        Effects of sperm insemination on the final meiotic maturation of mouse oocytes arrested at metaphase I after in vitro maturation

        Objective: The aims of this study were to investigate whether fertilization could induce the resumption of meiosis in mouse oocytes arrested at metaphase I (MI) after in vitro maturation (IVM), and to investigate the effect of $Ca^{2+}$ chelator treatment at the time of fertilization on the transition from MI to metaphase II (MII). Methods: MII-stage and arrested MI-stage mouse oocytes after IVM were fertilized, and then embryonic development was monitored. Blastocysts from each group were transferred into 2.5 days post-coitum pseudo-pregnant ICR mice. MI oocytes after IVM were treated with a $Ca^{2+}$ chelator to investigate the effect of $Ca^{2+}$ oscillations on their maturation. Results: As insemination time increased, the number of oocytes in the MI group that reached the MII stage also increased. The blastocyst rates and total cell numbers in the MII group were significantly higher than in the MI group. No pregnancy occurred in the MI group, but 10 pregnancies were achieved (10 of 12) in the MII group. The proportion of MI oocytes that matured to MII oocytes after fertilization was significantly higher in the non-treated group than in the $Ca^{2+}$ chelator-treated group. Conclusion: The findings that a higher proportion of MI-arrested oocytes progressed to MII after fertilization and that the MI-to-MII transition was blocked by $Ca^{2+}$ chelator treatments before fertilization indicate that the maturation of MI oocytes to MII oocytes is associated with intracellular $Ca^{2+}$ oscillations driven by fertilization.

        백서 심근경색모델에서 시간경과와 경색의 크기에 따른 심자도의 변화

        N/A Background: Magnetocardiogram (MCG), which records the changes of magnetic fields generated by the heart`s electrical activity, theoritically can provide unique data for clinical application. To date, MCG has been investigated only at a single time point after myocardial infarction (MI) with severe left ventricular dysfunction in rats. The purpose of the present study was to investigate sequential changes of MCG after MI and to evaluate effects of infarct size on MCG. Methods: Acute MI were induced by the permanent ligation of left coronary artery in 22 rats. Magnetic fields were recorded just above a rat with Nb Superconducting Quantum Interference Device (SQUID) gradiometer inside a magnetically shielded room. MCG was measured before and immediately after surgery and it was subsequently recorded at the time points of 1, 4 and 6 hours post operatively. MCG was also measured at 1, 3, 7 and 21 days after surgery. Results: Elevation of ST segment and appearance of pathological Q wave on the MCG were evident immediately after the ligation of coronary artery and persisted to 6 hours after MI. On MCG, ST segment was depressed and T wave was inverted from 1 day after MI. In rats with small-and moderate- sized MI (infarct size〈30%), ST depression returned to near the isoelectric level and Q wave disappeared from 7 days after MI. However, ST depression and Q wave were still present in rats with larger infarct (infarct size≥30%). Conclusion: Evolutional changes of MCG were well-recognized up to 21 days after MI. Furthermore, the infarct size can be expressed by the extent of Q wave and ST segment depression on MCG. Taken together, these data indicate that MCG is a helpful modality for the diagnosis, evaluation of infarct size and follow up after MI.(Korean J Med 62:42-48, 2002)

      • Histone deacetylase 3 is selectively involved in L3MBTL2-mediated transcriptional repression

        <P><B>Abstract</B></P><P>This is the first report that L(3)mbt-like 2 (L3MBTL2) specifically interacts with the histone deacetylase domain of histone deacetylase 3 (HDAC3) via its MBT domain. Here, we show that L3MBTL2 selectively interacts with HDAC3, but not other class I HDACs. An in vitro peptide-binding assay demonstrated the specific association of HDAC3 with methylated histone-K20 tail and L3MBTL2. Furthermore, depletion of HDAC3 resulted in a decrease of methylated K20-H4, as well as an increase in acetylated histone H3. Consequently, HDAC3 knock-down selectively suppressed L3MBTL2-mediated transcriptional repression.         한국인 직무 스트레스 측정도구의 개발 및 표준화

        Background and Purposes: Over the past three decades, numerous studies performed in Korea have reported that job stress is a determinant risk factor for chronic diseases and work disability. Every society has its own culture and occupational climate particular to their organizations, and hence experiences different occupational stress. An occupational stress measurement tool therefore needs to be developed to estimate it objectively. The purpose of this study is to develop and standardize the Korean Occupational Stress Scale (KOSS) which is considered to be unique and specific occupational stressors in Korean employees. Subjects and Methods: Data were obtained from the National Study for Development and Standardization of Occupational Stress (NSDSOS Project: 2002-2004). A total of 12,631 employees from a nationwide sample proportional to the Korean Standard Industrial Classification and the Korean Standard Occupational Classification were administered. The KOSS was developed for 2 years (2002-2004). In the first year, we collected 255 items from the most popular job stress measurement tools such as JCQ, ERI, NIOSH and OSI, and 44 items derived from the a qualitative study (depth interview). Forty-three items of KOSS, in the second year, were retained for use in the final version of the KOSS by using Delphi and factor analysis. Items were scored using conventional 1-2-3-4 Likert scores for the response categories. Results: We developed eight subscales by using factor analysis and validation process: physical environment (3 items), job demand (8 items), insufficient job control (5 items), interpersonal conflict (4 items), job insecurity (6 items), organizational system (7 items), lack of reward (6 items), and occupational climate (4 items). Together they explained 50.0% of total variance. Internal consistency alpha scores were ranged from 0.51 to 0.82. Twenty-four items of the short form of the KOSS (KOSS-SF) were also developed to estimate job stress in the work setting. Because the levels of the subscales of occupational stress were gender dependent, gender-specific standard norms for both the 43-item full version and the 24-item short form using a quartile for the subscales of KOSS were presented. Conclusion: The results of this study suggest that KOSS might be an appropriate measurement scale to estimate occupational stress of Korean employees. Further and more detailed study needs to be conducted to improve the validity of this scale.

