        한방 이화주의 미백 및 피부 주름 개선 효과

        본 연구에서는 한방이화주의 피부 생리기능 활성을 알아보기 위해 70% EtOH 추출물의 피부 미백, 주름 개선 및 항염증 효과를 조사하였다. HEE은 tyrosinase 활성 억제 및 tyrosine을 기질로 melanin이 형성되는 pathway에 관여하는 주요한 인자인 TRP-1과 TRP-2를 저해하는 작용 기전을 통해 피부 색소침착의 주요 원인 물질인 melanin 생합성을 농도 의존적으로 저해하는 것을 확인하였다. HEE은 피부진피 내 피부 탄력을 유지하는 elastin을 분해하는 효소인 elastase의 활성을 저해하였고, 피부의 keratinocyte가 생성분비하는 MMP-2와 MMP-9의 단백질 발현과 proteolytic 활성을 억제하여 노화에 따른 피부 주름 생성 억제할 수 있는 가능성을 확인하였다. 또한 세포 독성 없이 LPS에 의해 유도된 염증 반응을 50% 저해하는 HEE의 농도(IC50)는 24.9 μg/mL이며, 50 μg/mL 농도로 처리하였을 때 염증 반응 저해 효과가 70%로 높은 효과를 가지는 것을 확인하였다. 이상의 결과를 종합하면 HEE의 피부 미백, 주름 개선 및 항염증에 우수한 효과를 나타내고 있으므로 기능성 화장품의 주요한 소재로 이용 가치가 높을 것으로 사료된다. Extrinsic skin aging is characterized by the loss of skin tone and resilience, irregular pigmentation, and deep wrinkles. The aim of this study was to investigate the effects of Ehwa Makgeolli containing oriental herbs (Glycyrrhiza uralensis Fisch., Lycium chinense MILL., Morus alba L., and Saururus chinensis Baill) on skin whitening and wrinkling in human skin cells. We prepared Makgeolli extracts (HEE) with 70% ethanol. HEE significantly inhibited in vitro mushroom tyrosinase activity and reduced the cellular and secreted melanin content of mouse melanoma melanocytes (B16F1 cells). HEE down-regulated the protein expression of tyrosinase related protein (TRP)-1/-2, a key player in melanogenesis. Treatment with HEE in human keratinoctyes (HaCaT cells) inhibited the proteolytic activities of matrix metalloproteinase (MMP)-2/-9 in a dose-dependent manner and dramatically reduced the expression of MMP-2/-9. In addition, HEE attenuated lipopolysaccharide (LPS)- induced nitric oxide production in murine macrophages (RAW264.7 cells). These results indicate that HEE may be a great cosmeceutical ingredient for its whitening, anti-wrinkle, and anti-inflammatory effects.

        Background: Electromagnetic fields (EMF) are ubiquitous in modern society including medical field. As the technology of medical instruments and telecommunications has developed rapidly, it has influenced on our lives in many ways. Modem medical practice requires high quality medical equipments, which have a great deal of electromagnetic interference and susceptibility. The purpose of this study were to evaluate electromagnetic condition under usual clinical condition and to suggest a practical guideline in general hospital. Materials and Methods: The actual state of the electromagnetic interference in the medical field was studied under usual clinical conditions including operating rooms, intensive care units, magnetic resonance imaging unit, and hyperthermia unit. Results: There was considerable noise as a result of electromagnetic fields from medical equipments including electrosurgical units and hyperthermia unit, and cellular phones, which could induce serious functional derangements of functioning medical devices. Conclusion: It will be necessary to evaluate the individual electromagnetic situations under various medical conditions and to define a limited zone for cellular phone as well as reposition medical equipments to secure a safer medical practice and to minimize electromagnetic interference.

