        운동형태의 차이가 Alloxan유발 당뇨쥐의 혈당 및 인슐린 농도에 미치는 영향

        The Purpose of this study was to investigate the effect of exercise intensity on blood glucose and insulin levels in diabetic rats with alloxan. Twenty sprague-dawley male rats were assigned to power training groups(n=10) and endurance training groups(n=10). Diabetes was induced by single injection of alloxan(50 mg/kg B.W) Blood glucose and insulin were determined every week for 4-weeks. The conclusion of this study follows as below: 1) There was significantly different on blood glucose levels in two groups. 2) There was significantly different on insulin levels in two groups These results suggest that proper exercise intensity in diabetic rats can significantly decrease blood glucose and insulin level and make good glycemic control. In conclusion, it has been found that regular prolonged endurance training with diabetic rats, improves insulin and blood glucose.

        Supine Bicycle Ergometer 운동이 뇌파와 기분상태에 미치는 영향

        In this study we investigated the effects of supine ergometer exercise on physiological and psychological relaxation. Seven healthy young men(aged 24.4±2.00years) volunteered for the experiment. The experiment consisted of the following three successive segments" A pre-exercise period of 10 min, during which the subjects rested in a supine posture with their eyes closed for the final 5 min; an supine ergometer exercise period of approximately 10 min, during which the subjects performed intensity of 50%HRmax exercises; A post-exercise recovery of 5 min, and a recovery of 10 min, which was similar to the pre-exercise rest period. The electroencephalo-graphic(EEG) activity of the central cortex of the brain were measured pre-exercise, post-exercise 5 min, and recovery phase 10min. Fast Fourier Transformation of the EEG was used to obtain power spectrum areas in the delta(0.5~4 Hz), theta(4~8 Hz), alpha(8~13 Hz) and beta(13~30 Hz) frequencies. We compared the relative power values(power %) of the electroencephalogram alpha bands(8~13 Hz) and korean edition of the profile of mood states(K-POMS) before and after the supine ergometer exercise. We also estimated the percentage of maximal heart rate(%HRmax) throughout the experiment to ascertain the intensity of the supine bicycle ergometer exercise. The results of %HRmax indicated that the intensity of supine exercises practised in the experiments ranged from low to moderate. The power % of EEG alpha bands had increased significantly after the supine ergometer exercise compared with the pre-exercise rest(p<0.05). From the POMS results, we observed that positive mood(vigour) increased and negative mood(tension, depression, and total mood)decreased significantly after the supine ergometer exercise(p<0.05). This study found that the subjects showed increased physiological and psychological indices of relaxation after supine bicycle ergometer exercise.

      • Retinoic acid에 의해 유발된 흰쥐의 구개열에서 Transforming Growth Factor-beta 1, 2, 3의 발현의 변화

        Objective : Craniofacial malformations are among the most frequent congenital birth defects in humans : cleft palate, that is inadequate fusion of the palatal shelves. Retinoic acid is teratogenic in many species, producing multiple malformations, including cleft palate. Also it plays an important role in embryogenesis, by regulating morphogenesis, cell proliferation, differentiation, and deposition of the extracellular matrix. The effects of retinoic acid which leads to cleft palate vary depending on the stage of development exposed. After exposure of embryonic mice to retinoic acid on gestational day(GD) 10, abnormally small palatal shelves form. And the elevation of palatal shelves is delayed. The growth and differentiation factor, transforming growth factor-beta(TGFβ) family is thought to play important roles in multiple developmental processes, especially, the development of the embryonic palate. The expression of the TGFβ family in embryonic palate suggests a functional role for these molecules. In this study, author has determined the tissue distributions of TGFβ family in murine embryonic palate, and compared them in terms of developmental stage and locations in normal and retinoic acid-induced cleft palate. Methods and Material : Retinoic acid was administered orally at gestational day(GD) 10 to ICR mice. The pregnant mice were sacrifeced on GD 12, 13. 14, 15 to obtain the fetuses and immunohistochemistry was performed. And using the scanning electron microscope, morphology of normal palate and cleft palate was studied. Results : In the retinoic acid-treated fetuses, palatal shelves did not elevate and cleft palate was induced. On GD 13 in the palates of the retinoic acid-treated fetuses, expression of the TGFβ1, 3 decreased dramatically, but that of TGFβ2, did not change. The expression of the TGFβ1, 3 in the palates of the retinoic acid-treated fetuses, increased on GD 14, 15, Significant differences in TGFβ family expression were observed with areas of the palate. Conclusion : These results suggest that retinoic acid inhibits the expression of TGFβ 1, 3, which is critical in the developing palate, and elevation of palatal shelves is delayed and impaired, therefore cleft palate is induced. These data demonstrate a role for retinoic acid and retinoic aicd-induced TGFβ family in the regulation of palatal development and suggest a role for TGFβ in retinoic acid-induced cleft palate.

