        This is to investigate the posibility of home manufacturing of heavy chemical industry machines, such as rotary kiln, crusher, blower, cooler, electrostatic precipitator and weighing feeder of cement plant. It is concluded that even though we can not make all of them (some of them are made now and some others are going to be made in the near future, some of them are made whole and some others are made partially), we can build or export the cement plant by importing the important machines which we can not make now and by substituting them with ours gradually.

        Efficient Interleukin-21 Production by Optimization of Codon and Signal Peptide in Chinese Hamster Ovarian Cells

        Interleukin-21 is a common γ-chain cytokine that controls the immune responses of B cells, T cells, and natural killer cells. Targeting IL-21 to strengthen the immune system is promising for the development of vaccines as well as anti-infection and anti-tumor therapies. However, the practical application of IL-21 is limited by the high production cost. In this study, we improved IL-21 production by codon optimization and selection of appropriate signal peptide in CHO-K1 cells. Codon-optimized or non-optimized human IL-21 was stably transfected into CHO-K1 cells. IL-21 expression was 10-fold higher for codon-optimized than non-optimized IL-21. We fused five different signal peptides to codon-optimized mature IL-21 and evaluated their effect on IL-21 production. The best result (a 3-fold increase) was obtained using a signal peptide derived from human azurocidin. Furthermore, codon-optimized IL-21 containing the azurocidin signal peptide promoted IFN-γ secretion and STAT3 phosphorylation in NK-92 cells similar to codon-optimized IL-21 containing original signal peptide. Collectively, these results indicate that codon optimization and azurocidin signal peptides provide an efficient approach for the high-level production of IL-21 as a biopharmaceutical.

        서울시 대기오염과 일별 사망의 상관성에 관한 시계열적 연구 (1998~2001년)

        This study was performed to examine the relationship between air pollution exposure and mortality in Seoul for the years of 1998~2001I. Daily counts of death were analyzed by general additive Poisson model, with adjustment for effects of seasonal trend, air temperature, humidity, and day of the week as confounders in a nonparametric approach. Daily death counts were associated with CO (current day), 0₃(current day), PM_(10) (current day), NO₂, (1 day before), SO₂, (1 day before). Increase of 41.711㎍/㎥ (interquartile range) in PM_(10) was associated with 1.3% (95% CI = 0.7~1.9%) increase in the daily number of death. 0₃concentrations resulted in an increased risk of 1.3% for 23.86 ppb in all-aged mortality [RR = 1.013 (1.004-1.023)]. This effect was greater in children (less than 15 aged) and elderly (more than 65 aged). After ozone level exceeds 25 ppb, the dose-response relationship between mortality and ozone was almost linear. We concluded that Seoul had 1~5% increase in mortality in association with IQR (interquartile range) in air pollutants. Daily variations in air pollution within the range currently occurring in Seoul might have an adverse effect on daily mortality. These findings also support the hypothesis that air pollution, at levels below the current ambient air quality standards of Korea, is harmful to sensitive subjects, such as children or elderly.

        반월상 연골-대퇴 인대의 반월상 연골 장력 유지에 대한 기능 : 돼지 슬관절을 이용한 실험적 연구 An Experimental Study Using the Knee Joint of the Pig

        목적: 후방 반월상 연골-대퇴 인대가 외측 반월상 연골의 장력을 유지하여 주는 기능이 있음을 돼지의 슬관절을 이용하여 실험적으로 증명하고자 하였다. 대상 및 방법: 실험용 재료로는 돼지의 슬관절을 이용하였으며 후방 반월상 연골-대퇴 인대가 정상 및 단열된 상태에서 각각 1) 정상 외측 반월상 연골, 2) 외측 반월상 연골 후각부의 방사상 파열, 3) 외측 반월상 연골 전절제술의 상태를 만들어 실험하였다. Instron기계를 이용하여 슬관절의 신전위에서 150kg의하중이 슬관절에 전달되도록 하였으며 pressure-sensitive film(저압용 Prescale film. Fuji)을 이용하여 가압 면적, 최대 압력 및 평균 압력을 측정하였다. 판독은 후지필름 Prescale 압력 화상 해석 시스템인 FDP-901E series를 이용하였다. 결과: 후방 반월상 연골-대퇴 인대가 정상이면 정상군과 방사상 파열군에서 가압 면적, 최대압력 및 평균 압력이 큰 차이를 보이지 않았다. 하지만 후방 반월상 연골-대퇴 인대의 단열된 경우에는 외측 반월 연골의 상태에 관계없이 압력이 한 곳에 집중되는 소견을 보여 후방 반월상 연골-대퇴 인대가 정상인 상태에서 외측 반월상 연골 전절제술의 경우와 결과가 비슷하였다. 결론: 돼지의 후방 반월상 연골-대퇴 인대가 축성 부하에 대하여 외측 반월상 연골의 장력을 유지하여 주는 기능을 담당한다는 것을 알 수 있었다. Purpose: To prove experimentally if the posterior meniscofemoral ligament (PML) has hoop tensioning function for the lateral meniscus using the knee joint of the pig. Materials and Method: Amputated stumps with the knee joint of the pig (Yorkshire) were used. The experiment performed under 6 conditions; 1) intact PML with intact lateral meniscus (LM) 2) intact PML with radial tear of posterior horn of LM, 3) intact PML with intact lateral meniscus (LM), 2) intact PML with radial tear of posterior horn of LM, 3) intact PML with total lateral meniscectomy, 4) cut PML with intact LM, 5) cut PML with radial tear of posterior horn of LM, and 6) cut PML with total lateral meniscectomy. The pressure-sensitive film (Prescale, Fuji) was inserted under the lateral femoral condyle and axial loads was transmitted to the knee joint with universal testing machine (Instron, model No. 4469, USA). The pressed area, maximum pressure and average pressure were measured by the Prescale imaging analysis system FDP-901E series. Results: With the intact PML, there was little difference in pressed area and maximum and average pressures between the conditions of intact meniscus and radial tear of posterior horn of LM. With the cut PML, regardless of the states of the LM, pressure concentration (much decreased pressed area, and increased maximum and average pressures) occurred, which had similar results to the condition of total lateral meniscectomy with intact PML. Conclusion: The PML of the pig had hoop tensioning function for the LM under axial loads.

