Objectives: Gastrodia elata (GE) has been used to treat cognition impairment, including Alzheimer’s disease (AD) in Korea. The purpose of this study was to investigate the effects of GE water extracts (GEE) on the lipopolysaccharide (LPS)-induced AD model in mice. (Aβ). Methods: We classified six groups as follow; group 1: control (CON), group 2: LPS (0.5 mg/kg/day, four times), group 3: 4 mg/kg donepezil (DP), group 4: 100 mg/kg GEE+LPS, group 5: 200 mg/kg GEE+LPS, group 6: 500 mg/kg GEE+LPS. Results: We found that GEE has an effect that inhibits decrease of discrimination index in object recognition test, as well as spontaneous alteration in the Y-maze test by LPS. Treatment with LPS increased amlyloid-β (Aβ) concentration, and decreased brain-derived neurotrophic factor (BDNF) in cerebral cortex of mice. However, GEE significantly protected against LPS-induced Aβ and BDNF changes. Our findings also showed that the inflammatory cytokines 〔tumor necrosis factor-alpha (TNF-α), interleukin- 6 (IL-6) and interleukin-1β (IL-1β)〕 mRNA and protein were up-regulated by the LPS injection. But GEE significantly suppressed LPS-induced inflammatory cytokines increase in a dose-dependent manner. Conclusions: This study suggests that the GEE may be an effective AD therapeutic agent, in treating neurodegenerative diseases including AD.

Herba houttuyniae has been used as a constituent of herval medicine prescriptions for the treatment present study, we investigated the celular efects of herba houttuyniae extract (HE) and the signal pathways of HHE-induced apoptosis in HL-60 hu-man promyelocytic leukemia cell line. HHE treat-ment caused apoptosis of cels as evidenced by discontinuous fragmentation of DNA, the los of mitochondrial membrane potential, release of mi-tochondrial cytochrome c into the cytosol, activa-tion of procaspase-9 and caspase-3, and proteo-lytic cleavage of poly(ADP-ribose) polymerase. inhibitor, or cyclosporin A, a mitochondrial per-meability transition inhibitor, completely abolished HHE-induced DNA fragmentation. Together, these results suggest that HE possibly causes mito-chondrial damage leading to cytochrome c release into cytosol and activation of caspases resulting in PARP cleavage and execution of apoptotic cel death in HL-60 cells.

Many naturally occurring plant extracts are studied for their beneficial effects for health and particularly on cancer. Apoptosis, or programmed cell death, occurs in both normal and pathological conditions, including cancer Dysregulation of apoptosis allows transformed cells to continually and uninhibitedly enter the cell cycle, thus perpetuating the sequence of mutation, genomic instability and, finally, oncogenesis. To investigate the apoptosis-inducing effect of the extract of Fructus Trichosanthis (EFT) on leukemic HL-60 cells and its mechanism, HL-60 cells in vitro in culture medium were given different doses of the extract. The inhibitory rate of cells were measured by microculture tetrazolium assay, cell apoptotic rate was detected by flow cytometry, morphology of cell apoptosis was observed by DAPI fluorescence staining, and the activations of caspases and PARP were detected using Western blotting analysis. The extract could activate the caspase-3 and caspase-8, induce PARP cleavage, inhibit growth of HL-60 cells, and cause apoptosis significantly The suppression was in dose-dependent manner. Marked morphological changes of cell apoptosis including condensation of chromatin and nuclear fragmentation were observed clearly by DAPI fluorescence staining especially. These results will provide strong laboratory evidence of EFT for clinical treatment of acute leukemia.

The aim of this study was to investigate the cytotoxic effect and its mechanism on Radix Aconiti(RA) extract in lung cancer cell lines. RA extract treatment decreased the cell viability in a dose-dependent fashions in lung cancer cells including A549, H460, H23 and H157 cells. Many investigators reported that A549 and H460 cells expressed wild-type p53, but H23 and H157 cells preserved mutated p53. After treatment with RA extract in A549 and H460 cells, we measured the expression of p53 protein levels using Western blot, analysis. In both cells treated with RA extracts, p53 protein expressions were increased in a dose-dependent manner. In our experiments, RA extracts also have cytotoxic effects in H23 and H157, which have mutated p53. Treatment with RA extract decreased bcl-2 protein expressions in both cells. These results suggest that RA extracts have cytotoxic effects via p53 expression increase and bcl-2 inhibitable pathways in A549, H460 cells and H23, H157 cells, respectively.

