        Phellinus baumii 자실체 열수 추출물의 Sarcoma-180에 대한 항암 및 면역효과

        국내에서 재배되고 있는 약용버섯인 상황(P. baumii) 자실체의 열수 추출물로 부터 건강 기능성 식품 및 의약품개발의 기초자료를 제공하고자 항암실험 및 면역효과에 대한 실험을 수행하였다. 본 연구에서 사용한 PMO-P4의 경우 rDNA의 ITS부위의 염기서열 분석을 통한 계통분석 결과 P. baumii로 판명 되었으며, PMO-P4 자실체로부터 열수 추출한 성분(HWE-P4)을 160mg/kg/day의 농도로 하여 Sarcoma-180 암세포를 유발한 마우스에 경구투여한 결과 A군의 경우, 종양억제효과는 35.3%로 대조군보다 유의성 있게 감소하였으며(p<0.05), 생명연장 효과는 156%증가한 것으로 나타났다. 시료의 경구투여 방법에 있어서도 시료를 먼저 2주간 투여한 후 암세포를 접종한 A군의 경우가 시료 투여와 암세포 접종을 동시에 실시한 B군의 경우 보다 종양억제효과 및 생명연장효과가 높았음을 알 수 있었다. A군의 경우 대조군에 비하여 CD4/CD8의 비율이 71.4%증가 하였으며, CD25(IL-2receptor chain)분자의 발현을 5배정도 증가시키는 것으로 관찰되었다. 이러한 결과로 보아 P. baumii로 밝혀진 PMO-P4 자실체로부터 열수 추출한 성분의 항암효과는 암세포를 직접 공격하여 항암효과를 나타내기 보다는 T cell등의 면역세포를 활성화시킴으로써 암세포를 억제 혹은 사멸시키는 것으로 생각된다. A hot water extract(HWE-P4) was separated from the fruit bodies of PMO-P4, and its antitumor and immunomodulatory activities against sarcoma-180 in ICR mice were investigated. The internal transcribed spacer(ITS) regions from PMO-P4 were amplified using polymerase chain reaction(PCR) and sequenced. The results revealed that PMO-P4 was belong to the Phellinus baumii. When oral administration at the dose of 160mg/kg/day in the mice until the end of the experiment with 2 week's pre-feeding of the HWE-P4, the survival rate of the mice was 152% for 50days after the inoculation of sarcoma-180 and the suppression rate of the tumor growth was 35.3%(p<0.05) for 28 days after inoculation of sarcoma-180. The HWE-P4 increased 71.4% of the CD4/CD8 ratio and 5-fold of the expression of CD25(IL-2 receptor chain) compared with the control. From these results, the antitumor activity of HWE-P4 is exerted through its immunomodulating activity on the host's immune system.

