        The blood compatibility of S(sulfur)-cyanoethylated keratin(SCEK) was studied. SCEK was obtained by rupture the cystine linkage of wool keratin followed by cyanoethylation. Platelet count, prothrombin time and activated partial thromboplastin time were measured by Coulter counter to investigate the behavior of blood on the surface of SCEK and other four synthetic materials. Enzyme linked immuno sorbent assay was adopted to measure platelet factor IV released from agranule in platelet. The contact an히e and gas permeation rate of SCEK were also measured.

        Seasonal variation in longitudinal connectivity for fish community in the Hotancheon from the Geum River, as assessed by environmental DNA metabarcoding

        Background: Longitudinal connectivity in river systems strongly affects biological components related to ecosystem functioning, thereby playing an important role in shaping local biodiversity and ecosystem health. Environmental DNA (eDNA)-based metabarcoding has an advantage of enabling to sensitively diagnose the presence/absence of species, becoming an efficient/effective approach for studying the community structure of ecosystems. However, little attention has been paid to eDNA-based biomonitoring for river systems, particularly for assessing the river longitudinal connectivity. In this study, by using eDNA we analyzed and compared species diversity and composition among artificial barriers to assess the longitudinal connectivity of the fish community along down-, mid- and upstream in the Hotancheon from the Geum River basin. Moreover, we investigated temporal variation in eDNA fish community structure and species diversity according to season. Results: The results of species detected between eDNA and conventional surveys revealed higher sensitivity for eDNA and 61% of species (23/38) detected in both methods. The results showed that eDNA-based fish community structure differs from down-, mid- and upstream, and species diversity decreased from down to upstream regardless of season. We found that there was generally higher species diversity at the study sites in spring (a total number of species across the sites [n] = 29) than in autumn (n = 27). Nonmetric multidimensional scaling and heatmap analyses further suggest that there was a tendency for community clusters to form in the down-, mid- and upstream, and seasonal variation in the community structure also existed for the sites. Dominant species in the Hotancheon was Rhynchocypris oxycephalus (26.07%) regardless of season, and subdominant species was Nipponocypris koreanus (16.50%) in spring and Odontobutis platycephala (15.73%) in autumn. Artificial barriers appeared to negatively affect the connectivity of some fish species of high mobility. Conclusions: This study attempts to establish a biological monitoring system by highlighting the versatility and power of eDNA metabarcoding in monitoring native fish community and further evaluating the longitudinal connectivity of river ecosystems. The results of this study suggest that eDNA can be applied to identify fish community structure and species diversity in river systems, although some shortcomings remain still need to be resolved.

        급성 A형 간염에 동반된 Gianotti-Crosti 증후군 1예

        Patient with Gianotti-Crosti Syndrome (GCS) presents with a distinctive self-limiting acral papular or papulovesicular eruption. Patient with GCS is associated with an underlying viral and bacterial illness. GCS is a self-limiting cutaneuous response to different infections. Its clinical differences are probably due to individual characteristics of each patient rather than the causative infections agents. GCS in patients infected with hepatitis A virus has not been reported previously in Korea. We report a case of 16-year-old girl who developed an erythematous papular eruption on her extremities a week after an epidemic of hepatitis A. The patient had the characteristics which resembled clinical and histopathologic findings of GCS. Hepatitis A virus RNA was detected by nested reverse transcription-polymerase chain reaction of tissue from lesion site of the patient.

        Major Compound Analysis and Assessment of Natural Essential Oil on Anti-Oxidative and Anti-Microbial Effects

