      • 光照射에 의한 콩나물의 質的 變化

        光의 照射에 의한 콩나물의 質的變化를 調査할 目的으로 光度 및 光質別 處理를 하여 成長狀態, Vitamin C, chlorphnyll, cellulose등의 含量과 質感에 對한 官能檢査를 行하였으며 chlorophyll과 vitamin C의 關係를 chloramphenicol 處理에 의하여 調査해 보았다. 그 結果 白色光의 경우 50 lux와 100 lux 處理로 vitamin C의 含量 增加와 cellulose등의 生成이 促進되었으며 質感이 暗所보다 良好하였다. 그러나 200 lux 하에서는 매우 不良하였다. blue光은 他光보다 cellulose와 vitamin C生成에 效果的 이었다. chlorophyll과 vitamin C의 含量과는 直接的인 關係가 없고 다만 光照射와의 關聯性이 있는 것으로 推定되었다. This study was carried out to evaluate the quality of green soybean sprouts grown under light. The soybean was germinated under light through blue, red and white polyethylene films at 25℃ for 10 hours a day, and light intensity was controlled 50 to 200 lux. The contents of Vitamin C. cellulose and chlorophyll were determined and checked up texture by sensory evaluation of the soybean sprouts soup. In addition, the relationship of chlorophyll and Vitamin C was measured by treatment of chloramphenicol which has inhibitory effect of chloroplast, chlorophyll accumulating site. Vitamin C and cellulose contents were all increased by irradiation of white light at 50 and 100 lux and also texture of the sprouts was better than it of the sprouts grown under darkness while, it was very tough under 200 lux. Blue light was more effective for biosynthesis of Vitamin C and cellulose than white and reg light. Biosynthesis of chlorophyll was not relative to Vitamin C content directly. but the relationship may be concerned with light irradiation.

        면접조사를 통한 초등학생의 영양교육 실태 및 실천도 조사

        This study investigated the experience and practice of elementary school students on nutrition education. The data were collected from 217 male and female students attending 5-6th grade elementary schools in Seoul and Kyunggi-Do from March to June 2007, interviewing face to face by a nutrition teacher and 3 interns of a nutrition teacher. The results were as follows: 86.5% of the subjects learned about ‘Table etiquette’, ‘Reasons for eating fruits and vegetables’ (78.7%), ‘Food waste and environment’ (72.3%), ‘Healthy snacks’ (55.7%), ‘Food sanitation’ (52.3%), ‘Food culture of foreign countries’ (48.1%). Nutrition education experience was significantly different by gender. A total of 43.5% boys responded that they never learned about ‘basic food preperation’ (p < 0.01). They had learned ‘Nutrients for body’ and ‘Food waste and environment’ in school, ‘Healthy weight loss’, ‘Food culture of foreign countries’, ‘Food circulation’ on television, Most content (‘Table etiquette’, ‘Simple cooking’, ‘Food sanitati n’, ‘Eating behaviors for health’, ‘Reasons for eating fruits and vegetables’, ‘Healthy snacks’) was learned from parents. The practice after nutrition education was higher in ‘Table etiquette’ (2.14), ‘Eating fruits and vegetables’ (2.07) than others compared with education experience. The most reason of non-practice on nutrition information was ‘Troublesome’. In ‘Nutrients for body’, a boy answered ‘Difficult for practice’ 20.0%, a girl answered ‘Difficult to understand’ 32.6%, showing a significant difference between the gender groups (p < 0.001). They remembered the ‘Nutrients for body’ (49.6%), ‘Food sanitation’ (44.5%) because of ‘important content’,‘Basic food preparation’ (40.6%), ‘Food culture of foreign countries’ (36.3%) because of ‘interesting content’, ‘Healthy weight loss’ (52.0%), ‘Eating behavior for health’ (44.5%) and ‘Healthy snacks’ (33.7%) because of ‘need for my health’.

        Clinical Utilization of Brain Magnetic Resonance Imaging-Based Artificial Intelligence Software in the Spectrum of Alzheimer’s Disease: Case Series

        Brain magnetic resonance imaging (MRI) is a key tool for diagnosing neurodegenerative diseases such as Alzheimer’s disease (AD). However, MRI analysis by visual interpretation and reading can be time-consuming and requires specialized expertise. Brain MRI-based artificial intelligence (AI) software has been developed to aid clinicians in diagnosing and managing neurodegenerative disorders, including AD. This study demonstrates the clinical application of the AI software for volumetric analysis of brain MRI scans in patients within the AD spectrum. In the current case series, four patients with memory impairment visited the memory clinic of Yeouido St. Mary’s Hospital. They underwent a series of assessments, including automated analysis of AI-based software for brain MRI volumetric measurements. The information provided by the software was highly accurate, consistent, and was especially valuable for the early diagnosis and monitoring of disease progression. The results imply that this technology potentially aids in the early detection and management of AD, making it a valuable tool for clinicians in the diagnosis of neurodegenerative diseases.

