      • 고속 파이프라인 A/D 변환기를 위한 연산 증폭기의 설계

        In the design specifications of operational amplifiers which are used in high-speed pipeline A/D converters is analyzed, the technique to design using a design tool of operational amplifiers is proposed. The CMOS operational amplifier usable in 10-b 25-MHz pipelined A/D converters is designed. The operational amplifier designed using 1.2 ㎛ CMOS technology exhibits a do gain of 71 dB, a unity-gain frequency of 195 MHz, a phase margin of 57^*, a slew rate of 186 V/㎲, a output voltage swing of ± 1.1 V, and a power dissipation of 4 mW at 5 V supply voltage and 0.6 pF load. And applying to the S/H amplifier with a gain of 2 and a capacitive load of 0.5 pF the settling time of 14 ns for output voltage swing of 1 ± V satisfied the design specifications.

        In this paper, we describes a 10-bit 40 MS/s pipelined A/D converter with 0.8 um double-poly double-metal CMOS technology. It achieves low power static disssipation of 36㎽ at 5 V power supply. A 1.5 b/stage pipelined architecture in the designed A/D Converter is used to allow large correction range for comparator offset and perform the fast interstage signal processing. In order to design high resolution and low power pipelined A/D converter, the sample and hold amplifier is designed using op-amp sharing technique and dynamic comperator. In addition, fully-differential folded cascode op-amp with gain boosting is designed by using SAPICE. At 10 ㎒ input SNR is 56.7 dB. The DNL and INL exhibit ±0.6 LSB, +1/-0.75 LSB respectively.

        Methylenetetrahydrofolate Reductase Polymorphisms and Risk of Recurrent Pregnancy Loss: a Case-Control Study

        <P>The balance between coagulation and fibrinolysis is an essential part in early pregnancy. Mutations in methylenetetrahydrofolate reductase (<I>MTHFR</I>) gene lead to decreased activity of the enzyme and hyperhomocysteinemia, which then induces platelet aggregation by promoting endothelial oxidative damage, possibly resulting in adverse effect on maintenance of pregnancy. We investigated the role of <I>MTHFR</I> single nucleotide polymorphisms (SNPs), C677T and A1298C, in Korean patients with recurrent pregnancy loss (RPL). We conducted a prospective case-control study in the Korean population. Subjects included 302 women with 2 or more consecutive, unexplained, spontaneous miscarriages before 20 weeks of gestation and 315 control women without a history of recurrent miscarriages. The genotyping for C677T and A1298C polymorphisms was performed using the TaqMan assay. Continuous variables were compared using Student's t-test, and χ<SUP>2</SUP> test was used to evaluate differences in the genotype distributions between the RPL and the controls. The genotype distribution of both polymorphisms in the RPL group did not differ from those of the controls. For further analysis, if RPL patients were divided according to the numbers of pregnancy losses (≥ 2 and ≥ 3) neither group was significantly different compared with controls. <I>MTHFR</I> gene C677T and A1298C polymorphisms are not associated with idiopathic RPL in Korean women, suggesting that those may not be susceptible allelic variants or be deficient to cause RPL.</P>

        Background: Asiatic acid is a putative anticancer agent in several types of cancer cells. Researches of its biological role in negative regulation of cell growth have focused on the extent of induction of apoptosis in a cell-type-specific manner. Objectives: To identify the mechanisms underlying asiatic acid-induced downregulation of BCL2. Methods: MicroRNA microarray analysis was used to investigate the effect of asiatic acid treatment of A549 cells on miRNA expression profiles. We used quantitative RT-PCR to confirm the asiatic acid-induced increase in miR-1290 level with primers specific for the mature miR-1290 sequence. We also assessed the effect of asiatic acid on the levels of BCL2 in A549 cells using western blot analysis. Results: We identified an important regulator of asiatic acid-induced cell death, microRNA (miR)-1290, which sensitizes cells to Asiatic acid-induced cytotoxicity and negatively regulates BCL2 expression. Asiatic acid significantly upregulated miR-1290, and asiatic acid-induced cell death was shown to be dependent on miR-1290 activity. Molecular assays demonstrated that BCL2 mRNA is a direct target of miR-1290-mediated RNA interference. Conclusion: The findings of the present study suggest that miR-1290 suppresses cell viability and cell cycle progression. Our results provide insight into miR-1290-mediated cellular mechanisms in asiatic acid-treated A549 non-small cell lung carcinoma cells.