        본 연구에서는 화장품산업에 이용 중인 천연 에센셜 오일 8종(라벤더, 티트리, 로즈마리, 주니퍼베리, 편백, 싸이프러스, 시더우드, 파인)을 가지고 물리 화학적 분석과 생리활성 및 항균효과를 실험하였다. 라벤더오일의 경우 에스테르 성분인 linalyl acetate가 48% 함유되어 있었으며, 5,000 ppm의 농도에서 22.36%의 항산화능을 보였다. 티트리 오일의 경우 0.5% 농도에서 43.94% 항산화능을 보였으며 항균효과 또한 뛰어났다. S. aureus, S. epidermidis, S. mutans, C. albicans 균에 각각 6, 3.5, 6.5, 5 mm의 저해환이 측정되어 매우 우수한 품질의 에센셜 오일임이 확인되었다. 편백 오일의 경우 pH가 2.64로 가장 산성도가 높았으며, 세스퀴테르펜 성분이 19.20%로 확인되었다. 시더우드 오일의 경우 샘플 중 비중과 굴절률이 가장 높았으며 전체성분의 99.73%가 세스퀴테르펜으로 구성되어 있었다. 또한 5,000 ppm 농도에서 39.68% 항산화능을 보였다. 특히 1% 농도에서 P. acnes, P. ovale, C.albicans 균에 각각 3.5, 6, 6 mm의 저해환이 측정되어 매우 탁월한 항균효과를 보였다. 반면 세스퀴테르펜 함량이 증가할수록 비교적 항산화능도 높게 나오는 경우도 있지만 그렇지 않은 경우도 있어서 성분조성보다는 각각의 항산화 성분 유무에 따라 항산화능이 결정된다고 판단되며, 마찬가지로 항균력 또한 테르펜의 성분조성보다는 각각의 항균성분의 유무에 따라 항균력이 결정된다고 사료된다. We studied the physical, chemical, biological, and antimicrobial effects of eight types of essential oils used in the cosmetics industry: lavender, tea tree, rosemary, juniper berry, Chamaecyparis obtusa, cypress, cedar wood, and pine. Lavender oil had a linalyl acetate (an ester chemical compound) content of 48% and radical scavenging activity of 22.36% at 5,000 ppm. Tea tree oil had radical scavenging activity of 43.94% at 5,000 ppm and antimicrobial activity against S. aureus, S. epidermidis, S. mutans, and C. albicans in each 6, 3.5, 6.5, and 5 mm, respectively. Chamaecyparis obtusa oil had the highest acidity (pH 2.64) compared with the other oils, and sesquiterpene compounds were found to have 19.20%. Cedar wood oil had the highest specific gravity and refractive index compared to the other oils and had a sesquiterpene content of 99.73%. The radical scavenging activity of cedar wood essential oil exceeded 39.68% at 5,000 ppm. The clear zone, indicating antimicrobial activity against P. acnes, P. ovale, and C. albicans, was 3.5, 6, and 6 mm, respectively, at a concentration of 1% cedar wood oil. Results showed that with a high sesquiterpene content, the antioxidant effect was generally, but not always, high, suggesting that this is determined according to composition of the compound rather than presence of each antioxidant. The results indicate that antimicrobial activity is determined by the existence of each antimicrobial ingredient rather than terpene composition.

      • LNG/LPG관련 해양사고 시나리오 구축에 대한 고찰

        LNG/LPG는 폭발가능성이 크기 때문에 매우 위험한 물질이다. LNG/LPG는 gas상태의 연료를 극 초저온에 고 압력 상태로 고농축한 액체 연료이다. 온도나 압력에 따라 물질의 상태가 변하기 때문에 폭발이나 화재가 일어날 수 있다. 하지만 무조건 사고가 일어나는 것은 아니며 현재까지도 화재나 폭발이 일어난 경우가 드물다. LNG/LPG선박은 충돌이나 파손 등 사고위기가 클수록 위험하다. 사고위기가 일어나는 요소로는 복잡한 항로, 많은 선박 수, 해마다 증가하는 LNG/LPG의 수요량 등이 있다. 본 연구에서는 LNG/LPG관련 해양사고 시나리오를 만들기 위해 사고 연계 고리(Accident chain)를 만들어 분석하였다. 해양사고의 연계 고리를 만들기 위해서는 story가 필요하다는 것을 알게 되었고, Risk를 통해 Peril과 Hazard를 분석 할 수 있었다. LNG/LPG의 위험성은 고압에 기인하는 위험, 화재위험, 동상위험, 화학반응의 위험, 질식 위험 등으로 분류 할 수 있었다. 아직까지는 LNG/LPG선박의 화재 및 폭발사고는 거의 일어나지 않았으나 매년 그 수요량이 증가하고 있고, 매우 위험한 물질임을 본 연구를 통해 알 수 있었다.

