New Antimicrobial Activity from Korean Radish Seeds (Raphanus sativus L.)

To isolate antifungal substances from Korean radish (Raphanus Sativus L.) seeds, various purification techniques such as DE52 cellulose anion exchange, SP-Sephadex C-25 cation exchange, and Sephadex G-50 gel filtration chromatographies were used. The molecular masses of two purified R. sativus antifungal proteins (RAPs) were estimated to be about 6.1kDa (RAP-1) and 6.2kDa (RAP-2) by SDS-PAGE, and 5.8kDa (RAP-1) and 6.2kDa (RAP-2) by a gel filtration chromatography, respectively. Purified proteins RAP-1 and 2 clearly exhibited different growth inhibitory activities against other microorganisms like Candida albicans and Saccharomyces cerevisiae. Although they have similar molecular masses, both RAP-1 and 2 proteins are not identical because their microbial inhibitory actions were different. Therefore, RAP-1 could be a new antifungal protein when compared with the antifungal activities of 2S albumins, Rs-AFPs, Mj-AMPs, chitinase, glucanase, permatin, and ribosome inactivating proteins, all of which are antifungal proteins of plants.

TSH secreting pituitary adenoma is a rare disorder and represents about 1% of all pituitary adenomas. About 80 cases have so far been reported. We experienced four cases of TSH secreting pituitary adenoma. Three of them were macroadenomas and 1 was microadenoma. Serum TSH levels varied between 4.1~185.6 uIU/ml. The serum -subunit levels of two patients tested were slightly elevated (0.87, 0.96 mIU/ml). Serum TSH and -subunit response to TSH, dexamethasone, T3, bromocriptine, and octreotide were variable when studied. Three patients underwent transsphenoidal microsurgery. One of them received radiotherapy. They were cured. Treatment with octreotide in 1 patient who did not received surgery resulted in normalization of serum TSH and thyroid hormone levels and reduction in tumor size. 포항 지역에서 생산되는 부추를 중심으로 부추의 식품기능성 분석을 한 결과 암예방 지표 효소인 QR의 유도능을 확인하였고 항산화 효소의 유도능은 음성대조구에 비해 약 2배 이상의 활성을 보여 앞으로 더 많은 연구를 기대하게 한다. 지역에 따른 QR 유도능과 SOD 유사 활성 유도능의 차이는 각 지역의 토질과 환경에 의한 차이에 기인하는 것으로 판단된다. 이에 대한 구체적인 연구는 토양분석 등 기후 특성에 대한 연구가 이루어져야 할 것으로 사료된다. 부추의 항비만기능성 식품으로서의 가능성을 지방세포 분해능에 의해 측정한 결과 50㎍/㎕ 농도에서는 음성 대조구의 약 3배 이상의 분해능을 나타내어 항비만 기능성 식품으로서 효과가 큰 것으로 기대된다. Leek(Allium tuberosum Rotter) is a korean traditional herb of which leaves are used for daily food. In order to improve the potency of leek as a functional food, we investigated the several biochemical functions in vitro. The quinone reductase activity, and anticarcinogenic enzyme, was induced 2 times at the concentration of 1㎎/㎖ of leek as compared to that of negative control(80% methanol). The induction of SOD(Superoxide dismutase) activity was also demonstrated. But the functional activities of leek were shown to be a little difference depending on the place where the leek was produced. The difference might to be due to the different environmental factors where affects the growth of leek. In an experiment to test the lypolytic activity of leek, the glycerol release was significantly increased(about 3 times to that of negative control). This was suggested that leek can be considered as a potential food for diet. The above results provide the potential food, and more profound research is requested.
Characterization and Purification of Anti-Complement Polysaccharide from Spirodela polyrhiza

We purified and characterized a crude polysaccharide from Spirodela polyrhiza with anti-complement activities. The crude polysaccharide fraction (SP-0) which had potential anti-complement activity was extracted in hot water for 4 hrs at 10$0^{\circ}C$. The ethanol-precipitate, the crude polysaccharide traction (SP-1), showed a potent anti-complement activity. Further purification of the crude polysaccharide (SP-1) was carried out by cetavlon, ion exchange chromatography and gel column chromatography. Among cetavlon fractions, SP-4 showed the most potent anti-complement activity. When 100 $\mu\textrm{g}$/mL of SP-4 was incubated with an equal volume of normal human serum (NHS), the TCH$_{50}$ was reduced by about 78%. When the SP-4 fraction was further purified by DEAE-Sepharose (Cl$^{[-10]}$ ), the SP-4IIa, SP-4IIb and SP-4IIc, absorbed fractions, were almost the same as the anti-complement activities of SP-4. SP-4IIc, having the greatest potential activation and the highest yield by ion exchange chromatography, was further purified by gel column chromatography on a Sepharose CL-6B column. Four polysaccharide fractions of SP-4IIc-1, SP-4IIc-2, SP-4IIc-3 and SP-4IIc-4 were obtained, consisting mainly of arabinose, rhamnose, galactose and glucose, with approximate molecular weights of about 305,000, 132,000, 64,000 and 12,000, respectively. Among these subfractions, SP-4IIc-1 had the most potent anti-complement activity. When the SP-4IIc-1 aggregate was applied to a gel column chromatography in 10 mM and 50 mM NaCl solution, the position of the peak fractions shifted to a low molecular weight region, and the molecular weight of SP-4IIc-1 decreased with increased NaCl concentration in the gel column chromatography. It was found that the self-aggregation formed spontaneously in void volume by gel column chromatography using Sepharose CL-6B in water and the self-aggregation significantly affected the anti-complement function.

The method to prepare multi-component random/block copolymers is one-shot (or partial) system which starting materials are mixed at one time. The random/block copolymers composed of diesters and 1,4-butanediol were synthesized and charaterized by sequence length, thermal property, and crystallization. Thus the melting point trends and crystallization behaviors of multicomponent random/block copolymers were also depended on aromatic unit in copolymers as well as reaction condition.

These experiments were undertaken to develope a simple and reliable method of nonsurgical ova collection under the field condition. Uterine horns of 37 cows were non-surgically flushed with Dulbecco`s phosphate buffered saline supplemented with 1% fetal calf serum by two different flushing procedures; the gravity and pressure flushing method using two way foley catheter. The results obtained in these experiments were summarized as follows: 1. It was possible to pass the catheter through the cervix to infuse the flushing medium into the uterine cavity in all cows tried, but of 37 cows tried to recover ova, 70.3% succeeded in recovery of 1 or more ova. 2. The highest success rate (87.5%) in ova recovery was obtained from the 1 or 2 calved cows and the lowest (57.1%) from 5 or more calved cows. 3. The recovery rates of flushing medium infused into the uterine cavity by gravity and pressure flushing method were 94.0 and 96.6%, respectively. 4. An average of 4.2 and 4.9 ova per cow were recovered yielding recovery rates of 56.7% and 59.5%, respectively, by gravity and pressure flushing methods. 5. It could be concluded from the above mentioned results that the pressure flushing method is more reliable and effective than the gravity flushing method in saving flushing medium and collecting ova.
