        Sanitizing radish seeds by simultaneous treatments with gaseous chlorine dioxide, high relative humidity, and mild heat

        <P>Sanitizing radish seeds intended for edible sprout production was achieved by applying simultaneous treatments with gaseous chlorine dioxide (ClO2), high relative humidity (RH, 100%), and mild heat (55 degrees C). Gaseous ClO2 was produced from aqueous ClO2 (0.66 ml) by mixing sulfuric acid (5% w/v) with sodium chlorite (10 mg/mL) in a sealed container (1.8 L). Greater amounts of gaseous ClO2 were measured at 23% RH (144 ppm after 6 h) than at 100% RH (66 ppm after 6 h); however, the lethal activity of gaseous ClO2 against naturally occurring mesophilic aerobic bacteria (MAB) on radish seeds was significantly enhanced at 100% RH. For example, when exposed to gaseous ClO2 at 23% RH, the number of MAB on radish seeds decreased from 3.7 log CFU/g to 2.6 log CFU/g after 6 h. However, when exposed to gaseous ClO2 at 100% RH for 6 h, the MAB population decreased to 0.7 log CFU/g after 6 h. Gaseous ClO2 was produced in higher amounts at 55 degrees C than at 25 degrees C, but decreased more rapidly over time at 55 degrees C than at 25 degrees C. The lethal activity of gaseous ClO2 against MAB on radish seeds was greater at 55 degrees C than at 25 degrees C. When radish seeds were treated with gaseous ClO2 (peak concentration: 195 ppm) at 100% RH and 55 degrees C, MAB were reduced to populations below the detectable level (<-0.7 log CFU/g) within 2 h without decreasing the seed germination rate (97.7%). The lethality of combined treatments against artificially inoculated Escherichia coli O157:H7 was also evaluated. When exposed to gaseous ClO2 at 100% RH and 55 degrees C for 6 h, the initial number of E. coli O157:H7 (3.5 log CFU/g) on radish seeds decreased to below the detection limit (0.7 log CFU/g) by direct plating but it was not eliminated from seeds. The germination rate of radish seeds was not significantly (P> 0.05) decreased after treatment for 6 h. The information reported here will be useful when developing decontamination strategies for producing microbiologically safe radish seed sprouts. (C) 2016 Published by Elsevier B.V.</P>

        Effect of substrate temperature on the nanostructural and chemical features of nc-Si:H thin films prepared by PECVD

        Hydrogenated nanocrystalline silicon (nc-Si:H) thin films were deposited by plasma enhanced chemical vapor deposition (PECVD) at a RF power of 100W; SiH<SUB>4</SUB> and H<SUB>2</SUB> were introduced into a reaction chamber at 25 and 40sccm, respectively, and the substrate temperature ranged from room temperature to 600<SUP>o</SUP>C. The effect of the substrate temperature on the formation of nanoscale Si crystallites (nc-Si) and their structural and optical features were investigated. The average size and concentration of the Si nanocrystallites varied with the substrate temperature; the former ranged from ∼1.0 to ∼5.0nm, and the latter reached up to ∼15.5% when the substrate temperature was 400<SUP>o</SUP>C. With increasing substrate temperature to 400<SUP>o</SUP>C, the relative fraction of Si-H bonds in the films, [Si-H]/@?<SUB>n=1</SUB><SUP>3</SUP>[Si-H<SUB>n</SUB>]<SUB>n=integer</SUB>, was increased up to ∼29.3%.

      • H3K27 methylation and H3S28 phosphorylation-dependent transcriptional regulation by INHAT subunit SET/TAF-Iβ

        Significant progress has been made in understanding the relationship between histone modifications and 'reader' molecules and their effects on transcriptional regulation. A previously identified INHAT complex subunit, SET/TAF-Iβ, binds to histones and inhibits histone acetylation. To investigate the binding specificities of SET/TAF-Iβ to various histone modifications, we employed modified histone tail peptide array analyses. SET/TAF-Iβ strongly recognized PRC2-mediated H3K27me½/3; however, the bindings were completely disrupted by H3S28 phosphorylation. We have demonstrated that SET/TAF-Iβ is sequentially recruited to the target gene promoter ATF3 after the PRC2 complex via H3K27me recognition and may offer additive effects in the repression of the target gene.

        Purpose : The purpose of this study is to identify satisfaction of adults who participate in healthy life practicing programs which are being provided by public health care centers. Method : Data were drawn from 129 adults who have utilized healthy life practicing programs regarding exercise, nutrition, and non-smoking offered by 2 public health centers in Seoul, using a structured questionnaire from 18th, October 2013 to 19th, November 2013. Descriptive statistics, t-test and ANOVA with Scheffe’s test were applied for data analyses using SPSS Statistics 21.0. Results : There were significant differences in satisfaction with healthy life practicing programs according to sex(F=6.848, p=.010), education level(F=5.289, p=.023), and age(F=5.950, p=.003). Also, satisfaction with healthy life practicing programs was significantly associated with degree of muscle strength exercise(F=4.881, p=.009). Satisfaction with exercise program was higher than satisfaction with nutrition healthy life practicing programs(t=5.997, p=.003). Conclusion : The degree of satisfaction with healthy life practicing programs was different according to participants’ socio-demographic characteristics. Therefore, providers of healthy life practicing programs need to consider sex, age, and educational level of program participants to improve satisfaction with healthy life practicing programs.