        마우스에서 Sarcoma-180에 대한 Bifidobacterium adolesentis ATCC-15703의 항암 효과

        ICR 마우스에서 Bifidobacterium adolesentis ATCC-15703 균주의 Sarcoma-180(S-180) 복수암과 고형암에 대한 항암효과 및 면역증강 효과에 대하여 연구하였다. S-180 세포(1×10 exp (6) cells/mouse)를 마우스의 복강에 접종하였을 때, S-180 복수암은 100% 유발되었으며, ICR 마우스의 체중은 암세포 접종 후 9일부터 급증하여 폐사 직전에는 정상 마우스의 1.4배였다. S-180 세포(2×10 exp (6) cells/mouse)를 ICR 마우스의 우측서혜부에 피하 접종한 후 21일에 100%의 고형암 유발을 확인하였으며, 적출된 암괴의 무게는 6.90±2.57g이었다. 균체를 복강에 100㎍ 투여한 마우스에서 50일간의 평균 생존율(T/C %)은 132%였으며, ICR 마우스의 체중증가는 억제되었다. S-180 세포 접종후 100㎍을 ICR 마우스의 우측 서혜부에 투여했을 때, 21일째의 종양성장 억제율은 62.3%였으며 시험군 8마리 중 2마리는 암괴가 형성되지 않았다. 균체지체는 S-180 암세포에 직접적인 독성을 나타내지 않는 것으로 확인되었다. 균체를 암세포 배양액에 100∼1000 ㎍/T-flask의 농도로 첨가하였을 때, S-180 암세포들은 대조와 거의 동일하게 성장하였다. 살균건조된 균체는 대식세포의 활성을 현저하게 증가시켜 주었다. 100 ㎍을 ICR 마우스의 복강에 투여한 후 1일째 대식세포 수가 대조에 비해 약 8배 증가되었으며, acid phophatase 활성도도 투여 후 2일째에 가장 높게 나타났다. Antitumor and immunoadjuvant activities of Bifidobacterium adolesentis ATCC-15703 were investigated against the ascites and solid forms of tumor induced by Sarcoma 180(S-180) in ICR mice. When 100 ㎍ of the bacterium was intraperitonealy administered in the mice, the survival rate(T/C %) of the mice was 132% for 50 days after the inoculation of S-180, however the increment of their body weights was suppressed. When the 100 ㎍ was subcutaneously administratered at the right grion, the suppresion rate of the tumor growth on the 21st day after inoculation of S-180 were 62.3%. Complete supression of the tumor growth was observed from 2 out of 8 test mice. The bacterium itself appeared to have noncytotoxic substance against the S-180 in vitro. The bacterial administration markedly enhanced the activity of peritoneal macrophages. The number of peritoneal macrophages at one day after the administration of the 100 ㎍ increased about 8 times as much as that of the control, and the highest activity of acid phosphatase appeared at 2 days.

      • Inhibition of Tumor Formation and Changes in Hepatic Enzyme Activities by Kimchi Extracts in Sarcoma-180 Cell Transplanted Mice

        Inhibitory effects of the methanol extract, hexane extract, methanol soluble fraction (MSF) and juice from 3 weeks fermented kimchi on the tumor formation in sarcoma-180 cell transplanted mice were studied. Effects of the solvent extracts and juice of the kimchi on the levels of lipid peroxide, glutathione, and the enzyme activities of the liver were also investigated in normal and sarcoma-180 cell transplanted mice. At 32 days following transplantation, MSF reduced the tumor formation by 54% compared with the control group, resulting in the smallest tumor weight. Lipid peroxide content in liver increased by the transplantation of sarcoma-180 cells. However, it decreased when MSF of kimchi was treated to the mice. MSF also suppressed xanthine oxidase activity in cytosol of the liver cells in mice transplanted by sarcoma-180 cells. Kimchi extracts had no inhibitory effect on hepatic aminopyrine-N-demethylase activity in sarcoma-180 cell transplanted or normal mice. Methanol extract and hexane extract of kimchi slightly increased hepatic glutathione contents in sarcoma-180 teated mice. The injection of MSF from kimchi markedly increased glutathione levels in the liver of sarcoma-180 treated mice compared to the controls. The MSF recovered the activities of hepatic glutathione reductase and glutathione S-transferase that decreased by the injection of sarcoma-180 cells. These results showed that MSF of kimchi could suppress the growth of tumors, inhibiting lipid peroxide production and xanthine oxidase activity, in mice. We also suggested that kimchi extract might play an important role in the prevention of cancer by enhancement of the glutathione level itself as well as via glutathione reductase and glutathione S-transferase.

