        Tissue-specific expression and subcellular localization of ALADIN, the absence of which causes human triple A syndrome

        Triple A syndrome is a rare genetic disorder caused by mutations in the achalasia-addisonianism-alacrima syndrome (AAAS ) gene which encodes a tryptophan aspartic acid (WD) repeat-containing protein named alacrima-achalasia-adrenal insufficiency neurologic disorder (ALADIN). Northern blot analysis shows that the 2.1 kb AAAS mRNA is expressed in various tissues with stronger expression in testis and pancreas. We show that human ALADIN is a protein with an apparent molecular weight of 60 kDa, and expressed in the adrenal gland, pituitary gland and pancreas. Furthermore, biochemical analysis using anti-ALADIN antibody supports the previous finding of the localization of ALADIN in the nuclear membrane. The mutations S544G and S544X show that alteration of S544 residue affects correct targeting of ALADIN to the nuclear membrane.         <P><B>A class of imprintable, bendable, and shape‐conformable polymer electrolyte</B> with excellent electrochemical performance in a lithium battery system is reported. The material consists of a UV‐cured polymer matrix, high‐boiling point liquid electrolyte, and Al<SUB>2</SUB>O<SUB>3</SUB> nanoparticles, formulated for use in lithium‐ion batteries with 3D‐structured electrodes or flexible characteristics. The unique structural design and well‐tuned rheological characteristics of the UV‐curable electrolyte mixture, in combination with direct UV‐assisted nanoimprint lithography, allow the successful fabrication of polymer electrolytes in geometries not accessible with conventional materials.</P>

        비윤리적 언어 표현의 의미 자질에 따른 하위 유형 연구: -대화형 인공지능의 윤리적 언어 표현을 위한 기초 연구(2) -

        본고는 대화형 인공지능의 윤리적 언어 표현을 위한 기초 연구의 일환으로 비윤리적 언어 표현을 의미 자질을 기준으로 하위 유형으로 분류하고 각 하위 유형의 대표 사례와 특징을 살펴보는 것을 목적으로 한다. 이를 위해 본고는 먼저 욕설 또는 비속어의 하위 유형 분류에 대한 기존 연구들을 비판적으로 검토하고, 비윤리적 언어 표현의 의미 자질을 기준으로 좀 더 체계적이고 객관적인 하위 유형 분류의 틀을 제시하였다. 본고에서 적용한 의미 자질 대립쌍은 [±인간], [±사회성], [±내면성], [±행위성], [±지시성], [±상하], [±나이], [±선택], [±지능], [±부분], [±질환], [±요구], [±실체성], [±감탄] 등 14가지이며, 이를 통해 ‘연령’, ‘지위’, ‘직업’, ‘출신’, ‘지력’, ‘행태’, ‘부위’, ‘장애’, ‘외모’, ‘명령’, ‘행동’, ‘구체물’, ‘추상물’, ‘감정’, ‘성상’ 등 총 15개의 하위 유형을 분류하였다. 본고는 이러한 하위 유형 분류 분류가 지금까지 수집한 비윤리적 언어 표현들은 물론이고 앞으로 추가적으로 수집할 표현들을 일관성 있게 정리하는 데에 큰 도움이 될 것이라고 본다. 그리고 궁극적으로는 대화형 인공지능이 비윤리적 언어 표현을 좀 더 객관적이고 체계적으로 인식하는 데에도 도움이 될 것으로 기대한다. The purpose of this study is to classify unethical linguistic expressions(ULEs) into subtypes based on semantic features as a basic research for ethical linguistic expression of interactive artificial intelligence, and to examine representative cases and characteristics of each subtype-classification. For this purpose, this study first critically examined the existing studies on the subtype-classification of 'swear word', 'vulgar word' or 'slang', and suggested a more systematic and objective subtype-classification framework based on the semantic features of ULEs. There are 14 pairs of semantic features applied to this study:[±human], [±sociality], [±interiority], [±activity], [±referentiality], [±hierarchy], [±age], [±choice], [±intelligence], [±part], [±disease], [±demand], [±substantiality], [±admiration]. And through these, 15 subtypes were classified: 'age', 'status', 'occupation', 'origin', 'intelligence', 'behavior', 'region', 'disability', 'appearance', 'command', 'action', 'material', 'abstraction', 'emotion', ‘attribute’. This subtype-classification will be a contribute to organize the ULEs collected so far, as well as the additional expressions to be gathered in the future in a consistent manner. And ultimately, it is expected that this research will help the interactive artificial intelligence to recognize the ULEs more objectively and systematically.