      • CD133-induced TM4SF5 expression promotes sphere growth via recruitment and blocking of protein tyrosine phosphatase receptor type F (PTPRF)

        <P><B>Abstract</B></P> <P>CD133 is a surface marker of liver cancer stem cells. Transmembrane 4 L six family member 5 (TM4SF5) promotes sphere growth and circulation. However, it is unknown how CD133 and TM4SF5 cross-talk with each other for cancer stem cell properties. Here, we investigated the significance of inter-relationships between CD133, TM4SF5, CD44, and protein tyrosine phosphatase receptor type F (PTPRF) in a three-dimensional (3D) sphere growth system. We found that CD133 upregulated TM4SF5 and CD44, whereas TM4SF5 and CD44 did not affect CD133 expression. Signaling activity following CD133 phosphorylation caused TM4SF5 expression and sphere growth. TM4SF5 bound to CD133 and promoted c-Src activity for CD133 phosphorylation as a positive feedback loop, leading to CD133-mediated sphere growth that was inhibited by TM4SF5 inhibition or suppression. TM4SF5 also bound PTPRF and promoted paxillin phosphorylation. Decreased sphere growth upon CD133 suppression was recovered by TM4SF5 expression and partially by PTPRF suppression. TM4SF5 inhibition enhanced PTPRF levels and abolished PTPRF suppression-mediated sphere growth. Altogether, CD133-induced TM4SF5 expression and function were important for liver cancer sphere growth and may be a promising target to block metastasis.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The role of TM4SF5 and CD133 cross-talk in the CSC properties of HCC is unknown. </LI> <LI> CD133-induced c-Src, Akt, and β-catenin signaling mediates TM4SF5 induction. </LI> <LI> TM4SF5 drives CD133 phosphorylation via a bidirectional positive feedback loop. </LI> <LI> TM4SF5 binds PTPRF to regulate cellular signaling for anchorage-independent growth. </LI> <LI> CD133/TM4SF5/PTPRF and CD44 are potential biomarkers for HCC metastasis. </LI> </UL> </P>

      • Frequent expression of follicular dendritic cell markers in Hodgkin lymphoma and anaplastic large cell lymphoma

        <P><B>Aims</B></P><P>Although the tumour cells of Hodgkin lymphoma (HL) are derived from mature B-cells, the lineage infidelity of Hodgkin/Reed–Sternberg cells (HRSs) often causes diagnostic problems. Recently introduced HRS markers are also positive for follicular dendritic cells (FDCs). We investigated the expression of several FDC markers in HL and anaplastic large cell lymphoma (ALCL) and evaluated their diagnostic efficacy.</P><P><B>Methods</B></P><P>Eighty-five cases of HL and 52 cases of ALCL were included in this study. Immunohistochemistry was performed for glioma-associated homologue (GLI) 3, class III β-tubulin (TUBB3), fascin, clusterin, γ-synuclein, podoplanin, syntenin, CD21, CD35 and EGFR.</P><P><B>Results</B></P><P>HRSs were diffusely positive for GLI3, fascin and TUBB3; the mean positivity rates per case were 94% for GLI3, 82% for fascin, 69% for TUBB3, 17% for clusterin, 17% for γ-synuclein and 14% for syntenin. Podoplanin, CD21, CD35 and EGFR were almost negative. However, the frequency of marker expression was not associated with the histologic subtype or the presence of Epstein–Barr virus (EBV). ALCL showed a similar pattern to HL, but the overall frequency of positivity was lower than that observed in HL. The mean positivity rates were 56% for GLI3, 62% for fascin, 58% for TUBB3 and 21% for clusterin. The other markers were nearly negative. Anaplastic large cell lymphoma kinase positivity did not affect the expression rates.</P><P><B>Conclusions</B></P><P>This study confirmed the frequent expression of FDC markers in HL and ALCL. Especially, GLI3, fascin and TUBB3 are the most sensitive markers. Further studies are required to evaluate the association between FDCs, HRSs and ALCL cells.</P>

      • Variations in the number of CCL3L1 gene copies and Kawasaki disease in Korean children.

        <P>High-dose intravenous immunoglobulin (IVIG) therapy is the highly effective and standard treatment for Kawasaki disease (KD). However, similar to 20 % of KD patients have persistent fever or recurrence of fever after the initial IVIG treatment, which increases the risk for coronary artery lesions (CALs). Furthermore, the mechanism of IVIG resistance in KD patients still is unknown. The number of CC chemokine ligand 3-like 1 (CCL3L1) gene copies is reported to be associated with KD and IVIG resistance in Japanese patients. In addition, the authors observed significant upregulation of the CCL3L1 gene expression after in vitro immunoglobulin treatment in B cell lines derived from KD patients. Therefore, this study of 459 KD patients and 496 healthy control subjects tested whether the number of CCL3L1 gene copies is associated with a risk of KD, CALs, and/or IVIG resistance in Korean KD patients. However, the number of CCL3L1 gene copies was not associated with KD (P = 0.18), CAL formation (P = 0.062), or the IVIG resistance (P = 0.90). Therefore, the results indicate that the number of CCL3L1 gene copies does not have a role in susceptibility to KD or CALs nor with IVIG resistance in Korean KD patients.</P>

