6J using a transcription activator-like efector nuclease (TALEN) system in comparison to the reported phenotypes of three other p19arf-defcient mouse lines generated using homologous recombination. Results Ninety-fve percent of FVB/N-p19arf KO mice showed ocular opacity from week 4 after birth which worsened rapidly until week 6, while such abnormality was absent in C57BL/6J-p19arf KO mice up to the age of 26 weeks. Histopathological analysis revealed retrolental masses and dysplasia in the retinal layer in FVB/N-p19arf KO mice from week 4. Besides these, both strains developed normally from birth to week 26 without increased tumorigenesis except for a subcutaneous tumor found in a C57BL/6J-p19arf KO mouse. Conclusion Our fndings demonstrated surprisingly variable manifestation of p19arf-linked phenotypes between FVB/N and C57BL/6J mice, and furthermore between our mouse lines and the established lines, indicating a critical impact of genetic background on functional study of genes using gene targeting strategies in mice.

초등교사의 열정에 대한 인식: 의미연결망 분석의 활용

본 연구에서는 초등교사가 가지고 있는 열정에 대한 인식구조를 파악하기 위해 현직 초등교사 131명을 대상으로 설문조사를 실시하였다, 연구에서는 열정, 열정의 장단점, 교사열정에 대한 인식구조를 알아보기 위해 의미연결망 분석을 활용하여 근접 중심성, 연결중심성, 매개중심성, 아이겐벡터 중심성을 위주로 살펴보았다. 연구결과 첫째, 열정에서는 ‘힘’,‘강렬’과 같은 높은 강도나 에너지를 나타내는 단어들이 중요한 역할을 하였으며 커뮤니티 분석 결과 열정의 행동, 정서, 동기 특성에 대한 주제가 분류되었다. 둘째, 열정의 장점에서는 ‘삶’,‘원동력’,‘에너지’등이, 열정의 단점에서는 ‘타인’,‘피로’,‘살피지 못함’등이 중요한 역할을 하는 것으로 보아 기존 이론에 나타난 열정의 특성을 설명하고 있었다. 셋째, 교사열정에서는 ‘학생’,‘수업’과 같은 교수활동의 실천을 나타내는 단어들이 중요한 역할을 하였으며 커뮤니티 분석 결과 교육활동 전반, 학생과 수업, 전문성 개발에 대한 주제가 분류되었다. The purpose of this study was to explore the meaning of passion perceived by elementary school teachers. In order to understand the structure of perception of passion, we surveyed 131 in-service elementary school teachers. Responses to passion, the positive and negative role of passion, and teacher passion were analyzed using a semantic network analysis. We focused on closeness centrality, degree centrality, betweenness centrality and eigenvector centrality. Results showed that first, in passion, words indicating high strength and energy such as‘power’and‘intensity’played an important role in the structure of perception. Themes on behavioral, emotional, and motivational characteristics of passion were classified as sub-factors of passion as a result of community analysis. Secondly,‘life’,‘motivation’, and‘energy’appeared in key-word analysis in the positive role of passion while‘others’,‘fatigue’, and‘not caring for’appeared in the negative role of passion, reflecting the theory on the dualistic model of harmonious and obsessive passion. Third, in teacher passion, words indicating teaching activities and their practices such as‘student’and‘instruction’played an important role in the structure of perception. Themes related to educational activities in general, students and instruction, and professional development were classified as sub-factors of teacher passion.

7β-Hydroxycholesterol이 사람 백혈암에 대한 세포 독성 효과

The cytotoxic activity of 7β-hydroxycholesterol (7β-OHC) has been evaluated using various leukemia cell lines by MTT test. Among the tested cell lines, 7β-OHC showed to be more cytotoxic to THP-1, human monocytic leukemia cell line. IC50 value of 7β-OHC on THP-1 was 10μM. Kinetic of on THP-1 showed that growth of THP-1 began to decrease at 36 h after treatment 7β-OHC. 7β-OHC induced apoptosis shown by DNA fragmentation by gel electrophoresis. 7β-OHC induced apoptosis through down-regulation of Bcl-2 expression. Taken together, these results suggested that treatment with 7β-OHC inhibits the proliferation of THP-1 cells through apoptosis.