        한국전쟁기 김말봉의 『별들의 故鄕』 연구

        This paper takes a close look at Kim, Mal-bong's Hometown of Stars released during the Korean War period, in order to relate the writer's literary characteristics seen after the liberation from the Japanese colonial rule to the characteristics in social melodrama. Hometown of Stars adopts the formula of social melodrama that gives its unique double effect by combining basic plot of love stories among characters with social and historical events as a background. First, in the Hometown of Stars, the Licensed Prostitution Abolition Movement after the Liberalization plays a role in diversifying love relationship among characters and gives an opportunity for self-reflection. In particular, the Movement is used to highlight the changing social views of the characters. The writer indirectly sheds light on the inhumane treatment of prostitutes and the problems of residual values of the feudalistic way of thinking based on the class system. However, his intention to put the behind story of social and historical facts into a realistic context, rather than to investigate the Movement in itself helps intensify the pleasure to read this novel. Next, the social and historical background of the Korean War serves as a vehicle to provide plausibility to the plot and to raise readers' awareness of the reality through the hidden fact of history. The writer's views that the government's misinformation at the time of the Korean War helped increase people's casualties are represented in a natural fashion through characters' behaviors, evoking a sense of reality on the historical facts from readers. Moreover, the War, which serves as a defining moment in dramatizing love conflicts of the characters, boosts the interest in reading the story. The writer sets the Korean War as a turning point to convert the ideological confrontation after the Liberalization to the punitive confrontation structure in which virtue is rewarded and vice is punished. The ending of Hometown of Stars shows both melodramatic limit and social melodramatic characteristics that go beyond the limit, because the clash between the prevailing ideology and the moral faith forms the typical punitive confrontation structure, while transcending the formula to reach the universal value of humanity, especially religious ethics. The writer is not swayed by conventions or ideology and he uses “star” as a metaphor of human and spirit, hoping that they can be reborn in the “hometown”, the source of new hope. In this symbolic fashion, he suggests a “wise” way to overcome wartime confusion to readers. The formula of social melodrama in Hometown of Stars not only provides pleasure of reading popular novels, but also puts social and historical facts into a realistic context and deals a measurable blow to covered historical facts and ignored social thinking frame. In that sense, Kim, Mal-bong could succeed in strengthening literature's impact by the means of social melodrama that contains popular and social elements at the same time. This is related to his direct involvement and hard work in abolishing the Licensed Prostitution System after the Liberation. It is also notable that the Licensed Prostitution Abolition Movement is a main theme in Hometown of Stars, as well as in his other works after the Liberation, such as Splendid Hell, Blue Wings and Life.

        오리부산물과 한약재를 이용한 추출액의 영양성분 및 중금속 노출에 대한 피해 완화 효과 : (Ⅱ) 중금속 노출에 대한 오리추출액의 피해 완화 효과 (Ⅱ) Damage Mitigation Effects on Heavy-metals Exposure of Peking-Duck By-Product Extracts

        This study was performed to investigate the effects of peking-duck extracts added with medicinal herbs(DJ) on the intoxication of heavy metals (Hg, Pb, Cd, As) in rats. Sprague-Dawley rats weighing 150±15g, were randomly assigned to 5 groups: basal diet only in normal control group(NCG); basal diet and heavy metals without DJ injection in heavy metal control group(HMC); basal diet, heavy metals and DJ(3㎎/㎖) injection in heavy metal low duck juice group(HMLD), basal diet, heavy metals and DJ(30㎎/㎖) injection in heavy metal middle duck juice group(HMMlD); basal diet, heavy metal and DJ(300㎎/㎖) injection in heavy metal high duck juice group(HMHD). Hg and As was injected by 50ppm and Cd and Pb by 25ppm for 17days. Also DJ oral feeding was conducted for 28days. The result of this study were as follows: Food intake and body weight gain in heavy metal administered groups were lower than those of NCG. Liver, kidney and testis weights were not significantly different among 5 groups. GOT, GPT and BUN activities were significantly reduced in DJ treated groups as compared to HMC. DJ showed the suppressing effect on the accumulation of Hg, Pb and Cd in serum, liver and kidney. Fecal Hg and Cd excretions increased with DJ feeding. The results suggested that DJ may have some protective effects on Hg, Cd and Pb intoxication by reducing the accumulation m tissues and increasing excretion. This study also showed the effective way of using duck-extract and its application to the oriental medicine.

        경막 내 colistimethate 투여로 치료한 다제 내성 cinetobacter baumannii 뇌실염

        Acinetobacter species is a non-fermentative aerobic gram-negative coccobacillus that is an important pathogen found in nosocomial infections. Recently, multi-drug resistant Acinetobacter baumannii (MDR-AB) infections have been increasing and pose a serious problem. Most such infections present as bacteremia, pneumonia, or a wound infection; however, CNS infections are very rare. We herein present a case of ventriculitis caused by MDR-AB in a 37-year old man after a neurosurgical intervention. The patient was successfully treated with intrathecal colistimethate.
