DJ-1/park7 modulates vasorelaxation and blood pressure via epigenetic modification of endothelial nitric oxide synthase

<P><B>Aims</B></P><P>DJ-1/park7, a multifunctional protein, may play essential roles in the vascular system. However, the function of DJ-1/park7 in vascular contractility has remained unclear. The present study was designed to investigate whether the DJ-1/park7 is involved in the regulation of vascular contractility and systolic blood pressure (SBP).</P><P><B>Methods and results</B></P><P>Norepinephrine (NE) elevated contraction in endothelium-intact vessels in a dose-dependent manner, to a greater extent in DJ-1/park7 knockout (DJ-1/park7<SUP>−/−</SUP>) mice than in wild-type (DJ-1/park7<SUP>+/+</SUP>) mice. Acetylcholine inhibited NE-evoked contraction in endothelium-intact vessels, and this was markedly impaired in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP>. Nitric oxide (NO) production (82.1 ± 2.8% of control) and endothelial NO synthase (eNOS) expression (61.7 ± 8.9%) were lower, but H<SUB>2</SUB>O<SUB>2</SUB> production (126.4 ± 8.6%) was higher, in endothelial cells from DJ-1/park7<SUP>−/−</SUP> mice than in those from DJ-1/park7<SUP>+/+</SUP> controls; these effects were reversed by DJ-1/park7-overexpressing endothelial cells from DJ-1/park7<SUP>−/−</SUP> mice. Histone deacetylase (HDAC)-1 recruitment and H3 histone acetylation at the eNOS promoter were elevated and diminished, respectively, in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP> controls. Moreover, SBP was significantly elevated in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP> controls, but this elevation was inhibited in mice treated with valproic acid, an inhibitor of Class I HDACs including HDAC-1.</P><P><B>Conclusion</B></P><P>These results demonstrate that DJ-1/park7 protein may be implicated in the regulation of vascular contractility and blood pressure, probably by the impairment of NO production through H<SUB>2</SUB>O<SUB>2</SUB>-mediated epigenetic inhibition of eNOS expression.</P>

비매너 주차 단속시스템

주차장에서 일어날 수 있는 비매너 주차로 인한 충돌을 사전에 예방하기 위한 단속시스템이다. 주차장에서 일어날 수 있는 장애인, 전기차 전용 주차구역에 일반차량 주차, 2개 차선을 물고 주차하는 차량이 있다. 위와 같은 차량을 딥러닝 객체인식 기능을 통해 비매너 주차를 감지하여 알려준다. 비매너 주차 상황이 찍힌 사진이나 영상을 학습데이터로 사용하여 상황을 인식할 수 있도록 학습데이터를 제작하고 그 상황을 인식하여 비매너 주차 유무를 판단한다. 주차장의 환경을 좀 더 쾌적하게 함으로써 주차장 이용자간 충돌을 줄이는데 목적이 있다. It is a enforcement system to prevent collisions caused by unmanageable parking that may occur in parking lots. There are handicapped people who can get up in parking lots, general vehicles parked in electric vehicle parking areas, and vehicles parked in two lanes. The vehicle above is detected and notified through the deep learning object recognition function. By using a picture or video of an unmanageable parking situation as learning data, the learning data is produced so that the situation can be recognized, and the situation is recognized to determine the presence or absence of unmanageable parking. The purpose is to reduce collisions between parking lot users by making the environment of the parking lot more comfortable.

