        당뇨병환자에서 뇨증 Fibronectin농도의 증가에 관한 연구

        Plasma fibronectin is an α₂-glyoprotein, which is produced by vascular endothelial cells. Raised level of plasma fibronectin has been observed in diabetic patients particularly in the presence of microvascular complications. However, no available data exist about urine level of this glycoprotein in diabetic patients. We measured urine fibronectin level by ELISA methods in 54 diabetic patients who have microvascular comlications or not. The following results were obtained. 1) Urine fibronectin level(㎍/g creatinine) in diabetic group(1740.0±678.0) is increased compaired with those in normal control group(471.0±59.0). but this results were not significantly different among two groups(P>0.05). 2) Urine fibronectin level(㎍/g creatinine) was significantly increased in patients with nephropathy (6188.0±3144.0) compared with those in normal control group(471.0±59.0) and patients without nephropathy(645.0±251.0)(P<0.001). 3) There were a significant correlation between BUN, creatinine, creatinine clearance, 24hr urine total protein and urine fibronectin level in diabetic patients. Our data suggest that urine fibronectin excretion level might be used as a sensitive guide for diabectic nephropathy.

        This study's objects are to survey the reason and background of name change from Science of Physical Education to Sport Science, and the problems of system. This study also comprehends the situation and the research trend of these problems in Germany and Japan, and wants to find the question and the needs at Sport Science. the results are as follows: First, we came to know the trend of world physics changes and advances very fast and innovafively. Under the notion of Sport Science, School physics has changed from Physical Education to Sport Education. Not only the curriculum title but also the curriculum meaning, the treat of teaching aids and the scope of teaching aids have changed, and the contents of sport has been various. Second, Sport Science is authorized and settled, systemically after outgrowing the physics which is the son of Pedagogics. But in scientific structure, paradigm and theory, Sport Science has many works to do. From the results of above study, we can get some solutions. - We should discuss and take an interest in the research matter and academic character for the Sport Science to stand for itself. - We should recognize the need of integrated theory and the importance of interdisciplinary approach. - We should watch out the research trend and world communication. - We should endeavor to use strict special words including the name problem, and to get the originality of Sport Science.

      • 인체 대장암에서 72kDa 제 4형콜라겐 분해효소의 발현

        수술 절제한 대장암 45예에서 Northern blot analysis와 gelatin zymography를 시행하여 72kDa 제 4 형콜라겐 분해효소의 mRNA치와 활성도를 조사하였다. 대장암에서 주위 정상조직에 비해 이 효소의 mRNA치가 증가하였으며 단백질 합성단계에서 효소의 활성도와도 연관되었다. 특히 활성형인 62kDa의 활성도가 크게 증가하였다. 암진행에 따른 이 효소의 활성도 변화는 T2와 T3에서 암조직과 정상조직사이에서 mRNA치와 효소활성도에 차이가 있었으나 전반적으로 암의 침윤도, 림프절 전이, 듀크스 병기에 따른 유의한 차이는 볼 수 없었다. 암침윤과 전이에는 제 4 형콜라겐 분해효소 외에 딴 metalloproteinase들, plasminogen activator등 여러 가지 다른 단백질 분해효소도 관여하며, 암종류에 따라 이들 효소의 작용정도가 차이가 날 수 있다. 또 조직 억제제인 tissue inhiitors of metalloproteinase(TIMPs)와의 균형관계도 고려해야 한다. 그러므로 대장암에서 72kDa 제 4 형콜라겐 분해효소의 증가된 활성양상을 이들 다른 인자들과 함께 관찰하면 암의 진행정도를 나타낼 수 있는 보다 뚜렷한 지표를 만들 수 있을 것으로 생각된다. Type Ⅳ collkagenases are thought to play a role in the proteolysis of the basement membrance and the extracellular matrix by the malignant cells that occur early in the development of invasive tumors. In several human tumors, including breast, colonic, ovarian, and gepatocellular carcinomas, Type Ⅳ collagenase expression has been positively correlated with tumor invasiveness. In this study, we examined the expression and the activation of 72kDa Type Ⅳ collagenase in cancerous and normal tissues of the colon and, we attempted to clarify its clinical significance in 45 cases of human colorectal cancer by using northern blot analysis and gelatin zymography. This study showed that the level of 72kDa Type Ⅳ collagenase mRNA was increased in colorectal cancer and that this increase was associated with increased enzymatic activity. While there was a difference in the expression and the activation of this enzyme between T2 and T3, this enzyme activity did not show a definite correlation with cancer progress as measured by the depth of tumor invasion and lymph node metastasis.

