        Method Validation of 16 Types of Structural Amino Acids using an Automated Amino Acid Analyzer

        An analytical method for the simultaneousdetermination of 16 types of structural amino acids ininfant formula using an automated amino acid analyzerwas validated through tests of precision, accuracy andlinearity. The automated analysis of the amino acids wasperformed on an ion-exchange packed column with avisible detector. The certificated reference material (CRM)1546 from NIST was used as the test sample to determinethe precision and accuracy of the analytical method. Theregression analyses revealed good correlations [correlationcoefficient (r2)] that were greater than 0.99. The recoveryvalues of the amino acids were ranged from 87.18 to118.08%. The limits of detection and limits of quantificationwere less than 0.059 mg/100 g and less than 0.198 mg/100 g, respectively. The intra- and inter-day precisionswere less than 14.62% in the CRM sample.

        Hanbok had the history of more than 2,000 years, so Hanbok represents the traditional Korean style of dress design. Since 1980's, Hanbok has been changed to be suitable for contemporary society with diverse titles such as reformed Korean clothes, living Korean clothes, contemporary Korean clothes, and fashion Korean clothes, etc. However, Korean clothes of such a new style have not overcome a limitation in traditional styles in terms of design. In this regard, it is necessary for us to try to have the world fashion industry recognize the fashion look of 'Korean style' through the contemporary fashion of Korean image and to pursue the globalization of Korean style through this fashion. This study is firstly to review the characteristics of Korean beauty, secondly to examine the concept in Korean fashion design, and thirdly to iud out the characteristics of contemporary Korean fashion design through the study of successful cases. The characteristics of Korean beauty can be classified into natural beauty, abstract beauty, and humorous beauty. Korean beauty shows the nature and the life of Korean people, and it should make people aware of the character and the tradition of Korean culture. The Korean fashion design is the one that utilize the Korea's unique sentiment or sense of Korean beauty that has been succeeded from the ancient to the present into fashion design by way of visual element like design, color and texture. And the successful Korean fashion designs were the universal fashion designs that enable Korean culture to be hewn to the world through fashion and that enable the entire world market to agree and to accept Korean style. In order to create the global Korean design, it is necessary to assimilate the Western style to Korean style and newly create it into a compromised form. And it is necessary to look for a universal sense of Korean beauty that could be accepted by the people of the world.

        Analysis of E,E-farnesol and squalene in makgeolli using stir bar sorptive extraction coupled with gas chromatography-mass spectrometry

        The aim of this study was to establish an analytical method for the determination of E,E-farnesoland squalene in makgeolli, which is a traditional type of Korean fermented rice wine. E,E-farnesol and squalenein makgeolli were extracted using stir bar sorptive extraction (SBSE) coupled with gas chromatography-massspectrometry. SBSE was found to be an effective method for analyzing the E,E-farnesol and squalene levelsin makgeolli. The linear dynamic range of the SBSE method for detecting E,E-farnesol and squalene rangedfrom 0.5 to 200 ng/mL with R2=0.9974 for E,E-farnesol and 100 to 50000 ng/mL with R2=0.9982 for squalene. The limit of detection and the limit of quantification using the SBSE method were 0.1 and 0.5 ng/mL for E,Efarnesoland 15.0 and 40.0 ng/mL for squalene, respectively. The average recoveries obtained were, quantitatively,101-107% for E,E-farnesol and 98-103% for squalene, respectively, supporting the accuracy of the SBSE-GCMSmethod.

        Method Validation of Analytical Method for 12 Flavonol Glycosides in Foods Using Ultra High-performance Liquid Chromatography Coupled with Photodiode Array Detection

        An analytical method for the simultaneous determination of 12 flavonol glycosides in buckwheat, black tea, and wild parsley using ultra high-performance liquid chromatography (UHPLC) coupled with a simple liquid extraction method using dimethyl sulfoxide (DMSO) was validated in precision, accuracy, and linearity. The UHPLC separation of target compounds was performed on a C18 column using a photodiode array (PDA) detector and the wavelength was fixed at 350 nm. The recovery values for flavonol glycosides ranged from 85.44 to 108.79%. The limits of detection and limits of quantification were less than 0.32 mg/kg and less than 0.97 mg/kg, respectively. The intraday and interday precisions were less than 13.69% for all the test samples. This method coupled with UHPLCPDA detection could be expected to provide more convenient sample preparation than conventional methods in the tested foods.

