        This research is based on the drastically increased occurrence and death rate of diabetes recorded within the past ten years and the analysis of the Korean government's Diabetes Mellitus Management system in Korea in handling this alarming problem. The information used for this analysis is based on documents and statistics from the Ministry of Health and Welfare Statistics(2001, 2002) and also, health insurance statistics and documents from the National Health Insurance Corporation, 2001. The analysis of Health Plan 2010 will also be a key issue discussed in this paper. The Korean government's Diabetes Mellitus Management Plan's aim of increasing the life and healthy life expectancy while decreasing the medical costs at the same time is considered as a vital link in the government's Chronic Disease Management Plan. The main aim is to decrease the death rate, due to diabetes, from 22.6 people in 2000 to 19.0 by the year 2010. Health Plan 2010 plans to increase the role of public health centers. It also plans to form a system that will be used as a device in decreasing chronic diseases such as high blood pressure, diabetes and increasing general health by using the strategies of Health Plan 2010 against diabetes -to increase the government's role in public health. Korea's death rate, due to diabetes, is 2.2 times higher than that of Japan. Korea's being 22.6% compared to that of Japan which is only 10.1%. From now on our concerns should be directed in education and manpower. We need to have more specialized personnel who can be responsible for the nutrition, exercise, prevention and care of the public. Education is the fundamental basis for the control of Diabetes and is known throughout the world as the most cost-effective treatment. From this point on, the Government should concentrate on educating the people on the importance of healthy life styles. To do this it needs to provide the necessary funds needed in Health promotion and health education, and also to provide for professional personnel(eg. health educator).

      • 남자 초·중학생의 무산소성 작업능력과 무산소성 파워의 관계

        The critical power(CP) is the highest intensity to sustain for a long time without a fatigue. The anaerobic work capacity(AWC) is the highest anaerobic capacity to be provided by anaerobic energy system without the infection of hypoxia. The purpose of this study was to determine the relationship between AWC and AnP and to verify the validity of AWC. The subjects were consisted of 10 elementary and 10 middle school male students(12.8±1.3yrs, 159.3±12.1㎝, 49.4±11.8㎏). The critical power and anaerobic work capacity were calculated by work-time relationship after four cycle ergometry test to fatigue from 1 to 10 minute. The anaerobic power included Margaria-Kalmen test, Sargent jump and Wingate test. The main finding were as follows. The AWC of elementary and middle school students. were 6.47±2.63KJ and 11.10±3.01KJ. The AWC of elementary students was 58.3% of the middle school students's one. The AnP of elementary students were 47.2∼66.5% of the middle school students's one. And there were high correlation between AWC and AnP(0.64∼0.87, P<0.01). This study showed that AWC is useful index on AnP both elementary and middle school students.

        Health Risk Appraisal(HRA) is usually defined as a process by which we expect of individual's chances of acquiring specific diseases within a defined period of time. The concept of worksite as an area of health maintenance and promotion is newly developing. Our movement for health promotion of employees has been increasingly known in the worksite. The aim of this study is to develop health risk appraisal tools about health promotion at the worksite for employees. We performed this study by two steps: one step was to develop a HRA questionnaire of worksites in Korea, another was to evaluate the reliability of the questionnaire. For developing HRA questionnaire, we reviewed scientific examples at first, and then weighted the score by delphi. To evaluate the reliability of developed questionnaire, we carried out survey by test-retest method. A total of 131 employees completed HRA questionnaire on two times. The results are as follows. The total score of health risk in unhealthy workers was higher than that of healthy workers. The range of test-retest reliability of responses to the questionnaire was 0.57-0.94. Therefore it seems that this questionnaire was very suitable to assess the health behavior of workers. In conclusion, the developed HRA questionnaire can be used as a tool for evaluating health behavior and for providing health counseling materials.

        Acinar cell carcinoma is a rare tumor that represents 1~2% of al1 pancreatic cancers. Clinical and radiologic findings are inconclusive in this disease Acinar cell carcinoma is characterized by rapid progression and early metastasis, which lead to its poor prognosis. A 41-year-o1d man was admitted to our hospital for abdominal pain. Abdominal computed tomography (CT) and positron emission tomography-computed tomography (PET-CT) showed a splenic mass, which was being invaded by a pancreatic tail mass and which had increased ^(18)F-fluorodeoxyglucose (FDG) uptake Primary radical distal pancreatectomy and splenectomy were performed. Pathologic findings revealed an acinar cell carcinoma of the Pancreas The patient underwent a total gastrectomy three months later because of gastric recurrence Four months later, multiple hepatic metastases were discovered, and the patient underwent a left hepatectomy During treatment with capecitabine, there was no evidence of tumor progression for 14 months. We report a case of metastatic pancreatic acinar cell carcinoma, which did not progress for an extended period while the patient was being treated with capecitabme.