        야뇨증 환아들의 심리사회적 특성에 대한 다기관 연구 : 행동 및 정서 문제를 중심으로

        Objectives : The aim of this study was to examine the behavioral and emotional problems associated with nocturnal enuresis in Korean children. Methods : Three hundred eighteen children with nocturnal enuresis, together with their parents, completed the Child Behavior Checklist (CBCL), Disruptive Behavior Disorder Scale according to DSM-IV (DBDS), Children's Depression Inventory (CDI), State-Trait Anxiety Inventory for Children (STAIC), and Piers-Harris Children's Self-Concept Scale (PHCSC). Ninety-three normal students were selected as the control group. Results : Compared to the normal control group, the mean scores with regard to the withdrawn, social problems, attention problems, delinquent behavior, aggressive behavior, externalizing problems and total problems profiles were significantly higher in the nocturnal enuresis group according to the CBCL results. The nocturnal enuresis group also scored significantly higher in the ADHD and ODD profiles of the DBDS. The nocturnal enuresis group was more depressed and anxious than the control group according to the results of the CDI and STAI. The mean score of the PHCSC was significantly lower in the nocturnal enuresis group when compared to the normal control group. Conclusion : The results of this study suggest that children with nocturnal enuresis in Korea have clinically relevant behavioral and emotional problems. The findings support the link between nocturnal enuresis and psychopathology in Korean children.

        Phosphorylation of Hepatitis C Virus RNA Polymerases Ser29 and Ser42 by Protein Kinase C-Related Kinase 2 Regulates Viral RNA Replication

        <P>Hepatitis C virus (HCV) nonstructural protein 5B (NS5B), an RNA-dependent RNA polymerase (RdRp), is the key enzyme for HCV RNA replication. We previously showed that HCV RdRp is phosphorylated by protein kinase C-related kinase 2 (PRK2). In the present study, we used biochemical and reverse-genetics approaches to demonstrate that HCV NS5B phosphorylation is crucial for viral RNA replication in cell culture. Two-dimensional phosphoamino acid analysis revealed that PRK2 phosphorylates NS5B exclusively at its serine residues <I>in vitro</I> and <I>in vivo</I>. Using <I>in vitro</I> kinase assays and mass spectrometry, we identified two phosphorylation sites, Ser29 and Ser42, in the Δ1 finger loop region that interacts with the thumb subdomain of NS5B. Colony-forming assays using drug-selectable HCV subgenomic RNA replicons revealed that preventing phosphorylation by Ala substitution at either Ser29 or Ser42 impairs HCV RNA replication. Furthermore, reverse-genetics studies using HCV infectious clones encoding phosphorylation-defective NS5B confirmed the crucial role of these PRK2 phosphorylation sites in viral RNA replication. Molecular-modeling studies predicted that the phosphorylation of NS5B stabilizes the interactions between its Δ1 loop and thumb subdomain, which are required for the formation of the closed conformation of NS5B known to be important for <I>de novo</I> RNA synthesis. Collectively, our results provide evidence that HCV NS5B phosphorylation has a positive regulatory role in HCV RNA replication.</P><P><B>IMPORTANCE</B> While the role of RNA-dependent RNA polymerases (RdRps) in viral RNA replication is clear, little is known about their functional regulation by phosphorylation. In this study, we addressed several important questions about the function and structure of phosphorylated hepatitis C virus (HCV) nonstructural protein 5B (NS5B). Reverse-genetics studies with HCV replicons encoding phosphorylation-defective NS5B mutants and analysis of their RdRp activities revealed previously unidentified NS5B protein features related to HCV replication and NS5B phosphorylation. These attributes most likely reflect potential structural changes induced by phosphorylation in the Δ1 finger loop region of NS5B with two identified phosphate acceptor sites, Ser29 and Ser42, which may transiently affect the closed conformation of NS5B. Elucidating the effects of dynamic changes in NS5B phosphorylation status during viral replication and their impacts on RNA synthesis will improve our understanding of the molecular mechanisms of NS5B phosphorylation-mediated regulation of HCV replication.</P>
