        톨루엔 노출 근로자에서 유전자적 요인, 생활습관 및 식이가 요중 마뇨산 배설에 미치는 영향

        목 적 : 본 연구는 생물학적 지표 특성 연구를 위한 단면조사 연구로서 톨루엔 노출과 톨루엔의 생물학적 모니터링으로 이용되는 요중 마뇨산 농도간의 양-반응 관계에서 효과 변경인자로서 작용하는 유전자 ALDH2, CYPIAI, CYP2El의 다양성 분포를 조사하고, 이들 유전자가 요중 마뇨산 배설 농도에 미치는 영향을 알아보고자 하였다. 방 법 : 연구대상자는 경인지역에 위치한 22개의 사업체에서 페인트 제조 72명, 제조업에서 도장작업 50명, 인쇄업 12명, 접착작업 12명, 벽지의 코팅작업 14명등 160명의 근로자이었다. 성, 연령, 유기용제 작업장 근무경력, 흡연습관, 음주습관, 검사일 이전 밤에 음주 여부, 시료 채취전 6시간 이내에 섭취한 식품조사, 측정일 개인보호구의 착용 여부에 대하여 설문조사를 시행하였다. 톨루엔에 대한 개인노출량은 passive air sampler로 측정하였고, 요중 마뇨산은 요중 크레아틴으로 보정하여 측정하였다. 또한 aldehyde dehydrogenase 2 (ALDH2) , cytochrome P45O IAI(CYPIAI), 그리고 cytochrome P45O 2El(CYPfEl)의 유전적 다양성은 혈액으로부터 백혈구의 DNA를 추출하여 제한 효소 절단 길이 다양성 (restiction fragment length polymorphism, RFLP)법으로 검사하여 다음의 결과를 얻었다. 결 과 : 연구대상자의 요중 마뇨산 농도의 기하평균은 0.44g/g creatinine, 기하표준편차 2.80이었다. 톨루엔 노출 농도, 개인 보호구 착용여부, 안식향산이 함유된 식품의 섭취여부중 요중 마뇨산 배설농도에 영향을 미치는 인자는 톨루엔 노출 농도뿐이었다. ALDH2, CYPIAI, CYPfEl, 연령, 흡연, 근무기간에서 회귀계수간에 차이를 보이는 경향이 있었다. 요중 마뇨산 농도에 대한 다중회귀분석에서 톨루엔, ALDH2, CYPIAI, CYP2El유전자형의 회귀계수가 통계적으로 유의하였다. 결 론 : 톨루엔 노출로 인한 요중 마뇨산 배설 농도는 ALDH2, CYPIAI, CYP2El유전자 다양성에 의해 영향을 받기 때문에, 유전자형의 대사능의 차이를 고려하여 마뇨산 기준 농도를 찾는 연구가 이루어져야 하겠다. Objectives The purpose was to investigate the distributions and the effects of genetic polymorphism of aldehyde dehydrogenase 2(ALDH2), cytochrome P45O IAI (CYPIAI), and cytochrome P45O 2El (CYPfEl) on the toluene metabolism. Methods : The subacts consisted of 160 workers who were exposed to toluene in different industries such as paint manufacturing, painting on steel and wood products, printing, bonding, and coating. The exposed toluene level was monitored by passive air sam paler, and the questionnaire variables were age, sex, smoking, drinking, previous night's drinking, use of personal protectlve equipment, work duration, and taking benzoic acid containing food. The urinary hippurric acid collected in the end of shift was corrected by urinary creatinine concentration. The genotypes of ALDH2, CYPIAI, and CYP2El were investigated using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) methods with DNA extracted from venous blood. Results'The geometric mean and the geometric standard deviation of urinary hippuric acid concentration were 0.44 g/g creatinine and 2.80. The urinary hippuric acid concentration was significantly related to personal exposed toluene level among personal exposed toluene level, use of personal protective equipment, and benzoic acid containing food diet. The slope differences of the regression for ALDH2, CYPIAI, and CYP2E1 genetic polymorphism, age, smoking, and work duration tended to be significant. In multiple regression analysis, the regression coefficient of toluene, ALDH2, 7YPIAl, CYP2E1 genetic polymorphism were significant. Conclusions : From the above results, urinary hippuric acid level after toluene exposure was significantly affected by the genetic polymorphism of ALDH2, CYPIAI, CYPfEl. It is needed further investigation of the urinary hippuric acid level considering the effect of genetic polymorphism.

