        Protective Effects of Puerarin on Carbon Tetrachloride-Induced Hepatotoxicity

        스포츠 메가 이벤트의 그랜드슬램 달성이후 한국 스포츠외교의 방향과 과제

        올림픽이나 월드컵축구대회 같은 스포츠 메가 이벤트는 인종, 정치, 종교, 이념의 경계를 넘어 전 세계인이 참가하는 축제인 동시에 국위를 드높이는 수단이기 때문에 국제관계에서도 그 영향력은 날로 커지고 있다. 우리나라는 스포츠 강국의 경기력 못지않게 스포츠 메가 이벤트 유치와 성공적인 대회 개최를 통해 스포츠외교 측면에서도 큰 성과를 거두었다. 2018년 동계올림픽 개최지로 평창이 확정되면서 스포츠 메가 이벤트의 그랜드슬램을 달성한 우리나라는 스포츠외교에 있어서 새로운 방향모색과 질적 성장을 위한 몇 가지 과제를 던져주고 있다. 특히 2012년 런던하계올림픽에서 심판의 오심과 축구 경기장에서의 독도 세리머니 등은 한국 스포츠외교의 실상과 앞날에 대한 새로운 과제를 던져주었다. 본고는 스포츠외교를 ``스포츠`` 라는 수단을 통해 국가나 집단(개인)간의 상호관계를 증진하려는 속성으로 파악하여, 스포츠 메가 이벤트 그랜드슬램 달성 이후 한국 스포츠외교의 새로운 방향과 과제를 모색 하는데 그 논제의 핵심을 삼았다. 이를 위해 본고는 스포츠 외교가 하나의 독립적 학문으로 성립되어야 하며, 공공외교와 스마트파워의 콘텐츠로서의 유기적 연관성을 보다 공고히 해 나가야 할 뿐만 아니라, 학제 간 통섭, 경계 간 조화 그리고 범정부적 협력 등이 필요함을 제안하고자 한다. Korea Sports has developed much enough since have successfully held diverse Mega Sports Event, Kept staying in top 10 in Olypic medal standings. Especially 2012 London Olypic Games, Korea ended up in fifth place with 13golds, eight silvers and sevent bronze medals. In the medal stadings, it was Korea`s second-best result after a fourth-place finish at the 1988 Seoul Olympics. Nowadays, Enter a globall village phase, Sports play significant role in international relations. A large number of affirmative points of sports that contribute to increasing nation`s pride and enhacing national prestige. Sports play important roles in international politics as a Smart Power. Comsidering scholars` opinions that people`s morale is a principal factor of national power including economic and military power, sports can be a proper alternative for raising the morale as well as a measure of uniting the society and diplomatic tool in international politics. Pyeongchagn was chosen to host the 2018 Winter Olypics. Korea hosted the Summer Olympics in 1988, co-hosted the World Cup 2002 with Japan, and then 2011, We held the IAAF Athletic Championships in Daegu. Korea became the sixth in the world to complete the sports Mega Events` Grand Slam. In particular, referees` misjudges and so called ``dok-do`` ceremony (a celebration of football game victory of korea claiming ownership of ``dok-do``, a disputed area between Korea and Japan) at the London 2012 Olympic Games, they cuased what our focus should be. In that sense, the purpose of this study is to deals with possibility of sports diplomacy after Grand Slam. This study stressed that sports diplomacy has to be dealt with as an independent studies. Also it needs to be more interdisciplinary harmony, public diplomacy, smart power`s contents, and intergovernmental cooperation to create infrastructures enhancing sports diplomacy.

        체육인재 육성사업 중장기 발전을 위한 정책 제언

        The purpose of this study is to indicate problems of the existing sports-talent development projects and suggest long-term development plans for those projects. In order to proceed with this study first, a sports talent, the subject of the sports-talent development projects, was defined and categorized from the view of political and economic dimensions. Second, the current sports-talent programs were scrutinized to find out what the obstacles and difficulties resulting from operating those programs are in terms of process and content. Finally, the long-term development plans for the sports-talent projects were suggested. The results of the study indicates first, a well-organized governance for the sports-talent projects needs to be established in order to coordinate the similar and overlapped projects efficiently, second, a sports coaching academy that provides various courses according to participants’level and career interest is to be newly established, third, a process of discovering talents and a follow-up service for the beneficiaries of the projects are to be emphasized and included as a part of the projects, fourth, a comprehensive career development system for sports-talents is to be developed, fifth, an intensive global program for sports- talents is to be provided, and finally an independent training school customized for sports-talents are to be built. This study suggests development directions for the future sports-talent projects by comprehending of the study results.