        Primary retroperitoneal mucinous cystadenocarcinoma is a very rare malignancy, and little is known concerning its Pathogenesis, optimal treatment, and prognosis. A 29-year-o1d pregnant woman (21 weeks) Presented with abdominal discomfort CA 19-9, CA 125, and CEA were normal Abdominal CT scanning revealed a 19x15x13 cm retropentoneal tumor Exploratory laparotomy and tumor excision were performed. Mucinous retropentoneal implants were removed as completely as possible Histologically, the tumor showed focal areas of capsular invasion, but free resection margins The uterus and both ovaries were normal in appearance No adjuvant therapy was pursued. Six months later, Peritoneal and bilateral ovarian metastases were discovered. Hence, we report the details of this case of primary retroperitoneal mucinous Cystadeno-carcinoma and present a review of the literature.

        Induction of apoptosis in MCF-7 and MDA-MB-231 breast cancer cells by Oligonol is mediated by Bcl-2 family regulation and MEK/ERK signaling

        Oligonol is a novel catechin-rich biotechnology product. The role of oligonol in modulating intracellular signaling mechanisms was investigated with the view of demonstrating its potential chemopreventive effect and the ability to inhibit cell proliferation using the estrogen-responsive MCF-7 and the estrogen-unresponsive MDA-MB-231 human breast cancer cell lines. Cell survival assay indicated that Oligonol was cytotoxic to both cells. Oligonol triggered apoptosis as revealed by the morphological features typical of nucleus staining and the accumulation of sub-G1 peak. Treatment with 25 μg/ml Oligonol resulted in an activation of caspase-7 and up-regulation of Bad on MCF-7 cells, while the Oligonol (20 μg/ml) induced up-regulation of Bcl-2 protein in a time–response manner on MDA-MB-231 cells. ERK1/2 in both cells were inactivated after Oligonol treatment in a time-dependent manner, and also inactivated upstream MEK1/2. Oligonol triggers apoptosis in MCF-7 and MDA-MB-231 cells through the modulation of pro-apoptotic Bcl-2 family proteins and MEK/ERK signaling pathway.

        Ubiquitin E3 ligases in cancer: somatic mutation and amplification

        Defects in DNA double-strand break (DSB) repair signaling permit cancer cells to accumulate genomic alterations that confer their aggressive phenotype. Nevertheless, tumors depend on residual DNA repair abilities to survive the DNA damage induced by genotoxic stress. This is why only isolated DNA repair signaling is inactivated in cancer cells. DNA DSB repair signaling contributes to general mechanism for various types of lesions in diverse cell cycle phases. DNA DSB repair genes are frequently mutated and amplified in cancer; however, limited data exist regarding the overall genomic prospect and functional result of these modifications. We list the DNA repair genes and related E3 ligases. Mutation and expression frequencies of these genes were analyzed in COSMIC and TCGA. The 11 genes with a high frequency of mutation differed between cancers, and mutations in many DNA DSB repair E3 ligase genes were related to a higher total mutation burden. DNA DSB repair E3 ligase genes are involved in tumor suppressive or oncogenic functions, such as RNF168 and FBXW7, by assisting the functionality of these genomic alterations. DNA damage response-related E3 ligases, such as RNF168, FBXW7, and HERC2, were generated with more than 10% mutation in several cancer cells. This study provides a broad list of candidate genes as potential biomarkers for genomic instability and novel therapeutic targets in cancer. As a DSB related proteins considerably appear the possibilities for targeting DNA repair defective tumors or hyperactive DNA repair tumors. Based on recent research, we describe the relationship between unstable DSB repairs and DSB-related E3 ligases. [BMB Reports 2023; 56(5): 265-274]

      • Introduction to research facility of laboratory animal center, Daegu-Gyeongbuk Medical Innovation Foundation (DGMIF)

        Daegu-Gyeongbuk Medical Innovation Foundation (DGMIF) consists of four research facilities (new drug development center, medical device development center, laboratory animal center, and clinical drug manufacturing center). The laboratory animal center is a facility to support the development of new drugs and medical devices, and plays a key role in evaluating the efficacy and safety tests of new drugs and medical devices through the management of laboratory animals, production of animal disease model, biometric analysis, experimental animal resource management, and in-vivo tests. The laboratory animal center is advancing image analysis equipment and surgical techniques to support high-quality animal experiments requiring high technology, and is improving the welfare of experimental animals through professional animal management, health monitoring, IACUC approval, post approval monitoring, and veterinary care to achieve reliable research results. DGMIF LAC has facilities for rodents including mouse and rats, rabbits, dogs, pigs, and monkeys, and the rodent animal facility is using IVC and ISO-N cage system and areas are divided into four areas including species conservation area, small animal testing area, re-entry area for imaging analysis, and ABSL-2 area for infection tests using ABSL-2 level microorganism. For high-quality animal management, DGMIF LAC acquired ICLAS PEP (Performance evaluation program for diagnostic laboratories, International Council for Laboratory Animal Science) accredited for the first in South Korea in 2017, KESAF (Korean Excellent Laboratory Animal Facility) in 2016, and AAALAC (The Association for Assessment and Accreditation of Laboratory Animal Care) International Full Accreditation in 2020. It is the only institution in Daegu and North Gyeongsang Province that that has received these two accreditations. Since the operation of the animal facility in 2014, the number of laboratory animals has been steadily increasing. Furthermore, additional facility for mini-pigs was completely built. DGMIF LAC will play a major role in the evaluation of the effecacy tests for new drugs and medical devices through the maintenance and development of high-quality laboratory animals that comply with international standards.
