        혈당 측정용 스트립 개발에 관한 연구

        과산화 효소와 포도당 산화효소의 효소반응을 이용하여 혈액 중 당을 측정하는 혈당측정용 스트립을 개발하였다. 멤브레인에 포도당 산화효소와 과산화 효소, 색원체를 건조 처리하면 혈당이 멤브레인에 처리된 포도당 산화효소와 즉각 반응하여 과산화 수소를 발생하고 발생된 과산화 수소가 과산화 효소와 반응하여 착색물을 형성한다. 제조된 스트립은 혈액과 접촉하면 반응시간 2∼3분 이내에 0∼800mg/dl의 혈당에 대하여 농도에 따라 연 녹, 청녹, 짙은 청색을 나타내며 이때 민감도는 40 mg/dl이었다. 육안용 혈당 스트립을 이용하여 정상을 포함한 당뇨 환자의 혈당 농도를 측정한 결과 Ames사와 BM사에서 시판하고 있는 제품과 유사한 결과를 얻을 수 있었다. 본 연구에서 개발된 스트립이 자동 분석기기의 재료로 활용될 수 있는지 여부를 알아보기 위하여 분광 비색계로 발색 반응의 최적 파장을 분석하고 최적 파장에서 각 혈당 농도별 반사 밀도를 측정하여 검정선을 얻었다. 이 검정선에 의해 임상 혈청시료의 혈당 농도를 측정한 결과 일본의 Kyoto Daiichi사의 혈액 분석시스템과 유사한 결과를 얻었다. 이로서 본 연구에서 개발한 혈당 스트립을 이용하여 혈액 중 혈당량을 육안으로 측정할 수 있음은 물론 자동 분석기기의 기본 시료로 이용될 수 있음을 확인할 수 있었다. We have developed a simple and accurate strip test that measures the blood glucose level semiquantitatively by visual observation, or qualitatively by using UltraScan spectrocolorimeter. The strip has solid phase reagents, including glucose oxidase, peroxidase, chromogen, affixed to a plastic support. The strip test is capable of measuring blood glucose level in the range of 0∼800 mg/dl and generating the results within 2 to 3 minutes. Human blood specimens obtained from normal individuals and the diabetic patients were evaluated by the new blood glucose strip and by the kit supplied by other commercial products. The test results exhibit the correlation coefficient of 0.964. The new test strip is proven simple and accurate, and it offers an alternative to the commercially available glucose tests.

      • 석면함유 슬레이트 지붕 물받이 퇴적물 중 석면 섬유 함유율

        This study attempted to determine the degree of asbestos release from the aging slate roof by comparing the asbestos content in the slate roof rain gutter with a colored steel plate (tin plate) over the slate roof. Four slate roof houses located in Haengmok-ri, Asan-si, Chungcheongnam-do, and one house constructed with a colored steel plate on the slate roof were selected to collect the sediment of the roof rain gutter. The asbestos fiber content was calculated by a point counting method using a polarization microscope after pretreatment with conversion treatment and hydrochloric acid treatment. The average asbestos content of the four slate roof rain gutter were 1.89%. However, asbestos was not detected in the Slate covering roof rain gutter, which were constructed on the slate roof. Asbestos fiber content was the highest at 2.89% in the slate roof rain gutter installed in 1976, followed by 2.44% in 1953. From the above results, it is necessary to minimize secondary damage as asbestos fibers released from slate roof houses to the surrounding atmosphere or leaked from slate roofs as rainwater may cause soil pollution and seriously affect residents' health. Although covering with colored steel plates (tin plates) has been shown to prevent the leakage of asbestos fibers to some extent, it is believed that a policy alternative to remove the slate roof as soon as possible is needed to solve the fundamental problem.

      • 내시경적 조임근절개술후에 발생한 췌십이지장 가성동맥류 1예

        We reported a case of pancreatic pseudoaneurysm, involving pancreaticoduodenal artery, developed after endoscopic retrograde cholangiopancreatography(ERCP) with endoscopic sphinterotomy. A 54 year old woman was transferred to our hospital due to sustained hematochezia and hypovolemia. A week before admission, she underwent ERCP with endoscopic sphinterotomy for acute cholangitis in local clinics. She developed abdominal pain and hematochezia at 12 hours after the procedures. A post procedure contrast enhanced computed tomography of abdomen demonstrated a pseudoaneurysm in inferior portion of pancreatic head. This was not present on the pre procedure contrast enhanced computed tomography of abdomen and was thought to arise from the pancreaticoduodenal artery as a complication of the pre sphincterotomy. She was brought to our emergency room in a state of hypovolemia. A upper endoscopy showed active bleeding from ampula of vater. An celiac angiography demonstrated pancreatic pseudoaneurysm involving inferior pancreaticoduodenal artery and selective coil embolization of was performed.