        GT rotary file을 이용한 만곡 근관형성시 작업 효율 및 file 변형 발생에 관한 연구

        Root canal preparation process is of utmost importance in successful treatment of root canal. Also, one of the most important purpose of the root canal preparation is to enlarge the root canal three dimensionally without changing the curvature of the root canal. However, as the curvature of the root canal increases, there are many difficulties involved in formation of optimum root canal. Therefore, in order to solve the above mentioned problems, new developments in methods of root canal preparation and equipments for such purposes were made. Recently, vigorous studies about newly introduced engine-driven nitckel-ti-tanium rotary file are conducted. As shown in research results to dates, it is well established that the use of nickel-titanium file is better suited for curved root canal than stainless steel file in maintaining the curvature of root canal and reducing the deformation of root canal. However, it is also acknowledged that there are a few discrepancies in research results according to protocol, due to failure to remove variables in experiments. In addition, although it is recommended by the manufacturer that the GT rotary file should maintain a low rotational speed of 150∼350rpm and 'light pressure' as light as not to break the lead of a pencil, academic studies about the vertical force which is not yet standardized are not sufficiently explored. Therefore, this research devised and utilized a special research equipment to standardize the appropriate range of vertical force for GT rotary file through experiments by breaking of the lead of a pencil as expressed by the manufacturer and to accurately measure factors involved through repeating and recreating the environment of root canal preparation. Forming nine experimental groups by varying the vertical forces (150g, 220g, 300g) and rpm (150rpm, 250rpm, 350rpm), the effects of changing vertical forces and rpm on working efficiency were measured in terms of time expended in root canal preparation by crown-down method using a transparent resin block with 35 degree curvature and GT rotary file (z-test). The following research using this special research equipment that involved nine experimental groups and varying the vertical force for root canal preparation from 300g which is within the normal vertical force range to 700g and 1000g which fall outside the normal rpm range. The results were as follows : 1.Analysis of the experiment results revealed that the tome spent in root canal preparation decreased as the vertical forces and rpm increased (p<0.05). Also, the effects of rpm were greater than those of the vertical forces within the normal vertical force range (β-weight test) . 2.Observation of the deformation of GT rotary file revealed that deformation increases in a direct correlation with the vertical force increase and in a reverse correlation with the rpm decrease, In the case of the vertical forces close to the normal range, the probability of GT rotary file deformation were quite different depending on the rpm changes. In the case of◎eater tier◎cal forces, the occurrences of deformation of the file were more frequent regardless of the rpm changes. 3.Deformation and breakage of file were also commonly observed in the expended time measurement experiments and GT rotary file deformation experiments in which low speed rpm (150rpm) was used and at the curved portion of the resin block.         근관감염균에 대한 polyphosphate의 항균효과

        This study was performed to observe the antibacterial effect of polyphosphate (polyP) with various chain lengths (P3~P75) on virulent. invasive strains of P. gingivalis A7A1 28 and W50, and multidrug resistant E. faecalis ATCC29212. P. gingivalis strains were grown in brain heart infusion broth (BHI) containing hemin and vitamin K with or without polyP. PolyP was added at the very beginning of the culture or during the exponential growth phase of the culture. Inhibition of the growth of P. gingivalis was determined by measuring the absorbancy at 540nm of the grown cells. Viable cell counts of the culture and release of intracellular nucleotide from P. gingivalis were measured. E. faecalis was grown in plain BHI with antibiotics alone or in combination with polyP(calgon: 0.1-1.0%) and the bacterial absorbancy was measured. The overall results suggest that polyP has a strong antibacterial effect on the growth of the virulent strains of P. gingivalis and the antibacterial activity of polyP seems largely bactericidal. accompanying bacteriolysis in which chelation phenomenon is not involved. Although polyP does not exert antibacterial activity against E. faecalis, it appears to increase antibacterial effect of erythromycin and tetracycline on the bacterium. Therefore, polyP alone or in combination with antibiotics may be developed as a candidate for the agent controlling oral infections including endodontic infection.

        The Purpose of this study is the provide basic data for bowlers by analyzing the kinematical variables of throwing motion to the male pro bowler using 3-dimensional image analysis method. This researcher took the pictures with a video camera, analyzed them using the Kwon 3D program. 1×4×2m control object installed to get a 3 dimensional space coordinates. Results were as follows. 1. It was indicated that the trunk angle did not change left and right. Back and forth from stance to release motions of three subjects, and the average angle of trunk angle at the stance and that at the tabular side were 19.5˚ and 14.6˚ respectively, which showed they did not deviated from well-known range of forward direction, 15-20˚ and lateral direction 10-15˚. Also the trunk angle of release motion was average 17.1˚ for thalamus side, 41.3˚ for tabular side. 2. It was indicated that the knee angle repeated the increase and decrease to a small degree from the first to the fourth phase, the average angle of left knee at the release motion, the sixth phase was 110.8˚, which was higher than generally known 90˚.

        Compound of the structure of <benz-[f]-indole>-4.9-dione are known to have an antibacterial activity against Gram-positive bacteria. New kinds of 2-amino-<benz-[f]-indole>-4.9-diones are prepared via intramolecular cyclization. 2-Chloro-3-(α-cyano-α-ethoxycarbonyl-methyl)-1.4-naphthoquinone was reacted with some alkylamines (methylamine, ethylamine, ethanolamine, isopropylamine, cyclohexylamine, benzylamine) to yield 2-amino-3-ethoxycarbonyl-N-alcyl-<benz-[f]-indole>-4.9-diones.