      • 선인장 양심실 보조장치의 설계 및 임상적용을 위한 평가

        현재 개발되고 있는 맥동형 이식형 인공심장은 완전이식형 인공심장과 좌심실보조장치 뿐이다. 좌심실보조장치를 장착한 환자의 10~15%가 우심실의 보조를 필요로하고 완전이식형 인공심장의 장착을 위해서는 자연심장을 제거해야하는 상황에서 이식형 양심실보조장치에 대한 필요성이 대두되고 있다. 본 연구진은 완전이식형 인공심장으로 개발된 한국형 인공심장을 개선하여 양심 실 보조장치를 개발하였다. 양심실보조장치는 이동형 작동기식 미케니즘을 이용하고 있으며 에너지 변환장치와 감속기로 구성된 작동기와 혈액주머니, 그리고 내장형 제어기로 구성되어 있다. 선인장 펌프로 명명된 KAH350은 캐뉼라의 연결을 위한 커넥터를 포함하여 길이 177mm, 폭 164mm, 높이 67 이고 무게 780g이며 최대 심박출량은 5L/min 이다. 양심실보조장치로 개발된 선인장펌프는 좌심실보조장치로도 응용이 가능하다. 이식적합성과 생체적합성을 평가하기 위하여 좌심실보조장치로 5회, 양싱실보조장치로 6회의 동물실험을 수행하였다. 각각 최장 28일간 생존하였으며 11회의 동물실험중 장치의 결함에 의한 것이 3회 있었는데 이들은 모두 전자장치의 결함과 방수처리문제였다. 현재 이식적합성과 일박출량을 향상시킨 KAH400모델이 개발중이다. The types of pulsatile implantable artificial hearts that are on the way of development and have been developed are totally implantable artificial heart and implantable left ventricular assist device (LVAD). Approximately 10% to 15% of all patients Implanted with wearable VADs have required right heart support with another device. And it is reluctant to patient who should remove his or her own heart to be implanted with total artificial heart. These situations drive the development of implantable bi-ventricular assist device (BVAD). The Korean BVAD was develophed by modifying the moving actuator type Korean artificial heart. This electro-mechanical BVAD comprises actuator including energy converter and reduction gear train, blood sacs, and internal motor and energy controllers. The KAH350 which is named as 'Cactus Pump' is 177mm in length, 164mm in width, and 67mm in thickness including connectors and nuts. The weight and maximum cardiac output of Cactus Pump is 780g and 5L/min, respectively. The Cactus Pump that was developed as BVAD could be used as LVAD by attaching compliance caps on the 2 ports of one ventricle. The animal experiments were undergone 5 times for LVAD, and 6 times for BVAD. The best records were 28 days survival in both applications. There were 3 times of device failure and they were all associated with electrical connection and hermetic sealing. The KAH400 that has improved anatomical fitting characterisitcs and stroke volume is on development.

        하악골에 발생한 골막 골육종

        Periosteal osteosarcoma is a distinct entity of malignant bone tumor with characteristic clinical, morphological, and histological features within the group of juxtacortical osteosarcoma. Periosteal osteosarcoma is predominantly located in the tubular long bones, and extremely rarely involving the jaws. A case of periosteal osteosarcoma of the right mandible is presented. A 27-year-old woman complained of the gingival swelling and bleeding tendency of the right posterior mandible. Clinical examination revealed a reddish brown strawberry-like swelling on the affected mucosa, which measured 1.5㎝×1.5㎝. The tumor was located on the lingual cortex of the mandible and extended into the surrounding soft tissues. Microscopically, the tumor consisted exclusively of atypical chondroblastic cells with a small osteoblastic area. A minimal bone marrow involvement was noted and the adjacent cortex was free of tumor. These findings suggested that the tumor originated from the periosteal cambium layer, which lies between the periosteal fibrous layer and the cortex of mandible.

        Recent advances in physiologically based pharmacokinetic and pharmacodynamic models for anticancer nanomedicines

        Nanoparticles (NPs) have distinct pharmacokinetic(PK) properties and can potentially improvethe absorption, distribution, metabolism, and elimination(ADME) of small-molecule drugs loaded therein. Owingto the unwanted toxicities of anticancer agents in healthyorgans and tissues, their precise delivery to the tumor is anessential requirement. There have been numerous advancementsin the development of nanomedicines for cancertherapy. Physiologically based PK (PBPK) models serveas excellent tools for describing and predicting the ADMEproperties and the effi cacy and toxicity of drugs, in combinationwith pharmacodynamic (PD) models. The recent preliminaryapplication of these modeling approaches to NPsdemonstrated their potential benefi ts in research and developmentprocesses relevant to the ADME and pharmacodynamicsof NPs and nanomedicines. Here, we comprehensivelyreview the pharmacokinetics of NPs, the developed PBPK models for anticancer NPs, and the developed PDmodel for anticancer agents.

      • Proteomic Analysis of Tumor Necrosis Factor-Alpha (TNF-α)-Induced L6 Myotube Secretome Reveals Novel TNF-α-Dependent Myokines in Diabetic Skeletal Muscle

        <P>There is a strong possibility that skeletal muscle can respond to irregular metabolic states by secreting specific cytokines. Obesity-related chronic inflammation, mediated by pro-inflammatory cytokines, is believed to be one of the causes of insulin resistance that results in type 2 diabetes. Here, we attempted to identify and characterize the members of the skeletal muscle secretome in response to tumor necrosis factor-alpha (TNF-α)-induced insulin resistance. To conduct this study, we comparatively analyzed the media levels of proteins released from L6 skeletal muscle cells. We found 28 TNF-α modulated secretory proteins by using separate filtering methods: Gene Ontology, SignalP, and SecretomeP, as well as the normalized Spectral Index for label-free quantification. Ten of these secretory proteins were increased and 18 secretory proteins were decreased by TNF-α treatment. Using microarray analysis of Zuker diabetic rat skeletal muscle combined with bioinformatics and Q-PCR, we found a correlation between TNF-α-mediated insulin resistance and type 2 diabetes. This novel approach combining analysis of the conditioned secretome and transcriptome has identified several previously unknown, TNF-α-dependent secretory proteins, which establish a foothold for research on the different causes of insulin resistance and their relationships with each other.</P><P>In the current study, we used a reliable proteomic approach to comparatively analyze skeletal muscle secretomes in the presence and absence of tumor necrosis factor-alpha (TNF-α) induced insulin resistance. Our results introduce a novel population of TNF-α dependent secretory proteins derived from skeletal muscle.
