상피종양의 keratin 생성능에 대한 전자현미경적 연구

Keratins are characteristically present in all keratinizing and nonkeratinizing epithelial cells and their neoplasms. They consist of at least 19 different polypeptides ranging from 40 to 69 kilodaltons, which are developmentally related to specific epithelial cell types. Keratins of high and intermediate molecular weights are readily demonstrated in squamous cell carcinomas, which are characteristically rich in tonofilaments and tonofilament-desmosome complexes. Keratins of low molecular weights are expressed in adenocarcinoma including renal cell carinoma, hepatocellular carcinoma, small cell carcinoma, and carcinoid tumor. These tumors do not reveal any tonofilaments and sacrcely show discrete filaments on electron microscopy. An immunoperoxidase technique employing antibody to keratin was used to study distribution and staining pattern of keratin filaments in benign and malignant epithelial tumors (20 squamous cell carcinomas, 20 gastric adenocarcinomas, 20 transitional cell carcinomas and 15 non-neoplastic epithelia). These immunohistochemical results were compared with ultrastructural features of neoplastic diagnosis of squamous cell carcinoma, adenocarcinoma and transitional cell carcinoma, and estimation of degree of differentiation. The results are as follows : 1. The squamous epithelium, glandular epithelium and transitional epithelium disclosed the positive reaction for keratin. 2. Squamous cell carcinoma and transitional cell carcinoma exhibited positive reaction for keratin. 3. Gastric adenocarcinoma showed uniformly negative or weakly positive reaction for keratin. 4. On electron microscopy, heavy bundles of tonofilaments and desmosomes were more frequently encountered in well differentiated squamous cell carcinoma, In poorly differentiated squamous cell carcinoma, a few tonofibrils and desmosome were noted. 5. In the adenocarcinomas, the intermediate filaments were arranged randomly as nonaggregated, short filaments spread throughout the cytoplasm, although occasionally they formed a perinuclear whorl. These filaments did not become aggregated to form tonofilaments. 6. On the immunogold labeling for keratin of low molecular weight areas in gastric adenocinoma, in tense labeling of intermediate filaments for keratin was noted. From the above result, ultrastructural and immunohistochemical study on malignant epithelial cell were useful in differential diagnosis of squamous cell carcinoma, adenocarcinoma and transitional cell carcinoma, and estimation of degree of differentiation. It was also proved that immunogold labeling technique was especially valuable for diagnosis of vague case which showed negative or weakly positive reaction in immunohistochemical stain.

This study was designed to exam the aged's needs for spiritual nursing care. The purpose was to serve as a basis for the development of spiritual nursing practice. The major findings are as follows : 1. The degree of needs for spiritual nursing care as area was that needs of love and relationship mean 22.0, needs of meaning and object mean 28.2, needs of forgiving mean 13.5. Total needs for spiritual nursing care mean 63.7, which was on the upper middle level. The needs of meaning and object was rated highest. 2. Among the general characteristics of the subjects, needs of love and relatiohship wasn't significanlty different. 3. Among the general characteristics of the subjects, needs of meaning and object was significantly different according to two factors : age(F=7.260, p=0.001), religion(F=5.275, p=0.001). Higher needs of meaning and object was possessed by the older than the younger, by the one who have religion than the other. 4. Among the general characteristics of the subjects, four factors made a significantly difference to needs of forgiving : sex(t=-2.851, p=0.006), age(F=8.201, p=0.001), religion(F=6.928, p=0.000), disease(t=2.327, p=0.024). Higher needs of forgiving was possessed by man than woman, by the older than the younger, by the one who have religion than the other, by the one who have disease than the other.