        개회충 유충 감염에서 발생되는 호산구성 폐질환의 특성

        연구배경: 개흡충증은 한국에서 호산구증다증과 호산구성 폐질환을 일으키는 흔한 원인의 하나이다. 우리는 개흡충증에 의한 호산구성 폐질환의 특장을 분석하였다. 방법: 2001년 9월 1일부터 2006년 3월 30일까지 개회충 2기유충에 의한 호산구증다증으로 진단 받은 141명의 환자들을 대상으로 하였다. 본 연구를 위해 단순 흉부 방사선 촬영. 흉부 컴퓨터단층촬영, 기관지내시경, 기관지폐포세척술과 각종 혈액학적 검사를 시행하였다. 개회충증의 진단은 개회충 2기유충의 순수 분비배설 항원을 사용하였다. 결과: 141명의 환자 중 32명이 개회충에 의한 호산구성 폐질환으로 진단되었다. 젖빛유리양상이 컴퓨터단층촬영에서 지배적으로 23/32명(71.9%)이었다. 13/32명(40.6%)은 무작위적 구역적 분포를 보였다. 흉수액이 9/32명(28.1%)에서 관찰되었다. 28/32명(87.5%)이 호흡기 증상을 호소하였다. 11/32명(34.4%)은 호흡기 증상과 소화기 증상을 동시에 호소하였다. 12/32명(37.5%)에서 간에 국소의 저음영의 침윤을 보였다. 결론: 개회충에 의한 호산구성폐질환의 임상적, 방사선적 특성은 다발성의 결절성 또는 경화성의 젖빛유리음영이 양폐 상하엽에 무작위적 분포를 하는 것이다. 앞으로 우리나라에서 호산구 증다증과 폐침윤을 보이는 성인에서 감별진단을 위해 개회충증의 가능성을 고려해야 할 것으로 판단된다. Background: Toxocariasis is a common cause of eosinophilia and eosinophilic lung disease in Korea. We analyzed the characteristics of eosinophilic lung disease in toxocariasis. Method: One hundred and forty one patients with eosinophilia caused by a toxocara larval infection were evaluated from September 1, 2001 through March 30, 2006. The plain chest x-ray, chest CT(computed tomography), and bronchoalveolar larvage(BAL) were examined. A diagnosis of toxocariasis was made by ELISA using that secretory-excretory antigen from the T. canis larvae. Results: Toxocarial eosinophilic lung diseases was diagnosed in 32 out of 141 patients. Ground glass attenuation was the main feature on the CT scans in 23 out of 141 patients (71.9%). Thirteen patients (40.6%) had a random in zonal distribution on CT. Pleural effusion was observed in 9 patients (28.1%). Twenty eight patients (87.5%) complained of respiratory symptoms. Eleven patients (34.4%) had gastrointestinal symptoms and 12 patients (37.5%) had liver infiltration. Conclusions: The most common findings of the chest CT in patients with toxocariasis was a randomly distributed ground grass attenuation. A toxocara infection should be considered in a differential diagnosis of patients who exhibit pulmonary infiltration with eosinophilia in Korea. (Tuberc Respir Dis 2007; 62: 19-26)

        SMP30-mediated synthesis of vitamin C activates the liver PPARα/FGF21 axis to regulate thermogenesis in mice

        The vitamin-C-synthesizing enzyme senescent marker protein 30 (SMP30) is a cold resistance gene in Drosophila, and vitamin C concentration increases in brown adipose tissue post-cold exposure. However, the roles of SMP30 in thermogenesis are unknown. Here, we tested the molecular mechanism of thermogenesis using wild-type (WT) and vitamin C-deficient SMP30-knockout (KO) mice. SMP30-KO mice gained more weight than WT mice without a change in food intake in response to short-term high-fat diet feeding. Indirect calorimetry and cold-challenge experiments indicated that energy expenditure is lower in SMP30-KO mice, which is associated with decreased thermogenesis in adipose tissues. Therefore, SMP30-KO mice do not lose weight during cold exposure, whereas WT mice lose weight markedly. Mechanistically, the levels of serum FGF21 were notably lower in SMP30-KO mice, and vitamin C supplementation in SMP30-KO mice recovered FGF21 expression and thermogenesis, with a marked reduction in body weight during cold exposure. Further experiments revealed that vitamin C activates PPARα to upregulate FGF21. Our findings demonstrate that SMP30-mediated synthesis of vitamin C activates the PPARα/FGF21 axis, contributing to the maintenance of thermogenesis in mice.