      • Sarcoma 180 이식마우스에서 Interleukin-2, 4 10의 변화

        Experiments were undertaken to investigate the effect of cancer cell, sarcoma 180 (S180) on production of interleukin(IL)-2, 4, and 10 in BALB/c mice. Splenocytes of normal mice were incubated with S 180 cells. The culture supernatants were collected at 2, 4, 6, 24, and 72 hr and assessed for IL-2, 4, and IL-10 production by enzyme-linked immunosorbent assay. The production of IL-2 in the culture supernatant of adherent cells exposed with S 180 was not detected, but it increased at 24 and 72 hr in culture with non-adherent or whole splenocyte. The production of IL-4 in the culture supernatant of S 180, adherent, non-adherent or whole splenocyte was detected, but by co-culture with S 180 it showed significant increase at 72 hr. Also, the production of IL-10 in the culture supernatant of S 180, adherent, non-adherent, or whole splenocytes was detected. By exposure with S 180 it increased to 10-80 times in comparison with the control. Normal mice were transplanted with S 180 into peritoneum. Sera and peritoneal fluids(PF) were collected at 2, 4, 6, and 24 hr. The production of IL-2 in sera was not detected, but in PF it was a little. The production of IL-4 in sera or PF was detected, but it showed the significant decrease in comparison with the control. The production of IL-10 in sera was detected in transplantation or control group, and it increased in transplantation group. The production of IL-10 in PF showed the increase at 24 hr. By exposure of S 180 tumor cells in immuno-competent cells or transplantation with S 180 into mouse, IL-4 decreased in sera and PF, but IL-10 showed the remarkable increase in the culture supernatant of splenocytes and in sea. These data suggest that the unbalanced conditions such as suppression of IL-4 production and overproduction of IL-10 in vivo may be responsible for dysfunction of immune system.

      • Sarcoma-180 세포를 이용한 고등균류의 항암효과

        상황버섯과 버들송이 균사체를 중심으로 하여 이들 균사체로부터 열수 추출된 세포내다당류와 배양액에서 회수된 다당류를 이용하여 sarcoma180에 대한 항암효과를 실험한 결과는 다음과 같다. Sarcoma-180 cell-line으로 복수임을 유발시킨 mice에 균사체와 배양액으로부터 회수한 다당체를 50㎍/㎏ 투여시의 체중증가량은 대조군에 비하여, 현저하게 체중증가량이 감소하였으며, 균사체에서 보다 배양액에서의 다당류에서 더욱 효과 있는 것으로 나타났다. 수명연장 효과를 살펴본 결과는 대조군에 비하여 상황과 버들송이 균사체의 세포외단백다당체에서 각각 47.8%, 69.6%, 32.2% 및 23.9%의 생존율을 나타내었다. In vitro에서의 S-180에 대한 증식 억제효과는 대조군에 비하여 모든 군에서 억제효과가 나타났으며, 시료의 농도에 비례하여 억제되는 것으로 나타났다. This study examine antitumor effects, the intracellular and extracellular, extracts from the liquid culture of the Phellinius igniarius and the Agrocybe cylindracea by using sarcoma180 tumor cells. The experimented was carred out by mice body weight increation, the life prolongation and tumor cell groth inhibition in vitro. The resultes obtained from the experiment as follows; Total body weight of mice, inoculated with sarcoma-180ascites tumor cells challenge, were decreased significantly in experimental groups, intracellular and extracellular polysaccharide extracts from the liquid culture of the Phellinius igniarius and the Agrocybe cylindracea than control groups. The life prolongation effects were 47.8%, 69.6%, 32.2% and 23.9%, respectively when intracellular and extracellular, extracts from the liquid culture of the Phellinius igniarius and the Agrocy cylindracea were administrated to the mice. Inhibitory efect on the growth rate of the sarcoma-180 was stronger in intracellular and extracellular, extracts from the liquid culture of the Phellinius igniarius and the Agrocybe cylindracea than control group. The growth rates of the sarcoma-180 in medium containing samples were inhibited gradually to a significant degree in proportion to the increase of the samples concentration in vitro.