<P><I>Ophiocordyceps sinensis</I> is a natural fungus that has been valued as a health food and used in traditional Chinese medicine for centuries. The fungus is parasitic and colonizes insect larva. Naturally occurring <I>O. sinensis</I> thrives at high altitude in cold and grassy alpine meadows on the Himalayan mountain ranges. Wild <I>Ophiocordyceps</I> is becoming increasingly rare in its natural habitat, and its price limits its use in clinical practice. Therefore, the development of a standardized alternative is a great focus of research to allow the use of <I>Ophiocordyceps</I> as a medicine. To develop an alternative for wild <I>Ophiocordyceps</I>, a refined standardized extract, CBG-CS-2, was produced by artificial fermentation and extraction of the mycelial strain <I>Paecilomyces hepiali</I> CBG-CS-1, which originated from wild <I>O. sinensis</I>. In this study, we analyzed the <I>in vitro</I> immune-modulating effect of CBG-CS-2 on natural killer cells and B and T lymphocytes. CBG-CS-2 stimulated splenocyte proliferation and enhanced Th1-type cytokine expression in the mouse splenocytes. Importantly, <I>in vitro</I> CBG-CS-2 treatment enhanced the killing activity of the NK-92MI natural killer cell line. These results indicate that the mycelial culture extract prepared from <I>Ophiocordyceps</I> exhibits immune-modulating activity, as was observed <I>in vivo</I> and this suggests its possible use in the treatment of diseases caused by abnormal immune function.</P>

<P><B>Abstract</B></P><P>The H<SUB>2</SUB>O<SUB>2</SUB>-catabolizing peroxidase activity of human peroxiredoxin I (hPrxI) was previously shown to be regulated by phosphorylation of Thr<SUP>90</SUP>. Here, we show that hPrxI forms multiple oligomers with distinct secondary structures. HPrxI is a dual function protein, since it can behave either as a peroxidase or as a molecular chaperone. The effects of phosphorylation of hPrxI on its protein structure and dual functions were determined using site-directed mutagenesis, in which the phosphorylation site was substituted with aspartate to mimic the phosphorylated status of the protein (T90D-hPrxI). Phosphorylation of the protein induces significant changes in its protein structure from low molecular weight (MW) protein species to high MW protein complexes as well as its dual functions. In contrast to the wild type (WT)- and T90A-hPrxI, the T90D-hPrxI exhibited a markedly reduced peroxidase activity, but showed about sixfold higher chaperone activity than WT-hPrxI.</P>

<P><I>Context</I>: <I>Ginkgo biloba</I> L. (Ginkgoaceae) leaves have been used as an herbal medicine that has a complex range of biological activities. However, when we consider that biological activity of plant extracts is highly variable according to the source, location, and harvest season, technology to obtain the natural products with homogeneity is extremely important.</P><P><I>Objective</I>: We established the technology to obtain the cambial meristematic cells (CMCs) of <I>Ginkgo biloba</I>, which were expanded <I>in vitro</I> with homogeneity through a suspension culture and then determined the anti-inflammatory activity of fractionated samples prepared from the ethanol extract of CMCs.</P><P><I>Materials and methods</I>: We determined the anti-inflammatory activity of samples using lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophage cells. Especially, influence of sample treatment on the expression of various indicators, such as nitric oxide (NO), inducible nitric oxide synthase (iNOS), cyclooxygenase (COX)-2, mitogen-activated protein (MAP) kinases, transcription factor, and cytokines, involved in inflammatory activity was assessed.</P><P><I>Results</I>: A fractionated sample demonstrated 53.4% inhibition of LPS-induced NO production from the cells. Additionally, when fractionated samples were treated, iNOS and COX-2 expressions were almost completely suppressed. Fractionated samples also inhibited the phosphorylation of LPS-induced extracellular signal-regulated (ERK) and p38 MAP kinases more than 60%. IκB phosphorylation and subsequent nuclear factor (NF)-κB activation were also suppressed by fractionated samples. The expression of pro-inflammatory cytokines, IL-6 and tumor necrosis factor (TNF)-α, was significantly inhibited by the sample treatment.</P><P><I>Discussion and conclusion</I>: Fractionated samples from the ethanol extract of <I>Ginkgo biloba</I> CMCs could potentially be the source of a powerful anti-inflammatory substance.</P>