      • 악성 혈액질환에서 성공적인 동종골수이식 후 숙주 기질 미세환경의 구축

        인체의 골수는 간엽모세포를 함유하고 있으며 이들은 골수미세환경의 주된 세포들로 분화가 가능하여 조혈기능을 지지한다. 본 연구에서는 성별이 다른 동종조혈모세포이식 환경에서 조혈모세포의 완전 생착을 보이고, 이식 후 1년에서 8년이 지난 11예의 재생불량성 빈혈 및 백혈병 환자들을 대상으로 하여 골수에서 MSC를 분리하고 체외 확장을 통해 배양된 MSC에서 X 염색체 탐식자를 이용한 FISH 및 microsatellite polymorphism PCR 기법으로 그 기원을 확인하였다. 그 결과 조혈모세포는 완전히 공여자 기원으로 대치되었음에도 불구하고 MSC는 모두 수여자 기원임을 알 수 있어, 동종조혈모세포이식에서 미세환경의 구축은 수여자의 자가 생산에 의한 골수 간질세포에 의한 것으로 생각된다. Background: Human bone marrow (BM) contains mesenchymal stem cells (MSC) that can differentiate into various cells of mesenchymal origin. It remains a matter of controversy whether donor-derived stromal cells are capable of engraftment following hematopoietic stem cell transplantation (HSCT) or not. Methods: To determine if donor-derived stromal cells are transferred to the recipients of allogeneic HSCT, we investigated the characterization of MSC in 11 patients 1 to 8 years after sex mis-matched allogeneic HSCT in severe aplastic anemia and leukemia. Results: All patients had complete engraftment with donor- derived stem cells as shown by detection of donor type DNA in peripheral blood mononuclear cells. Following culture, MSC showed the expression of SH2 and SH4, but none of the hematopoietic markers of CD14, CD34, or CD45. MSC which can be differentiated to osteogenic lineage showed the genotype of recipient completely using FISH or PCR analysis. Conclusion: This study confirmed that MSC isolated from recipients of allogeneic HSCT in severe aplastic anemia and leukemia are not of donor genotype despite of full hematopoietic engraftment with donor type. Donor cells did not contribute to reconstitute the marrow microenvironment.

        광주시 대기오염과 일별 사망의 상관성에 관한 시계열적 연구 : 1998~2001년

        This study is performed to examine the relationship between air pollution exposure and mortality in Gwangju for the years of 19% - 2001. Daily counts of death were analyzed by general additive Poisson model, with adjustment for effects of seasonal trend, air temperature, humidity, and day of the week as confounders in a nonparametric approach. Daily death counts were associated with CO(current day), 03(6 day before), PMdcurrent day), NO₂(1 day before), SO₂(2 day before). Increase of 29.74 pg/m (interquartile range) in PM_(10)was associated with 1.4 % (95% CI = 0 % - 2.8 %) increase in the daily number of death. This effect was greater in children(1ess than 15 aged) and elderly(more than 66 aged). We concluded that Gwangju had 1 - 24 % increase in mortality in association with IQR in air pollutants. Daily variations in air pollution within the range currently occurring in Gwangju might have an adverse effect on daily mortality. These findings also support the hypothesis that air pollution, at levels below the current ambient air quality standards of Korea, is harmful to sensitive subjects, such as children or elderly.