        A novel Epstein–Barr virus-latent membrane protein-1-specific T-cell receptor for TCR gene therapy

        <P><B>Background:</B></P><P>Adoptive transfer of genetically engineered T-cells to express antigen-specific T-cell receptor (TCR) is a feasible and effective therapeutic approach for numerous types of cancers, including Epstein–Barr virus (EBV)-associated malignancies. Here, we describe a TCR gene transfer regimen to rapidly and reliably generate T-cells specific to EBV-encoded latent membrane protein-1 (LMP1), which is a potential target for T-cell-based immunotherapy.</P><P><B>Methods:</B></P><P>A novel TCR specific to LMP1 (LMP1-TCR) was isolated from HLA-A*0201 transgenic mice that were immunised with the minimal epitope LMP1<SUB>166</SUB> (TLLVDLLWL), and LMP1-TCR-transduced peripheral blood lymphocytes were evaluated for functional specificities.</P><P><B>Results:</B></P><P>Both human CD8 and CD4 T-cells expressing the LMP1-TCR provoked high levels of cytokine secretion and cytolytic activity towards peptide-pulsed and LMP1-expressing tumour cells. Notably, recognition of these T-cells to peptide-pulsed cells was maintained at low concentration of peptide, implying that the LMP1-TCR has high avidity. Infusion of these engineered T-cells revealed remarkable therapeutic effects and inhibition of tumour growth in a preclinical xenogeneic model. We observed explosive <I>ex vivo</I> proliferation of functional TCR-transduced T-cells with artificial antigen-presenting cells that express co-stimulatory molecules CD80 and 4-1BBL.</P><P><B>Conclusions:</B></P><P>These data suggest that the novel TCR-targeting LMP1 might allow the potential design of T-cell-based immunotherapeutic strategies against EBV-positive malignancies.</P>

        Cryptococcus neoformans is an organism that mainly causes opportunistic infection in immunocompromised patients. It can also cause various infections in immunocompetent patients including pulmonary cryptococcosis, but it is rare. Steroids are used in clinical practice for treatment of a wide array of disease, because of their anti-inflammatory or immunosuppressive action. But, steroids use predisposes patients to a variety of infection by interfering with host defense mechanism. Infections associated with steroids are dependent on the route of administration, dose, and duration of therapy. Generally, the rate of infection is not increased in patients receiving low dose and limiting periods administration. We presented a case of pulmonary cryptococcosis obscured by cryptogenic organizing pneumonia (COP) in 68-year old female with an immunocompetent patient after low dose and short duration of steroid therapy and review all published casese.

      • Identification of Exosomal lncRNA Based Panel as Potential Diagnostic Marker of Early Stage Hepatocellular Carcinoma

        Aims: Currently, a reliable serum biomarker for hepatocellular carcinoma (HCC) has not been established, particularly for very early stage HCC (single tumor < 2 cm). We aimed to investigate diagnostic serum exosomal long non-coding RNA (exo-lncRNA) panel for very early stage HCC. Methods: Driver oncogenic lncRNA (onco-lncR) candidates were selected by integrative analysis of lncR expression profiles from two different RNA sequencing datasets of human HCC (Catholic_ LIHC and TCGA_LIHC). Expressions of selected onco-lncRs in serum exosome were measured using quantitative real-time PCR (RT-qPCR). Diagnostic performances of serum exo-lncRs for early stage HCC were evaluated in the test cohort (N=44) and validation cohort (N=139). The sensitivity, specificity, and area under the receiver operating characteristic curve (AUC) were calculated for candidate lncRNAs and the current HCC biomarker, alpha-fetoprotein (AFP). Serum exo-lncR panels were developed using a logistic regression model, and their diagnostic performance was evaluated. Results: Six promising driver onco-lncRs, DLEU2, HOTTIP, MALAT1, NEAT1, SNHG1, and TUG1, were identified. were markedly up-regulated in HCC in two different RNA sequencing datasets, and were also known to play important role in various caner by Lnc2Cacner 2.0 database. Among the six candidates, four serum exo-lncRs (DLEU2, HOTTIP, MALAT1, and SNHG1) showed promising performance in the test cohort with area under the receiving operator curve (AUROC) > 0.8. In our validation study, serum exo- MALAT1 could diagnose HCC in all stages (AUROC = 0.908), even in very early stages (AUROC = 0.920), with a greater accuracy than candidate serum exo-lncRs and serum AFP. Conclusions: In conclusion, exo- MALAT1-based serum panel is a promising diagnostic marker for very early stage HCC.