        Radicicol이 신경세포에서 베타 아밀로이드 전구단백질의 대사에 미치는 영향

        Alzheimer’s disease (AD) is characterized pathologically by the presence of intracellular neurofibrillary tangles and deposition of ${\beta}-amyloid $ (A ${\beta}$) peptides, which are generated by processing of amyloid precursor protein (APP). It is urgent to develop effective therapies for the treatment of AD, since our society rapidly accelerate aging. A${\beta}$ peptides have been believed to be neurotoxic and now are also considered to have effects on the mechanism of memory formation. In this study, effects of radicicol on the metabolism of APP were analyzed. Radicicol inhibited the secretion of A${\beta}$ from the Neuro2a cell line (APPswe cell) expressing APPswe. Beta-site APP cleaving enzyme (BACE) fluorescence resonance energy transfer (FRET) assay revealed that it inhibited BACE activity in a dose dependently manner. Immunoblotting study showed that it inhibited intracellular heat shock protein (HSP)90 and it increased the secretion of HSP90 from the APPswe cells. We suggest that radicicol inhibits APP metabolism and Ap generation by the means of HSP90 inhibitory mechanism and partially BACE inhibitory mechanism. This is the first report that radicicol inhibits the secretion of A${\beta}$ peptides from neuroblastoma cells.

        콜레마니진디벌(Aphidius colemani) 촉각에 분포하는 감각기의 종류 및 외부미세구조적 특징

        Several species of the genus Aphidius are used in biological control programs against aphid pests throughout the world and their behavior and physiology are well studied. While the importance of sensory organs in their behavior is understood, their antennal structure remains largely unknown. In this study, the external morphology and distribution of the antennal sensilla on the antennal of both female and male adults of A. colemani were described using scanning electron microscopy (SEM). Generally, the filiform antennae of males ($l,515.2{\pm}116.5\;{\mu}m$) are longer than females ($l,275.1{\pm}103.4\;{\mu}m$). Antennae of this species are made up of radicle, scape, pedicel and flagellomeres. Male and female antennae differed in the total number of flagellomeres as 15 in males and 13 in females. The result of SEM observation was characterization and grouping of seven types of receptors into morphological classcs: Bohm bristles, Sensilla placodea, 2 types of S. coeloconica and S. trichodea, and S. basiconica as with a tip pore and with wall pores. In addition, the possible functions of the above sensilla types arc discussed in light of previously published literature; mechanoreception (Bohm bristles, S, coeloconica I & II and S. trichodea) and chemoreception (S. placodea, S. basiconica type I & II). Aphidius속(Hymenuptera: Braconidae)에 속하는 일부 종은 주로 생물학적 방제 인자로서 사용되고 있으며, 행동학적 생태에 대하여 연구가 많이 되어있다. 행동에 있어서 신경기관을 중요성을 알고 있음에도 불구하고 촉각의 구조는 널리 알려져 있지 않다. 본 연구에서는 콜레마니진디벌의 암수 성충의 촉각에 분포하는 감각기를 주사전자현미경(SEM)을 이용하여 분류하고, 외부형태를 관찰하였다. 일반적으로 콜레마니진디벌의 촉각은 염주상(filiform)의 형태로 수컷($1,515.2{\pm}116.5\;{\mu}m$)이 암컷($l,275.1{\pm}103.4\;{\mu}m$)보다 더 길다. 촉각은 촉각근절(radicle), 밑마디(scape), 자루마디(pedicel, 채찍마디(flagellomeres)로 구분된다. 암 수컷의 채찍마디의 수에서 수컷은 15마디, 암컷은 13마디로 차이가 나타났다. 콜레마니진디벌의 촉각을 SEM을 이용하여 관찰한 결과, 총 7 종류의 감각기 즉; Bohm bristles, Sensilla placodea와 두 가지 타입의 S. coeloconica, S. trichodea, 또 tip pore 또는 wall pores가 있는지 여부에 띠라 두 가지 S. basiconica를 확인할 수 있었다. 이전에 연구들을 바탕으로 각 감각기들의 기능을 추측해볼 때 Bohm bristles과 S. coeloconica type I & II, S. trichodea 등은 기계적감각을, S. placodea, S. basiconica type I & II는 회학적감각을 담당할 것으로 추정된다.