      • 이종재료간 Ⅴ-노치문제에 대한 응력강도계수의 광탄성 실험적 해석

        접합 구조물에서는 이종재료간의 V-노치(Notch)부가 있게 되고, V-노치선단부에서 응력집중현상이 생겨 접합계면균열이 발생하게 된다. 따라서 V-노치부의 강도평가가 중요하나 강도평가법이 아직 확립되지 못한 것이 현실이다. 이종재료간 V-노치부의 응력장은, σ_{ii} ∝ Kr^{p-1}의 관계로 알려지 있고, K는 응력강도계수에 해당되고, p-1은 응력특이성 지수이다. 그런데, 이종재료간 V-노치부의 응력장에서 노치선단에서 거리 r가 o으로 되면 응력이 무한대로 되는 응력특이성지수 p-1이 2가지 이상인 경우가 되고, 균열일 때처럼 응력특이성지수 p-1이 -0.5로 고정되지 않으므로 응력강도계수 K만의 파괴법칙이 성립할 수 없다. 따라서, 이종재료간 V-노치부의 노치각도 및 재료의 종류에 따른 응력특이성지수를 구해야 되고 그때의 응력강도계수를 구해야 한다. 이종재료간 V-노치부의 노치각도 및 재료의 종류에 따른 응력특이성지수와 응력강도 계수 해석에 각각 뉴톤-랍슨법(Newton-Raphson Method), 뉴톤-랍슨법과 최소자승법을 이용한 선점법(Collocation method)이 응용될 수 있음을 수치해석을 통하여 저자의 일부가 확인한 바 있다. 광탄성 실험을 통하여 이종재료간 V-노치부의 노치각도 및 재료의 변화에 따른 응력강도계수의 해석이 뉴톤-랍슨법과 최소자승법을 이용한 선점법(Collocation method)에 의해 가능한 지를 검토한 결과 두 재료를 접합시킬 때 잔류응력이 발생하지 않을 때는 가능함을 확인하였으나, 광탄성 실험결과의 데이터를 체취할 때의 오차와 응력특이성의 강약에 따른 영향 그리고 두 재료를 접합할 때에 발생한 잔류응력의 영향이 있을 때는 이러한 오차와 영향을 고려한 많은 연구가 있어야 된다. Sress singularities and stress intensity factors of two materially V-notched problems are analyzed by the Newton-Raphson method which is an efficient numerical method for solving a non-linear equation and by the collocation method using the Newton-Raphson and least squares method. The photoelastic isochromatic fringe patterns by the experiment are compared with those by computer graphics. The problems of experiment or analysis for stress intensity factors of two materially V-notched plate are discussed.

        서울 성동구 지역 미세먼지의 화학적 조성에 관한 연구

        To investigate the chemical characteristics of PM_(2.5) in Seoul, Korea, atmospheric particulate matters were collected using a PM_(10) dichotomous sampler including PM_(10) and PM_(2.5) inlet during the period of October 2000 to September 2001. The Inductively Coupled Plasma-Mass Spectromety (ICP-MS), Ion Chromatography (IC) methods were used to determine the concentration of both metal and ionic species. A statistical analysis was performed for the heavy metals data set using a principal component analysis (PCA) to derived important factors inherent in the interactions among the variables. The mean concentrations of ambient PM_(2.5) and PM_(10) were 24.47 and 45.27 ㎍/m³, respectively. PM2 5 masses also showed temporal variations both yearly and seasonally. The ratios of PM_(2.5)/PM_(10) was 0.54, which similar to the value of 0.60 in North America. Soil-related chemical components (such as Al, Ca, Fe, Si, and Mn) were abundant in PM_(10), while anthropogenic components (such as As, Cd, Cr, V, Zn and Pb) were abundant in PM_(2.5). Total water soluble ions constituted 30~50 % of PM_(2.5) mass, and sulfate, nitrate and ammonium were main components in water soluble ions. Reactive forms of NH₄^(+) were considered as NH₄NO₃ and (NH₄)₂SO₄ during the sampling periods. In the results of PCA for PM_(2.5), we identified three principal components. Major contribution to PM_(2.5) seemed to be soil, oil combustion, unidentified source. Further study, the detailed interpretation of these data will need efforts in order to identify emission sources.

        Primary systemic amyloidosis (AL amyloidosis) is an uncommon disease characterized by the accumulation in vital organs of a fibrillar protein consisting of monoclonal immunoglobulin light chains. It is a plasma cell dyscrasia usually accompanied by a bone marrow plasmacytosis and an M component in the serum or urine. The presenting features of AL amyloidosis are protean and frequently nonspecific, but the diagnosis is rarely made until symptoms or signs referable to a particular organ appear. The amyloid deposits cause cardiac or renal dysfunction and, ultimately, death. We report a case of AL amyloidosis, presented with unexplained diarrhea and abdominal pain for eight months, documented by a renal biopsy, and review the previous literatures