이명직의 성서강해 연구

The purpose of this paper is to describe and analyse several Bible expositions (Matthew, Daniels, Songs of Solomon, Romans, Ruth, Hosea, Revelation) written by Lee, Myung-Chick during 1920s. Rev. Lee has been called the father of the Holiness Church for long time. He had been the chief administrator of the Holiness Church and the principal of the Kyung-Sung Bible School (now, Seoul Theological University) for several decades. However, he was also the famous Bible expositor in Korean Church. He wrote many Bible expositions in his life and published them in monthly the Living Water. His exposition of the Bible had influenced the Holiness Church as well as the Korean church in general. In 1920s, Korean society confronted secularization of western culture, communism from Russia, and liberal theology of western church. In this context, Lee had tried to maintain the Full (or pure) Gospel of the Bible. He had criticized secularization very strongly, considered communism as anti-Christ, and attacked liberal theology as a threat of Christianity. After description and analysis of the Bible exposition, the writer concludes the main characteristics of the Bible expositions by Lee, Myung-Chick as followings. First, Rev. Lee tried to read the Old Testament from Christological perspectives. He found many Christological figure in the Old Testament. Second, he stood in the tradition of the Wesleyan Holiness movement, key teaching of which is the Holiness experience by the Baptism of the Holy Spirit. Third, his framework of the Bible interpretation is premillennialism. For Rev. Lee, Chiristian should live in the shadow of the second coming of Christ. Fourth, his emphasis on holiness and the second coming of Christ led to attack secularization as anti-Christ. Christian should not follow this world. Here Holiness movement considered as anti-secularization movement. Finally, Rev. Lee explained the Bible from woman’s perspectives. Of course, Rev. Lee is not feminist. However, all Christian should be a bride of the coming Christ, the bridegroom of all believers. For him, Daniels, Ruth, and the woman of Solomon in the Songs were the examples of true bride of Christ. Church history sometimes considers as history of the Bible interpretation. The Bible interpretation principles of Rev. Lee had heavily influenced the Korean Holiness Church.

Proceeding of International Symposium on East-West Medicine Seoul. 187-200, 1999.-Twirling the needle (TN) is one of several needing methods frequently used for acupuncture in the oriental medicine. The present study was conducted to see if TN enhanced the antinociceptive effect produced by plain-or electroacupuncture in the rat tail flick test. Rats (Sprague-Dawley, 200-300g)lightly anesthetized with thiopental sodium(40mg/kg, i.p.) were used in the tail-flick test. The basal reaction time for tail-flick was 2 sec. For "plain" acupuncture(PA), a needle was inserted for 20 min into a Chock-samni (ST36) acupuncture point which (is located at the anterior tibial muscle and about 10mm below the knee joint). For electroacupuncture(EA), train-pulses(3 Hz,0.3-ms pulse width, 0.2-0.3mA) were applied to the inserted needle for the period of insertion. For combining TN with either PA or EA, TN was performed during the each acupuncture period. Two methods for TN were used; twirling 3 times for 1 second persisted for 1 min in every 5 mins(long-duration and long-interval;LDLI) and twirling 3 times for 1 second persisted for 10 sec in every 1 min(short-duration and short-interval;SDSI). PA and EA produced an increase in tail flick latency (TFL); peak increases were 21.3±5.1% and 42.1±17.3% of the pre-acupunture control, respectively, and occurred immediately after cessation of 20min-acupuncture. Performing TN combined with PA increased TFL more than PA itself by showing a greater peak increase in TFL when performed with a SDSI-TN than with a LDLI-TN(60.6±12.5% and 42.7±22.7% of the pre-acupuncture control, respectively). Performing TN combined with EA also increased TFL more than EA itself and a greater peak increase in TFL was observed with a SDSI-TN as compared to a LDLI-TN(77.5±13.8% and 67.3±14.0% of the pre-acupuncture control, respectively). These results indicate that TN enhances both PA-and EA-produced antinociception where an greater enhanced antinociception than a LDLI-TN. It is suggested that performing a SDSI-TN combined with EA seems to be one of the most effective ways to produce analgesia.

Background and Purposes: Over the past three decades, numerous studies performed in Korea have reported that job stress is a determinant risk factor for chronic diseases and work disability. Every society has its own culture and occupational climate particular to their organizations, and hence experiences different occupational stress. An occupational stress measurement tool therefore needs to be developed to estimate it objectively. The purpose of this study is to develop and standardize the Korean Occupational Stress Scale (KOSS) which is considered to be unique and specific occupational stressors in Korean employees. Subjects and Methods: Data were obtained from the National Study for Development and Standardization of Occupational Stress (NSDSOS Project: 2002-2004). A total of 12,631 employees from a nationwide sample proportional to the Korean Standard Industrial Classification and the Korean Standard Occupational Classification were administered. The KOSS was developed for 2 years (2002-2004). In the first year, we collected 255 items from the most popular job stress measurement tools such as JCQ, ERI, NIOSH and OSI, and 44 items derived from the a qualitative study (depth interview). Forty-three items of KOSS, in the second year, were retained for use in the final version of the KOSS by using Delphi and factor analysis. Items were scored using conventional 1-2-3-4 Likert scores for the response categories. Results: We developed eight subscales by using factor analysis and validation process: physical environment (3 items), job demand (8 items), insufficient job control (5 items), interpersonal conflict (4 items), job insecurity (6 items), organizational system (7 items), lack of reward (6 items), and occupational climate (4 items). Together they explained 50.0% of total variance. Internal consistency alpha scores were ranged from 0.51 to 0.82. Twenty-four items of the short form of the KOSS (KOSS-SF) were also developed to estimate job stress in the work setting. Because the levels of the subscales of occupational stress were gender dependent, gender-specific standard norms for both the 43-item full version and the 24-item short form using a quartile for the subscales of KOSS were presented. Conclusion: The results of this study suggest that KOSS might be an appropriate measurement scale to estimate occupational stress of Korean employees. Further and more detailed study needs to be conducted to improve the validity of this scale.

