      • DJ-1/park7 modulates vasorelaxation and blood pressure via epigenetic modification of endothelial nitric oxide synthase

        <P><B>Aims</B></P><P>DJ-1/park7, a multifunctional protein, may play essential roles in the vascular system. However, the function of DJ-1/park7 in vascular contractility has remained unclear. The present study was designed to investigate whether the DJ-1/park7 is involved in the regulation of vascular contractility and systolic blood pressure (SBP).</P><P><B>Methods and results</B></P><P>Norepinephrine (NE) elevated contraction in endothelium-intact vessels in a dose-dependent manner, to a greater extent in DJ-1/park7 knockout (DJ-1/park7<SUP>−/−</SUP>) mice than in wild-type (DJ-1/park7<SUP>+/+</SUP>) mice. Acetylcholine inhibited NE-evoked contraction in endothelium-intact vessels, and this was markedly impaired in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP>. Nitric oxide (NO) production (82.1 ± 2.8% of control) and endothelial NO synthase (eNOS) expression (61.7 ± 8.9%) were lower, but H<SUB>2</SUB>O<SUB>2</SUB> production (126.4 ± 8.6%) was higher, in endothelial cells from DJ-1/park7<SUP>−/−</SUP> mice than in those from DJ-1/park7<SUP>+/+</SUP> controls; these effects were reversed by DJ-1/park7-overexpressing endothelial cells from DJ-1/park7<SUP>−/−</SUP> mice. Histone deacetylase (HDAC)-1 recruitment and H3 histone acetylation at the eNOS promoter were elevated and diminished, respectively, in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP> controls. Moreover, SBP was significantly elevated in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP> controls, but this elevation was inhibited in mice treated with valproic acid, an inhibitor of Class I HDACs including HDAC-1.</P><P><B>Conclusion</B></P><P>These results demonstrate that DJ-1/park7 protein may be implicated in the regulation of vascular contractility and blood pressure, probably by the impairment of NO production through H<SUB>2</SUB>O<SUB>2</SUB>-mediated epigenetic inhibition of eNOS expression.</P>

      • Development of a Mobile Application, "Wild Flowers of Bukhansan National Park (version 1.0)", for Identification of Plants in Bukhansan National Park

        We developed the educational purpose mobile application, named "Wild Flowers of Bukhansan National Park (version 1.0)", aiming for easy identification of wild flowers for students and visitors in the park. When visitors find a flower or part of plant in the park, visitors can search for its name utilizing the pictures and characters provided in their own smartphone mobile devices or tablet PCs. The application provides pictures of wild flowers in the park and character-based searching system based on 12 diagnostic features (e.g., growth form, leaf arrangement, flower symmetry, petal color, petal number, sepal number, etc). We adopted the complete floristic survey of Chung and Lee (1962) and added species that we confirmed their distribution in the park during the development of this application. In summary, number of vascular plants in this park was estimated to be 428 taxa including 100 families, 280 genera, 327 species, 1 subspecies, 50 varieties, and 5 formas. We provided a total of 588 pictures representing 358 taxa and each taxon includes multiple pictures in many cases. Included identification quizzes can be an efficient educational tool as well as fun activity for students and visitors who are learning plant species in Korea. Our next step will include GPS function in the application for indicating visitor's location and for providing previously reported sites of the species that we interested in the map of the park. The future application which includes GPS function will be a valuable tool for the monitoring of rare plants, plant researches related to the climate changes, etc. We currently provide Korean iPhone version only, and English version and both of android versions will be serviced soon.

        DJ-1 protects cell death from a mitochondrial oxidative stress due to GBA1 deficiency

        Background GBA1 mutations are the most common genetic risk factor for development of Parkinson’s disease (PD). The loss of catalytic activity in GBA1, as well as the reduction of the GBA1 protein in certain cellular compartment, may increase disease progression. However, the mechanisms underlying cellular dysfunction caused by GBA1 deficiency are still mostly unknown. Objective In this study, we focus on the genetic interaction between GBA1 deficiency and PD-causing genes, such as DJ-1, in mitochondrial dysfunction. Methods GBA1 knockout (KO) SH-SY5Y cells were used to assess DJ-1 functions against oxidative stress in vitro. The levels of cellular reactive oxygen species were monitored with MitoSOX reagent. The expression of the PARK7 gene was analyzed using the quantitative real-time PCR (qRT-PCR). To understand the mechanism underlying DJ-1 upregulation in GBA1 KO cells, we assess ROS levels, antioxidant protein, and cell viability in GBA1 KO cells with treatment of ROS inhibitor N-acetyl-cysteine or miglustat, which is an inhibitor of glucosylceramide synthase. Dopaminergic degeneration was assessed from Gba1 L444P heterozygous mice mated with Park7 knockout mice. Results We find that DJ-1 is significantly upregulated in GBA1 KO cells. Elevated levels of DJ-1 are attributed to the transcriptional expression of PARK7 mRNA, but not the inhibition of DJ-1 protein degradation. Because DJ-1 expression is highly linked to oxidative stress, we observe cellular reactive oxygen species (ROS) in GBA1 KO cells. Moreover, several antioxidant gene expressions and protein levels are increased in GBA1 KO cells. To this end, GBA1 KO cells are more susceptible to H2O2-induced cell death. Importantly, there is a significant reduction in dopaminergic neurons in the midbrain from Gba1 L444P heterozygous mice mated with Park7 knockout mice, followed by mild motor dysfunction. Conclusion Taken together, our results suggest that DJ-1 upregulation due to GBA1 deficiency has a protective role against oxidative stress. It may be supposed that mutations or malfunctions in the DJ-1 protein may have disadvantages in the survival of dopaminergic neurons in the brains of patients harboring GBA1 mutations.       • RAW264.7 세포에서 interferon-r 및 LPS에 의해 유도되는 NO생성에 미치는 TALT-35의 영향

