Tumor-infiltrating lymphocytes ('1°ILs) interact most closely with tumor cells and thus are more likely to reflect tumor host interactions accurately. But it is unknown whether such T cells are nonspecific inflammatory cells or a subset of specific host immune responses. In this study, there was no clear correlation between the infiltration of T lymphocytes in stomach cancer and the overexpression of c-ErbB-2 or increasing class I MHC expression on tumor cells. A positive correlation was seen between the presence of TILs in the tumor and tumors with diploidy by flow cytometric DNA analysis. The proliferative responses of Ills stimulated with IL-2, anti-CD3 mAb, or both were examined. When compared to normal mucosal-associated lymphoid tissue lymphocytes, the proliferative response of TILs to high dose IL-2 was minimal. A similarly poor response to anti-CD3 mAb plus IL-2 was also observed. The freshly isolated TILs exhibit reduced ability to proliferate in response to IL-2, anti-CD3 mAb or both. The microenvironment of the tumor suppresses the proliferative capacity of the TILs. The mechanism of this suppression remains unknown. It could be mediated by suppressor cells, by soluble substances within the tumor, or both. To examine this question, supernatants of stomach cancer cells (SNSNU-1) were tested for the presence of immunosuppressive factors. Human peripheral blood T-cells and tumor-draining lymph node lymphocytes (TDLNL) were incubated for 3 days with SNSNU-1 and then assessed for proliferative responses to PMA, anti-CD28 mAb, or both and for the inducibility to express IFN- r or IL-4 mRNA to PMA. Peripheral blood T-cells pretreated with SNSNU-1 were unable to proliferate in response to PMA, anti-CD28 mAb or both. SNSNU-1 also produces inhibitory activities of TDLNL proliferative response to PMA or anti-CD28 mAb and PMA (49%, 52%, respectively). In contrast, culture supernatants obtained from HEp-2, K562 or Daudi showed normal proliferative responsiveness of peripheral blood T-cells and TDLNL by PMA, anti-CD28 mAb or both.
거대 점오염원이 주변 대기질에 미치는 영향에 관한 연구

In order to show the effect of a vast point pollutant source on air quality of Pusan Thermoeletric Power Plant and its surrounding area, air quality around Pusan Thermoeletric Power Plant was simulated by ISCLT-2 which was supplied by EPA. For this purpose the emission amount of SO_2, NO_2 and TSP was calculated and atmospheric stability was classified for a recent decade(1985~1994) in Pusan. A result of the emission amount showed that much amount of NO_2, NO_2 and TSP are emitted from industrial area. It was clear that NO_2 is much emitted from line source and industrial area. And as a result of classification of atmospheric stability, neutral, stable and unstable state were 58%, 24.1% and 17.9%, respectivly. The result of ai quality simulation by ISCLT-2 showed that Pusan Thermoeletric Power Plant is affecting on the increse of 2.0ppb, 3.0ppb and 5.0㎍/㎥, SO_2, NO_2, and TSP respectively at its surrounding area, site A-3 which was located westward 2.2㎞ distance from Plant

<P>Plant-virus-based expression vectors have been used as an alternative to the creation of transgenic plants. Using a virus-based vector, we investigated the feasibility of producing the endoglucanase D (EngD) from Clostridium cellulovorans in Nicotiana benthamiana. This protein has endoglucanase, xylanase, and exoglucanase activities and may be of value for cellulose digestion in the generation of biofuels from plant biomass. The EngD gene was cloned between the nuclear inclusion b (NIb)- and coat protein (CP)-encoding sequences of pSP6PepMoV-Vb1. In vitro transcripts derived from the clone (pSP6PepMoV-Vb1/EngD) were infectious in N. benthamiana but caused milder symptoms than wild-type PepMoV-Vb1. RT-PCR amplification of total RNA from non-inoculated upper leaves infected with PepMoV-Vb1/EngD produced the target band for the CP, partial NIb and EngD-CP regions of PepMoV-V1/EngD, in addition to nonspecific bands. Western blot analysis showed the CP target bands of PepMoV-Vb1/EngD as well as non-target bands. EngD enzymatic activity in infected plants was detected using a glucose assay. The plant leaves showed increased senescence compared with healthy and PepMoV-Vb1-infected plants. Our study suggests the feasibility of using a viral vector for systemic infection of plants for expression of heterologous engD for the purpose of digesting a cellulose substrate in plant cells for biomass production.</P>
Rapid HPLC determination of gastrodin in Gastrodiae Rhizoma

Gastrodin is a major biologically active ingredient in members of the genus Gastrodia. For this reason, there are many reports on the quantification of gastrodin in Gastrodiae Rhizoma (GR) and in GR-containing herbal preparations. HPLC, HPLC-MS, and TLC are the major approaches for gastrodin quantification; however, they usually require complicated pre-treatment, lengthy analysis, and expensive instruments. Therefore, a rapid and reliable method for determining gastrodin in GR is necessary. Optimal HPLC separation was achieved using a Chromolith Performance RP-18e ($4.8{\times}100mm$, $5{\mu}m$) stationary phase. The optimal mobile phase was a mixture of water (A) and acetonitrile (B) with a gradient of 1 % B at 0-8 min, 20 % B at 8-10 min, 80-100 % B at 10-12 min, and 100 % B at 12-13 min, followed by equilibration with 1 % B for 2 min at a flow rate of 1.5 mL/min. The detection wavelength was UV220 nm. Gastrodin appeared within 4 min under the above conditions. The calibration curve of gastrodin showed good linearity in the 0.25-10.0 range ($r^2=0.9998$). The limit of detection ($0.13{\mu}g$), limit of quantification ($0.25{\mu}g$), and reproducibilities of area and retention time (0.04 and 3.24 %, respectively) were within acceptable ranges. In addition, the intra-day precision and accuracy of gastrodin were 0.74 and $100.63{\pm}0.04%$, respectively, while the inter-day precision and accuracy were 0.06 and $99.25{\pm}0.05%$, respectively. The range of mean gastrodin content in six GR samples, which were cultivated at different sites, was 0.37-0.79 %. This result may be a guideline for the quality control of GR and GR-containing medicinal preparations.
Antireflective grassy surface on glass substrates with self-masked dry etching

<P>Although recently developed bio-inspired nanostructures exhibit superior optic performance, their practical applications are limited due to cost issues. We present highly transparent glasses with grassy surface fabricated with self-masked dry etch process. Simultaneously generated nanoclusters during reactive ion etch process with simple gas mixture (i.e., CF<SUB>4</SUB>/O<SUB>2</SUB>) enables lithography-free, one-step nanostructure fabrication. The resulting grassy surfaces, composed of tapered subwavelength structures, exhibit antireflective (AR) properties in 300 to 1,800-nm wavelength ranges as well as improved hydrophilicity for antifogging. Rigorous coupled-wave analysis calculation provides design guidelines for AR surface on glass substrates.</P>