      • FOXP1: valuable target for development of treatment of cancer stem cells in ovarian cancer

        목적: Ovarian cancer has the highest mortality rate of all gynecological cancers with a high recurrence rate. It is important to understand the nature of recurring cancer cells to terminally eliminate ovarian cancer. The winged helix transcription factor Forkhead box P1 (FOXP1) has been reported to function as either oncogene or tumor-suppressor in various cancers. In the current study, we show that FOXP1 promotes cancer stem cell- like characteristics in ovarian cancer cells. 방법: Knockdown of FOXP1 expression in A2780 or SKOV3 ovarian cancer cells decreased spheroid formation, expression of stemness-related genes and epithelial to mesenchymal transition-related genes, cell migration, and resistance to Paclitaxel or Cisplatin treatment, whereas overexpression of FOXP1 in A2780 or SKOV3 ovarian cancer cells increased spheroid formation, expression of stemness-related genes and epithelial to mesenchymal transition-related genes, cell migration, and resistance to Paclitaxel or Cisplatin treatment. 결과: Overexpression of FOXP1 increased promoter activity of ABCG2, OCT4, NANOG, and SOX2, among which the increases in ABCG2, OCT4, and SOX2 promoter activity were dependent on the presence of FOXP1-binding site. In xenotransplantation of A2780 ovarian cancer cells into nude mice, knockdown of FOXP1 expression significantly decreased tumor size. 결론: These results strongly suggest FOXP1 functions as an oncogene by promoting cancer stem cell-like characteristics in ovarian cancer cells. Targeting FOXP1 may provide a novel therapeutic opportunity for developing a relapse-free treatment for ovarian cancer patients.

      • A case of disseminated intravascular coagulation developed after surgical management of corpus luteal hemorrhage in a patient with Klippel-Trenaunay syndrome

        Klippel-Trenaunay syndrome (KTS) is a rare congenital vascular malformation characterized by the clinical triad of bony or soft tissue hypertrophy, usually affecting one extremity; hemangiomas and/or lymphangiomas, and varicosities or venous malformations. Vascular malformation of KTS can involve several organs and be a source of significant morbidity and even mortality. Clinical manifestations of KTS range from occult to massive, life-threatening hemorrhage. Several cases of lifethreatening gastrointestinal bleeding in KTS patients have been reported. In contrast, large venous malformations can be associated with low-grade consumptive coagulopathy. Spontaneous hemoperitoneum may occur in various gynecological conditions. The most common gynecological causes of spontaneous hemoperitoneum in women of childbearing age are ectopic pregnancy and ruptured corpus luteal cyst . The treatment of spontaneous hemoperitoneum of gynecological causes is well established. However in patients with coagulopathy, the management has to be planned differently. The authors present a case of a 29-year-old female with KTS who presented with a spontaneous hemoperitoneum by corpus luteal hemorrhage and subsequent life-threatening disseminated intravascular coagulation (DIC) after ovarian cystectomy.

      • HMGA1: a therapeutic target for ovarian cancer via regulation of cancer stem cell characteristics

        Cancer stem cells are a subpopulation of cancers cells, characterized by self-renewal ability, tumorigenesis, and drug resistance. HMGA proteins are architectural factors constituting critical hubs in the chromatin network and abundantly expressed during embryogenesis and in many different cancers. The aim of this study was to investigate the role of HMGA1 in regulation of cancer stem cells in ovarian cancer. Spheroid-forming cancer stem cells were isolated from A2780 ovarian cancer cells by three-dimensional spheroid culture. Significantly higher HMGA1 expression was observed in spheroid cells along with higher expression of stemness-related genes, such as SOX2, KLF4, ALDH, ABCB1, and ABCG2, and higher resistance to chemotherapeutic agents such as paclitaxel and doxorubicin, compared with adherent A2780 ovarian cancer cells. HMGA1 knockdown in spheroid cells reduced proliferative advantage, spheroid-forming efficiency, and expression of stemness- related genes. HMGA1 overexpression in adherent A2780 cells increased cancer stem cell properties, including proliferation, spheroid forming efficiency, and expression of stemness-related genes. In addition, HMGA1 regulated ABCG2 promoter activity through HMGA1 binding sites. Knockdown of HMGA1 in spheroid cells reduced resistance to chemotherapeutic agents whereas overexpression of HMGA1 in adherent ovarian cancer cells increased resistance to chemotherapeutic agents in vitro. Furthermore, HMGA1-overexpressing A2780 cells showed a significant survival advantage after chemotherapeutic agent treatment in xenograft tumorigenicity assay. Taken together, our results provide novel insight regarding a critical role of HMGA1 in regulation of cancer stem cell characteristics of ovarian cancer cells, thus suggesting that HMGA1 can be an important target in development of therapeutics to cure ovarian cancer patients.