        In vitro Fertilization and Development of Pig Oocytes Inseminated with Boar Sperm by Different Sperm Washing Media after Thawing of the Frozen Straws

        This study was carried out to investigate in vitro fertilization and development of in vitro matured pig oocytes inseminated with the Duroc boar sperm by different sperm washing media after thawing of the 5 ml frozen straws. Immature follicular oocytes (30-40) were transferred into each well of a Nunc 4-well multidish containing $500{\mu}l$ mTCM199 maturation medium. The sperm rich portion of ejaculates was collected into a 250 ml insulated vacuum bottle and gradually cooled 22 to $24^{\circ}C$ over a 2 h period. Semen was centrifuged at 800 g for 10 min and the seminal plasma discarded. Sperm were esuspended in a lactose-egg yolk and N-acetyl-Dglucosamine (LEN) diluent to contain $1{\times}10^{9}$ sperm/ml and cooled to $5^{\circ}C$ over a 2 h period. Immediately before freezing, semen was rediluted with an equal volume of LEN+4% glycerol and packed into 5 ml straws. After thawing of the 5 ml straw, the 5 ml semen was diluted with 20 ml Beltsville thawing solution (BTS) at room temperature. Oocytes were inseminated with untreated (unwashed and nonpreincubated) or treated sperm (washed two times in BTS, mTLP-PVA and mTBM media, respectively and nonpreincubated) with $2{\times}10^{7}$ sperm concentration. Oocytes were coincubated for 6 h in $500{\mu}l$ mTBM fertilization. At 6 h after IVF, oocytes were transferred into $500{\mu}l$ NCSU-23 culture medium for further culture of 6 h. Sperm penetration, polyspermy and male pronuclear formation of oocytes at 12 h after IVF and developmental ability of oocytes at 48 h after IVF were evaluated. Sperm penetration rate, male pronuclear formation and rate of cleaved embryos were higher in the BTS, mTLP-PVA and mTBM treatments than the unwashed treatment (p<0.05). The rate of blastocysts from the cleaved oocytes (2-4 cell stage) were higher in the mTLP-PVA treatment than in the unwashed, BTS and mTBM treatments. In conclusion, we recommend the washing of frozen-thawed sperm with mTLP-PVA medium before in vitro fertilization of oocytes in mTBM medium.

      • Combined steam and carbon dioxide reforming of methane and side reactions: Thermodynamic equilibrium analysis and experimental application

        <P>Thermodynamic equilibrium analysis of the combined steam and carbon dioxide reforming of methane (CSCRM) and side reactions was performed using total Gibbs free energy minimization. The effects of (CO2 + H2O)/CH4 ratio (0.9-2.9), CO2:H2O ratio (3:1-1:3), and temperature (500-1000 degrees C) on the equilibrium conversions, yields, coke yield, and H-2/CO ratio were investigated. A (CO2 + H2O)/CH4 ratio greater than 1.2, a CO2:H2O ratio of 1:2.1, and a temperature of at least 850 degrees C are preferable reaction conditions for the synthesis gas preparation in the gas to liquid process. Simulated conditions were applied to the CSCRM reaction and the experimental data were compared with the thermodynamic equilibrium results. The thermodynamic equilibrium results were mostly consistent with the experimental data, but the reverse water gas shift reaction rapidly occurred in the real chemical reaction and under excess oxidizing agent conditions. In addition, a long-term stability test (under simulated conditions) showed that the equilibrium conversion was maintained for 500 h and that the coke formation on the used catalyst was not observed.       • Organic-inorganic hybrid photocatalyst for carbon dioxide reduction

        <P>Efficient hybrid photocatalysts for carbon dioxide reduction were developed from dye-sensitized TiO2 nanoparticles and their catalytic performance was optimized by ternary organic/inorganic components. Thus, the hybrid system consists of (E)-2-cyano-3-(5'(5 ''-(p-(diphenylamino)phenyl)thiophen-2 ''-yl)thiophen-2'-yl)-acrylic acid as a sensitizer and fac-[Re(4,4'-bis(diethoxyphosphorylmethyl)-2,2'-bipyridine)(CO)(3)Cl] as a reduction catalyst (ReP), both of which have been fixed onto TiO2 semiconductors (s-TiO2, h-TiO2, d-TiO2). Mott-Schottky analysis on flat-band potential (E-fb) of TiO2 mesoporous films has verified that Efb can be finely modulated by volume variation of water (0 to 20 vol%). The increase of added water resulted in substantial positive shifts of Efb from -1.93 V at 0 vol% H2O, to -1.74 V (3 vol% H2O), to -1.56 V (10 vol% H2O), and to -1.47 V (20 vol% H2O). As a result, with addition of 3-10 vol% water in the photocatalytic reaction, conversion efficiency of CO2 to CO increased significantly reaching a TON value of similar to 350 for 30 h. Catalytic activity enhancement is mainly attributed to (1) the optimum alignment of Efb by 3-10 vol% water with respect to the E-ox* of the dye and E-red of ReP for smooth electron transfer from photo-excited dye to ReP via the TiO2 semiconductor and (2) the water-induced acceleration of chemical processes on the fixed ReP. In addition, the energy level was further tuned by variation of the dye and ReP amounts. We also found that the intrinsic properties of TiO2 sources (morphology, size, agglomeration) exert a great influence on the overall photocatalytic activity of this hybrid system. Implications of the present observations and reaction mechanisms are discussed in detail.</P>

        Purpose: This study aimed to know whether there is a comparison of perception, knowledge and health promoting behaviors about particular matter between nursing students and non-health departments college students. Methods: A quantitative research is used. Data were collected from August 1 to 19. The study sample was nursing students and non-health departments students. For the similar condition of samples, this study excluded whose first language is not Korean. Independent T-test and pearson’s correlation was used for this study. Results: A significant positive correlation did not existed between difference of major and perception(t=0.017, p=0.986), knwledge(t=0.652, p=.516) and health promoting behaviors(t=-0.114, p=.910) about particular matters. Conclusion: Based on the study results, there is no significant correlation of perception and actions about particular matters between nursing students and non-health department college students.