        Hepatoprotective Effects of an Anthocyanin Fraction from Purple-Fleshed Sweet Potato Against Acetaminophen-Induced Liver Damage in Mice

        The present study was undertaken to examine the protective effects of an anthocyanin fraction (AF) obtained from purple-fleshed sweet potato on acetaminophen (paraceptamol [APAP])-induced hepatotoxicity in mice and to determine the mechanism involved. Mice pretreated with AF prior to APAP administration showed significantly lower increases in serum alanine aminotransferase and aspartate aminotransferase activities and hepatic malondialdehyde formation than APAP-treated animals without AF. In addition, AF prevented hepatic glutathione (GSH) depletion by APAP, and hepatic GSH levels and GSH S-transferase activities were up-regulated by AF. APAP-induced hepatotoxicity was also prevented by AF, as indicated by liver histopathology findings. In addition, the effects of AF were examined on cytochrome P450 (CYP) 2E1, the major isozyme involved in APAP bioactivation. Treatment of mice with AF significantly and dose-dependently reduced CYP2E1-dependent aniline hydroxylation and CYP2E1 protein levels. Furthermore, AF had an antioxidant effect on FeCl2/ascorbate-induced lipid peroxidation in mouse liver homogenates and had superoxide radical scavenging activity. These results suggest that AF protects against APAP-induced hepatotoxicity by blocking CYP2E1-mediated APAP bioactivation, by up-regulating hepatic GSH levels, and by acting as a free radical scavenger.

        Artemisia annua extract ameliorates high-fat diet-induced fatty liver by activating AMPK

        Non-alcoholic fatty liver disease (NAFLD) is emerging as the most common liver disease in industrialized countries. Recently, natural compounds that may be beneficial for improving NAFLD have received increasing attention. Artemisia annua L. is the source of antimalarial phytomolecule, artemisinin, which has been reported to prevent obesity. However, the effect of A. annua extract on hepatic lipid metabolism remains unclear. This study was performed to determine the protective effect of Artemisia annua extract (AAE) on high-fat diet (HFD)-induced hepatic lipid accumulation, and elucidate the molecular mechanisms behind its effects in vivo and in vitro. We found that HFD-fed mice with AAE administration (50 mg/kg/day) for 8 weeks dramatically reduced hepatic lipid accumulation compared to the control mice taken with HFD alone. The body and liver weights of AAE group were significantly lower than those of HFD group, and oral administration of AAE remarkably suppressed the serum levels of triglyceride (TG), total cholesterol (TC), fasting glucose, alanine transaminase (ALT) and aspartate transaminase (AST) in HFD-fed mice. AAE significantly increased the phosphorylation of AMP-activated protein kinase (AMPK) and acetyl-CoA carboxylase (ACC) in the liver of HFD-fed mice and HepG2 hepatocytes. Moreover, AAE downregulated the hepatic expression of sterol regulatory element-binding protein-1c (SREBP-1c) and fatty acid synthase (FAS) in HFD-fed mice and high glucose-treated HepG2 cells. In addition, the inhibitory effects of AAE on the overexpression of SREBP-1c and FAS were attenuated by compound C, which is the specific AMPK inhibitor, in high glucose-treated HepG2 cells. These results indicated that AAE may represent a promising approach for the prevention and treatment of obesity-related NAFLD via the activation of AMPK and the regulation of AMPK-dependent lipogenic genes.

        Micronized, Heat-Treated Lactobacillus plantarum LM1004 Alleviates Cyclophosphamide-Induced Immune Suppression

        The present study investigated the immunomodulatory activity and associated mechanisms of heat-treated Lactobacillus plantarum LM1004 (HT-LM1004) in a cyclophosphamide (CTX)-induced mouse model of immunosuppression. HT-LM1004 induced phagocytic activity and nitric oxide production in RAW264.7 macrophages and stimulated the release of tumor necrosis factor (TNF)-α, interferon (IFN)-γ, interleukin (IL)-2, and IL-12p70. In mice with CTX-induced immunosuppression, oral HT-LM1004 administration restored thymus and spleen indices, including spleen weight. Consistent with the in vitro results, HT-LM1004 increased TNF-α, IFN-γ, IL-2, and IL-12p70 levels in mice after 14 days of treatment and enhanced the natural killer (NK) cell activity of splenocytes from mice with CTX-induced immunosuppression against YAC-1 lymphoma cells. The method of HT-LM1004 generation influenced this activity: L. plantarum LM1004 grown in a membrane bioreactor, which reduced the size of the cells to <1.0 μm through physical stress (micronization), promoted NK cell cytotoxicity to a greater extent than LM1004 subjected to heat treatment alone. These findings indicate that HT-LM1004 without or with micronization can reverse CTX-induced immunosuppression without adverse side effects by potentiating NK cell function.

        Hepatoprotective Effects of Streptococcus thermophilus LM1012 in Mice Exposed to Air Pollutants

        In this study, we explored whether the use of Streptococcus thermophilus LM1012 (TL-LM1012) as a safe probiotic exerts hepatoprotective effects by suppressing oxidative stress and inflammation in vitro and alleviating aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactate dehydrogenase (LDH) production in vivo. In a series of safety tests, TL-LM1012 was found to have a negative response to hemolysis and biogenic amines, as well as susceptibility to antibiotics. TL-LM1012 protected cell viability and suppressed cytotoxicity by inhibiting oxidative stress and induced heme oxygenase-1 and superoxide dismutase activity in a dose-dependent manner in diesel exhaust particulate matter (DEPM)-treated HepG2 cells. Moreover, proinflammatory cytokines, including tumor necrosis factor-α, interleukin (IL)-6, and IL-1β, were suppressed in DEPM-treated splenocytes. In DEPM-treated mice, oral administration of TL-LM1012 regulated AST, ALT, and LDH production in the serum after 14 days of treatment. These findings indicate that TL-LM1012, a safe probiotic, provides a potent preventive or therapeutic effect against liver disease caused by air pollution.