      • 급성 심근경색 초기에 도플러 심초음파를 이용한 좌심실 이완기 기능의 평가

        Background : Congestive heart failure after acute myocardial infarction relate to left ventricular systolic sysfunction. Also, left ventricular diastolic function contribute to heart failure. The aims of this study were, firstly to characterize left ventricular diastolic function by transmitral flow velocity, and secondly to study the significance of Doppler measurements in relation to the development of heart failure in the early phase of myocardial infarction. Method : Pulsed Doppler echocardiography of transmitral flow was assessed in 34 patients with acute myocardial infarction. According to the Doppler transmitral flwo velocity profile, the study patients were assigned to the three groups: normal filling pattern (DT > 140ms, IVRT < 100ms), relaxation abnormality (DT > 140ms, IVRT ≥ 100ms), pseudonormal or restrictive pattern (DT ≤ 140ms). Also, on the basis of the presence of heart failure during first week of hospitalization, the patients were divided into two groups: patients with no sign of heart failure (Killip class Ⅰ) and heart failure (Killip class Ⅱ-Ⅳ). Results : Of the 34 patients studied, 10(29%) were normal filling pattern, 17(50%) abnormal relaxation and 7(21%) were restrictive patterns. The left ventricular ejection fraction(EF, 40 ±6%) of restrictive pattern was significantly lower than that of normal filling pattern of abnormal relaxation(p < 0.01). And, the IVRT, DT, E/A and left ventricular EF were reduced in patients with heart failure(14 patients) compared to no sign of heart failure(20 patients)(p<0.05). Of the patients with heart failure, IVRT and left ventricular EF were reduced in patients with DT≤140 ms compared to DT> 140ms(p=0.001). Conclusion : Left ventricular diastolic dysfunction is present early after onset of symptoms of acute myocardial infarction. Assessment of left ventricular diastolic function complements measurements of systolic function in the evaluation of cardiac function. DT, IVRT, E/A and left ventricular EF are useful predictors for development of congestive heart failure following acute myocardial infarction.

      • 최근 광주지역에서 분리된 황색포도상구균의 항생제 내성

        본 연구를 통해 얻어진 연구결과를 요약하면 광주지방에서 분리된 황색포도상구균의 항생제 내성율은 AMXCCV, CPFX, CLM, EM, GM, OXAC, PCV, SXT, VM, CTX, IMPM에 대해 각각 61.0, 65.3, 50.0, 76.5, 76.6, 66.9, 96.7, 15.6, 0, 87.5, 95.5%로 조사되었다. 각각의 항생제에 대해 내성을 보이는 균과 감수성을 보이는 균으로 양분한 다음 다른 항생제들에 대한 감수성 정도를 조사한 결과에 의하면 각각의 항생제에 대해 내성을 보이는 균들은 감수성을 보이는 균들에 비해 다른 항생제에 대해서도 감수성이 낮았다. MRSA는 약 67%로 비교적 높은 비율로 분리되었으나 최근 5년 동안 비율이 크게 변화되지 않았다. MRSA는 사용한 항생제 중 SXT와 VM에 대해서만 높은 감수성을 보였을 뿐 효과가 있는 것으로 알려졌던 다른 항생제들에 대해서는 높은 내성율을 보였다. Objectives : The emergence of Staphylococcus aureus showing multi-drug resistance emphasizes the importance of the prudent use of antibiotics. To survey and obtain the basic information for reappraisal of practical usefulness, we investigated the profiles of antimicrobial resistance of S. aureus isolated recently in Gwangju area. Materials and Methods : S. aureus was isolated from community-hospitals in Gwangju and finally identified by coagulase test. The standard disc diffusion method was used for antimicrobial susceptibility test. Results : The resistance ratio of S. aureus against amoxicillin/clavulanic acid, ciprofloxacin, clindamycin, erythromycin, gentamicin, oxacillin, phenoxymethyl penicillin, trimethoprim/sulphamethoxazole, vancomycin, cefotaxime, and imipenem/cilastatin was 61.0, 65.3, 50.0, 76.5, 76.6, 66.9, 96.7, 15.6, 0, 87.5, and 95.5%, respectively. Resistant strains to each antibiotic were more resistant to the other antibiotics than susceptible strains. The ratio of oxacillin-resistant S. aureus (ORSA) in this area has not increased for recent five years. However, the resistance of ORSA against cefotaxime and ciprofloxacin has increased significantly. ORSA was still susceptible to trimethoprim/sulphamethoxazole (78.3%) and vancomycin (100%) Conclusion : These results offer the latest information about the antimicrobial resistance of S. aureus in Gwangju area and will be used hr reappraisal and selection of antibiotics effective against S. aureus.

        녹차 건분이 고지방식이 급여에 의한 흰쥐의 지질 농도에 미치는 영향

        The effects of dried green tea leaf powders on serum lipid concentrations were evaluated in rats. Sixty male Sprague-Dawley rats weighing 100±10g were divided into six groups and fed high fat diets for six weeks. Experimental groups were administered with following diets: Normal fat diet and normal and high fat diets with 1% dried green tea leaf powders. Tissue weights of live, lung, stomach, heart, kidney and spleen of high fat diet exposed rats were reduced by dried green tea leaf powders groups. The concentrations of serum triglyceride in rats fed the dried green tea leaf powers were lower than those in other groups. The concentrations of total cholesterol in green tea leaf powders the were lower powders green tea were significantly higher than those of other groups. The levels of LDL-cholesterol in serum of the dried green tea leaf powders groups were tended to be lower than those of other groups. GPT and GPT were decreased in dried green tea leaf powders groups and than in the high fat groups. LDHase was lower in the dried green tea leaf powders groups than in the high fat group. These results suggest that dried leaf powders green tea groups may reduce elevated levels of serum lipid concentrations in rats fed high fat diets.