        알츠하이머질병 모델동물인 Tg2576 마우스를 이용한 미나리 알코올추출물의 기억력 개선 효능

        본 연구는 천연 식물이 기억력 개선에 미치는 영향을 검토하기 위하여 총 7가지 식물에 대하여 아세틸콜린분해효소 활성 억제력을 측정하였다. 특히 미나리 알코올추출물(18.76%)의 억제력이 가장 우수하였으며, 미나리 알코올추출물에 대한 추가 연구를 수행하였다. Tg2576 마우스의 기억력에 미치는 영향을 검토하기 위하여 미나리 알코올추출물 50 mg/kg으로 3개월간 경구투여 후 수동회피테스트로 인지기능변화를 측정하였고, 뇌 속의 아세틸콜린분해효소 활성 억제, 베타아밀로이드1-42 단백질 생성 억제력을 측정하였다. 그 결과, 수동회피 테스트에서 미나리추출물을 투여한 Tg 마우스군은 181.77초로 생리식염수를 투여한 Tg 마우스군과 비교하여 머무름 시간이 유의적으로 증가하게 나타났다. 베타아밀로이드1-42 단백질 농도 측정 시 미나리 알코올추출물에 의하여 축적 농도가 985.19 pmol/g으로 감소하였으며, 생리식염수를 투여한 Tg 마우스군과 유의적 차이가 있었다. 추가적인 효소억제력 실험 결과, 미나리 알코올추출물의 아세틸콜린분해효소 활성억제에 대한 50% 활성억제농도(IC<SUB>50</SUB>)값은 991.77 μg/mL로 나타났으며, Lineweaver-Burk Plot 결과, 무경쟁적 저해로 나타났다. 따라서 미나리 알코올추출물은 Tg2576 형질전환 마우스의 인지기능을 개선시키며, 콜린성 신경시스템을 보호하는 물질로 판단된다. 미나리 알코올추출물은 기억 및 학습 증진에 효과적으로 작용하는 천연식물로써 이상의 결과를 근거로 한 미나리 소재의 다양한 기능성제품 개발과 부가가치 향상이 가능할 것으로 판단된다. This study was conducted to investigate the effect of herbs on memory improvement by focusing on their cholinergic functions in Tg2576 mice. Seven herbs were used to obtain extracts by using alcohol and water. In screening test for cholinergic activities of the extracts, acetylcholinesterase (AChE) activity was highly inhibited in Oenanthe javanica alcohol extract (OJAE, 18.76%) as compared with the others. The OJAE-treated Tg2576 (Tg-OJAE) groups showed the statistically significant increases of latency time in passive avoidance test. Also, it was found that the concentration of Aβ1-42 was significantly reduced in Tg-OJAE groups compared to non-treated Tg2576 groups. In the additional enzyme test, it was found that IC<SUB>50</SUB> of OJAE was 991.77 μg/mL and OJAE acted as an uncompetitive inhibitor of AChE. Therefore, it seemed that OJAE can be used for the development of processed foods for memory improvement.