1,1,1,2-Tetrafluoroethane(HFC-134a)과 Propane(HC-290)의 2성분계 기-액 평형 측정

혼합물을 냉매로 사용할 경우, 냉매의 성능 평가를 위해 기-액 상평형 데이터가 필수적으로 요구된다. 본 연구에서는 HFC-134a와 propane의 이성분계 상평형 데이터를 얻기 위해 273.15, 283.15, 283.15, 303.15, 313.15, 323.15K의 조건에서 실험을 실행하였다. 상평형 실험 장치로는 circulation type의 장치를 사용하였다. 실험데이터는 Peng-Robinson-Stryjek-Vera(PRSV)상태방정식과 NRTL excess Gibbs free energy model을 결합한 식에 다섯 개의 혼합법칙(Huron-Vidal original and modified equation)들을 이용하여 상관관계를 알아보았다. PRSV 상태방정식을 이용하여 얻은 데이터는 실험 데이터와 만족할 만한 일치를 볼 수 있었다. 실험은 실행한 모든 온도에서 공비점이 형성하는 것을 확인하였다. Vapor-liguid equilibrium(VLE) data are essential when using mixtures as a refrigerant. The VLE data of binary mixture containing 1, 1, 1, 2-tetrafluoroethane and propane were measured at 273.15, 283.15, 293.15, 303.15, 313.15 and 323.15K using circulation type experimental apparatus. The experimental data were correlated with five mixing rules combined with the Peng-Robinson-Stryjek-Vera equation of state. We compared the data obtained by using the PRSV EOS with experimental data and a satisfactory consistency of model predictions with experimental data were obtained. Azeotropic behavior has been observed in all these mixtures.

EDF 기대손실에 기초한 로버스트 공정능력지수

This paper presents a robust process capability index(PCI) based on the expected loss derived from the empirical distribution function(EDF). We propose the EDF expected loss in order to develop a PCI that does not depends on the underlying process distribution. The EDF expected loss depends only on the sample data, so the PCI based on it is robust and it does nor require complex calculations. The inverted normal loss function(INLF) is employed in order to overcome the drawback of the quadratic loss which may increase unboundedly outside the specification limits. A comprehensive simulation study was performed under various process distributions, in order to compare the accuracy and the precision of the proposed PCI with those of the PCI based on the expected loss derived from the normal distribution. The proposed PCI turned out to be more accurate than the normal PCI in most cases, especially when the process distribution has high kurtosis or skewness. It is expected that the proposed PCI can be utilized in real processes where the true distribution family may not be knwon.

효소적 방법으로 제조된 키토산 올리고당의 첨가에 따른 김치의 숙성 및 저장 기간 연장에 관한 연구

효소로 처리된 키토산 올리고당의 첨가에 의한 김치의 숙성 기간 연장 효과를 알아보기 위하여 배추김치에는 0.2-0.4%, 2일째에 0.4%를 첨가하여 20℃에서 10일간 관찰한 결과 0.2% 첨가시 대조군에 비해 2일, 0.4% 첨가시는 4일, 2일째에 0.4%를 첨가한 경우는 8일 이후로 연장 효과가 2-4배 있었으며, 0.4% 이상에서는 미생물 생육자체가 억제되므로 풍미 및 유기산의 생성을 고려할 때 2일째 첨가된 0.4%가 젖산균의 수는 어느정도 유지하면서 stationary phage를 연장시킴으로서 더 좋은 결과를 나타냈다. 갓 김치의 경우 0.1%-0.5% 첨가시 대조군에 비해 1.2-1.4배의 연장 효과가 있어서 배추 김치에 비해서는 낮은 효과를 나타냈다. 따라서, 효소로 처리된 키토산 올리고당의 첨가에 의해 배추 김치는 2-4배의 연장 효과, 갓김치는 1.2-1.4배의 연장 효과를 나타내어 미생물 증식 억제에 의한 김치의 숙성 및 저장 기간 연장 효과를 기대할 수 있었다. To prolong the fermenting and preservation period of Kimchi, chitosanoligosaccharide was prepared by chitosanase. Various concentrations of chitosanoligosaccharide were added into Chinese cabbage Kimchi and Gat Kimchi. Chinese cabbage Kimchi was formulated with various chitosanoligosaccharide concentrations(0.2-0.4%) and fermented at 20℃. And Gat Kimchi was prepared with various chitosanoligosaccharide(0.1-0.5%) concentrations during 26 days at 4℃. In this results, the fermentation period was increased 2-2.5 and 1.2-1.4 folds more than control(0% of chitosanoligosaccharide) for Chinese cabbage Kimchi and Gat Kimchi, respectively. Conclusively, adding of chitosanoligosaccharide into Kimchi significantly prolonged the fermentation and preservation period.