        한천분해효소를 생산하는 해양세균 Thalassomonas sp. SL-5의 분리 및 특성

        A novel agar-degrading bacterium SL-5 was isolated from seashore of Homigot at Kyung-Buk province, and cultured in marine broth 2216 media. The bacterium SL-5 was identified as Thalassomonas genus by 16S rDNA sequencing with 96% identity. Growth rate was faster at $27^{\circ}C$ than at $37^{\circ}C$ and agarase was produced as growth-related. The optimum pH of the enzyme activity was 7.0 and the optimum temperature for the reaction was $40^{\circ}C$. Although the enzyme had no thermostability, the enzyme activity was remained over 80% at $60^{\circ}C$. The enzyme hydrolyzed neoagarohexaose to yield neoagarobiose as the main product, indicating that the enzyme is $\beta-agarase$. Thus, the enzyme would be useful for the industrial production of neoagarobiose. 포항 호미곶 해수에서 한천분해활성을 보이는 해양성 세균 SL-5을 분리하였으며 16S rDNA 염기서열분석으로 해양기원의 Thalassomonas 속과 가장 유사한 균주임을 확인하였다. SL-5 균주가 생성하는 한천분해효소(agarase)는 성장의존성인 것으로 확인되었고 효소활성을 위한 최적 pH는 pH 7.0(20 mM sodium phosphate 완충용액)이고 최적 온도는 $40^{\circ}C$로 나타났다. 한천분해효소는 $80^{\circ}C$까지 65%의 효소활성을 보이는 호열성 효소이지만, 내열성은 그리 높지 않았다. 한천 분해효소의 분해산물에 대한 TLC 분석결과, 본 효소가 $\beta-agarase$라는 사실을 확인할 수 있었다. 분리한 Thalassomonas sp. SL-5 균주가 생산하는 한천분해효소를 이용하여 한천으로부터 다양한 기능성 한천올리고당을 생산하는데 있어 유용하게 사용될 것으로 기대된다.

      • 생쥐 뇌조직 체외배양법을 활용한 향산화제의 향독성효과에 관한 연구

        In an effort to establish an in vitro model of Parkinson's disease (PD), we prepared slice cultures of midbrain from newborn mouse. The slice section containing substantia nigral region was cultured for 10days. The striatal culture was grown to preserve the distribution of interneurons with minimal contamination of any other brain tissue. Using this explant culture containing both glial cells and nitro-striatal dopamine neurons, the protective effect of antioxidant pineal hormone and melatonin on 1-methyl-4-phenyl-1, 2,3,6-tetrahydropyridine (MPTP) -induced loss of the dopamine neurons was investigated. MPTP selectively damaged dopaminergic neurons, as indicated by immunoblot and immunohistochemistry for glial fibrillary acidic protein (GFAP) and tyrosine hydroxylase, the marker enzymes for glial and dopaminergic neurons, respectively. Twenty-four-hour treatment of MPTP (50 μM) in the explant culture induced 37% loss of dopaminergic neurons while no apparent damage was observed in glial region. Type B monoamine oxidase (MAO-B) that resides in glial cells, produces free radicals during catecholamine metabolism and also oxidizes MPTP to its active form, MPP+, was activated by 2.2 times after MPTP treatment. Melatonin was able to recover both the dopaminergic neuronal loss and the elevated MAO-B activity level fully to the untreated control level. Similar observation was made bur the co-treatment of vitamin E with MPTP, suggesting the generation of oxygen radical species by MPTP. On behalf of pursuing molecular mechanisms of Parkinsonism, the data illustrate the proposed mechanism of MPTP-induced dopaminergic cell death ; namely, MPP+ that is processed by MAO-B in glial cells is taken up avidly by dopaminergic neurons and causes a toxic effect at least partly, if not mostly, through oxidative stress. The culture system, therefore, can serve as an experimental model of PD in which the putative neuroprotective effects of antioxidants can be investigated. This model is also useful for screening novel drugs and growth factors without a massive sacrifice of animals, and ultimately for developing neuroprotective therapies.