        대장내시경으로 확인된 악성 림프종의 내시경적 분류와 임상병리학적 고찰

        Background/Alms: There have been only a few endoscopic s0dies of lower intestinal lesions of malignsnt lymphoma. The aims of this study were to classify the ileocolonic 1ymphomas according to the colonoscopic fhgdings and to reveal the dinicopathological relationship according to the classes. Methods: Subjects were 24 cases of ileocolonic lymphomas. We evaluated the clinicopathological features according to their endoscopic fmdings. Results: The chief complaints were abdominal pain, bleeding, and abdomina1 mess, Twenty-six lesions from 22 patients, excluding 2 multiple lymphomatous polyposis, were endoscopically classfled as follows. Fungating (10 lemons), ulcerofungating (7 lesions), inflltrative (5 lesions), ulceroinfiltrative (3 lesions), and ulcerative (1 lesion). The 1ocafim of the lesions was the terminal ileum in 11 lesions, the colon in 10 lesions, and both regions in 5 lesions. Within the 1arge bowel, the cecum was involved most frequently, followed by the ascending colon and the remainder. Most of pathological types were the diffuse large cell and the large cell immunoblastic. There was no relationship between the endoscopic findings and the histologic types. Eight eases were manifested as intussusceptlon. Conclasions: Ileocolonic lymphomas can be classified endoscopically into five types. Among the types, fungating and ulcerofungating types axe the most frequent.

        Natural Killer T (NKT) Cells Attenuate Bleomycin-Induced Pulmonary Fibrosis by Producing Interferon-γ

        <P>Pulmonary fibrosis is a progressive illness characterized by interstitial fibrosis. Although the precise mechanism for pulmonary fibrosis is not completely understood, an immune response involving interferon (IFN)-γ appears to play a role. Therefore, we examined the functional roles of natural killer T (NKT) cells, which produce IFN-γ and interleukin-4 on activation, in bleomycin-induced pulmonary fibrosis. In NKT cell-deficient mice, pulmonary fibrosis was worse in terms of histology, hydroxyproline levels, and mortality than in control mice. The transforming growth factor (TGF)-β1 levels were higher in the lung after injecting bleomycin, and blockade of TGF-β1 by neutralizing monoclonal antibody attenuated the pulmonary fibrosis in CD1d<SUP>−/−</SUP> mice. In contrast, the production of IFN-γ was reduced in lungs from CD1d<SUP>−/−</SUP> mice. Moreover, the adoptive transfer of NKT cells into CD1d<SUP>−/−</SUP> mice increased IFN-γ and reduced TGF-β1 production, attenuating pulmonary fibrosis. An <B><I>in vitro</I></B> assay demonstrated that IFN-γ was involved in suppressing TGF-β1 production in cells collected from bronchoalveolar lavage. The adoptive transfer of NKT cells from IFN-γ<SUP>−/−</SUP> mice did not reverse pulmonary fibrosis or TGF-β1 production in lungs of CD1d<SUP>−/−</SUP> mice whereas NKT cells from B6 control mice attenuated fibrosis and reduced TGF-β1 production. In conclusion, IFN-γ-producing NKT cells play a novel anti-fibrotic role in pulmonary fibrosis by regulating TGF-β1 production.</P>

      • NKT cells promote antibody-induced joint inflammation by suppressing transforming growth factor β1 production

        <P>Although NKT cells has been known to exert protective roles in the development of autoimmune diseases, the functional roles of NKT cells in the downstream events of antibody-induced joint inflammation remain unknown. Thus, we explored the functional roles of NKT cells in antibody-induced arthritis using the K/BxN serum transfer model. NKT cell–deficient mice were resistant to the development of arthritis, and wild-type mice administrated with α-galactosyl ceramide, a potent NKT cell activator, aggravated arthritis. In CD1d<SUP>−/−</SUP> mice, transforming growth factor (TGF)-β1 was found to be elevated in joint tissues, and the blockade of TGF-β1 using neutralizing monoclonal antibodies restored arthritis. The administration of recombinant TGF-β1 into C57BL/6 mice reduced joint inflammation. Moreover, the adoptive transfer of NKT cells into CD1d<SUP>−/−</SUP> mice restored arthritis and reduced TGF-β1 production. In vitro assay demonstrated that interleukin (IL)-4 and interferon (IFN)-γ were involved in suppressing TGF-β1 production in joint cells. The adoptive transfer of NKT cells from IL-4<SUP>−/−</SUP> or IFN-γ<SUP>−/−</SUP> mice did not reverse arthritis and TGF-β1 production in CD1d<SUP>−/−</SUP> mice. In conclusion, NKT cells producing IL-4 and IFN-γ play a role in immune complex–induced joint inflammation by regulating TGF-β1.</P>