글루코만난을 첨가하여 제조한 패티가 고지방 식이로 유도된 비만 흰쥐의 체중 감소 및 영양상태에 미치는 영향

This study was carried out to investigate the effect of body weight reduction of patty containing glucomannan in high fat diet-induced obese rats. Male Spraque-Dawley rats were randomly assigned to control and high fat diets groups for 4 weeks. After 4 weeks, the control and high fat diet groups were rearranged into 5 groups by different levels of patty containing glucomannan (60%). After 5 weeks, serum and whole blood levels were analyzed. Obesity index was significantly lower in the group fed patty containing glucomannan than that of high fat diet groups. The status of white blood cells in hematological examination was significantly higher in rats fed high fat diet and not significantly different from those fed patty. Serum albumin levels were not significantly different, while serum glucose levels were significantly different among groups. Serum Triglyceride and total cholesterol levels were the highest in rats fed high fat diets, while the lowest in rats fed patty containing glucomannan.

해조류 에탄올 농도별 추출물의 항돌연변이원성 및 항균성

톳. 미역 및 다시마의 에탄올 농도별 추출물이 MMS. ICR191 및 sodium azide에 의해 유도되는 돌연변이원성의 억제효과 및 E. coli, B. subtilis 및 S. serevisiae에 대한 항균효과를 검정하였다. 톳의 90% 에탄올 추출물은 MMS와 ICR191에 대한 항돌연변이 원성이 각각 70 및 95% 이상으로 매우 높았으나, sodium azide에 대한 항돌연변이원성은 없었다. 미역의 에탄올 농도별 추출물은 30∼90% 에탄올 추출물에서 50% 이상의 항돌연변이 효과를 보았으나 MMS와 sodium azide에 대한 항돌연변이 효과는 없었다. 다시마의 30∼90% 에탄올 추출물의 ICR191에 대한 항돌연변이 효과는 70% 이상이었으나, MMS와 sodium azide에 대한 효과는 없었다. 에탄올 농도별 톳의 추출물들에 대한 항균성 검색 결과 70과 90% 추출물들이 E. coli와 B. subtilis에 대하여 뚜렷한 항균효과는 미역 추출물 보다 다시마의 추출물이 더욱 요과적이었다. 미역 추출물은 50% 추출물만이 S. cerevisiae에 대한 성장저지효과를 나타낸 반면, 다시마 추출물은 90% 에탄올 추출물에서는 시험된 모든 공시균주에 대하여, 그리고 70% 에탄올 추출물에서는 E. coli와 B. sutilis에 대하여 높은 성장저지효과를 나타내었다. Desmutagenic against MMS-, ICR191- and sodium azide-induced mutations, and antimicrobial effects against Escherichia coli, Bacillus subtilis and Saccharomyces cerevisiae by the extracts from Hizikia fusiforme. Laminaria japonica and Undaria pinnatifida with ethanol of a different concentrations were investigated. Desmutagenic effects against MMS and ICR191 of 90% ethanol extracts from Hizikia fusiforme were up 70 and 95%. respectively. no against sodium azide. Desmutagenic effects against ICR191 of 30∼90% ethanol extracts from Undaria pinnatifida and Laminaria japonica were up to 50 and 70%. respectively. but no against sodium azide and MMS. Antimicrobial effect of 70 and 90% ethanol extracts from Hizikia fusiforme had potent antimicrobial activities against E. coli and B. subtilis. Ethanol extracts from Laminaria japonica showed more effective antimicrobial effects than those from Undaria pinnatifida. Only 50% ethanol extract from Undaria pinnatifida showed growth inhibition effect against S. cerevisiae. Seventy percent ethanol extract from Laminaria japonica showed growth inhibition effect against all strains tested. while 90% ethanol extract showed against E. coli and B. subtilis.