        운송업 종사 근로자의 건강가치 인식 및 건강증진 행위와 건강진단 결과와의 관련성

        목 적 : 건강진단 결과 건강인과 요관찰자 및 일반질환자를 포함한 비건강인에서 건강가치인식 및 건강증진행위 실천정도를 알아보고, 건강인과 비건강인으로 층화한후 건강가치인식이 높은 군과 낮은 군에서 건강증진행위 실천에 차이가 있는지를 비교함으로써 바람직한 건강증진 프로그램 개발과 적용을 위한 기초자료를 제공하고자 수행되었다. 방 법 : 1998년도 건강진단을 받은 철도운송업 종사 근로자 4,919명중에서 자기기입식 설문에 응답한 건강인 233명, 요관찰자 172명, 일반질환자 160명을 연구대상으로 하였다. 결 과 . 삶의 가치 중 건강을 가장 가치있게 인식하고 있는 근로자가 건강인에서 37.3%, 비건강인에서 36.1%로 두 군간에 차이가 없었다. 건강증진 행위 실천에 있어 비 건강인이 건강인에 비하여 운동영역의 실천정도가 유의하게 낮았으며 대인관계 지지영역은 유의하게 높았다. 그 외의 영양, 스트레스관리, 자아실현 및 건강책임 관련 영역의 실천정도는 건강인과 비건강인 사이에 유의한 차이가 없었다. 건강인에서의 연령 변수를 제외하고 건강인 및 비건강인 모두 일반적 특성에 따른 건강가치 인식도에 차이가 없었다. 건강가치 인식도에 따른 건강증진행위 실천 정도는 건강인의 경우 건강가치 인식이 높은 군이 낮은 군에 비하여 자아실현 및 건강책임 영역의 실천 정도가 유의하게 높았으며, 비건강인의 경우 유의한 차이를 보이는 영역은 없었다. 결 론: 건강가치 인식, 건강증진행위 실천 정도와 건강진단 결과와의 관련성은 매우 낮게 나타나 건강증진 프로그램 개발과 적용에 있어 인식보다는 실천에 촛점을 두어야 될 것으로 판단된다. Objectives : This study was performed to compare health value cognition and health promotion behavior compliance between healthy group and not being healthy group, and to evaluate the difference of health promotion behaviors compliance between high group and low group in health value cognition. Methods . The subjecus of this study were 565 wor17ers, 233 healthy workers, 172 observation cases and 160 disease cases, selected from 4,919 transit coporation's workers. Results : In health promoting behavior compliance, not belng healthy group showed lower level than healthy group in exercise and showed higher level than healthy group in interpersonal support significantly. A worker who replied on "health" as the most value in life was consisted in 37.3% of healthy group and 36.1% of not belng healthy group, Health value cognition according to general characteristics showed no significant difference except only variable of age in healthy group. Health promoting behavior compliance according to cognition of health value showed in healthy group that a high level group of health value cognition was more significant difference than a low level group in self aclualization and health responsibility of health promoting behavior and didn't show difference significantly in not being healthy group. Conclusions : In conclusion, the relationship between health value cognition, health promotion behavior compliance and periodic health examination results showed weakly. Therefore, in order to develop and apply spontaneous health promotion program, it was considered that should emphasize compliance than cognition.

        ARABON과 CARBON 섬유 여과포의 코팅에 의한 내열성 향상에 관한 연구

        고온에서 집진 성능이 우수하고 저렴하게 사용할 수 있는 합성섬유 여과포의 코팅기술을 개발하기 위한 내열성 코팅 실험을 수행하였다. 코팅된 여과포의 내열실험을 300℃ 및 400℃에서 수행한 후, 여파포의 무게변화, 두께변화, 수축률을 측정하여, 내열 특성을 측정하였다. 가열 전후의 무게 변화를 측정한 결과 ARABON 여과포는 300℃에서 36.3% 감소하였고, CARBON 여과포는 38.9%로 감소하였으며, 수축현상은 ARABON이 13.7%로 약간 변화되었지만 CARBON 여과포는 21.7%로 조금 더 변화되었다. 고온에서 합성섬유 여과포의 내열성능을 나타내기 위하여 변화지수, CI를 정의하여 사용하였다. 300℃와 400℃에서 CARBON 여과포의 가장 우수한 코팅물질은 Al(NO₂)₃이었다. This study has been accomplished by the coating experiments to develope the synthetic fabrics with thermal resistance, which are the economic and efficient filter at high temperature. The thermal resistance of the fabric filter was experimentally determined by the changes of the weight, thickness, and contraction of the coated and non-coated filters. The tested temperatures were 300℃ and 400℃. The weight losses of the ARABON and CARBON fabrics are 36.3% and 38.9%, and the contractions are 13.7% and 21.7% at 300℃ compared to 400℃ respectively. The change index, Cl, was defined and used for providing the thermal resistant information of the tested fabrics. The most coating material is Al(NO₂)₃ for CARBON fabrics at 300℃ and 400℃.

