忍冬 열수 추출물의 항산화 효과 및 HCl-Ethanol로 유도된위염 동물 모델에서의 위 점막 손상 보호 효과

Objective : Gastritis is a major complication of gastrointestinal disease. Lonicera japonica is used in folk medicine to treat different diseases such as exopathogenic wind-heat, epidemic febrile diseases, sores, carbuncles and some infectious diseases. Therefore, this study examined the effects of Lonicera japonica water extract (LJE) on HCl/ethanol- induced acute gastric ulceration and anti-oxidants properties. Methods : LC-ESI-IT-TOF MS was employed for rapid identification of major compound from LJE. The antioxidant activities were evaluated through total polyphenol and flavonoid contents and radical scavenging assays and superoxide dismutase (SOD)-like activity. SD rats were randomly divided into five different groups including the normal group, ulcer group, positive group (20 ㎏/㎎ of omeprazole, ip), and experimental groups (100 ㎏/㎎ and 500 ㎏/㎎ of LJE, ip). Results : 4,5-Dicaffeoyl quinic acid, loganic acid, secologanic acid, sweroside, loganin, vogeloside were identified based on the detection of the molecular ion with those of literature data. The LJE was possessed free radical scavenging activities such as DPPH (IC50=189.7 ㎍/㎖), ABTS (IC50=164.5 ㎍/㎖), and SOD-like activity (IC50=405.02 ㎍/㎖). Macroscopic and histological analyses showed LJE treated group were significantly reduced to an extent that it allowed leukocytes penetration of the gastric walls compared with the ulcer group. In addition, an ulcer inhibition rate and prostaglandin E2 levels were increased in rats treated with LJE. Conclusion : The present study has demonstrated the antioxidantive and gastroprotective effect of LJE, these findings suggested that LJE has the potential for use in treatment of gastric disorders.

Objective : To establish a reliable tool between for the distinction of original plants of Sanguisorbae Radix, we analyzed the complete chloroplast genome sequence of Sanguisorbae Radix and identified single nucleotide polymorphisms (SNPs). Materials and methods : The chloroplast genome sequence of Sanguisorba officinalis, Sanguisorba tenuifolia f. alba (Trautv. & Mey.) Kitam and Sanguisorba tenuifolia Fisch. ex Link obtained using next-generation sequencing technology were described and compared with those of other species to develop specific markers. Candidate genetic markers were identified to distinguish species from the chloroplast sequences of each species using Modified Phred Phrap Consed and CLC Genomics Workbench programs. Results : The structure of the chloroplast genome of each sample that had been assembled and verified was circular, and the length was about 155 kbp. Through comparative analysis of the chloroplast sequences, we found 220 nucleotides, 158 SNPs, and 62 Indel (insertion and/or deletion), to distinguish Sanguisorba officinalis, Sanguisorba tenuifolia f. alba (Trautv. & Mey.) Kitam and Sanguisorba tenuifolia Fisch. ex Link. Finally, 15 specific SNP genetic markers were selected for the verification at positions. Avaliable primers for the dried herb, which is used as medicine, were used to develop the PCR amplification product of Sanguisorbae Radix to assess the applicability of PCR analysis. Conclusion : In this study, we found that Fendel-qPCR analysis based on the chloroplast DNA sequences can be an efficient tool for discrimination of Sanguisorba officinalis, Sanguisorba tenuifolia f. alba (Trautv. & Mey.) Kitam and Sanguisorba tenuifolia Fisch. ex Link.

The objective of this study is to evaluate the emotional change of a driver according to the change of temperature, humidity, $CO_2$ gas concentration and $C_4H_{10}$ gas concentration in the automobile by electroencephalogram(EEG) spectrum analysis. The experiment was performed in an automobile simulated shielding room with healthy volunteers(age : 18-31, male : 14, female : 7). The subject's emotion at every different condition was indexed 11-point scale(+5 : most pleasant, -5 : least pleasant). The emotion index at each condition resulted in as follows; in temperature $1.00(23.5^{\circ}C)$, $-2.33(30.5^{\circ}C)$, in humidity 1.50(45%), -1.50(65%), in $CO_2$ gas concentration 0.67(500ppm), -0.57(6,000ppm), in $C_4H_{10}$ gas concentration -0.25(0ppm), -2.75(1,200ppm). From EEG spectrum analysis, the average mean power frequency(MPF) value at each different condition was shown as follows; in temperature $5.48(23.5^{\circ}C)$, $4.06(30.5^{\circ}C)$, in humidity 11.03(45%), 3.03(65%), in $CO_2$ gas concentration 7.16(500ppm), 6.38(6,000ppm), in $C_4H_{10}$ gas concentration 10.21(0ppm), 2.87(1,200ppm). When the environment became unpleasant to the subject, all of the obtained MPF values were decreased. It was found that there is close agreement between subjective evaluation by subject's scaling and objective evaluation by EEG spectrum analysis at different environment. These results indicated that the EEG spectrum analysis is a proper method to assess emotional response of a driver in the changing automobile environment.

We designed to investigate the relationship the cold-hot theory of herbology and body temperature in experimental model. we prepared four kinds of oriental medicine, which consisted of two cold herbs, Daehwang and Seokgo, and two hot herbs, Buja and Padu. Decrease of body temperature by cold exposure for 12 hour was not inhibited by four herbs oral administration for two weeks. Thermogenesis in mammals is an essential physiological function to maintain the body temperature. Mitochondrial uncoupling proteins(UCPs), which have a potential to generate heat by uncoupling oxidative phosphorylation, apper to play a crucial role in thermogenesis. Therefore UCP is commonly recognized as a key molecule in metabolic thermogenesis and its dysfunction contributes to the development of obesity. In these experiments, Daehwang water extracts inhibited the UCP1 mRNA expression increase by cold exposure in brown adipose tissue. But other herbs did not significantly influence on UCPs mRNA expression in white adipose tissue and seleus muscle tissue. Based on this experiment, we will try to clarify the effects of Daehwang water extracts on UCP1 expression and function.

Objectives : To learn the effects of CV12·CV9 and LI4·LI11 electroacupuncture stimulation on high fat diet induced-obese mouse. Methods : The control group was provided with high fat diet without electroacupuncture stimulation, while the experimental group CV12·CV9 and LI4·LI11 with three frequencies(10㎐, 100㎐, 200㎐) for 5 times a week during five week periods with the high fat diet. We investigated the body weight, weight gain, and food intake. Results : After the electroacupuncture stimulation, a significant loss of increasement in weight was found in the experimental group compared with the high fat diet group. But there was no significant dif- ference made by the change of acupoint combinations and frequencies. After the electroacupuncture stimu- lation, no significant change was found in the food intakes, but the weight gain a week was decreased compared with high fat diet group. Conclusions : These results suggest that electroacupuncture stimulation on CV12·CV9 and LI4·LI11 might be effective in treatments of obesity to high fat diet.

A new human sensibility (kansei) meter that can measure human sensibility at the indoor environment is developed in this paper. Four sensors that can measure temperature, humidity, CO₂ and C₄H_(10) concentrations are used. Among these sensors, the first three are used to determine the human sensibility, And the last to protect human from the harmful gas. First of all, human sensibilities are defined for each sensor datum, and then those are linearly combined to make a final human sensibility (kansei). The efficiency and usefulness of the meter are verified using a simulator on Windows 95 and a stand-alone system constructed using a microprocessor.