        TALP-35 purified from human term placenta is known to increase microtubule polymerization and stabilize the polymerized microtubule. To examine the effect of TALP-35 on immune system this study was performed. MTT assay was performed to investigate the effect of TALP-35 on the proliferation of RAW264.7 cells. TALP-35 dose dependently suppress the proliferation of RAW264.7 cells at high concentration (above 1 μM) in unstimulated cells, in case of 10 μM TALP-35 treated cells the suppression was 25% but in stimulated cells it was only 15%. Cosedimentation assay was carried out to investigate whether TALP-35 can bind to tubulin of RAW264.7, monocyte/macrophage lineage of mouse, and polymerize it. TALP-35 polymerize the tubulin of RAW264.7 cells and sedimented in dose-dependent manner. To investigate the effect of TALP-35 on the expression of iNOS protein western blotting was performed. The expression level of iNOS was decreased dose dependently in high concentration of TALP-35 treatment. To examine the activity of iNOS, secreted NO was determined by method based on Griess reaction. Interferon-γ and LPS-stimulated production of NO from RAW264.7 cells was decreased dose dependently above 0.1 μM concentration of TALP-35 and 50% is decreased at 10μM of it. This study shows TALP-35 can control cytokine induced-iNOS expression therefore it might control inflammatory diseases.

      • 암수동체성 어류 Rivulus marmoratus의 치어에 미치는 몇가지 화학적 발암원의 급성 독성에 관하여

        어독성 검출계로서 암수동체성 어류 Rivulus marmoratus의 가치성을 살펴보고자 본종의 치어에 미치는 N-methyl-N'-nitro-N-nitrosoguanidine(MNNG), N-nitroso-N-methylurea(MNU)와 aflatoxin B₁(AFB₁)의 급성독성을 조사한 결과는 아래와 같다. 1. MNNG의 24hr-??값은 37.7ppm이었고 95% 신뢰한계의 상한치는 38.5ppm, 하한치는 37.0ppm이었다. MNU의 24hr-?? 값은 96.0ppm이었고 상한치는 97.9ppm, 하한치는 94.2ppm이었으며 AFB₁의 24hr-?? 값은 0.94ppm이었고 95% 신뢰한계의 상한치는 0.98ppm, 하한치는 0.90ppm이었다. 2. 모든 처리화합물은 본종에 대해 좁은 농도범위 내에서 급성 독성 효과를 나타내었으며 아울러 뚜렷한 농도반응을 보여주었다. 이상의 결과를 본종이 지닌 어독성 연구 실험동물로서의 가치성과 연관시켜 논의하였다. The acute toxicities of N-methyl-N'-nitro-N-nitrosoguanidine(MNNG), N-nitroso-N-methylurea(MNU) and aflatoxin B₁(AFB₁) to the hermaphroditic fish Rivulus marmoratus larvae were examined to evaluate the possibility of the use of this fish as the assay model of the aquatic toxicology. The results obtained were as follows; 1. The 24hr-?? to MNNG was 37.7 ppm with 95% confidence range value of 38.5~37.0ppm. The ?? for 24hours to MNU and AFB₁were found to be 96.0ppm and 0.94ppm, respectively. Their 95% confidence ranges were 97.9~94.2ppm in MNU and 0.98~0.90ppm in AFB₁. 2. The R. marmoratus larvae responded to narrow range of concentration of all tested chemicals, and toxic effects of the chemicals were clearly dose dependent. The above results were discussed in relation to the usefulness of R. marmoratus as a suitable model for aquatic toxicology.

      • 좌심실 장축 운동의 기능적 의의 (Ⅰ)

        To assess the functional significance of long axis movement which would reflect the fiber orientation of the ventricular muscle, we examined the mitral annulus motion and compared it with standard minor axis movement. M-mode echocardiogram were performed in 24 healthy individuals and 17 mitral valve replaced (MVR) patients with simultaneous EKG and phonocardiogram recordings. The results were as follows : 1. In the controls, the onset of shortening of long axis preceeds that of minor axis (phase difference : 20±3msec, mean± SEM) but in MVR patients this phase difference was reversed (-33±8msec). It means that the spherical shape changes which can be observed in normal controls during isovolumic contraction period were lost in MVR patients. 2. In MVR patients, the time intervals from A2 to the end of shortening of long axis were significantly delayed compared with normal controls (54±3 VS 90±8 msec, P<0.01). 3. In MVR patients, the delayed shortening of long axis was observed in amplitude as well as in time. In summary. we observed the regional nonuniformity between long axis and short axis in normal control and interpreted that it could play a role in modulating ventricular function which was lost in MVR patients.

      • 목련과 수종과 부위에 따른 견직물의 천연염색변이

        In order to establish a dyeing method with Magnoliaceae tree species dyeing was examined with 4 different parts, leaf, stem bark, root, flower of 4 Magnohaceae species, Magnolia kobus Dc., Magnolia denudata Desr, Magnolia obovata Thunb, Liriodendron tulipifera L Mordants used were chemicals, AIK(SO_4)_2 24H_2O, Ca(OH)_2, FeSO_4 7H_2O. All four species showed similar coloring among four different parts. Leaves and petals showed dark yellow, stem bark, grayish yellow, root bark, dark red or yellow. Al mordant made more brighter colour than Fe mordant. Stem bark showed less difference in color by mordants, whereas petals did the biggest.