To investigate the protective effect of Gamdu-tang(GDT) and its constituents, Radix Glycyrrhizae(RG) and Semen Glycine(SG) on the damage of cardiac endothelial cells by xanthine oxidase (XO)/hypoxanthine (HX)-induced oxygen free radical, Neutral Red (NR) and DNA synthesis assay were used. The results were obtained as follows ; Cardiac endothelial cells treated with XO/HX showed the cytotoxicity such as decreases in viability and DNA synthesis. Cardiac endothelial cells pretreated with GDT extracts were not showed the decrease of DNA synthesis by XO/HX. These results show that XO/HX elicits toxic effects in cultured cardiac endothelial cells derived from neonatal rat, and suggest that GDT extract is very effective in the prevention of XO/HX-induced toxicity.

We designed to investigate the relationship the cold-hot theory of herbology and body temperature in experimental model. we prepared four kinds of oriental medicine, which consisted of two cold herbs, Daehwang and Seokgo, and two hot herbs, Buja and Padu. Decrease of body temperature by cold exposure for 12 hour was not inhibited by four herbs oral administration for two weeks. Thermogenesis in mammals is an essential physiological function to maintain the body temperature. Mitochondrial uncoupling proteins(UCPs), which have a potential to generate heat by uncoupling oxidative phosphorylation, apper to play a crucial role in thermogenesis. Therefore UCP is commonly recognized as a key molecule in metabolic thermogenesis and its dysfunction contributes to the development of obesity. In these experiments, Daehwang water extracts inhibited the UCP1 mRNA expression increase by cold exposure in brown adipose tissue. But other herbs did not significantly influence on UCPs mRNA expression in white adipose tissue and seleus muscle tissue. Based on this experiment, we will try to clarify the effects of Daehwang water extracts on UCP1 expression and function.

산화적 스트레스에 대한 천마 추출물의 신경세포 보호 및 항산화 효과

We recently reported that Gastrodia elata extracts (GEE) had an effects to protect against lipopolysaccharide-induced cognitive impairment in vivo model. In this study, we investigated the neuroprotective effects and the mechanism of action of GEE in hydrogen peroxide (H2O2)-induced cell death of SH-SY5Y human neuroblastoma cell. The SH-SY5Y cells were divided into five groups, including control(non-treated group), 100 μM H2O2, 100, 200, 500 ㎍/㎖ GEE+ 100 μM H2O2 groups. Pre- and co-treatment with GEE prevented cell death induced by 100 μM H2O2 for 24 h in SH-SY5Y cells. Our findings also showed that anti-oxidants enzymes (Cu/Zn superoxide dismutase, Mn superoxide dismutase, catalase) were up-regulated by 100 μM H2O2. But GEE suppressed H2O2-induced anti-oxidants enzymes decrease in a dose-dependent manner. Treatment with GEE also inhibited phosphorylation of eukaryotic initiation factor-2α (eIF-2α) and p38 by H2O2. Taken together, the neuroprotective effects of GEE in terms of recovery of antioxidant enzymes expression, down-regulation of eIF-2α and p38 phosphorylation, and inhibition of cell death are associated with reduced oxidative stress in SH-SY5Y cells.
苦蔘 추출물이 XO/HX에 의해 손상된 血管內皮細胞에 미치는 영향(Ⅰ)

To investigate the protective effect of Sophorae Radix (SR) on the damage by pulmonary vascular endothelial cells by xanthine oxidase (XO)/hypoxanthine (HX)-induced oxygen free radical, Neutral Red (NR) and c-fos immunopositive cell assay were used. The results were obtained as follows ; The viability of vascular endothelial cells treated with XO/HX was decreased. And c-fos immunopositive cells represented a maximal increase in group treated with XO/HX for 2 hour in pulmonary vasvular endothelial cells. But pretreated groups with SR extracts were not inhibited the increase of c-fos, immunopositive cells by XO/HX in a dose-dependent manner. These results show that XO/HX elicits toxic effects in cultured pulmonary vascular endothelial cells, and suggest that SR extract is very effective in the prevention of XO/HX-induced increase of c-fos immunopositive cells.