      • 성견에서 수직적 골결손부에 골이식후 교정력 적용시기에 따른 치주조직의 반응

        The aim of this study was to investigate the periodontal response according to the timing of orthodontic force application after bone graft into the angular bony defect. Nine dogs were divided into three groups, 2, 4, and 6 weeks, according to the timing of orthodontic force application after bone graft. Periodontal angular bony defects were created surgically at the distal aspect of both maxillary third incisors. Two weeks later, flap operation was performed to eliminate inflammation and reference notch was made on the root surface at the level of the bottom of each defect. Demineralized freeze-dried bone was implanted on the left side whereas only debridement was done on the other side. Experimental tooth movement was executed during 8 weeks on both graft and non-graft sides. After 2 weeks of retention period, animals were sacrificed for histologic specimens. The results were obtained as follows: 1. New bone formation was more pronounced in the graft side than in the non-graft side in all experimental animals. 2. In the 6-week group, new bone and cementum formation was observed in more than half from the notch to the cemento-enamel junction, and the zone of connective tissue attachment was found without apical migration of junctional epithelium. 3. In the 4-week group, the amount of new bone formation was smaller than in the 6-week group whereas the overall remodeling pattern was similar. 4. New bone formation was confined to around the notch and the junctional epithelium migrated apically to the level of the notch with no connective tissue attachment and cementum formation in the 2-week group. The results of the present study suggest that periodontal response may be influenced by the timing of orthodontic force application after bone graft into angular bony defect.

        중학교 2학년 학생들의 활동과제 해결과정 연구

        본 연구의 목적은 활동과제를 해결하는 동안 학생들이 나타내는 수학적 사고와 수학적 창의성을 분석하여 활동과제를 활용한 수학 수업에 대한 교육적 시사점을 찾는 것이다. 중학교 2학년 남학생 2명,여학생 2명을 대상으로 매 차시 120분씩 총 8차시의 실험 수업을 진행하는 사례연구를 실시하였다. 실험 수업의 결과 활동과제는 학생틀에게 그 자체로 동기유발이 되었고,교사에게 의존하지 않으면서 자신의 특성과 수준에 맞는 자연스러운 사고를 경험하게 하여 학습자 중심의 수학 교육을 가능하게 한다는 결론을 내릴 수 있었다. 그리고 학생들이 나타낸 수학적 사고와 수학적 창의성을 Polya의 문제해결과정 4단계에 따라 분석하여,활동과제를 활용한 수학수업에서 수학적 사고와 수학적 창의성을 향상시키기 위한 교수학적 제안점을 도출하였다. The purpose of this research is to provide the fundamental information about mathematics classes based on activism by analyzing mathematical thinking and mathematical creativity students display while solving tasks based on activism. A case study was conducted with two male middle school 2nd graders and two female middle school 2nd graders as subjects, and was composed of a total 8 experimental teaching sessions, each lasting 120 minutes. As result of analyzing mathematical thinking and mathematical creativity students display based on Polya's four Phase of problem solving process/ the type of mathematical thinking were intuitive insight, systematization of information, spatialization/ visualization, mathematical abstraction, inductive reasoning, deductive reasoning, generalization/application, reflective thinking and they were usually displayed collectively, but also independently. 8 types of mathematical thinking at the question understanding phase were displayed, 19 ones at the question understanding phase, 24 ons at the plan-execution phase, 9 ons at the reflection phase. The type of mathematical creativity were fluency, fluency, originality, ingenuity, and they were displayed collectively, but also independently. At the question understanding phase, the type of mathematical creativity were not displayed, 2 types of mathematical creativity at the plan-execution phase were displayed, 3 ones at the plan-execution phase, 4 ones at the reflection phase.

        Since Kyung-Bu High Speed Line has been introduced, a railway electrification has been proceeded rapidly. Therefore, in this study, a specification and application standard for an overhead catenary supporting thin walled steel pole using STKT590 material are proposed in order to improve an economical efficiency. To decide the specification, strength analysis, deflection analysis, deflection test and fracture test are performed. If the supporting structures using high strength materials such as STKT590 are installed, the advantages of cost by the materials price reduction and demand and supply are expected.

      • 항진균물질을 생산하는 Streptomyces fimbriatus AF21-3의 분리 및 동정

        Actinomycetes were isolated from soil to find new antibiotics. An active antimicrobial compound was isolated from the culture filtrate of Actinomycetes AF 21-3. The Actinomycetes strain which showed strong antifungal activity against Candida tropicalis and Microsporum gypseum was identified as Streptomyces fimbriatus and named as Streptomyces fimbriatus AF 21-3.