        영화 <내 어깨 위 고양이, 밥>에 나타난 반려동물의 의미와 펫로스 증후군 상담의 필요성 연구

        본고에서는 반려동물에 대한 사회적 관심이 높아지는 상황에서 그에 따라 등장한 펫로스 증후군과 관련하여 심리상담의 필요성을 강조하는 것을 목표로 하였다. 이에, 실화를 바탕으로 제작된 영화 <내 어깨 위 고양이, 밥>에 나타난 반려인과 반려동물의 서사를 심리학적 시각에서 분석해 본 후, 현대 사회에서 반려동물이 인간에게 갖는 의미가 무엇인지 우선적으로 살펴보았다. 그리고 영화의 내용과 최근 반려동물과 관련된 연구 결과를 종합해 본 결과 다음과 같은 특성들을 확인 할 수 있었다. 첫째, 반려동물은 더 이상 인간에게 종속적인 의미를 지니고 있지 않으며, 동등한 가족으로서의 권리를 인정받고 있다. 둘째, 인간은 반려동물과의 관계를 통해 치유의 경험을 할 수 있으며, 대인관계능력 또한 향상시킬 수 있다. 영화 <내 어깨 위 고양이, 밥>에서 나타난 제임스와 밥의 만남과 관계 형성 과정, 그리고 제임스의 가족관계 회복 및 마약 중독 치료의 성공 등의 모습에서 이러한 내용을 충분히 확인 할 수 있다. 끝으로, 영화 <내 어깨 위 고양이, 밥>이 실화를 기반으로 하고 있고, 실제 에세이는 30개국에서 베스트 셀러가 되며 많은 사람들의 공감을 얻기도 했는데, 이와 같은 성과가 결국 반려동물이 우리 사회에 얼마나 중요한 존재가 됐는지 알 수 있게 해주며, 반려동물이 반려인의 인생에 미치는 영향력이 얼마나 큰지 짐작할 수 있게 해주었다. 또한 반려동물 관련 서적이나 영화들이 오랜 기간 사람들에게 사랑받아왔고, 반려동물 작품 특유의 따뜻하고 온정적인 분위기가 보는 사람들로 하여금 치유를 받는 경험을 하게 해준다. 그렇기 때문에 반려인구 증가와 함께 나타난 펫로스 증후군 문제와 관련하여 문학치유는 상당히 중요한 역할을 수행할 수 있을 것으로 예상되며, 펫로스 증후군과 관련된 심리상담 프로그램 연구가 상당히 부족한 현시점에서 해당 분야에 대한 활발한 연구가 꼭 필요하다고 피력하고자 한다. This study aims to highlight the significance of pet loss counseling as society is trending to pay more attention to pets and many people begin to experience pet loss syndrome. Therefore, a movie based on a true story, “A Christmas Gift from Bob” is qualitatively reviewed for the stories of pet owners and pets from a psychological perspective, and it prioritizes to think about what pet could mean to humans in the modern world. After integrally carefully contemplating the movie and the recent research studies related to pets, the study was able to confirm the following points. First, pets are no longer subordinate to humans, and their rights are being recognized as if they are members of the family. Second, humans can have a healing experience through building a relationship with the pet, and they can improve social skills. This is reassured in the movie, “A Christmas Gift from Bob,” that how the relationship between James and Bob is formed, James’s family is healing, and successfully getting better with the drug addiction. Lastly, the movie “A Christmas Gift from Bob” is based on a true story, and the original essay had become a bestseller in 30 countries by winning hearts from people worldwide, and such a successful result shows how pet becomes important in our lives, and how much influence pet can have in pet owners’ lives. On top of that, people love books or movies about pets, and a typical compassionate or warm impression gives a healing experience to the audience. Therefore, an increasing number of people is choosing to spend their life with pets, and the issue of pet loss syndrome and bibliotherapy could take significant roles. Unfortunately, information about pet loss counseling is lacking at this point, more research would be necessary for this area of the study.

        Tissue-specific expression and subcellular localization of ALADIN, the absence of which causes human triple A syndrome

        Triple A syndrome is a rare genetic disorder caused by mutations in the achalasia-addisonianism-alacrima syndrome (AAAS ) gene which encodes a tryptophan aspartic acid (WD) repeat-containing protein named alacrima-achalasia-adrenal insufficiency neurologic disorder (ALADIN). Northern blot analysis shows that the 2.1 kb AAAS mRNA is expressed in various tissues with stronger expression in testis and pancreas. We show that human ALADIN is a protein with an apparent molecular weight of 60 kDa, and expressed in the adrenal gland, pituitary gland and pancreas. Furthermore, biochemical analysis using anti-ALADIN antibody supports the previous finding of the localization of ALADIN in the nuclear membrane. The mutations S544G and S544X show that alteration of S544 residue affects correct targeting of ALADIN to the nuclear membrane.         A Comparative Study on the Synthesis of Al-Doped Li<sub>6.2</sub>La<sub>3</sub>Zr<sub>2</sub>O<sub>12</sub> Powder as a Solid Electrolyte Using Sol—Gel Synthesis and Solid-State Processing

        <P>In this study, we fabricated and characterized two kinds of Al-doped Li6.2La3Zr2O12 (LLZO) powders with the modified sol-gel synthesis and solid-state processing, respectively. Moreover, the two kinds of LLZO powders were compared in detail examining the powder size and processing window for the cubic phase. In the case of the sol-gel synthesis, the processed powder exhibited a pure cubic phase with little secondary phases such as Li-deficient pyrochlore or LaAlO3 at calcination temperatures above 900 degrees C and a mixed phase of cubic and tetragonal structure at 800 degrees C. However, in the solid-state processed powder, the tetragonal phase was converted to a cubic one at a temperature of 1000 degrees C. When the amount of Li addition was varied, sol-gel synthesized LLZO powder also had a much wider range for cubic phases than that of the solid-state processed one. The sol-gel LLZO powder with a Li addition of 7.13 mol. showed an average grain size of 429 nm and a specific BET surface area of 5.34 m(2)/g. The formation of the cubic phase with few secondary ones was sensitive to Al doping amount. The results of this study suggest that the sol-gel process is the more optimum method for obtaining the cubic phase of LLZO powder for highly dense and conductive solid electrolyte ceramics.</P>