        Mature cystic teratoma (MCT) is a common benign neoplasm of the ovary. Complications occur in approximately 20% of cases. Clinical manifestations, laboratory findings, and imaging studies can assist in making a diagnosis of ovarian torsion of MCT. Furthermore, serum tumor markers may be helpful for diagnosing MCT and its torsion, and thus, can lead to early surgical intervention. A 56-year-old woman presented with a huge pelvic mass and pelvic pain. Serum CA19-9, CA125, and CEA levels were abnormally elevated at >700 U/ml, 282.5 U/ml, and 3.94 U/ml, respectively. The tumor was surrounded by extensive adhesions and showed inflammatory changes. The serum levels of these markers returned to normal levels after surgery.

      • Differential expression of reproductive hormone receptors and contraction associated genes in porcine uterus during the estrous cycle

        Objective: Contraction of uterus tissue frequently occurs throughout the estrous cycle and is regulated by several endogenous factors, including estradiol, progesterone, luteinizing hormone, follicle stimulating hormone, oxytocin (OXT) and contraction associated proteins (CAPs). Contraction activity of uterus tissue according to the estrous cycle is important, due to the fact that it is directly associated with balanced implantation and stable pregnancy. Methods: Uterine tissues were fixed with 10% formalin, embedded in paraffin wax, routinely processed, then sectioned into 4-m-thick slices. Tissue sections were deparaffinized and rehydrated through graded alcohol using standard procedures and then stained with hematoxylin & eosin (H&E; Sigma-Aldrich; Merck Millipore). Images were captured at a magnification of x40 using a model BX50F-3 optical microscope (Olympus Corporation, Tokyo, Japan) and examined for histological analysis. Results: To investigate regulation of contraction associated factors according to the estrous cycle, mRNA and protein expression levels of reproductive hormonal receptors, including estrogen receptors, progesterone receptor and luteinizing hormone/choriogonadotropin receptor in addition to CAPs including OXT, OXT receptor (OXTR), hydroxyprostaglandin dehydrogenase 15 (NAD) and gap junction 1 protein, were examined in the porcine uterus according to the follicular and luteal phases. For the results, hormonal receptors and CAPs were dynamically regulated depending on the estrous cycle. Conclusion: In conclusion, genes associated with uterine contraction and its regulatory hormonal receptors in the porcine uterus were differently regulated in the follicular and luteal phases, suggesting that these genes are critically involved in the remodeling and contraction of uterine tissue and may be required to modulate the physiological status of the uterus.

        <P>Intrinsically unfolded proteins (IUPs) do not obey the golden rule of structural biology, 3D structure = function, as they manifest their inherent functions without resorting to three-dimensional structures. Absence of a compact globular topology in these proteins strongly implies that their ligand recognition processes should involve factors other than spatially well-defined binding pockets. Heteronuclear multidimensional (HetMulD) NMR spectroscopy assisted with a stable isotope labeling technology is a powerful tool for quantitatively investigating detailed structural features in IUPs. In particular, it allows us to delineate the presence and locations of pre-structured motifs (PreSMos) on a per-residue basis. PreSMos are the transient local structural elements that presage target-bound conformations and act as specificity determinants for IUP recognition by target proteins. Here, we present a brief chronicle of HetMulD NMR studies on IUPs carried out over the past two decades along with a discussion on the functional significance of PreSMos in IUPs.</P>

        한강 유역 간흡충 고위험군의 감염과 관련요인

        Purpose: This study was done to investigate the prevalence and related factors of Clonorchis Sinensis for inhabitants of the Han riverside area and to identify knowledge related to Clonorchis Sinensis and intended behavioral changes to decrease risk of infection. Method: The data were collected from December 16, 2008 to January 10, 2009. Participants were 555 people who responded to a questionnaire. Frequency, percentage, $X^2$-test, and t-test were used to analyze the data. Results: The prevalence of Clonorchis Sinensis was 7.9% in this population. Related factors for Clonorchis Sinensis were gender (male=10.8%, female=3.6%, p=0.002) and smoking (p=0.007). but habits related to ingestion of alcohol and raw fish were not significant. As for knowledge of Clonorchis Sinensis, the Clonorchiasis group had a mean score of 10.09 $({\pm}3.95)$ of a possible 17 compared to a score of only 8.27$({\pm}4.60)$ for the negative group (p=0.011). The intended behavioral change related to risk of infection, according to presence of infection or not, was significant (p=0.004). Conclusion: These results suggest that Clonorchiasis is still highly endemic in all risk areas such as the Han riverside, indicating that it is necessary to set up effective management programs for patient care and prevention of Clonorchis Sinensis.