        액체크로마토그래피를 이용한 계란 중 테트라사이클린계 항생물질의 동시분석법 개발

        An analytical method for the simultaneous determination of four tetracycline (oxytetracycline, tet-racycline, chlortetracycline, doxycycline) in egg samples was developed and validated using liquid chromatographywith ultraviolet detection. Egg samples were extracted by the liquid-liquid extraction based on acetonitrile. The chro-matographic separation was achieved on a reverse phase C8 column with gradient elution using a mobile phase of 20mM oxalic acid (pH 1.5)/acetonitrile. The procedure was validated according to the Food Drugs Administrationguideline determing accuracy, precision, and limit of detection. Mean recovery of tetracyclines from spiked eggsamples (50, 100, 200, 400, and 800 g/kg) were 78.8-109.3%. Linearity in concentration range of 50-800 g/kg wasobtained with the correlation coefficient (r2) of 0.994-0.999. The intra- and inter-day precision (relative standard devi-ation; RSD) was between 0.3-12.8 and 0.2-11.7%, respectively. Limit of detection (LOD) and limit of quantification(LOQ) for the investigated tetracyclines were 30 and 50 g/kg depending on eg samples, respectively. This methodwas reliable, sensitive, economical and suitable for routine monitoring of tetracycline residues in dairy egg.

        액체크로마토그래피를 이용한 광어 및 계란 중 퀴놀론계의 동시분석법 개발

        An analytical method for the simultaneous determination of nine quinolones (QNs) namely, marbofloxacin, norfloxacin(IS), ciprofloxacin, danofloxacin, enrofloxacin, sarafloxacin, difloxacin, oxolinic acid, and flumequine in flatfish and egg was developed and validated using liquid chromatography with fluorescence detection (LC-FD). The samples were extracted using a traditional liquid-liquid extraction process; deproteinization was accomplished by the addition of trichloroacetic acid and acetonitrile (ACN), and defatting was performed with hexane. Chromatographic separation was achieved on a reverse phase C8 column with gradient elution using a mobile phase of 200 mM ammonium acetate buffer (pH 4.5) and ACN. The proposed method was validated according to the CODEX guideline. Mean recoveries of QNs from flatfish and egg were 89.6-106.5 % with relative standard deviations (RSDs) below 15 % at three different concentrations of 50, 100 and 500 μg/kg. Linearity was obtained with a correlation coefficient (r²) of 0.9989-1.0000. The LOD for the investigated QNs was 1-16 μg/kg depending on flatfish and egg. The present method can be applied simultaneously to determine QNs in muscle of flatfish and egg.         Determination of alcohols in various fermented food matrices using gas chromatography-flame ionization detector for halal certification

        In this study, an analytical method for the determination of alcohols by gas chromatography-flame ionization detection was applied to fermented foods. The limit of detection and limit of quantitation were 0.79 and 2.40 mg/kg for methanol, 0.55 and 1.66 mg/kg for ethanol, 0.51 and 1.56 mg/kg for n-propanol, 0.35 and 1.05 mg/kg for n-butanol, and 0.38 and 1.16 mg/kg for n-pentanol, respectively. The recoveries from the matrices of gochujang, soy sauce, and kimchi were 93.80–102.03%, 93.27–99.69% and 89.06–102.17%, respectively, and the corresponding intra- and inter-day relative standard deviations were below 5.33%, 5.35% and 4.10%. In 95 fermented foods, ethanol showed the highest mean, median and maximum values among the five alcohols. The detection rate of ethanol was 86.3% among all samples and 100% in gochujang and gochujang-based sauces. A total of 22 samples had an alcohol content above 0.5%, of which 16 were gochujang.