장기 알콜투여가 생쥐의 간 및 신장, 위장 조직 손상에 미치는 영향

Alcohol is a major risk factor for several diseases and excessive, long-term alcohol consumption are cause physical alteration-fatty liver, hepatitis, cirrhosis, breaking down, Wernicke-karasakoffs syndrome, weight loss, and poor immunity-in virtually all organ and tissue, This study was observed that liver, kindney, and stomach were altered in mouse by the effect of chronic alcohol administration. The mouse were sacrificed to obtain the tissue after mouse were orally injected with 25% ethanol 18㎖/kg/day for 120days. The tissue were stained by hematoxylin and eosun and then obseved by light microscope. The results of this study were as follows : 1. The congestion was appeared in liver after 120days alcohol administration 2. The destruction of glomerulus were increased and parietal cell of Bowman's capsule were swelled such as cuboidal cell after 12days alcohol administration. The congestion was appeared in alcohol administrated group. 3. The mucosa and gasstric pit were destructed and the ulceration was appeared in stomach after 120days administration. The parietal cells and chief cells were damaged. Above results were shown that the tissue were damaged by chronic alcohol administration.

외상후 혈뇨환자에서 응급 초음파검사의 의의

The Ultrasonography is rapid, mobile, and noninvasive diagnostic tool in emergency center. As a result, we estimate it`s value as an advocated primary diagnostic modality in the immediate evaluation of patients with multiple trauma. We investigated 60 patients with traumatic hematuria(microscopic hematuria or gross hematuria) who visited our emergency department from 1995, July to 1996, March. To evaluate the injury of urinary systems, emergency physician examined patients with hematuria by ultrasonography, and comapared the results with those of excretory urography and computed tomography. We demonstrated 21 positive findings for abnormal fluid collection and renal parenchymal injury and bladder rupture on ultrasonography. All bladder injuries were diagnosised by ultrasonography, and all major renal injury(grade Ⅲ,Ⅳ,Ⅴ) were also diagnosised by that. The accuracy of emergency ultrasound examination in traumatic hematuria was 97.0%, and sensitivity was 95.0% with specificity of 97.4%. The ultrasonography was an accurate diagnostic adjunct in detecting renal injury and bladder injury in traumatic hematuria patients. Ultrasonography provides an excellent noninvasive diagnostic tool for the initial screening study of patients with traumatic hematuria.

3-D 동작 분석 장치를 이용한 한국성인(20대, 30대)의 동작 범위에 관한 연구

Devices, such as a goniometer, have been used to measure a human being's active range of motion, but interferences between subjects and testers have affected the result precision. In this study, 56 dynamic motion ranges were measured using a 3-D motion analysis system which is quite accurate (2mm error for 1m). The 3-D motion analysis system consists of VICON 140, data acquisition system, and data analysis program (KRISSMAS). 80 Korean subjects (40 males, 28±6.1 aged and 40 females, 28.4±6.9 aged) participated. Subjects were divided into 4 groups by Ro¨hrer's index based on National Anthropometric Survey of Korea 1997. Basic statistical analysis were performed using SPSS (ver. 8.0) programs after refining data. Various comparisons were performed for the right and left side, male and female, age groups (20's, 30's. and 40's-60's) using previous studies. The results show that female subjects have more flexible joints in the neck, hip, ankle, and wrist, while male subjects have more flexibility in their waist. But most of the significances were not high (>0.05). The prediction that the right side of Korean bodies would be mort flexible was not a good hypothesis. And the joints flexibilities are not correlated with Ro¨hrer's Index.

대전 3, 4 공단지역의 대기중 중금속의 농도

This research centered on seasonal analysis for the concentration of heavy metals in 5 sites within Taejon 3rd and 4th industrial Complexes. Total suspended particulates were collected on glass microfibre filters (Whatman EPM 2000) by high volume air samplers. About 20 metal elements including 13 toxic ones were analyzed by ICP-MS and ICP-AES after the pretreatment of mixed acid extraction. The following results were summarized from the research : 1) the concentrations of TSPs were 17.7∼219.6㎍/㎥ while the arithmetic mean concentration was 101.7㎍/㎥ ; 2) the arithmetic mean concentration of human carcinogen such as arsenic, hexavalent chromium and nickel subsulfide was 9.99, 6.68 and 6.13 ng/㎥ while that of probable human carcinogen such as beryllium, cadmium and lead was 0.12, 3.37, 97.65 ng/㎥, respectively.
