      • Phase I/II study of S-1 combined with weekly docetaxel in patients with metastatic gastric carcinoma

        We designed a phase I/II trial of S-1 combined with weekly docetaxel to determine the maximum tolerated dose (MTD) and recommended dose (RD) and to evaluate the efficacy and toxicity in metastatic gastric carcinoma (MGC). Patients with measurable disease received S-1 orally b.i.d. on days 1–14 and docetaxel intravenously on days 1 and 8 every 3 weeks. In phase I (n=30), each cohort received escalating doses of S-1 (30–45 mg m<SUP>−2</SUP> b.i.d.) and docetaxel (25–40 mg m<SUP>−2</SUP>); MTD was 45 mg m<SUP>−2</SUP> b.i.d. S-1/35 mg m<SUP>−2</SUP> docetaxel and RD was 40 mg m<SUP>−2</SUP> b.i.d. S-1/35 mg m<SUP>−2</SUP> docetaxel. Dose-limiting toxicities included grade 3 elevated liver enzymes, gastric perforation, grade 3 diarrhoea/fatigue, febrile neutropenia with grade 3 anorexia/fatigue, and neutropenic infection with grade 3 stomatitis/anorexia. In phase II (n=52), the overall response rate was 66.7% (95% confidence interval (CI): 53.8–79.6%) and the median time to progression and overall survival were 6.5 months (95% CI: 4.9–8.1) and 13.7 months (95% CI: 9.9–17.5), respectively. The most common grade 3/4 toxicity was neutropenia (29.4%), and febrile neutropenia/neutropenic infection occurred in 19.6% of patients. Non-haematological toxicities were generally mild. There was one treatment-related death due to pneumonitis.         Resistive switching characteristics of ZnO-graphene quantum dots and their use as an active component of an organic memory cell with one diode-one resistor architecture

        We investigated the resistive switching characteristics of a polystyrene:ZnO-graphene quantum dots system and its potential application in a one diode-one resistor architecture of an organic memory cell. The log-log I-V plot and the temperature-variable I-V measurements revealed that the switching mechanism in a low-current state is closely related to thermally activated transport. The turn-on process was induced by a space-charge-limited current mechanism resulted from the ZnO-graphene quantum dots acting as charge trap sites, and charge transfer through filamentary path. The memory device with a diode presented a ~10<SUP>3</SUP>I<SUB>ON</SUB>/I<SUB>OFF</SUB> ratio, stable endurance cycles (10<SUP>2</SUP>cycles) and retention times (10<SUP>4</SUP>s), and uniform cell-to-cell switching. The one diode-one resistor architecture can effectively reduce cross-talk issue and realize a cross bar array as large as ~3kbit in the readout margin estimation. Furthermore, a specific word was encoded using the standard ASCII character code.

        산란계 사료내 CLA 함유 Oil (CLAzen 80) 첨가가 난황내 지방산 조성에 미치는 영향

        본 연구는 oil 형태의 conjugated linoleic acid(CLAzen 80)를 산란계 사료에 수준별로 첨가 급여하였을 때 산란율과 난황내 지방산 조성의 변화를 조사하기 위하여 수행하였다. 59주령의 산란계 72수를 완전임의배치법으로 4개 처리구에 6주간 공시하였다. 처리구는 CLAzen 80를 첨가하지 않은 대조구와 각각 1, 2 및 $3\%$를 첨가구를 두었다. 연구 결과를 살펴보면 산란율은 처리구별 통계적 유의차가 없었지만, 난황내 지방산 조성은 CLAzen 80 첨가에 의하게 크게 영향을 받았다. 난황내 C16:0과 C18:0과 같은 포화 지방산 함량은 CLAzen 80 첨가에 의하여 증가하였으나, 일가불포화지방산인 C18:1 함량은 오히려 감소하였다. 한편, 난황내 C18:2와 C18:3와 같은 다가불포화지방산은 CLAzen 80 급여 2$\~$4주사이에는 모든처리구들에 있어서 그 함량이 일정하게 유지되었다. 그러나, 대조구와 비교하여 CLAzen 80 급여 6주 째에는 난황내 C18:2 함량이 감소하였다. 불포화지방산:포화지방산 비율과 n-6:n-3 불포화지방산 비율은 2$\~$4주 사이에는 처리구별간에 통계적 유의차가 없었고, 6주째 불포화지방산:포화지방산 비율이 CLAzen 80 첨가에 의하여 감소하였다. 한편, 난황내 CLA 함량은 CLAzen 80 첨가수준에 비례하여 증가하였다. 따라서, 산란계 사료 내 CLAzen 80 첨가는 난황내 CLA 함량을 증진시키는 것으로 요약할 수 있다. The objectives of the present study were to investigate the effects of varying levels of dietary oil containing conjugated linoleic acid (CLA) on the egg production and fatty acid composition of egg yolk. Seventy-two 59-wk-old ISA Brown laying hens were randomly allotted to four dietary treatments, each consisting of three replicates with six birds per replicate. There were four treatments that consist of diets containing 0, 1, 2, or $3\%$ commercial CLA-containing oil. Egg production was not significantly different among the dietary treatments at 0, 2, 4, and 6 week. The proportion of saturated fatty acids such as C16:0 and C18:0 in egg yolk were increased, but that of monounsaturated fatty acid C18:1 was decreased by feeding CLA-containing oil supplementation. However, the proportion of polyunsaturated fatty acids such as C18:2 and C18:3 in egg yolk were not different among dietary treatments at 2 and 4 wk of the experiment. At 6 week, the proportion of C18:2 in egg yolk was decreased by feeding CLA-containing oil compared with the control. Polyunsaturated fatty acid:saturated fatty acid (P:S) ratio and n-6:n-3 polyunsaturated fatty acid ratio were similar across the treatments between 2 and 4 week. The P:S ratio was decreased by dietary CLA-containing oil supplementation at 6 week. The proportion of CLA in egg yolk was linearly increased with increasing levels of CLA-containing oil supplementation. In conclusion, dietary supplementation of CLA-containing oil to laying hens increased beneficially increased CLA content in their egg yolk.

