은행엽 물추출물이 흰쥐의 흉부대동맥 혈관이완에 미치는 영향

We have examined the relaxational response of the Ginkgo Folium water extract on isolated thoracic aorta from sprague dawley rat (300~350g) in the presence and absence of endothelium and the vasodilator nitric oxide. Segments of thoracic aorta obtained from rats immediately after delivery were mounted in organ baths superfused on a polygraph. We found that the thoracic aorta segments responded to the Ginkgo Folium water extract with a dose-dependent vasorelaxation. At 2.0 ㎎/㎖ Ginkgo Folium water extract, the relaxational response with and without endothelium were 75.8% and 6.9% of the maximum KCI-response. The Phenylepinephrine and 5-hydroxytrptamine - induced contraction at 10^-4M were inhibited by 77.3% and 75.8% after addition(2.0 ㎎/㎖) of the Ginkgo Folium water extract. After pre-treatment of the thoracic aorta with N^G-monomethyl-L-arginine(10^-4M), the vessels contracted in response to the nitric oxide synthase inhibitor. The present results show that nitric oxide may act as a crucial signal in the vasodilative action. In conclusion, Ginkgo Folium water extract-induced relaxation in the isolated rat thoracic aorta was composed of endothelium-dependent and dose-dependent relaxation. Endothelium-dependent relaxation may be mediated by release of endothelium derived relaxing factor, nitric oxide.

Background: Dipeptidyl peptidase-4 (DPP-4) inhibitors are widely used in the treatment of patients with type 2 diabetes and have proven protective effects on diabetic kidney disease (DKD). Whether DPP-4 inhibitors have renoprotective effects on insulin- deficient type 1 diabetes has not been comprehensively examined. The aim of this study was to determine whether gemigliptin, a new DPP-4 inhibitor, has renoprotective effects in streptozotocin (STZ)-induced type 1 diabetic mice. Methods: Diabetes was induced by intraperitoneal administration of a single dose of STZ. Mice with diabetes were treated without or with gemigliptin (300 mg/kg) for 8 weeks. Morphological changes of the glomerular basement membrane (GBM) were observed by electron microscopy and periodic-acid Schiff staining. In addition, we measured blood glucose and urinary albumin excretion and evaluated fibrotic markers using immunohistochemical staining, quantitative reverse transcription polymerase chain reaction analysis, and Western blot analysis. Results: Gemigliptin did not reduce the blood glucose levels of STZ-treated mice. In gemigliptin-treated mice with STZ, a significant reduction in urinary albumin excretion and GBM thickness was observed. Immunohistological examination revealed that gemigliptin attenuated renal fibrosis induced by STZ and decreased extracellular matrix protein levels, including those of type Ι collagen and fibronectin, and Smad3 phosphorylation. In cultured rat renal cells, gemigliptin inhibited transforming growth factor β-stimulated type Ι collagen and fibronectin mRNA and protein levels via down-regulation of Smad3 phosphorylation. Conclusion: Our data demonstrate that gemigliptin has renoprotective effects on DKD, regardless of its glucose-lowering effect, suggesting that it could be used to prevent DKD, including in patients with type 1 diabetes.
Lin28a attenuates TGF-β-induced renal fibrosis

Lin28a has diverse functions including regulation of cancer, reprogramming and regeneration, but whether it promotes injury or is a protective reaction to renal injury is unknown. We studied how Lin28a acts in unilateral ureteral obstruction (UUO)-induced renal fibrosis following unilateral ureteral obstruction, in a mouse model. We further defined the role of Lin28a in transforming growth factor (TGF)-signaling pathways in renal fibrosis through in vitro study using human tubular epithelium-like HK-2 cells. In the mouse unilateral ureteral obstruction model, obstruction markedly decreased the expression of Lin28a, increased the expression of renal fibrotic markers such as type I collagen, α-SMA, vimentin and fibronectin. In TGF-β-stimulated HK-2 cells, the expression of Lin28a was reduced and the expression of renal fibrotic markers such as type I collagen, α-SMA, vimentin and fibronectin was increased. Adenovirus-mediated overexpression of Lin28a inhibited the expression of TGF-β-stimulated type I collagen, α-SMA, vimentin and fibronectin. Lin28a inhibited TGF-β-stimulated SMAD3 activity, via inhibition of SMAD3 phosphorylation, but not the MAPK pathway ERK, JNK or p38. Lin28a attenuates renal fibrosis in obstructive nephropathy, making its mechanism a possible therapeutic target for chronic kidney disease.

본 논문에서는, 거리공간 상에서 양립사상, 타입 (A)의 양립사상과 타입 (P)의 양립사상의 개념을 소개하고, 타입 (P)의 양립사상들의 성질들을 이용하여 거리공간에 정의된 사상들의 공통 부동점 정리를 증명하였다.