        Although cationic surfactants can cause eye irritation at high concentration, it has some merits in hair conditioning function and has been commonly used in hair conditioning product as a main component. For these reasons, the selection and use of milder cationic surfactant is important in safety of cosmetics. With the use of the hen`s egg test-chorioallantoic membrane (HET-CAM) assay, we examined the eye irritation potential for several cationic surfactant according to chemical structure properties. The eye irritation potential of cationic surfactant was increased according to increasing number of alkyl chain, alkyl chain length. And the cationic surfactant that neutralized to methosulfate causes lower irritation than cationic surfactant of chloride. From these results, we can determine the eye irritation potentials of cationic surfactant according to chemical structure.

        Restoration of senescent human diploid fibroblasts by modulation of the extracellular matrix

        <P><B>Summary</B></P><P>Human diploid fibroblasts have the capacity to complete a finite number of cell divisions before entering a state of replicative senescence characterized by growth arrest, changes in morphology, and altered gene expression. Herein, we report that interaction with extracellular matrix (ECM) from young cells is sufficient to restore aged, senescent cells to an apparently youthful state. The identity of the restored cells as having been derived from senescent cells has been confirmed by a variety of methods, including time lapse live cell imaging and DNA finger print analysis. In addition to cell morphology, phenotypic restoration was assessed by resumption of proliferative potential, growth factor responsiveness, reduction of intracellular reactive oxygen species levels, recovery of mitochondrial membrane potential, and increased telomere length. Mechanistically, we find that both Ku and SIRT1 are induced during restoration and are required for senescent cells to return to a youthful phenotype. These observations demonstrate that human cellular senescence is profoundly influenced by cues from the ECM, and that senescent cell plasticity is much greater than that was previously believed to be the case.</P>

        Antidepressant-Like Effects of <i>Vaccinium bracteatum</i> in Chronic Restraint Stress Mice: Functional Actions and Mechanism Explorations

        <P>The fruit of <I>Vaccinium bracteatum</I> Thunb. (VBF) is commonly known as the oriental blueberry in Korea. The aim of this study was to evaluate the antidepressant-like effects of water VBF extract (VBFW) in a mouse model of chronic restraint stress (CRS) and to identify the underlying mechanisms of its action. The behavioral effects of VBFW were assessed in the forced swim test (FST) and open field test (OFT). The levels of serum corticosterone (CORT), brain monoamines, in addition to the extracellular signal-regulated kinases (ERKs)/protein kinase B (Akt) signaling pathway were evaluated. VBFW treatment significantly reduced the immobility time and increased swimming time in FST without altering the locomotor activity in unstressed mice. Furthermore, CRS mice treated with VBFW exhibited a significantly decreased immobility time in FST and serum CORT, increased locomotor activity in OFT, and enhanced brain monoamine neurotransmitters. Similarly, VBFW significantly upregulated the ERKs/Akt signaling pathway in the hippocampus and PFC. In addition, VBFW may reverse CORT-induced cell death by enhancing cyclic AMP-responsive element-binding protein expression through the up-regulation of ERKs/Akt signaling pathways. In addition, VBFW showed the strong antagonistic effect of the 5-HT<TEX>$ _{2\text{A}}$</TEX> receptor by inhibiting 5-HT-induced intracellular Ca<TEX>$ ^{2+}$</TEX> and ERK1/2 phosphorylation. Our study provides evidence that antidepressant-like effects of VBFW might be mediated by the regulation of monoaminergic systems and glucocorticoids, which is possibly associated with neuroprotective effects and antagonism of 5-HT<TEX>$ _{2\text{A}}$</TEX> receptor.</P>