        원발성 간암에서 PIVKA-II 및 Lens Culinaris Agglutinin-A 반응성 Alpha-fetoprotein(AFP-L3)의 임상적 유용성

        Background/Aims: Des-γ-carboxy prothrombin(DCP), a protein induced by vitamin K absence or antagonist-II(PIVKA-II), and Lens culinaris agglutinin-A reactive AFP-L3 have been reported to be useful markers for the diagnosis of hepatocellular carcinoma (HCC). In the present study, both the PIVKA-II and AFP-L3 were analyzed and compared with a conventional AFP to determine its usefulness, specificity and sensitivity in the diagnosis of HCC. Methods: Sera were collected from 108 patients consisting of 17 patients with chronic hepatitis, 22 patients with liver cirrhosis and 69 patients with HCC. The AFP-L3 was determined by an lectin affinity electrophoresis coupled with an antibody-affinity blotting. Level of DCP was measured by an enzyme immunoassay with an anti-DCP monoclonal antibody. Results: The sensitivity and specificity of PIVKA-II and AFP L3 were 49.3% and 89.5%, and 32.5% and 85.7%, respectively. No significant correlation was found between the PIVKA-II or AFP L3 and serum AFP. No correlation was found etween the PIVKA-II or AFP L3 and the characteristics of HCC.       • Loss of NDRG2 promotes epithelial-mesenchymal transition of gallbladder carcinoma cells through MMP-19-mediated Slug expression

        Background & Aims: Gallbladder carcinoma (GBC) is the most common malignancy of the biliary tract and one of the most lethal forms of human cancer. However, there is limited information about the molecular pathogenesis of GBC. Here, we examined the functional role of the tumor suppressor N-myc downstream-regulated gene 2 (NDRG2) and the underlying molecular mechanisms of disease progression in GBC. Methods: Clinical correlations between NDRG2 expression and clinicopathological factors were determined by immunohistochemical analysis of tumor tissues from 86 GBC patients. Biological functions of NDRG2 and NDRG2-mediated signaling pathways were determined in GBC cell lines with NDRG2 knockdown or overexpression. Results: Loss of NDRG2 expression was an independent predictor of decreased survival and was significantly associated with a more advanced T stage, higher cellular grade, and lymphatic invasion in patients with GBC. GBC cells with loss of NDRG2 expression showed significantly enhanced proliferation, migration, and invasiveness in vitro, and tumor growth and metastasis in vivo. Loss of NDRG2 induced the expression of matrix metalloproteinase-19 (MMP-19), which regulated the expression of Slug at the transcriptional level. In addition, MMP-19-induced Slug, increased the expression of a receptor tyrosine kinase, Axl, which maintained Slug expression through a positive feedback loop, and stabilized epithelial-mesenchymal transition of GBC cells. Conclusions: The results of our study help to explain why the loss of NDRG2 expression is closely correlated with malignancy of GBC. These results strongly suggest that NDRG2 could be a favorable prognostic indicator and promising target for therapeutic agents against GBC.

      • Degradation pattern prediction of a polymer electrolyte membrane fuel cell stack with series reliability structure via durability data of single cells

        The insufficient long-term durability of polymer electrolyte membrane fuel cell (PEMFC) stacks has been blocking commercialization of PEMFC technologies. An accelerated degradation test (ADT) is needed to facilitate the PEMFC development process by reducing the testing time. We propose an ADT procedure for a PEMFC stack with the concept of series reliability structure under startup-shutdown cycling testing conditions. The acceleration factor is estimated to fit the degradation paths of individual cells consisting of the PEMFC stack under normal use conditions via the accelerated degradation data of a single cell. We employ a nonparametric regression method to smooth the degradation curves observed from accelerated operating conditions. We illustrate the methodology for estimating the lifetime of the PEMFC stack using the theory of the smallest-order statistics. We propose a three-parameter Weibull distribution in fuel cell technology to fit the failure data of cells in a PEMFC stack.

        Beam production of <sup>18</sup>Ne with in-flight method for alpha scattering at CRIB

        <P><B>Abstract</B></P> <P>We conducted a measurement of <SUP>18</SUP>Ne + α scattering in inverse kinematics, in order to search for experimental evidence of α -cluster structure in <SUP>22</SUP>Mg above the alpha threshold ( <SUB> E thr </SUB> = 8 . 14 MeV ) and determine the astrophysical rates of the <SUP>18</SUP>Ne( α , p)<SUP>21</SUP>Na reaction under the conditions of break-out from the Hot-CNO cycle. A high intensity <SUP>18</SUP>Ne beam at 2.54 AMeV was successfully produced at CRIB (CNS Radioactive Ion Beam separator) of the Center for Nuclear Study, the University of Tokyo. This paper presents the in-flight production of the radioactive <SUP>18</SUP>Ne beam by using the production reaction <SUP>3</SUP>He(<SUP>16</SUP>O, <SUP>18</SUP>Ne)n with a windowed cryogenic gas target, which was employed in the mentioned resonant α -scattering experiment.</P>