Saponin-SOD가 SNUC-4 Cell에 의해 유발된 대장암 Nude Mice의 암 성장 억제 효과

This study was performed to measure the antioxidant effects of red ginseng extracts which antioxidation had been promoted through enzyme hydrolysis. In order to observe their tumor-suppressing effects, an anti-cancer medicine and Saponin-SOD, which was a highly antioxidant beverage made from red ginseng saponin adding SOD-like rice (with embryo buds) extracts, were administered to nude mice with large intestine cancer induced. There was a significant increase in the content of phenolic compounds as the enzyme was added. The red ginseng extracts showed a high electron-donating ability with the passage of time. The electron-donating ability was particularly high in the enzyme-treated red ginseng extract, and also observed as high in Saponin-SOD. The lipid-peroxide generation was inhibited depending on the concentration of Saponin-SOD added; the addition of 0.625% Saponin-SOD served to decrease the inhibition level up to 65% compared with the case of no addition(100%). As a result, it could be assumed that Saponin-SOD would strongly inhibit the oxidation of ghost membrane. After the cancer was induced in nude mice into which Saponin-SOD were injected; the growth of tumors was gradually decreasing with the passage of time after the cancer induction. In particular, when Saponin-SOD was administered together with an anti-cancer medicine, the synergic effect was observed. In conclusion, Saponin-SOD, when used with an anti-cancer medicine, is expected to reduce the amount of free radical and lipid peroxide, which are known to cause harmful effects occurring from the internal application of medicine.

일제하 한국독립운동의 정치사상적 主潮

From the Opening of Ports till annexation by Japan prevailed nationalism like the Confucian traditional morals and the Civilization, and under Japanese colonial rule principle of national self-determination, nationalism, principle of national improvement, socialism, communism. While each thought took each part in Korean nationalist movements, nationalism influenced upon the movements as a basic ideas. Its aim of getting independence from Japan and building up a modern national state was accepted by other thoughts but differently in actual process. Before 3 1 independence Movement, nationalism was expressed by the Righteous Army movements, the Enlightenment and activities of independence Club: the former one was resistant and uncompromising, and the latter two unresistant and compromising. At the Movement, nationalism acted unhesitantly and peacefully, based on the principles of national self-determination, but didn't fulfill its aims. It resulted in re-examining on nationalism and division into the nationalistic rightists and leftists, and filtrating of socialism and communism. In early 1920's nationalistic rightists still led the Enlightenment and the National Improvement movement, insisting on self-strength. But because of Japanese policy of dividing Korean power some of them turned to pro-Japanese ones and consequently some others tended toward being anti-Japanese. A few pro-Japanese rightists were even blamed for being anti-nationalist by the leftists. The leftists, as others, aimed at the national independence and liberation but insisted on resistant and uncompromising activities like people's movements by labors, peasants of students. Generally, characteristics of nationalist movement were reflected in the righteous army movement: unresistant before the annexation, and then resistance. In the meantime, socialists thought that only they could make Korea independent. With basic theories of national liberation and finally establishing socialistic state, they advanced to get independence from Japan through positive and intransigent resistance, namely armed struggle. It was possible for them to join nationalist movements because they criticized imperialism logically when wanted for theories of anti-imperialism, and appeared to overthrow capitalism identically regarded with Japanese imperialism, which recognized as one of important roles in the nationalist movements. At that time, struggle for national liberation based on a kind of theory of socialistic reform but all who jointed it were not socialist or Marxist-Lennist. Some were socialistic nationalists and the others nationalistic socialists. Anyway, even though socialists or communists seemed to play their roles in the national movements, they failed in integrating nationalist movements. As above, each thought influenced upon the national independence movement in each way, but those days Socialists and Communists did not hand out theories specific to Korean situation but took in Russian ones. Finally speaking, Korean political thought under Japanese imperialism have been divided, largely, into nationalism and socialism and had the orientation of political thoughts respectively-unresistant or resistant, moderate of radical, compromising or uncompromising, peaceful or armed. Socialists and Communists regarded themselves as much of national liberation. However, not assessing properly strength of Korean labor and not having specific theories and methods, they failed in associating with nationalists. Anyway, we should notice that they have played a role in national independence.
