Lactic acid bacteria are well-known probiotics, conferring several health benefits. In this study, we isolated lactobacilli from human breast milk and identified Lactobacillus reuteri LM1071 (RR-LM1071) using 16S rDNA sequencing. We tested the hemolytic activity, biogenic amine production, and antibiotic susceptibility of this strain to assess its safety. RR-LM1071 was found to be negative for hemolytic activity and biogenic amine production, as well as was measured in susceptible level for antibiotics in the minimal inhibitory concentration (MIC) test. The adhesive properties of RR-LM1071 were higher than those of LGG in HT-29 cells, and showed a greater hydrophobicity than LGG in hexadecane solvent. Under inflammatory conditions, RR-LM1071 suppressed the mRNA expression of IL-6, TNF-α, and IL-4 produced in IL-1β-induced HT-29 cells. Our results suggest that RR-LM1071 is a safe and valuable probiotic that can be used for the treatment of inflammatory bowel disease.

PMO-P4, being cultivated as “Sanghwang”in Korea, was proved to be P. baumii based on ITS (internal transcribed spacer) sequencing and RFLP (Restriction Fragment Length Polymorphism) patterns along with some Phellinus species including P. linteus. The similaraty of ITS sequencing between PMO-P4 and other Phellinus species was given the range of 48.6%~72.2%, showing the highest homology from P. linteus and the lowest from P. gilvus
Hepatoprotective Effects of Streptococcus thermophilus LM1012 in Mice Exposed to Air Pollutants

In this study, we explored whether the use of Streptococcus thermophilus LM1012 (TL-LM1012) as a safe probiotic exerts hepatoprotective effects by suppressing oxidative stress and inflammation in vitro and alleviating aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactate dehydrogenase (LDH) production in vivo. In a series of safety tests, TL-LM1012 was found to have a negative response to hemolysis and biogenic amines, as well as susceptibility to antibiotics. TL-LM1012 protected cell viability and suppressed cytotoxicity by inhibiting oxidative stress and induced heme oxygenase-1 and superoxide dismutase activity in a dose-dependent manner in diesel exhaust particulate matter (DEPM)-treated HepG2 cells. Moreover, proinflammatory cytokines, including tumor necrosis factor-α, interleukin (IL)-6, and IL-1β, were suppressed in DEPM-treated splenocytes. In DEPM-treated mice, oral administration of TL-LM1012 regulated AST, ALT, and LDH production in the serum after 14 days of treatment. These findings indicate that TL-LM1012, a safe probiotic, provides a potent preventive or therapeutic effect against liver disease caused by air pollution.
화랑곡 나방의 성유인 물질인(Z, E)-9, 12-Tetradecadienyl Acetate의 입체 선택적 합성

A stereoselective synthesis for the (Z, E)-9, 12-tetradecadienyl acetate, the sex pheromone of the Indian meal moth(Plodia interpunctella), was described. (E)-3-Pentenoic acid was prepared from malonic acid and propionaldehyde in the presence of triethanolamine as a catalyst. This acid was treated with lithium aluminiumhydride(LAH) to give (E)-3-Pentenol. This alcohol was tosylated with p-toluenesulfonyl chloride(p-TsCl), followed by bromination with lithium bromide(LiBr) to provide (E)-1-Bromo-3-Pentene. (E)-1-Bromo-3-pentene thus obtained was treated with triphenylphosphine(TPP) to furnish (E)-3-pentenetriphenylphosphonium bromide. And 9-acetoxy-1-nonanal was prepared by the pyridinium chlorochromate(PCC) oxidation of 9-acetoxy-1-nonanol, which in turn was obtained from the reaction of 1, 9-nonanediol and glacial acetic acid. Finally, (Z, E)-9, 12-tetradecadienyl acetate was afforded by Wittig reaction of (E)-3-pentenyltriphenylphosphonium bromide and 9-acetoxy-1-nonanal in the presence of K/hexamethylphosphrous triamide(HMPT).
Immunostimulatory activity of Lactococcus lactis LM1185 isolated from Hydrangea macrophylla

The lactic acid bacteria, Lactococcus lactis subsp. lactis LM1185 was isolated from Hydrangea macrophylla. Strain LM1185 showed 50.5% of acid tolerance at pH 2.5 for 2 h and 30.4% of 0.3% (w/v) bile salt tolerance for 24 h. The antioxidant activity of this strain was measured at 99.4% of 2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity. When RAW 264.7 macrophage cells were treated with strain LM1185, there was no observed cytotoxicity. This strain showed high nitric oxide production and mRNA expression levels of cytokines such as tumor necrosis factor-α and inducible nitric oxide synthase (iNOS). The nuclear factor-kB signaling pathway was activated by this strain resulting in the production of iNOS and cyclooxygenase-2 determined by western blotting. The present results indicated that L. lactis subsp. lactis LM1185 could be used as potential probiotics and may play a crucial role in the immunostimulatory effect on macrophages.

자작나무에서 자라는 버섯에는 차가버섯(Inonotus Obli-quus) 과 나스또야식 뚜르또빅(Fomes fomentarius (Fr.) Gill)과 로쥐느이 뚜르또빅(Phellinus igniarius(L. ex Fr) Quel)의 3종류가 있다. 이 중에서 차가버섯만 약효가 있고 독성이 없음이 알려져 있으나 다른 버섯은 아직 약효나 독성에 대해 확인 된 것이 없다. 차가버섯을 채취하는 사람들이 차가버섯과 함께 다른 버섯들도 같이 채취하여 주생산지인 러시아뿐만 아니라 국내에서 수입한 차가버섯을 구입할 때 다른 버섯이 섞여 있는지 확인하기가 어렵고 품질의 규격기준도 거의 외관에 의존하고 있는 실정이다. 본연구자들은 국내시장에 반입되어 유통 되고 있는 17종의 차가버섯을 수집하여 5.8S 함유 ITS (including ITS1, 5.8S and ITS2)부위를 이용한 PCR 증폭시험방법으로 DNA 염기서열을 비교분석 하였다. 차가버섯 시료의 DNA추출은 Chen 등(1999)이 사용한 LETS 방법 등으로 추출, 정제하여 사용하였다. 시험 결과 17개 샘풀 중 12종은 염기 서열 간 97% 이상 상동성이 매우 높아 차가버섯(약 70%)으로 확인되었고 5종은 PCR 증폭산물이 거의 이루어지지 않는 이종의 버섯으로 확인되었다. 본시험 결과, 시중의 차가버섯 중 일부는 차가버섯이 아니거나 이종의 버섯이 혼입된 채 유통되고 있다는 것을 확인 할 수 있었다. 향후 이와 같은 신속하고 정확한 유전자 감별법은 판별이 어려운 버섯류 품질관리에 유익하게 사용 될 것으로 판단된다.