        온백원이 암세포에 미치는 항암활성 효과

        Objective : The objective of this study was to investigate the experimental efficacy of anti-tumor activity of the complexed herbal formula, Onbaekwon (OBW), which was derived from the literature of Traditional Korean Medicine, Dongeuibogam. Methods : Nine Cancer cell lines, LoVo, MCF-7, HepG2, AGS, A549, NCI-H69, HL-60, Sarcoma 180, LL/2, were prepared and the cytotoxicity was assessed by 3-(4,5-dimethylthiazol-2yl)-2,5-dephenyl tetrazolium bromide (MTT) assay. Four of them, NCI-H69, HL-60, Sarcoma 180, and LL/2, showed strong cytotoxic activities and they were additionally undergone flow cytometry to find out their effects on apoptosis. ICR male mice were implanted with Sarcoma 180 intraperitoneally and divided into 8 species for each group. Control group was treated with normal saline, positive control group was treated with cyclophosphamide 8mg/kg, and experimental group was treated with OBW 1 g/kg. Results : Among 9 cancer cell lines, NCI-H69, HL-60, Sarcoma 180, and LL/2, expressed less than 0.10 mg/ml of $IC_{50}$ under 0.1~1mg/ml of OBW. NCI-H69, HL-60, Sarcoma 180, and LL/2, showed dose-dependent efficacy of apoptosis. When Sarcoma 180 cancer cell was implanted in ICR male mice and treated with the OBW, they prolonged the median overall survival for 0.8 days, from 17.5 to 18.3. Conclusion : OBW showed strong cytotoxicity to some cancer cells, which are NCI-H69, HL-60, Sarcoma 180, and LL/2, and its apoptotic activity was dose-dependent. OBW prolonged the median survival of mice implanted with Sarcoma 180. Further researches would be expected to support the efficacy of OBW.

        행인약침(杏仁藥鍼)의 급성(急性) 아급성(亞急性) 독성실험(毒性實驗) 및 Sarcoma-180 항암효과(抗癌效果)에 관(關)한 실험적(實驗的) 연구(硏究)

        Objective: The purpose of this study was to investigate acute and subacute toxicity and sarcoma-180 anti-cancer effects of Herbal acupuncture with Anneniacae amarum semen (Haeng-in) in mice and rats. Method: Balble mice were injected intraperitoneany with Haeng-In extract for $LD_{50}$ and acute toxicity test. Sprague-Dawley rats were injected intraperitoneally with Haeng-In extract for subacute toxicity test. TheAnneniacae amarum semen Herbal-Acupuncture was injected on Chung-wan (CV12) of mice with Sarcoma-180 cancer cell line. Results: 1. $LD_{50}$ was uncountable as none of the subjects expired from the treatment groups during the test. 2. The clinical signs and the body weight of mice treated with 0.1cc and 0.2cc Haeng-In extract were not affected during the acute toxicity test. 3. In acute toxicity test of serum biochemical values of mice, total protein and albumin were decreased in treatment group Ⅰ. Glucose was increased, and total cholesterol was decreased in treatment groups. GPT was increased in treatment group Ⅰ. 4. In subacute toxicity test, toxic symptoms were not detected in the treatment groups. 5. In subacute toxicity test, the body weight was increased in treatment groups on 14th and 21st day. 6. In subacute toxicity test. liver weight was increased in treatment group Ⅱ, and spleen weight was increased in treatment group Ⅱ. Lung weight was increased in an the treatment groups.(p<0.05) 7. In subacute toxicity test, severe tissue injury was found in lung and liver, especially treatment group Ⅰshowed more significant lung damage compared to treatment group l. 8. In subacute toxicity test, WBC. MCH and MCHC were increased in an the treatment groups, RBC, HGB and HCT were decreased in treatment group H(p<0.05). 9. In subacute toxicity test of serum biochemical values of rats, triglyceride was decreased in all the treatment groups. ALP was decreased in treatment group Ⅰ. and creatinine was decreased in treatment group Ⅱ. BUN/CR was increased in treatment group Ⅱ(p<0.05). 10. Median survival time of Sarcoma-180 cancer cell treated with Haeng-In was increased in all the treatment groups by twenty percent, compared to the control group(p<0.05). 11. Natural killer cell activity about the Sarcoma-180 cell was decreased at the ratio of 100:1 but was increased at the ratio of 10:1. In treatment group Ⅱ, increase was found at the ratio of 100:1 and 50:1 (p<0.05). 12. Interleukin-2 productivity of the Sarcoma-180 cell was decreased in treatment group I, but was increased in treatment group Ⅱ(p<0.05). Conclusion: According to the results, we can conclude Herbal-acupuncture with Anneniacae amarum semen caused toxicity, and had effects in Sarcoma-180 cancer cell.
